Regulatory roles of microRNA163 in responses to stresses in Arabidopsis.

MicroRNAs (miRNAs) are small regulatory RNAs that participate in various biological processes by silencing target genes. In Arabidopsis, microRNA163 (miR163) was found to be involved in seed germination, root development, and biotic resistance. However, the regulatory roles of miR163 remain unclear. In the current study, the mir163 mutant was investigated to comprehensively understand and characterize its functions in Arabidopsis. RNA-sequencing and Gene Ontology enrichment analyses revealed that miR163 might be involved in "response to stimulus" and "metabolic process". Interestingly, "respon

Critical roles of mitochondrial fatty acid synthesis in tomato development and environmental response.

Plant mitochondrial fatty acid synthesis (mtFAS) appears to be important in photorespiration based on the reverse genetics research from Arabidopsis (Arabidopsis thaliana) in recent years, but its roles in plant development have not been completely explored. Here, we identified a tomato (Solanum lycopersicum) mutant, fern-like, which displays pleiotropic phenotypes including dwarfism, yellowing, curly leaves, and increased axillary buds. Positional cloning and genetic and heterozygous complementation tests revealed that the underlying gene FERN encodes a 3-hydroxyl-ACP dehydratase enzyme invol

Dwarf and High Tillering1 represses rice tillering through mediating the splicing of D14 pre-mRNA.

Strigolactones (SLs) constitute a class of plant hormones that regulate many aspects of plant development, including repressing tillering in rice (Oryza sativa). However, how SL pathways are regulated is still poorly understood. Here, we describe a rice mutant dwarf and high tillering1 (dht1), which exhibits pleiotropic phenotypes (such as dwarfism and increased tiller numbers) similar to those of mutants defective in SL signaling. We show that DHT1 encodes a monocotyledon-specific hnRNP-like protein that acts as a previously unrecognized intron splicing factor for many precursor mRNAs (pre-mR

Lack of antagonism between salicylic acid and jasmonate signalling pathways in poplar.

Salicylic acid (SA) and jasmonic acid (JA) often play distinct roles in plant defence against pathogens. Research from Arabidopsis thaliana has established that SA- and JA-mediated defences are more effective against biotrophs and necrotrophs, respectively. These two hormones often interact antagonistically in response to particular attackers, with the induction of one leading to suppression of the other. Here, we report a contrasting pattern in the woody perennial Populus: positive SA-JA interplay. Using genetically engineered high SA lines of black poplar and wild-type lines after exogenous

Fibre-specific regulation of lignin biosynthesis improves biomass quality in Populus.

Lignin is a major component of cell wall biomass and decisively affects biomass utilisation. Engineering of lignin biosynthesis is extensively studied, while lignin modification often causes growth defects. We developed a strategy for cell-type-specific modification of lignin to achieve improvements in cell wall property without growth penalty. We targeted a lignin-related transcription factor, LTF1, for modification of lignin biosynthesis. LTF1 can be engineered to a nonphosphorylation form which is introduced into Populus under the control of either a vessel-specific or fibre-specific promot

Chloroplastic Os3BGlu6 contributes significantly to cellular ABA pools and impacts drought tolerance and photosynthesis in rice.

Cellular abscisic acid (ABA) concentration is determined by both de novo biosynthesis and recycling via β-glucosidase(s). However, which rice β-glucosidase(s) are involved in this process remains unknown. Here, we report on a chloroplastic β-glucosidase isoenzyme, Os3BGlu6, that functions in ABA recycling in rice. Disruption of Os3BGlu6 in rice resulted in dwarfism, lower ABA content in leaves, drought-sensitivity, lower photosynthesis rate and higher intercellular CO2 concentration. Os3BGlu6 could hydrolyze ABA-GE to ABA in vitro. The reversion and overexpression rice lines restored or increa

Computational solutions for modeling and controlling plant response to abiotic stresses: a review with focus on iron deficiency.

