The effect of heat shock on morphogenesis in barley : coordinated circadian regulation of mRNA levels for light-regulated genes and of the capacity for accumulation of chlorophyll protein complexes.

The effect of daily heat-shock treatments on gene expression and morphogenesis of etiolated barley (Hordeum vulgare) was investigated. Heat-shock treatments in the dark induced shortening of the primary leaves and the coleoptiles to the length of those in light-grown plantlets. In addition, the mRNA levels of the light-induced genes that were investigated were raised under these conditions and showed distinct oscillations over a period of at least 3 d. While the mRNA levels for chlorophyll a/b binding protein (LHC II), plastocyanin, and the small subunit of ribulose-1,5-bisphosphate carboxylas

The ezrin Gene Regulates Early Cardiac Morphogenesis and Contractile Function in Zebrafish Through the Coordinated Regulation of Apoptosis, Calcium Homeostasis, and the MAPK Signaling Pathway.

Ezrin, expressed by the EZR gene, is a member of the ERM protein family that connects the plasma membrane to the actin cytoskeleton, participating in processes such as cell adhesion, migration, and signaling. However, its role in cardiac morphogenesis remains incompletely understood. In zebrafish (Danio rerio), two ezrin homologs, ezra and ezrb, are present. CRISPR/Cas9 gene editing technology was used to generate ezra knockout lines, and the simultaneous knockdown of ezra and ezrb was induced via morpholino oligonucleotides (MOs). To investigate the molecular mechanisms, transcriptome sequenc

Down-regulation of the rice HRS1 HOMOLOG3 transcriptional repressor gene due to N deficiency directly co-activates ammonium and phosphate transporter genes.

Rice HRS1 HOMOLOG3 (OsHHO3) acts as a transcriptional repressor of AMMONIUM TRANSPORTER1 (OsAMT1) genes in rice; thus, reduced OsHHO3 expression in nitrogen (N)-deficient environments promotes ammonium uptake. In this study, we show that OsHHO3 also functions as a repressor of a specific subset of phosphate (Pi) transporter (PT) genes involved in the uptake and root-to-shoot translocation of Pi, including OsPT2, OsPT4, and OsPHO1;1. Disruption of OsHHO3 increased Pi uptake and Pi contents in shoots and roots, while overexpression of OsHHO3 caused the opposite effects. Furthermore, phosphorus (

Comparison of host cell gene expression in cowpox, monkeypox or vaccinia virus-infected cells reveals virus-specific regulation of immune response genes.

BACKGROUND: Animal-borne orthopoxviruses, like monkeypox, vaccinia and the closely related cowpox virus, are all capable of causing zoonotic infections in humans, representing a potential threat to human health. The disease caused by each virus differs in terms of symptoms and severity, but little is yet know about the reasons for these varying phenotypes. They may be explained by the unique repertoire of immune and host cell modulating factors encoded by each virus. In this study, we analysed the specific modulation of the host cell's gene expression profile by cowpox, monkeypox and vaccinia

Regulation of gene expression in brain tissues of rats repeatedly treated by the highly abused opioid agonist, oxycodone: microarray profiling and gene mapping analysis.

Although oxycodone is the most often used opioid agonist, it remains one of the most understudied drugs. We used microarray analysis to better understand the global changes in gene expression in brain tissues of rats repeatedly treated with oxycodone. Many genes were significantly regulated by oxycodone (e.g., Fkbp5, Per2, Rt1.Dalpha, Slc16a1, and Abcg2). Validation of the microarray data by quantitative real-time-polymerase chain reaction (Q-PCR) indicated that there was a strong significant correlation (r = 0.979, p < 0.0000001) between the Q-PCR and the microarray data. Using MetaCore (a co

Gene expression profiling of lymphoblastoid cell lines from monozygotic twins discordant in severity of autism reveals differential regulation of neurologically relevant genes.

