Identification of PI3K alpha inhibitors through large-scale virtual screening and integrated molecular modeling, biophysical characterization, and ADMET profiling.

Lung cancer remains one of the leading causes of cancer-associated mortality globally, with non-small cell lung cancer (NSCLC) accounting for approximately 85% of all cases. The PI3K p110α catalytic subunit (PIK3CA) signaling pathway is often dysregulated in non-small cell lung cancer (NSCLC) and plays a vital role in promoting tumor cell proliferation, survival, and resistance to apoptosis, making it a clinically validated therapeutic target. Despite the approval of several PI3K p110α inhibitors, challenges including off-target toxicity, drug resistance, and suboptimal pharmacokinetic profile

The bitterness behind sweetness: an integrated 3D-QSAR and molecular modeling approach elucidates the structure determinants of taste purity in natural chalcones.

The bitter and astringent properties of plant-derived chalcone compounds result in lower sweet taste purity compared to that of sugars such as sucrose, thereby limiting their applications. To elucidate the relationship between the structure of chalcones and their sweet taste purity, a 3D-QSAR model was established according to the sensory experiments that evaluated the sweetness and aftertaste of 22 chalcone compounds. Based on Comparative Molecular Field Analysis (CoMFA) and Comparative Molecular Similarity Index Analysis (CoMSIA), key sites and functional groups affecting the sweet taste pur

Multi-modal machine learning and molecular modelling reveal structurally diverse inhibitors of Mycobacterium tuberculosis protein tyrosine phosphatase B.

Protein tyrosine phosphatase B (PtpB) is a virulence-associated phosphotyrosine phosphatase secreted by Mycobacterium tuberculosis (Mtb), known to disrupt host immune signaling by dephosphorylating key proteins. Targeting PtpB represents a rational strategy for anti-TB drug discovery. This study presents an integrative computational framework for identifying and evaluating small-molecule inhibitors of Mtb PtpB. QSAR models were constructed using four molecular fingerprint types, CDK, PubChem, MACCS, and AtomPairs2DCount, as regression models predicting pIC50 values. Multiple machine learning a

Slice-level vs. volumetric radiomics for molecular subtyping of breast cancer: impact of patient-level data partitioning on model performance.

INTRODUCTION: Molecular subtyping of breast cancer, particularly the discrimination between Luminal A and Basal-like tumours, is critical for guiding treatment decisions and establishing prognosis. Radiomics offers a promising complementary, non-invasive imaging biomarker to support biopsy-based subtyping; however, two methodological gaps limit the validity of existing studies: the absence of controlled 2D-vs.-3D feature comparisons, and the systematic underreporting of patient-level data leakage in multi-slice pipelines. METHODS: To address both limitations, this study benchmarks slice-level

Global spatiotemporal distribution characteristics and regional risk stratification model of acute myeloid leukaemia based on molecular genetics and cytogenetic features.

Genetic abnormalities in acute myeloid leukaemia (AML) patients are usually conducted in small or regional studies; there is currently a lack of research on whether genetic abnormalities in each continent have regional specificity. In this study, we collected and jointly analysed studies published between 1995 and 2023 that reported on molecular and cytogenetic abnormalities of AML patients. Our findings reveal that the incidence of molecular genetic abnormalities has been increasing annually, whereas cellular genetic abnormalities show an overall declining trend. We performed cluster analysis

Computational screening and molecular modeling of probiotic-derived peptides targeting the conserved HR1 domain of SARS-CoV-2 spike protein.

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), the causative agent of the global COVID-19 pandemic, relies on its spike glycoprotein to mediate host cell entry via binding to human angiotensin-converting enzyme 2 (hACE2). The S2 subunit of the spike protein contains heptad repeat regions (HR1 and HR2) that form a six-helix bundle fusion core, a critical step for viral-host membrane fusion. This study aimed to computationally screen a library of 318 probiotic-derived bacteriocins to identify potential inhibitors targeting the conserved HR1 domain. Evaluation included historical v

The developmental and inflammatory disease target protein ADAM17 is vulnerable to off-target interaction by the drug eltrombopag: Insights from molecular modeling.

The loss of proteolytic activity of ADAM17 causes birth defects and embryonic lethality. Conversely, inhibiting ADAM17 activity represents a potential strategy for treating inflammatory and autoimmune diseases. ADAM17 has an active site cleft with a divalent Zn ion and hydrophobic S1'/S3' subsites interconnected to form an L shaped cavity. However, it is currently unknown whether the active site of ADAM17 is susceptible to off-target inhibition by the small molecule drug eltrombopag, which contains metal-binding moieties and is classified as pregnancy category C by the FDA.
