Mathematical modeling of neural stem cell migration within brain using multi-fiber tractography.

An agent-based model of therapeutic neural stem cell (NSC) migration is developed and used to predict the migration of NSCs in naïve mouse brain. The model utilizes generalized q-sampling imaging which resolves white matter fibers that cross in the brain and is shown to better account for variations in NSC migration patterns as compared to diffusion tensor imaging. In calibrating the model to experimental data, we show that the model is able to reproduce the spatial distribution of NSCs in the mouse brain. In addition, we show that the spatial distribution of NSCs in the mouse brain is sensiti

Spatial transcriptomics uncovers the hybrid molecular identity, ciliated phenotype, and immune signature of adenomyosis lesions.

Adenomyosis is a common gynecological condition characterized by ectopic endometrial-like tissue deep within the myometrium, causing debilitating symptoms. Treatment options are limited to hormones that primarily focus on symptom management only or invasive hysterectomy. Despite its prevalence, the pathogenesis of adenomyosis remains poorly understood, considerably impeding the development of targeted, true disease modifying treatments. Here, through spatial transcriptomics analysis of full-thickness human uterine wall biopsies (n = 10), we demonstrate that adenomyosis lesions exhibit a distin

Moebius syndrome in association with hypogonadotropic hypogonadism.

The association between hypogonadotropic hypogonadism and multiple CNS lesions in a variety of disorders suggests a possible causative link between these clinical findings. Neural afferent input into the hypothalamus from higher CNS centers modulates GnRH secretion and derangements of these neural pathways could potentially result in diminished gonadotropin secretion and hypogonadism. This report describes a patient with multiple CNS defects secondary to Moebius syndrome and hypogonadotropic hypogonadism whose clinical features support the hypothesis that his CNS and endocrine defects may be c

Nose-to-brain drug delivery: A new frontier in targeted therapeutics.

Delivering therapeutics to the Central Nervous System (CNS) remains a major challenge in drug development, largely because the blood-brain barrier (BBB) limits brain exposure for most systemically administered agents. Among alternative delivery routes, intranasal administration has attracted significant interest because it leverages direct anatomical connections between the nasal cavity and the brain, thereby reducing reliance on BBB permeability. This review examines the nasal route, focusing on how olfactory and trigeminal neural pathways contribute to drug transport into the CNS. Rather tha

Integrated stress response genes as novel biomarkers and therapeutic targets in ankylosing spondylitis: a transcriptomic and Mendelian randomization study.

INTRODUCTION: Ankylosing spondylitis (AS) is a chronic inflammatory disorder with poorly defined pathogenic mechanisms. The integrated stress response (ISR), an evolutionarily conserved signaling network, is implicated in AS development. This research endeavored to identify biomarkers for AS, thereby offering novel targets and approaches for therapeutic intervention. METHODS: Transcriptomic profiling of peripheral blood from AS patients identified differentially expressed genes. Mendelian randomization (MR) was applied to infer causal associations between ISR-related genes and AS susceptibili

Gas Recognition-Signal Transduction Binary Synergism in Bioinspired 2D Nanoconfined Ionic Membranes Enables Ultrasensitive NH3 Perception.

Conventional gas devices suffer from interfacial impedance limitations, resulting in limited sensitivity and increased power consumption due to their heterogeneous function architecture. Inspired by the uniquely integrated "recognition-transduction" mechanism discovered in insect odorant-binding proteins, 2D nanoconfined ionic membranes are developed through graphene oxide assembly-confined with ionic liquids, achieving monolithic integration of gas recognition and ion transduction. Graphene oxide nanosheets facilitate interlayer gas diffusion pathways, achieving response and recovery times of

Microbiome-emitted scents activate olfactory neuron-independent airway-gut-brain axis to promote host growth in Drosophila.

While it is now accepted that the microbiome has strong impacts on animal growth promotion, the exact mechanism has remained elusive. Here we show that microbiome-emitted scents contain volatile somatotrophic factors (VSFs), which promote host growth in an olfaction-independent manner in Drosophila. We found that inhaled VSFs are readily sensed by olfactory receptor 42b non-neuronally expressed in subsets of tracheal airway cells, enteroendocrine cells, and enterocytes. Olfaction-independent sensing of VSFs activates the airway-gut-brain axis by regulating Hippo, FGF and insulin-like growth fa

Intranasal Drug Delivery Technology in the Treatment of Central Nervous System Diseases: Challenges, Advances, and Future Research Directions.

