Ichthyosis: mechanisms of disease.

The disorders of cornification (ichthyoses) comprise acquired and inherited disorders characterized clinically by generalized scaling and histologically by hyperkeratosis. They may arise through defects in the production or maintenance of a normal cornified cell compartment, or both. The stratum corneum is composed of protein-enriched and lipid-depleted corneocytes ("bricks") surrounded by an intercellular domain ("mortar") composed of hydrophobic, lipid-enriched membrane bilayers, and containing desmosomes and a limited array of hydrolytic enzymes. Mechanisms whereby a genetic defect involvin

Synthesis and Fluorescence Profiling of Naphthylamine Analogs.

N-phenyl-1-naphthylamine (NPN) has been widely used to probe the integrity of the outer membrane of gram-negative bacteria. NPN fluoresces in the hydrophobic environment of lipid bilayers, whereas its fluorescence is quenched in the aqueous extracellular milieu. Because NPN does not intrinsically accumulate in intact gram-negative bacteria, it has been broadly applied as a reporter of outer membrane permeabilization. Despite its widespread utility, systematic derivatization of this probe has remained largely unexplored. Here, we investigated the structure-fluorescence relationships of NPN thro…

Nature's Power at Nanoscale: Isolation and Characterization of Exosome-like Nanovesicles from Murraya koenigii.

Plant derived exosome like nanoparticles (PDENs) are tiny vesicles that contain a variety of bioactive molecules. They can be used in delive … ility. Morphological analysis showed that the vesicles were intact and spherical, with well-defined lipid bilayers. Biochemical characterization showed the presence of functional groups such as -OH, -COOH and -NH₂ moieties. Proteomic analysis using LC-MS/MS revealed a diverse array of proteins, highlighting the functional potential of the isolated exosomes-like …

Proteomics Uncovers Enrichment Bias of Common Extracellular Vesicle Isolation Methods in HEK293T Cells.

Extracellular vesicles (EVs) are membranous structures consisting of lipid bilayers that are released by most cell types and serve as important mediators of intercellular communication. The HEK293T cell line model has gained considerable attention from the scientific community, particularly in the fields of engineering and drug delivery. Nevertheless, there is a dearth of systematic comparisons of the most prevalent EV isolation methodologies for HEK293T in terms of recovery and specificity. In this study, common EV isolation workflows using either ultracentrifugation (UC), ultrafiltration (UF…

A Pseudo-Surfactant Chemical Permeation Enhancer to Treat Otitis Media via Sustained Transtympanic Delivery of Antibiotics.

Chemical permeation enhancers (CPEs) represent a prevalent and safe strategy to enable noninvasive drug delivery across skin-like biological barriers such as the tympanic membrane (TM). While most existing CPEs interact strongly with the lipid bilayers in the stratum corneum to create defects as diffusion paths, their interactions with the delivery system, such as polymers forming a hydrogel, can compromise gelation, formulation stability, and drug diffusion. To overcome this challenge, differing interactions between CPEs and the hydrogel system are explored, especially those with sodium dodec…

Deuterated Arachidonic Acid Ameliorates Lipopolysaccharide-Induced Lung Damage in Mice.

Arachidonic acid (ARA) is a major component of lipid bilayers as well as the key substrate for the eicosanoid cascades. ARA is readily oxidized, and its non-enzymatic and enzymatic oxidation products induce inflammatory responses in nearly all tissues, including lung tissues. Deuteration at bis-allylic positions substantially decreases the overall rate of ARA oxidation when hydrogen abstraction is an initiating event. To compare the effects of dosing of arachidonic acid (H-ARA) and its bis-allylic hexadeuterated form (D-ARA) on lungs in conventionally healthy mice and in an acute lung injury m…

Common mechanisms involved in Alzheimer's disease and type 2 diabetes: a key role of chronic bacterial infection and inflammation.

Strong epidemiologic evidence and common molecular mechanisms support an association between Alzheimer's disease (AD) and type 2-diabetes. L … ection, inflammation and amyloidosis. Amyloid beta and the pancreatic amyloid called amylin bind to lipid bilayers and produce Ca(2+) influx and bacteriolysis. Similarly to AD, accumulation of amylin deposits in type 2 diabetes may result from an innate immune response to chronic bacterial infections, which are known to be associated with amyloidosis. Further re …

CCT chaperonin complex is required for efficient delivery of anthrax toxin into the cytosol of host cells.

