Synthesis, structural characterization, and cytotoxic evaluation of monofunctional cis-[Pt(NH3)2(N7-guanosine/2'-deoxyguanosine)X] (X = Cl, Br, I) complexes with anticancer potential.

A series of new monofunctional platinum(II) complexes of the type cis-[Pt(NH3)2(N7-guanosine/2'-deoxyguanosine)X] (X = Cl, Br, I) were synthesized and characterized using NMR spectroscopy, mass spectrometry, and ICP-atomic emission spectroscopy. These complexes are designed to address the limitations of conventional bifunctional platinum-based drugs, such as cisplatin, which include issues with cytotoxicity and selectivity towards cancer cells. By incorporating guanosine or 2'-deoxyguanosine ligands and varying halido substituents, the study investigated how structural modifications influence

Reversal of P-glycoprotein-mediated multidrug resistance by the novel tetrandrine derivative W6.

Overexpression of ATP-dependent efflux pump P-glycoprotein (P-gp) is the main cause of multidrug resistance (MDR) and chemotherapy failure in cancer treatment. Inhibition of P-gp-mediated drug efflux is an effective way to overcome cancer drug resistance. The present study investigated the reversal effect of the novel tetrandrine derivative W6 on P-gp-mediated MDR. KBv200, MCF-7/adr and their parental sensitive cell lines KB, MCF-7 were used for reversal study. The intracellular accumulation with P-gp substrates of doxorubicin was determined by flow cytometry. The expression of P-gp and ERK1/2

Enhanced anticancer efficacy of Pa-MAP 1.9 peptide-coated silver nanostars on breast cancer cells.

Breast cancer remains a major global health challenge due to its rising incidence and the limitations of conventional chemotherapeutic options. Antimicrobial peptides (AMPs) with dual antimicrobial and anticancer activities have emerged as promising alternatives; however, their clinical application is limited by issues such as stability, selectivity, and delivery. Nanocarrier systems, particularly silver nanostars (AgNSs), can address these challenges by improving peptide delivery and cellular interaction. This study explores the nanocomplex AgNSs-Pa-MAP 1.9 as a combinatory therapeutic platfo

Breast cancers utilize hypoxic glycogen stores via PYGB, the brain isoform of glycogen phosphorylase, to promote metastatic phenotypes.

In breast cancer, tumor hypoxia has been linked to poor prognosis and increased metastasis. Hypoxia activates transcriptional programs in ca … inhibition of glycogen utilization resulted in significantly decreased wound-healing capability in MCF-7 cells and a decrease in invasive potential of MDA-MB-231 cells. Thus, we identify PYGB as a novel metabolic target with potential applications in the management and/or prevention of metastasis in breast cancer.

An "off-on" fluorescent sensor based on FRET and magnetic beads for APE1 activity detection in breast cancer cell lysates.

The activity of apurinic/apyrimidinic endonuclease 1 (APE1) is a key biomarker in breast cancer. We report an "off-on" fluorescent nanosensor (OOFN) that combines fluorescence resonance energy transfer (FRET) with magnetic bead (MB) enrichment for simple and sensitive APE1 detection. The platform employs a MB-anchored FAM-labeled DNA strand (D1) hybridized with a BHQ1-labeled strand containing an AP site (D2). In the absence of APE1, close donor-acceptor proximity quenches fluorescence via FRET. APE1 cleaves the AP site, releases the quencher, and restores fluorescence, while MBs enable easy s

In vitro evaluation of ESE-15-ol, an estradiol analogue with nanomolar antimitotic and carbonic anhydrase inhibitory activity.

Antimitotic compounds are still one of the most widely used chemotherapeutic anticancer drugs in the clinic today. Given their effectiveness against cancer it is beneficial to continue enhancing these drugs. One way is to improve the bioavailability and efficacy by synthesizing derivatives that reversibly bind to carbonic anhydrase II (CAII) in red blood cells followed by a slow release into the blood circulation system. In the present study we describe the in vitro biological activity of a reduced derivative of 2-ethyl-3-O-sulphamoyl-estradiol (2EE), 2-ethyl-3-O-sulphamoyl-estra-1,3,5(10),15-

Macrocyclic histone deacetylase inhibitor-based near-infrared-responsive ionic nanomedicines for enhanced cancer therapy.

The integration of epigenetic therapy with photothermal treatment offers a promising strategy to enhance anticancer efficacy while minimizing systemic toxicity. Herein, we report the design and development of a novel class of ionic nanomedicines (INMs) synthesized from macrocyclic histone deacetylase inhibitors (mac-HDACis) and Near-Infrared (NIR) dye for combination cancer therapy. The mac-HDACi components, incorporating an N-(2-aminophenyl)acylamide Zinc-Binding Group (ZBG), were first protonated using acetic acid to yield the cationic compounds of mac-para HDACi and mac-meta HDACi. Ambient light drives verteporfin-induced protein cross-linking during standard laboratory sample processing.

Verteporfin (VP) is widely used as a light-independent pharmacologic inhibitor of the YAP-TEAD transcriptional complex, despite its potent photosensitizing properties. Here we show that VP-associated high-molecular-weight complexes and apparent depletion of key proteins on immunoblots can be substantially driven by ambient-light exposure during post-lysis handling, more than by light during cellular treatment. In this study, the hypothesis is tested independently using MEL270, HEK293, and MCF-7 cells treated with therapeutic and supra-therapeutic concentrations of VP under four systematically …