As population aging becomes an increasingly critical global issue, the incidence of central nervous system (CNS) diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), and stroke, has risen sharply. However, the blood-brain barrier (BBB) presents a significant obstacle to the effective treatment of these CNS disorders, limiting the ability of therapeutic agents to reach the brain. In this context, intranasal drug delivery, which bypasses the BBB, has attracted considerable attention in recent years. By utilizing pathways such as the olfactory and trigeminal nerves, intranasal

Pax6 affects Ras-Raf-ERK1/2 in mouse aging brain.

Pax6, a transcription factor and multifunctional protein, changes during aging. It also interacts with regulator proteins involved in cell metabolism and survival signalling pathways including Ras-GAP. Many forms of Ras, Raf and ERK1/2 are known but information on their region-specific expression patterns are unavailable from brain during aging. Therefore, it has been intended to evaluate expressions of Pax6 and forms of Ras, Raf, ERK1/2 in hippocampus, caudate nucleus, amygdale, cerebral cortex, cerebellum and olfactory lobe. Association of Pax6 with Ras, Raf and ERK1/2 was evaluated in co-cu

Potassium-dependent sodium-calcium exchanger (NCKX) isoforms and neuronal function.

K+-dependent Na+/Ca2+-exchangers (NCKX) are a relatively recently described five-member gene family of transporters which play a quantitatively significant role in neuronal Ca2+ transport. In this review we highlight the important individual contributions these transporters make to cellular Ca2+ homeostasis and neuronal function. Notably, different members of the family make distinct, non-redundant, contributions to critical behavioural pathways. In particular, NCKX proteins regulate the kinetics, termination and adaptation of Ca2+ signals in sensory transduction neurons in the olfactory and v

Canine distemper virus uses both the anterograde and the hematogenous pathway for neuroinvasion.

Canine distemper virus (CDV), a member of the Morbillivirus genus that also includes measles virus, frequently causes neurologic complications, but the routes and timing of CDV invasion of the central nervous system (CNS) are poorly understood. To characterize these events, we cloned and sequenced the genome of a neurovirulent CDV (strain A75/17) and produced an infectious cDNA that expresses the green fluorescent protein. This virus fully retained its virulence in ferrets: the course and signs of disease were equivalent to those of the parental isolate. We observed CNS invasion through two di

Sperm protein 17 expression defines 2 subsets of primary esthesioneuroblastoma.

Esthesioneuroblastomas (ENBs) are rare malignant tumors of the nasal vault, the origin, diagnosis, and management of which are still subjects of discussion. That there is no related prognostic factor or generally recognized therapeutic regimen highlights the need for further analyses of its underlying biologic features and investigations of new marker proteins that allow more reliable clinical testing. We here show that sperm protein 17 (Sp17) is expressed in the ciliated cells of the normal olfactory epithelium and in a proportion of primary ENB lesions. We found an association between Sp17 e

Psychotomimetics moderately affect dopamine receptor binding in the rat brain.

The hypothesis that psychotomimetics induce a rapid dopamine receptor regulation that could participate in the expression of the brain dopaminergic overactivation and in the early signs of psychotic-like behaviour, was checked by radioligand binding on rat brain cryosections. For this purpose, subchronic 7-day-d-amphetamine pretreatment was combined with acute amphetamine, phencyclidine or LSD challenge. Acute application of psychotomimetics affected only striatal and accumbens but not nigral and olfactory dopamine receptor binding after 40 min, while subchronic amphetamine expressed no effect

The neural cell adhesion molecule (NCAM) in development and plasticity of the nervous system.