Bacterial toxins have evolved successful strategies for coopting host proteins to access the cytosol of host cells. Anthrax lethal factor (LF) enters the cytosol through pores in the endosomal membrane formed by anthrax protective antigen. Although in vitro models using planar lipid bilayers have shown that translocation can occur in the absence of cellular factors, recent studies using intact endosomes indicate that host factors are required for translocation in the cellular environment. In this study, we describe a high-throughput shRNA screen to identify host factors required for anthrax le…

The species recognized by SPR generate ionic currents in artificial lipid bilayers and inhibit the physiological pharyngeal contractions in Caenorhabditis elegans, a new method for testing the toxic potential of Aβ oligomers. With these assays we found that the formation of biologically relevant Aβ oligomers is inhibited by epigal …

A translocation causing increased alpha-klotho level results in hypophosphatemic rickets and hyperparathyroidism.

Phosphate homeostasis is central to diverse physiologic processes including energy homeostasis, formation of lipid bilayers, and bone formation. Reduced phosphate levels due to excessive renal loss cause hypophosphatemic rickets, a disease characterized by prominent bone defects; conversely, hyperphosphatemia, a major complication of renal failure, is accompanied by parathyroid hyperplasia, hyperparathyroidism, and osteodystrophy. Here, we define a syndrome featuring both hypophosphatemic rickets and hyperparathyroidism due to parathyroid hyperplasia as well as other skeletal abnormalities. We…

Reversible block of the calcium release channel/ryanodine receptor by protamine, a heparin antidote.

Channel activity of the calcium release channel from skeletal muscle, ryanodine receptor type 1, was measured in the presence and absence of protamine sulfate on the cytoplasmic side of the channel. Single-channel activity was measured after incorporating channels into planar lipid bilayers. Optimally and suboptimally calcium-activated calcium release channels were inactivated by the application of protamine to the cytoplasmic side of the channel. Recovery of channel activity was not observed while protamine was present. The addition of protamine bound to agarose beads did not change channel a…

Point mutations in alpha bENaC regulate channel gating, ion selectivity, and sensitivity to amiloride.

We have generated two site-directed mutants, K504E and K515E, in the alpha subunit of an amiloride-sensitive bovine epithelial Na+ channel, alpha bENaC. The region in which these mutations lie is in the large extracellular loop immediately before the second membrane-spanning domain (M2) of the protein. We have found that when membrane vesicles prepared from Xenopus oocytes expressing either K504E or K515E alpha bENaC are incorporated into planar lipid bilayers, the gating pattern, cation selectivity, and amiloride sensitivity of the resultant channel are all altered as compared to the wild-typ…

pH-dependent permeabilization of the plasma membrane of mammalian cells by anthrax protective antigen.

Protective antigen (PA) of anthrax toxin forms ion-conductive channels in planar lipid bilayers and liposomes under acidic pH conditions. We show here that PA has a similar permeabilizing action on the plasma membranes of CHO-K1 and three other mammalian cell lines (J774A.1, RAW264.7 and Vero). Changes in membrane permeability were evaluated by measuring the efflux of the K+ analogue, 86Rb+, from prelabelled cells, and the influx of 22Na+. The permeabilizing activity of PA was limited to a proteolytically activated form (PAN) and was dependent on acidic pH for membrane insertion (optimal at pH…

Molecular mechanism of pore formation by Plasmodium Perforin-like Protein 2.

Malaria-causing Plasmodium parasites must pass through several host cell types to complete their life cycle. This cell traversal is facilitated by perforin-like proteins (PLPs), among which PLP2 is essential for erythrocyte rupture by gametocytes. However, the mechanism by which PLP2 forms pores is not yet understood. Here, we combine cryo-electron microscopy and tomography to reveal the structural basis of Plasmodium vivax PLP2-mediated membrane attack. PvPLP2 assembles on lipid bilayers into heterogeneous arc- and ring-shaped pores with variable stoichiometries. Among them, we determine the …

A dual-recognition fluorescence chip utilizing a substrate-borne DNA walker for precision profiling of small extracellular vesicles.

We report a fluorescence chip based on a membrane-anchored, bipedal DNAzyme walker assembled on small extracellular vesicles (sEVs) surfaces via catalytic hairpin assembly (CHA). Unlike previously reported single-arm or proximity-ligation-dependent walkers on nanoparticle supports, this CHA-triggered DNAzyme walker is designed with a bipedal architecture and operates directly on the sEVs membrane. In our design, CHA simultaneously generates two DNAzyme arms on the same sEVs, enabling faster substrate cleavage kinetics and superior signal amplification. The assay achieves a theoretical detectio

Design principles of the common Gly-X6-Gly membrane protein building block.

Protein behavior in lipids is poorly understood and inadequately represented in current computational models. Design and prediction abilities for bilayer-embedded molecular structures may be improved by characterizing membrane proteins' most frequent, favored structural features to glean both context-specific and general principles. We used protein design to proactively interrogate the sequence-structure relationship and stabilizing atomic details of two highly prevalent antiparallel transmembrane (TM) motifs with Small-X6-Small consensus sequences.
