[Mechanism of the analgesic effect of narcotic analgetics].

A study was made of the effect of morphine, promedol, phentanyl, pentazacine and psychostimulant d,l-amphetamine on the threshold of pain sensitivity and self-stimulation of the hypothalamus and the septum in rats. Electrical stimulation of the systems of positive reinforcement of the hypothalamus and the septum, and also analgetics increased the threshold of pain sensitivity, whereas d,l-amphetamine failed to influence it. D,l-amphetamine and morphine facilitated, promedol failed to influence, phentanyl decreased and pentazacine completely depressed the hypothalamic self-stimulation. The sept…

Development of Parkinson's disease in patients with Narcolepsy.

Although amphetamine drugs can damage dopaminergic axons, it is unknown whether chronic treatment with amphetamine increases the risk of developing Parkinson's disease (PD). Of 1,152 consecutive PD patients, 3 had a prior diagnosis of narcolepsy. This rate is five times higher than expected (p = 0.02). These patients had typical onset of narcolepsy and underwent treatment with amphetamine. Although preliminary, this observation raises the possibility that some factors intrinsic to narcolepsy or its treatment may be a risk factor for PD.

Priapism and quetiapine: a case report.

Priapism is a "persistent erection not accompanied by sexual desire or stimulation, usually lasting more than six hours and typically involving only the corpora cavernosa." Here we report on a gay male patient from our HIV/AIDS mental health clinic who developed serious priapism on quetiapine and recreational amphetamine. Gay men are at high risk for amphetamine use, and as such, this potential association between priapism, quetiapine, and amphetamine use should be considered in making prescription decisions with these patients.

[Routine case of "auto-erotic accident"--surprising turn in evaluation of cause of death after chemical-toxicologic analysis].

A man with indications of autoerotic manipulations was found dead in his flat and a homicide had to be excluded. Diverse material found in the flat suggested that the man was a consumer of cocaine. But the analyses of blood, urine and organs showed high concentrations of amphetamine and it was thought about an unintentionally mix-up with cocaine. A hair analysis was made and only amphetamine was detected. Therefore the utensils (mirror, razor-blade and sniff-pipe) belong to a rare nasal consumption of amphetamine.

[Mechanism of pantogam potentiation of certain effects of hexobarbital and sodium barbital and inhibition of phenamine hyperactivity].

Experiments on albino mice and rats have shown that pantogam, a derivative of pantothenic acid, potentiates the hypnotic effects of hexobarbital and barbital and enhances the effect of subthreshold doses of hexobarbital. The drug inhibits the amphetamine action in the amphetamine hyperaction test without affecting hexobarbital and amphetamine metabolism, or without increasing the blood-brain barrier permeability for these compounds and barbital. Pantogam does not influence the intensity of ethylmorphine N-demethylation in liver homogenates and the content of cytochrome P 450 and b5 in liver mi…

Impact of Recent Methamphetamine Use on Vancomycin Clearance.

PURPOSE: To determine whether recent methamphetamine use increases vancomycin clearance. METHODS: This was a multi-center, retrospective, IRB-approved study at two tertiary care medical centers. Adult patients with a urine drug screen, ≥3 consecutive vancomycin doses, and an appropriately drawn vancomycin trough were assessed and classified as amphetamine positive or amphetamine negative. The primary outcome was vancomycin clearance. RESULTS: 88 patients were included in the analysis, with 44 patients in each group. Vancomycin clearance was greater in the amphetamine positive group (94.54 vs…

Pharmacotherapy of methamphetamine addiction: an update.

Methamphetamine dependence is a serious public health problem worldwide for which there are no approved pharmacological treatments. Psychotherapy is still the mainstay of treatment; however, relapse rates are high. The search for effective pharmacological treatment has intensified in the last decade. This review will highlight progress in pharmacological interventions to treat methamphetamine dependence as well as explore new pharmacological targets. Published data from clinical trials for stimulant addiction were searched using PubMed and summarized, as well as highlights from a recent sympos…

Neurochemical and behavioral factors in the development of tolerance to anorectics.

Tolerance and cross-tolerance among several anorectic drugs were investigated in four separate tests. The same animals served as subjects in all four tests. In the first test, animals given milk shortly after injection of 3 mg/kg amphetamine (Cont group) developed tolerance to amphetamine anorexia, but animals given milk when amphetamine's anorectic effects had worn off (Noncon group) did not develop tolerance in spite of equal drug exposure. In the second test, Cont animals were tolerant to 2 mg/kg apomorphine, a drug with a neurochemical action related to amphetamine. No tolerance to 2 mg/kg…

Reinforcing and discriminative-stimulus effects of ephedrine isomers in rhesus monkeys.

Ephedrine is a sympathomimetic drug that is currently found in many over-the-counter preparations. This compound exists as four isomers which, in addition to a racemic mixture, were evaluated for their positive reinforcing effects and for their similarity to (+)-amphetamine as a discriminative stimulus. Rhesus monkeys (N=3) with intravenous cocaine (0.1 mg/kg/inj) or saline as a consequence for lever pressing were shown to self-administer all of the ephedrine compounds (range tested: 0.03-3.0 mg/kg/inj), with the exception of (-)-pseudoephedrine, when each drug/dose was substituted for cocaine…

Anorectic drugs which stimulate thermogenesis.

Two anorectic drugs commonly prescribed as adjuncts in weight control and a third experimental drug were studied in rats for their anorectic and possible thermogenic activities. Diethylpropion, a congener of amphetamine, mazindol, which is chemically unrelated to amphetamine, and ciclazindol, an experimental drug structurally similar to mazindol, were given in graded doses to determine their effect on food and oxygen consumption (VO2). Anorectic effects exhibited by diethylpropion and mazindol were similar and more potent than ciclazindol. Both resting and anesthetized VO2 measurements were do…

Site of action of anorectic drugs: glucoprivic- versus food deprivation-induced feeding.