The neural cell adhesion molecule (NCAM) is a member of the immunoglobulin superfamily and is strongly expressed in the nervous system. NCAM is found in three major forms, of which two--NCAM-140 and NCAM-180--are transmembrane proteins, while the third--NCAM-120--is attached to the membrane via a glycosylphosphatidyl inositol anchor. In addition, soluble NCAM forms exist in brain, cerebrospinal fluid, and plasma. NCAM mediates cell adhesion through homophilic as well as through heterophilic interactions. Following NCAM binding, transmembrane signalling is believed to be activated, resulting in

Molecular basis of the X-chromosome-linked Kallmann's syndrome.

Kallmann's syndrome combines hypogonadotropic hypogonadism and anosmia. The most frequent form of the disease is linked to the X chromosome and has been proposed to be due to a defect in the embryonic migration of GnRH neurons and olfactory axons from the nose to the brain. A candidate gene for the X-linked form of the disease has been isolated by positional cloning. Mutations in the open reading frame have been identified in several patients, providing convincing evidence that this gene is the actual gene, KAL, responsible for the X-linked Kallmann's syndrome. Correlations between molecular a

The thoracic muscular system and its innervation in third instar Calliphora vicina Larvae. II. Projection patterns of the nerves associated with the pro- and mesothorax and the pharyngeal complex.

We describe the anatomy of the nerves that project from the central nervous system (CNS) to the pro- and mesothoracic segments and the cephalopharyngeal skeleton (CPS) for third instar Calliphora larvae. Due to the complex branching pattern we introduce a nomenclature that labels side branches of first and second order. Two fine nerves that were not yet described are briefly introduced. One paired nerve projects to the ventral arms (VAs) of the CPS. The second, an unpaired nerve, projects to the ventral surface of the cibarial part of the esophagus (ES). Both nerves were tentatively labeled af

Central nervous system safety risks and evaluation strategies for mucosal vaccines: A review.

Mucosal vaccines administered via noninjectable routes, such as intranasal and oral delivery, elicit both systemic and local immunity. Concurrent breakthroughs in nanocarriers and adjuvant technologies have significantly enhanced the precision of antigen delivery and immune persistence. However, these innovations introduce unique risks to the central nervous system (CNS). Particles <200 nm may penetrate the blood-brain barrier (BBB), whereas proinflammatory adjuvants can activate intracerebral glial cells, thereby inducing neuroinflammation and potential neurotoxicity. Intranasal administratio

Effects of metal oxide inhalation on the transcription of some hormone receptors in the brain, examined in an in vivo mouse model.

Respirable metal oxide nanoparticles in welding fumes pose significant health risks upon inhalation, potentially leading to neurodegenerative diseases. While the exact mechanisms remain unclear, it is evident that metal oxide nanoparticles can disrupt cellular functions, including metabolism and inflammatory responses after crossing the blood-brain barrier (BBB). Our study investigates the impact of manual metal arc welding fumes on hormone receptor transcription in an in vivo mouse model. After collecting samples from six different brain regions at 24 and 96 h upon exposure, we focused on exp

Evaluation of the sense of taste and smell in patients with Neurofibromatosis Type 1.

OBJECTIVE: The objective of this study was to determine if patients with Neurofibromatosis Type 1 (NF1) have an impaired sense of smell or taste. Neurofibromin, the NF1gene protein product is ubiquitous in the body and is especially associated with the development of neurogenetic structures. Lately enlarged olfactory bulbs have been described in patients with NF1. Until now, there is no study to evaluate the sense of smell and taste in patients with NF1. METHOD: This study has been approved by the Hamburg Ethics committee. An evaluation of the sense of smell and taste was undertaken in 26 pat

Human olfactory neuroepithelial cells: tyrosine phosphorylation and process extension are increased by the combination of IL-1beta, IL-6, NGF, and bFGF.

Olfactory neuroepithelial cells (ONC) grown from biopsies of human donors are a novel cell culture system that may facilitate studies into normal and disease-related human neurobiology. We further characterized the expression of cell surface markers and intermediate filaments, and responses to neurotrophic factors by ONC. ONC are positive for cell surface markers N-CAM, PSA-N-CAM, neutral endopeptidase, N-aminopeptidase, NGF low-affinity receptor homologue (CD40), and transferrin receptor by flow cytometry for the intermediate filament proteins peripherin, vimentin, and NF-H by immunocytochemi
