Aqueous Co removal by mycogenic Mn oxides from simulated mining wastewaters.

Naturally occurring manganese (Mn) oxide minerals often form by microbial Mn(II) oxidation, resulting in nanocrystalline Mn(III/IV) oxide phases with high reactivity that can influence the uptake and release of many metals (e.g., Ni, Cu, Co, and Zn). During formation, the structure and composition of biogenic Mn oxides can be altered in the presence of other metals, which in turn affects the minerals' ability to bind these metals. These processes are further influenced by the chemistry of the aqueous environment and the type and physiology of microorganisms involved. Conditions extending to en

Broad susceptibility of Candida auris strains to 8-hydroxyquinolines and mechanisms of resistance.

The fungal pathogen Candida auris represents a severe threat to hospitalized patients. Its resistance to multiple classes of antifungal drugs and ability to spread and resist decontamination in healthcare settings make it especially dangerous. We screened 1,990 clinically approved and late-stage investigational compounds for the potential to be repurposed as antifungal drugs targeting C. auris and narrowed our focus to five Food and Drug Administration (FDA)-approved compounds with inhibitory concentrations under 10 µM for C. auris and significantly lower toxicity to three human cell lines. Alterations in the Gut Fungal Community in a Mouse Model of Huntington's Disease.

Huntington's disease (HD) is a neurodegenerative disorder caused by a trinucleotide expansion in the HTT gene, which is expressed throughout the brain and body, including the gut epithelium and enteric nervous system. Afflicted individuals suffer from progressive impairments in motor, psychiatric, and cognitive faculties, as well as peripheral deficits, including the alteration of the gut microbiome. However, studies characterizing the gut microbiome in HD have focused entirely on the bacterial component, while the fungal community (mycobiome) has been overlooked. The gut mycobiome has gained

The capsule of Cryptococcus neoformans.

The capsule of Cryptococcus neoformans is its dominant virulence factor and plays a key role in the biology of this fungus. In this essay, we focus on the capsule as a cellular structure and note the limitations inherent in the current methodologies available for its study. Given that no single method can provide the structure of the capsule, our notions of what is the cryptococcal capsule must be arrived at by synthesizing information gathered from very different methodological approaches including microscopy, polysaccharide chemistry and physical chemistry of macromolecules. The emerging pic

Spatial and temporal patterns of public transit aerobiomes.

BACKGROUND: Aerobiome diversity is extensive; however, species-level community structure remains poorly resolved. Likewise, microbiomes of public transit systems are of public interest due to their importance for health, though few studies have focused on these ecosystems whilst utilising shotgun metagenomics. Aerosol studies have focused predominantly on individual cities, with limited between-city comparisons suggesting specific community structures. Longitudinal studies show aerobiome diversity as dynamic, fluctuating during seasonal and daily cycles, though interannual cycles remains to be

Self-grown mycelium in confined geometries as nanofluidic devices.

Precise control of ion and molecular transport at the nanoscale underpins next-generation nanofluidic technologies. However, current approaches such as top-down fabrication and bottom-up assembly remain constrained by cost, scalability, or limited programmability. Fungal mycelium-the largest natural ion transport network in soil-offers a living bio-derived route to nanofluidics. Here, we harness mycelium's self-growth and hyphal anastomosis to construct nanofluidic structures that autonomously conform to confined geometries. With interconnected fibrous networks, nanoscale porosity, and negativ

Elucidation of a novel fungal biotransformation pathway of ibuprofen by Phanerochaete sordida YK-624 under non-ligninolytic conditions.

Ibuprofen, a widely used non-steroidal anti-inflammatory drug (NSAID), has been detected in the environment and is known to contaminate waterbodies. Here, we investigated the biotransformation of ibuprofen using the hyper-lignin-degrading fungus Phanerochaete sordida YK-624. This fungus exhibited excellent transformation capacity, removing 75% of ibuprofen within 5 h of incubation at a final concentration of 100 μM. Three novel metabolites of this transformation were isolated, and their structures were determined by nuclear magnetic resonance and high-resolution electrospray ionization mass sp

Immunosenescence and organoids: pathophysiology and therapeutic opportunities.

Immunosenescence is the gradual deterioration in the functionality of the immune system that has various clinical manifestations, such as a weakened response to vaccination, higher susceptibility to viral/bacterial/fungal infection, and higher incidence rates regarding the ageing-based disorders. Conventionally used models, like animal or 2D models for unraveling the interactions between ageing and immunity are unable to depict the complexity of bodily environments. However, this major limitation can be addressed via using the organoid technology. For clarification, organoids are tissue-resemb

Structural insights into mechanisms of zinc scavenging by the Candida albicans zincophore Pra1.

Candida albicans causes over 400,000 life-threatening, and an additional half a billion of mucosal infections annually. In response to infection, the host limits essential micronutrient availability, including zinc, to restrict growth of the invading pathogen. As assimilation of zinc is essential for C. albicans pathogenicity, limitation induces secretion of the zincophore protein Pra1 to scavenge zinc from the host. Pra1 also plays a number of important roles in host-pathogen interactions and is conserved in most fungi. However, the structure of fungal zincophores is unknown. Here, we present

Structural unification of diverse transmembrane acyltransferases reveals a conserved fold for the transmembrane acyl transferase (TmAT) superfamily.