Feeding induced by 2-deoxyglucose was compared with feeding induced by food deprivation in terms of antagonism by anorectic drugs and of anatomical site of action. Glucoprivic feeding was completely blocked by microinjection of amphetamine, fenfluramine, and mazindol into the paraventricular nucleus of the hypothalamus (PVN). Deprivation-induced feeding was not blocked by amphetamine, fenfluramine, or mazindol microinjected into the PVN. Neither the feeding induced by 2-deoxyglucose nor its reversal by amphetamine were blocked by pretreatment with the beta-adrenergic antagonist, propranolol. A…

Evidence for 5-HT2 receptor mediation in quipazine anorexia.

Doses of d-amphetamine (3.2 mg/kg), fenfluramine (10 mg/kg) and quipazine (8 mg/kg) cause a significant reduction in food intake during a 30-min daily feeding session in food-deprived rats. Pirenperone and ritanserin, 5-HT2 receptor antagonists, significantly blocked the anorectic effect of quipazine, while d-amphetamine and fenfluramine effects were not modified. Metergoline, a non-specific blocker of 5-HT receptors, significantly blocked the anorectic effects of fenfluramine and quipazine, but not the d-amphetamine effect. Pretreatment with alpha- and beta-adrenergic receptor antagonists (pr…

Is Hashimoto's Thyroiditis Associated with Insulin Resistance Markers?

Hashimoto's thyroiditis causes cell death, hypothyroidism, and insulin resistance. Cholecystokinin, orexin-A, and cocaine and amphetamine-regulated transcript are essential regulators of various endocrine and autonomic processes, including food intake. We aimed to determine the cocaine and amphetamine-regulated transcript, orexin-A, and cholecystokinin levels in Hashimoto's thyroiditis patients and assess the relationships between the metabolic parameters and Hashimoto's thyroiditis. The present study was carried out in 45 Hashimoto's thyroiditis patients and 40 healthy control groups. Serum c…

Altered visual perception in long-term ecstasy (MDMA) users.

RATIONALE: The present study investigated the long-term consequences of ecstasy use on visual processes thought to reflect serotonergic functions in the occipital lobe. Evidence indicates that the main psychoactive ingredient in ecstasy (methylendioxymethamphetamine) causes long-term changes to the serotonin system in human users. Previous research has found that amphetamine-abstinent ecstasy users have disrupted visual processing in the occipital lobe which relies on serotonin, with researchers concluding that ecstasy broadens orientation tuning bandwidths. However, other processes may have a…

Dopaminergic activity in Tourette syndrome and obsessive-compulsive disorder.

Tourette syndrome (TS) and obsessive-compulsive disorder (OCD) both are neuropsychiatric disorders associated with abnormalities in dopamine neurotransmission. Aims of this study were to quantify striatal D2/3 receptor availability in TS and OCD, and to examine dopamine release and symptom severity changes in both disorders following amphetamine challenge. Changes in [(11)C]raclopride binding potential (BP(ND)) were assessed using positron emission tomography before and after administration of d-amphetamine (0.3 mg kg(-1)) in 12 TS patients without comorbid OCD, 12 OCD patients without comorbi…

Adrenal medulla grafts in the hemiparkinsonian rat: profile of behavioral recovery predicts restoration of the symmetry between the two striata in measures of pre- and postsynaptic dopamine function.

Following unilateral striatal dopamine depletion, the hemiparkinsonian rat exhibits rotational behavior in response to amphetamine and apomorphine. The rotational behaviors induced by these drugs are thought to reflect an asymmetry in presynaptic striatal dopamine release and an asymmetry in postsynaptic striatal dopamine receptor function, respectively. Grafts of adrenal medulla cells in the lateral ventricle of hemiparkinsonian rats have been reported to reduce behavioral asymmetry. More than one profile of behavioral recovery, however, is observed. Some animals show a graft-induced decrease…

The global epidemiology and burden of psychostimulant dependence: findings from the Global Burden of Disease Study 2010.

AIMS: To estimate the global prevalence of cocaine and amphetamine dependence and the burden of disease attributable to these disorders. METHODS: An epidemiological model was developed using DisMod-MR, a Bayesian meta-regression tool, using epidemiological data (prevalence, incidence, remission and mortality) sourced from a multi-stage systematic review of data. Age, sex and region-specific prevalence was estimated for and multiplied by comorbidity-adjusted disability weightings to estimate years of life lost to disability (YLDs) from these disorders. Years of life lost (YLL) were estimated f…

Glucocorticoids involvement in the control of CNS excitability.

The effect of dexamethasone (DEX) and its interaction with psychostimulants agents (morphine, cocaine and amphetamine) has been studied on locomotor activity and straub reaction in mice: a) Morphine administration, (30-75-150 mg/kg,ip) induced a dose-related increase of the locomotor activity of mice, whereas DEX per se (0.1-1.0-10 mg/kg,ip) did not modify the activity of control mice. Pretreatment of mice with DEX 0.1 mg did not alter the hyperactivity produced by the three doses of morphine. In contrast, DEX administered at 1.0 mg reduced the morphine effects on locomotor activity, whereas D…

Diffuse Alopecia Areata Associated with Weight-Loss Pills.

Alopecia associated with ingestion of amphetamines has been reported occasionally. These drugs may act as a triggering factor to develop diffuse alopecia. We present a case of alopecia areata possibly linked to the ingestion of amphetamines. This case may provide evidence that amphetamines can also be a cause of alopecia areata in susceptible individuals. We intend to awaken an interest in the medication history of patients presenting with sudden diffuse alopecia areata.