BACKGROUND: The autism spectrum encompasses a set of complex multigenic developmental disorders that severely impact the development of language, non-verbal communication, and social skills, and are associated with odd, stereotyped, repetitive behavior and restricted interests. To date, diagnosis of these neurologically based disorders relies predominantly upon behavioral observations often prompted by delayed speech or aberrant behavior, and there are no known genes that can serve as definitive biomarkers for the disorders. RESULTS: Here we demonstrate, for the first time, that lymphoblastoi

The effect of cell density and specific growth rate on accessory gene regulator and toxic shock syndrome toxin-1 gene expression in Staphylococcus aureus.

A continuous culture technique utilising a variant of Staphylococcus aureus 8325-4 containing transcriptional gene fusions was used to investigate the relationships between cell density (OD(600)), steady-state specific growth rate (mu) and expression of both agr (accessory gene regulator) and tst (toxic shock syndrome toxin-1). The expression of these genes was assessed by two single-copy independently arranged chromosomal-based reporter systems, beta-galactosidase agr-P3 promoter fusion and a lux-tst promoter fusion. Cell density and specific agr expression were found to be positively correla

The Ectopic Expression of the MpDIR1(t) Gene Enhances the Response of Plants from Arabidopsis thaliana to Biotic Stress by Regulating the Defense Genes and Antioxidant Flavonoids.

The Defective in Induced Resistance 1 (DIR1) gene, a member of the lipid transferase proteins (LTPs), plays a crucial role in plant defense against pathogens. While previous transcriptomic studies have highlighted the significant expression of citrus LTPs during biotic stress, functional annotations of LTPs in the Citrus genera remain limited. In this study, we cloned the Murraya paniculata DIR1 (MpDIR1(t)) gene and overexpressed it in Arabidopsis thaliana to evaluate its stress response mechanisms against biotic stress. The transgenic Arabidopsis lines showed fewer disease symptoms in respons

Expression and regulation by thyroid hormone (TH) of zebrafish IGF-I gene and amphioxus IGFl gene with implication of the origin of TH/IGF signaling pathway.

Thyroid hormone (TH)/insulin-like growth factor (IGF) signaling pathway has been identified in all the vertebrates, but its evolutionary origin remains elusive. In this study we examined the expression profiles in vitro as well as in vivo of the IGF-I gene of fish Danio rerio (vertebrate) and the IGF-like gene (IGFl) of amphioxus Branchiostoma japonicum (protochordate) following T(3) treatment. Our results showed that T(3) was able to enhance hepatic IGF-I/IGFl gene expression in vitro in both zebrafish and amphioxus in a dose-dependent manner. This T(3)-induced hepatic expression of IGF-I/IGF

Insights into regulation and function of the major stress-induced hsp70 molecular chaperone in vivo: analysis of mice with targeted gene disruption of the hsp70.1 or hsp70.3 gene.

The murine hsp70 gene family includes the evolutionarily conserved hsp70.1 and hsp70.3 genes, which are the major proteins induced by heat and other stress stimuli. hsp70.1 and hsp70.3 encode identical proteins which protect cells and facilitate their recovery from stress-induced damage. While the hsp70 gene family has been widely studied and the roles of the proteins it encodes as molecular chaperones in a range of human pathologies are appreciated, little is known about the developmental regulation of hsp70.1 and hsp70.3 expression and the in vivo biological function of their products. Representational difference analysis of cDNA for the detection of differential gene expression in bacteria: development using a model of iron-regulated gene expression in Neisseria meningitidis.

Representational difference analysis of cDNA (cDNA RDA) provides a powerful technique for the identification of specific differences between two mRNA populations. The method has previously been used to analyse differential gene expression in eukaryotes, but until now has not been successfully applied to prokaryotes. A strain of Neisseria meningitidis with a deletion of the iron-regulated lactoferrin-binding protein A (IbpA) gene, grown under iron-replete conditions, and the isogenic parent strain, grown under iron limitation, were used as a model for developing cDNA RDA for use with bacteria.

yst gene expression in Yersinia enterocolitica is positively regulated by a chromosomal region that is highly homologous to Escherichia coli host factor 1 gene (hfq).

