Acute-on-chronic liver failure: consensus recommendations of the Asian Pacific association for the study of the liver (APASL): an update.

The first consensus report of the working party of the Asian Pacific Association for the Study of the Liver (APASL) set up in 2004 on acute-on-chronic liver failure (ACLF) was published in 2009. With international groups volunteering to join, the "APASL ACLF Research Consortium (AARC)" was formed in 2012, which continued to collect prospective ACLF patient data. Based on the prospective data analysis of nearly 1400 patients, the AARC consensus was published in 2014. In the past nearly four-and-a-half years, the AARC database has been enriched to about 5200 cases by major hepatology centers acr

Methamphetamine-Induced Cognitive Deficits and Psychiatric Symptoms Are Associated with Serum Markers of Liver Damage.

Cognitive deficits and psychiatric disorders have been regarded as the most common clinical symptoms of methamphetamine (MA) users. Accumulating evidence has shown that liver disease may be involved in cognitive deficits and psychiatric disorders. This study examines whether cognitive deficits and psychiatric symptoms are associated with serum levels of liver biomarkers in MA users. Cognition was assessed by the Repeatable Battery for the Assessment of neuropsychological Status (RBANS). Psychiatric symptoms were assessed by the Symptom Checklist-90 (SCL-90). Liver function was assessed by seru

[Excitatory effects of glucocorticoids on neuronal activity in the medial vestibular nucleus--mediation by glucocorticoid receptor on the membrane].

Although glucocorticoids are sometimes used for the treatment of vertigo in certain disorders such as Meniere's disease, the mechanism underlying anti-vertigo effect remains unknown. The present study was performed to examine the effects of a glucocorticoid, dexamethasone, on neuronal activity in the medial vestibular nucleus (MVN) to determine whether or not the drug acts directly on the MVN neuron using alpha-chloralose-anesthetized cats which were fixed in a stereotaxic instrument placed on a turn-table. Single neuronal activities in the MVN were extracellularly recorded with a glass-insula

Genotoxic evaluation of chemical fractions of Cymbopogon citratus (DC) Stapf using plasmid assay.

INTRODUCTION: Cymbopogon citratus (DC.) Stapf is a medicinal plant widely used in traditional medicine in Africa, Asia and Latin America, mainly consumed as aqueous decoctions or infusions to treat fever, colds, cough, gastrointestinal disorders, hypertension, inflammation and anxiety. Despite its extensive traditional use, information on the genotoxic and oxidative effects of its individual chemical fractions remains limited. METHODS: The DNA strand breaks and oxidative effects of the aqueous, butanolic and essential oil fractions obtained from the aqueous extract of C. citratus were evaluat

Downsizing Electrophilic MAX-Derived Helical Zipper Peptides to Covalently Bind and Degrade Oncogenic MYC.

MYC is an intrinsically disordered oncoprotein that drives cancer progression via widespread transcriptional dysregulation. Upon binding partner protein MAX, their basic-helix1-loop-helix2-Zip (bHLH-Zip) domains assemble into an ordered heterodimer that binds E-box DNA to promote cellular proliferation. This interface has been undruggable by small molecules. Here, we report a novel covalent downsizing strategy to inhibit MYC-MAX interaction by targeting the C-terminal Cys438 present in MYC, but absent in MAX and related proteins. Incorporating a mild acrylamide electrophile to covalently bond

Storage-Environment-Dependent Surface Evolution and Electrochemical Degradation of High-Nickel NCM Single-Crystal Cathodes.

High-nickel ternary single-crystal cathodes are promising for high-energy-density batteries in low-altitude-economy applications owing to their high specific capacity. However, surface residual lithium formation during storage substantially impairs their electrochemical performance. Herein, we investigate the formation mechanisms of surface residual lithium and the associated storage-induced degradation of three single-crystal compositions-NCM613, NCM8866, and NCM9442-under vacuum, humid air, argon, pure CO2, and ultrapure water. Potentiometric titration, x-ray diffraction, x-ray photoelectron

Advances in the Application of Amino Acids in the Preservation of Postharvest Fruits and Vegetables.

