The Obsessive-Compulsive Inventory-Revised: Replication of the psychometric properties in China.

The aim of this study is to replicate the findings from previous research about the psychometric properties of the Mandarin Chinese version of the Obsessive-Compulsive Inventory-Revised (OCI-R), which assesses the presence of symptoms obsessive-compulsive disorder and the distress associated with those symptoms. The final clinical sample included 80 participants from multiple psychiatric outpatient clinics in China. Participants completed the following questionnaires: the OCI-R, the Depression, Anxiety, and Stress Scale (DASS-21), the Yale-Brown Obsessive-Compulsive Scale-II (Y-BOCS-II), and t

Microvesicles from cerebrospinal fluid of patients with Alzheimer's disease display reduced concentrations of tau and APP protein.

Microvesicles are small membranous particles generated during cellular activation or stress. The analysis of the content and the surface of microvesicles allow conclusions about the cells they are originating from and the underlying pathology. Therefore, CSF microvesicles have been suggested to be promising targets to monitor the (etio)pathology of neurodegenerative diseases. Microvesicles in the CSF of 15 patients with Alzheimer's disease and 15 controls were analyzed by flow cytometry regarding the levels of CD3, CD4, CD45, CD64, BACE1, Aβ, APP and tau. The results were replicated in a secon

Stereotyped transcriptomic transformation of somatosensory neurons in response to injury.

In mice, spared nerve injury replicates symptoms of human neuropathic pain and induces upregulation of many genes in somatosensory neurons. Here we used single cell transcriptomics to probe the effects of partial infraorbital transection of the trigeminal nerve at the cellular level. Uninjured neurons were unaffected by transection of major nerve branches, segregating into many different classes. In marked contrast, axotomy rapidly transformed damaged neurons into just two new and closely-related classes where almost all original identity was lost. Remarkably, sensory neurons also adopted this

A Neural Circuit for Gut-Induced Reward.

The gut is now recognized as a major regulator of motivational and emotional states. However, the relevant gut-brain neuronal circuitry remains unknown. We show that optical activation of gut-innervating vagal sensory neurons recapitulates the hallmark effects of stimulating brain reward neurons. Specifically, right, but not left, vagal sensory ganglion activation sustained self-stimulation behavior, conditioned both flavor and place preferences, and induced dopamine release from Substantia nigra. Cell-specific transneuronal tracing revealed asymmetric ascending pathways of vagal origin throug

Integrative Analyses of De Novo Mutations Provide Deeper Biological Insights into Autism Spectrum Disorder.

Recent studies have established important roles of de novo mutations (DNMs) in autism spectrum disorders (ASDs). Here, we analyze DNMs in 262 ASD probands of Japanese origin and confirm the "de novo paradigm" of ASDs across ethnicities. Based on this consistency, we combine the lists of damaging DNMs in our and published ASD cohorts (total number of trios, 4,244) and perform integrative bioinformatics analyses. Besides replicating the findings of previous studies, our analyses highlight ATP-binding genes and fetal cerebellar/striatal circuits. Analysis of individual genes identified 61 genes e

Treatment of chronic hepatitis B patients with tyrosine-methionine-aspartate-aspartate mutations.

Lamivudine is an antiviral used for the treatment of chronic hepatitis B. Several studies have reported various mutations that are induced by lamivudine therapy. These mutations in the tyrosine-methionine-aspartate-aspartate (YMDD) motif are necessary and sufficient to confer high-level lamivudine resistance. During treatment with lamivudine, mutations develop in the YMDD motif of the hepatitis B virus (HBV) polymerase gene and lamivudine cannot prevent the replication of the mutant form. The virulence strain of developed mutation in the polymerase gene is lower than the original virus and the

A Brucella virulence factor targets macrophages to trigger B-cell proliferation.