Computational solutions enable plant scientists to model protein-mediated stress responses and characterize novel gene functions that coordinate responses to a variety of abiotic stress conditions. Recently, density functional theory was used to study proteins active sites and elucidate enzyme conversion mechanisms involved in iron deficiency responsive signaling pathways. Computational approaches for protein homology modeling and the kinetic modeling of signaling pathways have also resolved the identity and function in proteins involved in iron deficiency signaling pathways. Significant chang

Hydrogen sulfide acts downstream of jasmonic acid to inhibit stomatal development in Arabidopsis.

Main conclusion: Jasmonic acid (JA) negatively regulates stomatal development by promoting LCD expression and hydrogen sulfide (H2S) biosynthesis. H2S inhibits the initiation of stomata formation and acts upstream of SPEECHLESS. Abstract: Stomatal development is strictly regulated by endogenous signals and environmental cues. We recently revealed that jasmonic acid (JA) negatively regulates stomatal development in Arabidopsis thaliana cotyledons (Han et al., Plant Physiol 176:2871-2885, 2018), but the underlying molecular mechanism remains largely unknown. Here, we uncovered a role for H2S in

Formation of NPR1 Condensates Promotes Cell Survival during the Plant Immune Response.

In plants, pathogen effector-triggered immunity (ETI) often leads to programmed cell death, which is restricted by NPR1, an activator of systemic acquired resistance. However, the biochemical activities of NPR1 enabling it to promote defense and restrict cell death remain unclear. Here we show that NPR1 promotes cell survival by targeting substrates for ubiquitination and degradation through formation of salicylic acid-induced NPR1 condensates (SINCs). SINCs are enriched with stress response proteins, including nucleotide-binding leucine-rich repeat immune receptors, oxidative and DNA damage r

Divide et impera: boundaries shape the plant body and initiate new meristems.

485 I. 485 II. 486 III. 491 IV. 491 V. 495 495 References 495 SUMMARY: Boundaries, established and maintained in different regions of the plant body, have diverse functions in development. One role is to separate different cell groups, for example the differentiating cells of a leaf primordium from the pluripotent cells of the apical meristem. Boundary zones are also established during compound leaf development, to separate young leaflets from each other, and in many other positions of the plant body. Recent studies have demonstrated that different boundary zones share similar properties. They

Evolution of DNA methylation patterns in the Brassicaceae is driven by differences in genome organization.

DNA methylation is an ancient molecular modification found in most eukaryotes. In plants, DNA methylation is not only critical for transcriptionally silencing transposons, but can also affect phenotype by altering expression of protein coding genes. The extent of its contribution to phenotypic diversity over evolutionary time is, however, unclear, because of limited stability of epialleles that are not linked to DNA mutations. To dissect the relative contribution of DNA methylation to transposon surveillance and host gene regulation, we leveraged information from three species in the Brassicac

SlMAPK3-SlWRKY5-SlGR1 Module Mediates Thermotolerance Response in Tomato via Transcriptional and Post-Translational Mechanisms.

Heat stress severely impairs crop growth and production. WRKY transcription factors are key regulators of plant responses to abiotic stresses, but the mechanisms by which these transcription factors respond to heat stress in tomato are poorly understood. In this study, we identified a transcription factor SlWRKY5, which was significantly induced at high temperature (42°C). Phenotypic analyses showed that overexpression of SlWRKY5 positively regulated thermotolerance but suppressed growth of tomato plants, while its knockout exhibited opposite effects. Transcriptome analysis further demonstrate

The Trihelix transcription factor GT2-like 1 (GTL1) promotes salicylic acid metabolism, and regulates bacterial-triggered immunity.