Postharvest loss of fruits and vegetables remains a significant global challenge, while concerns over chemical residues, energy consumption, and storage-induced physiological disorders have accelerated the search for safe and eco-friendly preservation strategies. As key plant metabolites and signaling molecules, amino acids are gaining more attention for their roles in postharvest biology. This article reviews the primary postharvest issues of fruits and vegetables, the functional roles of representative amino acids, including γ-aminobutyric acid (GABA), glutamate (Glu), arginine (Arg), cystei

A nature-inspired peptide from the Boana cordobae frog as a potent and reversible AChE inhibitor with anti-amyloid and neuroprotective activities.

Alzheimer's disease (AD) is a multifactorial and progressive neurodegenerative disorder for which no effective treatment currently exists. The development of multitarget-directed ligands (MTDLs) capable of simultaneously modulating several pathological pathways represents a rational strategy to address its complex etiology. In this study, we report the isolation, chemical synthesis, and functional characterization of BcI-4, a short cationic peptide identified from the skin secretion of the Argentinean frog Boana cordobae. The peptide exhibited potent and reversible inhibitory activity against

Multi-target Triazole-Benzopyrone hybrids modulating cholinergic dysfunction, oxidative stress, and neuroinflammation through GFAP/NF-κB/APOE/NLRP3 axis in Alzheimer's disease.

Alzheimer's disease (AD) is a degenerative neurological disorder characterized by a deterioration in cognitive abilities, especially memory and learning. The main aim of this study is to evaluate the effects of our agents on oxidative stress, neuroinflammation, cognitive function, and behavioral performance in an LPS-induced AD animal model and comprehensive in vitro and in vivo assays. The synthesized compounds, namely 4b, 5b, 6, 8a-d, and 11a-c, revealed acetylcholinesterase inhibitory activity (3.50-5.91 nM) superior to that of the reference drug donepezil (6.33 nM). The IC50 value of 11a t

Altered molecular signaling pathways in the hippocampus of rhesus monkeys following chronic alcohol use.

Context-induced relapse is a significant factor limiting recovery from alcohol use disorder (AUD). However, the molecular processes in the hippocampus, critical for contextual memory, impacted by chronic alcohol use, remain poorly understood. We used a non-human primate model to test the hypothesis that chronic alcohol use impacts hippocampal molecular pathways that may serve as therapeutic targets for memory processing in chronic alcohol use. We conducted RNAseq profiling on hippocampal samples from adult male rhesus monkeys with chronic alcohol use (n = 7) and controls (n = 5). We identified

An induced pluripotent stem cell-based chemical genetic approach for studying spinal muscular atrophy.

INTRODUCTION: Spinal muscular atrophy (SMA) is a genetic disease characterized by degeneration of spinal cord motor neurons and neuromuscular junctions. Despite recent developments in therapies for SMA, treatment efficacy largely relies on the administration of drugs early in disease progression and is impacted by underlying patient genetics. Drug discovery for other diseases of the central nervous system (CNS) has also been hindered by heterogeneity in patient genetics and clinical presentations, as well as the need for early intervention. METHODS: To address these hurdles, we utilized a che

Multimodal Approach for Recalcitrant Melasma Using Picosecond Laser and Topical JAK Inhibition: A Case Report.

BACKGROUND: Melasma is a chronic hyperpigmentation disorder with high relapse rates, particularly in Fitzpatrick skin types III-V. Increasing evidence implicates cytokine-driven inflammation and visible light-induced melanogenesis as key contributors to disease persistence. These mechanisms support the rationale for exploring a multimodal therapeutic approach. CASE PRESENTATION: A woman in her early 30s with phototype IV presented with a 5-year history of recalcitrant centrofacial melasma. Previous treatments-including hydroquinone, oral tranexamic acid, and superficial chemical peels-produce

Central and peripheral plasticity after chemical unilateral labyrinthectomy: Glial responses, calyces dynamics, and behavioral consequences.

Peripheral vestibular disorders are highly prevalent, but the underlying neurobiological mechanisms remain poorly understood, particularly regarding bilateral peripheral plasticity following unilateral injury. Using a rat model of chemical unilateral labyrinthectomy (cUL) (SHAM, n = 14; cUL, n = 20), we combined behavioral analyses, auditory assessments, immunohistochemistry, and histological quantification to characterize central and peripheral adaptations from 1 to 30 days after vestibular deafferentation. cUL induced significant postural deficits from day 1, while hyperactivity and anxiety-