Yersinia enterocolitica produces heat-stable enterotoxin (Y-ST) as one of its virulence factors. The yst gene, however, frequently and spontaneously becomes inactive (silent) during storage, which is accompanied by concurrent changes in some biological properties such as colony morphology, growth rate, carbon fermentation and ornithine decarboxylase activity. Northern blot analysis revealed that the level of mRNA for yst was repressed. To investigate the regulatory region, we transformed a yst-silent strain with a chromosomal gene library of Y-ST producing an isogenic counterpart. Out of 3604

Crosstalk in Silence: PIWIL/piRNA in ncRNA Hubs for Immune Gene Regulation and Epigenetic Antiviral Defense.

Recent studies indicate that PIWI (P-element-induced WImpy testis) like proteins and their cognate PIWI-interacting RNAs (piRNAs), initially identified in germline cells for protecting genomic stability by transposon silencing, also exert multiple regulatory functions in somatic and immune systems. Additionally, PIWIL/piRNA pathways are linked to innate antiviral defense and immune signaling pathway regulation. This review systematically compiles the reports on the immunological functions of noncoding RNAs (ncRNAs) and delineates the potential molecular processes and functional characteristics

A novel cytochrome P450 gene, CYP12A2, regulates reproduction of Mythimna separata by modulating 20E biosynthesis.

BACKGROUND: Several P450 genes in the 20-hydroxyecdysone (20E) signaling pathway are known to critically regulate reproductive development in insects. However, the contribution of other P450 family members to insect reproduction remains poorly characterized. RESULTS: Here, we identified a novel P450 gene, CYP12A2, from the highly polyphagous pest Mythimna separata. Quantitative real-time polymerase chain reaction analysis revealed that CYP12A2 was upregulated during sexual maturation and predominantly enriched in the ovaries of 3-day-old female adults. Cathepsin G Promotes Hepatic Lipid Deposition by Regulating Key Genes Related to Lipid Metabolism.

Cathepsins play critical roles in various physiopathological processes, with several reported to be associated with nonalcoholic fatty liver disease (NAFLD). Herein, we investigated the expression patterns of the cathepsin family in human and mouse livers, cultured hepatocytes, and their roles in NAFLD. Public datasets of NAFLD patients and controls were analyzed to examine hepatic cathepsin expression in human livers. RT-qPCR assessed these genes in mouse livers, HepG2, Hepa1-6, and mouse primary cells. Despite different expression patterns, CTSA, CTSB, CTSD, CTSH, and CTSL were consistently

Sucrose synthase gene family analysis in loquat reveals EjSUS4 as a key regulator of sugar accumulation.

INTRODUCTION: Soluble sugar composition critically determines the commercial quality of loquat (Eriobotrya japonica), a prominent Rosaceae fruit. Sucrose synthase (SUS) plays a central role in carbohydrate partitioning and fruit sink strength, yet the SUS gene family in loquat remains uncharacterized. METHODS: Seven EjSUS genes were identified from the loquat genome and analyzed via phylogenetics, chromosomal mapping, synteny, and structural characterization. Spatiotemporal expression profiling was performed during fruit development in two cultivars ('YingShuang' and 'ZheHong 16'). Soluble su

A single-nucleus multi-omic atlas of gene regulation across 21 adult human tissues.

Diverse human cell types establish specialized functions through lineage- and context-specific regulatory programs. Interpreting non-coding genetic risk requires integrated multi-omic reference maps that directly connect regulatory DNA to cellular expression across human tissues. Here we present a single-nucleus multi-omic atlas comprising 459,856 transcriptomic and chromatin accessibility profiles from 21 adult human tissues and four donors, including paired measurements from 160,688 nuclei. The atlas resolves nine cell lineages, 61 broad cell types and 313 subclusters, and identifies 1,085,0
