OBJECTIVE: To establish a protocol for the simultaneous isolation and high-purity purification of primary astrocytes and microglia from neonatal rats cerebral cortex.
METHODS: Single-cell suspensions were prepared from cerebral cortices of postnatal day 2 (P2) rat pups. Fibroblasts were pre-removed using differential adhesion techniques. Mixed glial cultures were maintained with graded serum (from 10% to 5% to 2% FBS) to suppress fibroblast proliferation. On day 14, microglia were isolated by constant temperature shaking (200 rpm, 12 h, 37 °C), followed by manual agitation of remaining adhere…

Autism spectrum disorder (ASD) is a group of complex neurodevelopmental conditions characterized by persistent deficits in social interaction, communication, and repetitive behavior. Although the etiology of ASD has traditionally been linked to neuronal dysfunctions, emerging evidence highlights the significance of non-neuronal cells, particularly microglia and astrocytes, in the pathophysiology of ASD. Although both glial cell types exhibit diverse functional states, microglia are primarily associated with eliminating unused neurons and synapses during early brain development, whereas astrocy…

Beta-amyloid (Aβ) aggregates interact with glial cells, particularly astrocytes, leading to altered morphology, disrupted homeostasis, and ultimately contributing to neurodegeneration in Alzheimer's disease (AD). Astrocytes, once considered passive support cells, are now recognized as active participants in neuroinflammatory and neurodegenerative cascades. Recent research has highlighted profound transcriptomic and metabolomic disturbances in astrocytes under AD pathology, implicating them as key players in disease progression. Nobiletin (NOB), a polymethoxylated flavonoid abundantly found in …

Chemotherapy-induced neuropathic pain (CINP) can be triggered by microtubule-targeting agents, including plant-derived vinca alkaloids such … tivation. Furthermore, immunofluorescence double staining demonstrated TCF12 localization in spinal astrocytes of VCR-treated male mice. Knockdown of TCF12 via siRNA attenuated chemotherapy-induced mechanical allodynia and thermal hyperalgesia, and reduced MAVS expression. Methylmercury (MeHg) is a potent neurotoxin that can cross the blood-brain barrier and disrupt neurological function in animals. Emerging evidence suggests that MeHg neurotoxicity may originate from compromised astrocytes, as they are strategically positioned to metabolize and process substances that enter the central nervous system. Thus, a better characterization of the timely cellular responses of astrocytes upon MeHg exposure is key to delineating the toxicology of MeHg. Here, MeHg exposure caused a transient increase in reactive oxygen species (ROS) and intracellular calcium concentration…

BackgroundStudies suggest a strong association between astrocytes, neuronal DNA damage, elevated amyloid-β, and brain degeneration in Alzheimer's disease (AD).ObjectiveThis study aimed to show whether astrocytes with damaged DNA affect human neuronal progenitor cells (NPCs) or differentiated neurons in close proximity, dependent on astrocytic APOE allele expression.MethodsImmortalized human astrocytes (hTERT) expressing APOE were treated with etoposide to induce DNA damage and co-cultured in a transwell system with human NPCs or differentiated neurons. We used western blotting and immunostaini…

Alzheimer's disease (AD) is associated with synaptic and memory dysfunction. One of the hallmarks of AD is reactive astrogliosis, with reactive astrocytes surrounding amyloid plaques in the brain. Astrocytes have also been shown to be actively involved in disease progression, nevertheless, mechanistic information about their role in synaptic transmission during AD pathology is lacking. Astrocytes maintain synaptic transmission by taking up extracellular glutamate during synaptic activity through astrocytic glutamate transporter GLT-1, but its function has been difficult to measure in real-time…

Astrocytes play vital roles in regulating brain states across organisms. Specifically, they serve several roles in regulating breathing behaviors and associated brain states, including facilitating transitions between phases of breathing by sensing small changes in O2 and CO2 levels, regulating the sleep-wake cycle, and impacting arousal and wakefulness. Here, we tested the hypothesis that astrocytes in the ventral respiratory column (VRC) are important for arousal and sigh generation in alert mice (Aldh1l1Cre). Using calcium imaging we show that some Aldh1l1 cells are phase-locked with sigh g…

Pediatric stroke, a significant cause of long-term neurological deficits in children, often arises from disruptions within neurovascular unit (NVU) components. The NVU, a dynamic ensemble of astrocytes, endothelial cells, pericytes, and microglia, is vital for maintaining cerebral homeostasis and regulating vascular brain development. Its structural integrity, particularly at the blood-brain barrier (BBB), depends on intercellular junctions and the basement membrane, which together restrict paracellular transport and shield the brain from systemic insults. Dysfunction in this intricate system …

Astrocyte heterogeneity is closely associated with the pathophysiology of traumatic brain injury (TBI), particularly in the development of c … that correlates with clinical severity and outcomes in patients with TBI. Using Aldh1l1CreERT2; Irf1flox/flox mice, we demonstrated that astrocyte-specific deletion of Irf1 mitigates astrocyte-medi …

Astrocytes regulate synaptic transmission in healthy and pathological conditions, but their involvement in modulating synaptic transmission in chronic pain is unknown. Our study demonstrates that astrocytes in the anterior cingulate cortex (ACC) exhibit abnormal calcium signals and induce the release of glutamate in male mice. This leads to an elevation in extracellular glutamate concentration, activation of presynaptic kainate receptors, and an increase in synaptic transmission following neuropathic pain. We discovered that the abnormal calcium signals are caused by the reappearance of metabo…

BACKGROUND: The pathomechanism of blast traumatic brain injury (TBI) and blunt TBI is different. In blast injury, evidence indicates that a … lished the implication of PARP1 following blast overpressure (BOP) and its differential response on astrocytes and microglial cells. We found that the inhibition of PARP is proven beneficial by attenuating oxidative stress. In this study, we hypothesized the involvement of the PARP1-SIRT-NRF2 axis following induced blast-induced PARP over-activation in glial …

Fragile X syndrome (FXS), a leading inherited cause of autism spectrum disorder and intellectual disability, has been studied extensively using rodent models. More recently, human stem cell-derived model systems have also been used to gain mechanistic insights into the pathophysiology of FXS. However, these studies have focused almost exclusively on neurons. Further, despite growing evidence for a key role of glia in neuronal function in health and disease, little is known about how human astrocytes are affected by FXS. Therefore, in this study, we successfully developed a protocol that captur…