The movement of acyl groups across biological membranes is essential for many cellular processes. One major family of proteins catalysing this reaction are the acyl transferase family 3 (AT3) proteins, which form a pore to allow acyl-CoA to penetrate the membrane for transfer onto an extracytosolic acceptor molecule. Recent structures of the sequence-unrelated human heparan-α-glucosaminide N-acetyltransferase (HGSNAT) support a similar transmembrane acyl-group transfer mechanism. Here we demonstrate that both protein families contain a conserved 10-transmembrane helical fold with high structur

Expression and characterization of SARS-CoV-2 spike protein in Thermothelomyces heterothallica C1.

The COVID-19 pandemic demonstrated a pressing need for rapid, adaptive, and scalable manufacturing of vaccines and reagents. With the transition into an endemic disease and rising threats of other emerging pandemics, production of these biologicals requires a stable and sustainable supply chain and accessible distribution methods. In this study, we demonstrate the strength of an engineered filamentous fungal platform, Thermothelomyces heterothallica C1, for high volumetric productivity of the full-length spike glycoprotein. Spike protein produced in this system is highly thermostable and immun

Siderophores produced by marine microorganisms.

Siderophores are iron-chelating, low-molecular-weight compounds secreted by microorganisms in iron-deficient environments that enhance iron uptake. When compared to the siderophores produced by terrestrial microorganisms, marine siderophores have been shown to have diverse structures and physiological functions. Fifty-seven strains of marine microorganisms have been identified as siderophore producers so far. Among them, 54 bacterial strains belong to 23 genera of 19 families in five phyla, and three fungal strains belong to three genera of three families in two phyla. The most common source o

Contrasting the influences of phosphate and phosphite on growth of Aspergillus niger.

Phosphite serves as an alternative phosphorus material in terrestrial ecosystems. Aspergillus niger (A. niger), a prominent phosphate-solubilizing fungus (PSF), facilitates phosphorus and leaches heavy metal ions via organic acids and enzymes. With the synergistic effect of phosphate materials, heavy metal ions can be effectively immobilized by A. niger to achieve remediation of contaminated soils. This study investigated the structural distinctions between phosphite and phosphate compounds by using ATR-IR and Raman spectroscopy, while concurrently assessing the physiological impact of phosphi

Anti-fungal peptides: an emerging category with enthralling therapeutic prospects in the treatment of candidiasis.

Candida infections, particularly invasive candidiasis, pose a serious global health threat. Candida albicans is the most prevalent species causing candidiasis, and resistance to key antifungal drugs, such as azoles, echinocandins, polyenes, and fluoropyrimidines, has emerged. This growing multidrug resistance (MDR) complicates treatment options, highlighting the need for novel therapeutic approaches. Antifungal peptides (AFPs) are gaining recognition for their potential as new antifungal agents due to their diverse structures and functions. These natural or recombinant peptides can effectively

Naphthoquinones biflorin and bis-biflorin (Capraria biflora) as possible inhibitors of the fungus Candida auris polymerase: molecular docking, molecular dynamics, MM/GBSA calculations and in silico drug-likeness study.

A new worldwide concern has emerged with the recent emergence of infections caused by Candida auris. This reflects its comparative ease of transmission, substantial mortality, and the increasing level of resistance seen in the three major classes of antifungal drugs. Efforts to create a better design for structure-based drugs that described numerous modifications and the search for secondary metabolic structures derived from plant species are likely to reduce the virulence of several fungal pathogens. In this context, the present work aimed to evaluate in silico two naphthoquinones isolated fr

Design, Synthesis, in vitro and in silico Characterization of 2-Quinolone-L-alaninate-1,2,3-triazoles as Antimicrobial Agents.

Due to the ever-increasing antimicrobial resistance there is an urgent need to continuously design and develop novel antimicrobial agents. Inspired by the broad antibacterial activities of various heterocyclic compounds such as 2-quinolone derivatives, we designed and synthesized new methyl-(2-oxo-1,2-dihydroquinolin-4-yl)-L-alaninate-1,2,3-triazole derivatives via 1,3-dipolar cycloaddition reaction of 1-propargyl-2-quinolone-L-alaninate with appropriate azide groups. The synthesized compounds were obtained in good yield ranging from 75 to 80 %. The chemical structures of these novel hybrid mo

Peptide-Induced Biomineralization of Tin Oxide (SnO₂) Nanoparticles for Antibacterial Applications.

Recently, there has been growing attention and effort to search for new microbicidal drugs which present different mode of action from those already existing, as an alternative to the global threat of fungal and bacterial multi drug resistance (MDR). Here we propose biological synthesis of SnO₂ nanoparticles using mammalian cells as an economic and ecofriendly platform. This presents a novel biogenic method for SnO₂ synthesis using metal binding peptides extracted from MCF-7 human cancer cells, which induces the biomineralization of SnO₂ nanoparticles.