Brucella spp. and Trypanosoma cruzi are two intracellular pathogens that have no evolutionary common origins but share a similar lifestyle as they establish chronic infections for which they have to circumvent the host immune response. Both pathogens have a virulence factor (prpA in Brucella and tcPrac in T. cruzi) that induces B-cell proliferation and promotes the establishment of the chronic phase of the infectious process. We show here that, even though PrpA promotes B-cell proliferation, it targets macrophages in vitro and is translocated to the cytoplasm during the intracellular replicati

Engagement of NKp30 on Vδ1 T cells induces the production of CCL3, CCL4, and CCL5 and suppresses HIV-1 replication.

Natural cytotoxicity receptors (NCRs) were originally identified as specific natural killer cell activating receptors that, on binding to their endogenous ligands, trigger the killing of tumor cell targets. We recently described the differentiation of a novel subset of NCR(+) Vδ1 T cells characterized by a remarkably high cytolytic potential against cancer cells. Here we demonstrate that the engagement of NKp30, one of the NCRs expressed de novo on Vδ1 T cells after stimulation, triggers the production of high levels of CCL3/MIP-1α, CCL4/ MIP-1β, and CCL5/RANTES but not of CXCL12/SDF-1. Antiviral effect of edaphic cyanophytes on rabies and herpes-1 viruses.

Five cyanophyte species (Amorphonostoc punctiforme, Gloeocapsa turgidus, Sphaeronostoc coeruleum, Stratonostoc linckia f. spongiaeforme and Synechococcus cedrorum) were isolated and identified from sandy Egyptian soils. Polysaccharides extracted from these species showed a pronounced antiviral activity against Rabies and Herpes-1 viruses represented by the absence of the characteristic cytopathic effects of these viruses. It was found that 100 μg polysaccharide/ml induced 100% inhibition of the two viruses which, depending on the polysaccharide concentration. Both of Gloeocapsa turgidus and Sy

In vitro and in vivo genetic stability studies of a human adenovirus type 5 recombinant rabies glycoprotein vaccine (ONRAB).

Investigation into the genetic stability of a replication-competent human adenovirus rabies glycoprotein recombinant (ONRAB) developed for use as an oral vaccine for wildlife rabies prevention is of major importance due to the vaccine's intended placement in the environment. Using a collection of murine monoclonal antibodies directed to six distinct antigenic sites on the rabies glycoprotein, preservation of all main immunogenic epitopes of the protein after virus growth in vitro was established. A competition experiment which involved the in vitro passaging of a mixture of ONRAB and wild-type

Framing effects under cognitive load: the role of working memory in risky decisions.

Framing effects occur in a wide range of laboratory and natural decision contexts, but the underlying processes that produce framing effects are not well understood. We explored the role of working memory (WM) in framing by manipulating WM loads during risky decisions. After starting with a hypothetical stake of money, participants were then presented a lesser amount that they could keep for certain (positive frame) or lose for certain (negative frame). They made a choice between the sure amount and a gamble in which they could either keep or lose all of the original stake. On half of the tria

[Early events in chlamydial infection of host cells].

Chlamydiae, gram-negative, obligate intracellular bacteria, are major pathogens worldwide, causing several diseases including trachoma, respiratory diseases and sexually transmitted disease. Penetration of chlamydiae to epithelial cells, the environment which supports their growth and survival, leads to various events that begin with changes to the bacteria-containing vacuole, allowing for its progression from the endosomal to the exocytic pathway. The changes include fusion with vesicles carrying glycerophospholipids and sphingolipids and originating in the endoplasmic reticulum (ER) and the

Inactivation of expression of gene 4 of mouse hepatitis virus strain JHM does not affect virulence in the murine CNS.

The protein encoded by ORF 4 of mouse hepatitis virus (MHV) is not required for growth of some strains in tissue culture cells, but its role in pathogenesis in the murine host has not been defined previously in a controlled manner. MHV strain JHM causes acute and chronic neurological diseases in susceptible strains of rodents. To genetically manipulate the structural proteins of this and other strains of MHV, we have generalized an interspecies-targeted RNA recombination selection that was originally developed for the A59 strain of MHV. Using this approach, a recombinant MHV-JHM was constructe

Unusual intracellular trafficking of Salmonella typhimurium in human melanoma cells.