The Trihelix Transcription factor GT2-like 1 (GTL1) was previously shown to be a key regulator of ploidy-dependent trichome growth and drought tolerance. Here, we report that GTL1 plays an important role in coordinating plant immunity. We show that gtl1 mutants are compromised in the regulation of basal immunity, microbial pattern-triggered immunity (PTI) and effector-triggered RIN4-mediated immunity. Transcriptome analysis revealed that GTL1 positively regulates defense genes and inhibits factors that mediate growth and development. By performing hormonal measurements and chromatin-immunoprec

Elevated CO2 Modulates Hormonal Signaling and Galactose Metabolism to Improve Photosynthetic Performance and Water Use Efficiency in Peanut (Arachis hypogaea L.).

Atmospheric CO2 elevation promotes peanut biomass accumulation, yet the underlying molecular mechanisms remain unclear. This study investigated the physiological and molecular responses of two peanut cultivars (JH2 and JH54) to elevated CO2 (EC, 700 μmol·mol-1) versus ambient CO2 (CK, 400 μmol·mol-1) in open-top chambers, by analyzing photosynthetic traits, aboveground biomass, and leaf transcriptomic/metabolomic profiles. The results showed that EC significantly increased net photosynthetic rate, intercellular CO2 concentration, water use efficiency and biomass, but reduced stomatal conductan

Differential Regulation of Pre-Harvest Sprouting by OsERF1 and OsERF94 Through Hormone Signaling and Metabolic Reprogramming in Rice.

Pre-harvest sprouting (PHS), the premature germination of grains on the mother plant, causes substantial yield loss and grain-quality deterioration in rice under humid conditions. Although seed dormancy and germination are largely controlled by hormonal balance, the transcriptional mechanisms linking hormone signaling with metabolic adaptation during PHS remain unclear. In this study, we investigated the roles of two ethylene-responsive factor transcription factors, OsERF1 and OsERF94, in rice PHS regulation using CRISPR/Cas9-mediated knockout lines, together with physiological, gene-expressio

Dehydrogenase WRD1 regulates plant growth by modulating cell wall formation, ROS homeostasis, and auxin signaling in rice.

Nucleotide sugars serve as the substrates for the biosynthesis of cell wall polysaccharides. Although the enzymes such as uridine diphosphate (UDP)-glucose dehydrogenase (UGD) involved in nucleotide sugar biosynthesis have been biochemically characterized, their biological roles in plants remain largely unexplored. In this study, we identify a rice mutant weakness and root hair defective 1 (wrd1), which exhibits pleiotropic phenotypes including dwarfism, reduced tiller number, and abnormal root architecture. WRD1 encodes a UGD that catalyzes the conversion of UDP-Glc to UDP-GlcA. The WRD1 muta

Salicylic Acid Cooperates With Lignin and Sucrose Signals to Alleviate Waxy Maize Leaf Senescence Under Heat Stress.

Leaf senescence induced by high temperature (HT) has become a primary factor limiting maize yield, particularly during the filling stage. Exogenous salicylic acid (SA) has emerged as an effective strategy to mitigate leaf senescence and HT-induced damage, though its underlying mechanisms remain unclear. This study investigated the regulatory mechanism of SA application on waxy maize subjected to HT during the early filling stage. Compared to HT alone, exogenous SA alleviated the inhibition of photosynthesis and oxidative damage by enhancing the activities of enzymes involved in photosynthesis

The regulatory MaIAA1-like-MaERF003 module modulates the ripening of "Fenjiao" bananas.

Fruit ripening is a complex process mediated by plant hormones and transcriptional regulation. Ethylene and auxin are key regulators, but little is known about their crosstalk in fruit ripening. This study discovered the synergistic effect of auxin and ethylene during "Fenjiao" banana (Musa ABB Pisang Awak) fruit ripening, which is regulated by transcription factors MaIAA1-like and MaERF003. Both ethylene and auxin enhanced ripening and the expression of MaIAA1-like and MaERF003. However, 1-methylcyclopropene (1-MCP) treatment suppressed ripening and the expression of both genes. MaIAA1-like a