Salmonella spp. are enterobacteria capable of invading and replicating in both professional and non-professional phagocytes. Here, we investigate the fate of S. typhimurium in human melanoma MelJuSo cells. The bacterium entered MelJuSo cells by a trigger mechanism and resided within a unique organelle, the Salmonella-containing vacuole (SCV). The SCV acquired early endosomal markers transiently and then underwent a series of membrane modifications. In HeLa cells, vacuole maturation is characterized by the simultaneous acquisition of the lysosomal membrane glycoproteins (Lgps) Lamp1, CD63 and v

Bimanual finger tapping: effects of frequency and auditory information on timing consistency and coordination.

The authors' goal in this study was to probe the basis for an earlier, unexpected finding that preferred-frequency finger tapping tends to have higher frequencies and to be less stable for in-phase than for antiphase tasks. In follow-up experiments, 3 protocols were employed: a preferred-frequency replication in both coordination modes, a metronome-driven matching of the preferred frequencies to each of the coordination modes, and a frequency scaling of both modes. The original findings were affirmed for preferred frequency. Tapping to a metronome had a differential effect on in-phase and anti

Factor structure, reliability, and validity of the Pain Catastrophizing Scale.

The Pain Catastrophizing Scale (PCS; Sullivan et al., Psychol. Assess. 7, 524-532, 1995) has recently been developed to assess three components of catastrophizing: rumination, magnification, and helplessness. We conducted three studies to evaluate the factor structure, reliability, and validity of the PCS. In Study I, we conducted principal-components analysis with oblique rotation to replicate the three factors of the PCS. Gender differences on the original PCS subscales were also analyzed. In Study II, we conducted confirmatory factor analyses to evaluate the adequacy of fit of four alternat

On a failure to replicate: methodologically close, but not close enough. A response to hogben et al.

Williams, Brannan and Lartigue (1987) (Clinical Vision Science, 1, 367-371) reported that poor readers took significantly longer to search letter arrays for a target than did good readers. In addition, they reported that blurring the letter arrays leads to faster search times for poor readers and a loss of the significant differences between the groups seen with unblurred displays. In a recent attempt to replicate these findings, Hogben et al. (1996) (Vision Research, 36, 1503-1507) found no differences in search rates between good and poor readers using unblurred arrays, and no differences in

Similar effects of antidepressant and non-antidepressant drugs on behavior under an interresponse-time greater than 72-s schedule.

Antidepressant drugs were reported to decrease responses and increase reinforcements in water-deprived male albino rats pressing a lever for water on a schedule requiring a pause of at least 72 s between responses (IRT greater than 72). Subsequently other investigators, using food-deprived ovariectomized hooded rats pressing a lever for food, showed that antipsychotic drugs produced the same effect as antidepressants. Because methodologies differed somewhat, the present study was designed to replicate closely the experimental conditions of the original studies, e.g., same strain and sex, same

One, Two, Three, Four… What's More? Successor Relations, not Mappings to Approximate Magnitudes, Support Learning the Later-Greater Principle.

As we count up, number words that occur later in our count denote greater quantities. Here, we tested whether children's understanding of this "later-greater" principle originates from associating number words with approximate magnitudes, or if instead it arises from knowledge of successor relations between individual number words. In Experiment 1 (N = 60), we found that children can identify the greater of two small number words (e.g., "six" vs. "ten") based on associative mappings between number words and the ANS but do not rely on such mappings to compare larger numbers (e.g., "twenty-four"

Cross-order detection of bacteriophage transduction in microbial communities using RNA barcoding.

Bacteriophages (phages) facilitate gene transfer and microbial evolution in all ecosystems and have applications as tools for engineering microbiomes and as antimicrobials. Historic efforts to map phage hosts, such as plaque assays, are limited to cultured bacteria, are low throughput, and are hard to apply in microbial communities and environmentally-relevant contexts. To overcome these limitations, we integrate a synthetic ribozyme that stores information about participation in horizontal gene transfer in 16S ribosomal RNA (rRNA) into the phage-plasmid P1, and perform targeted 16S rRNA seque
