Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare and aggressive hematologic malignancy typically involving the skin, bone marrow, and central nervous system. Ocular adnexal involvement is exceedingly rare. We present a novel case of a 74-year-old man with BPDCN in remission following autologous stem cell transplantation who developed sequential ocular adnexal recurrences. He initially presented with a protuberant lesion of the right caruncle, followed by new lesions of the left lacrimal gland and right subconjunctival space. Histopathologic analysis confirmed BPDCN recurrence. No

Psoriasis is a chronic autoimmune skin disorder primarily driven by dendritic cells (DCs). However, the full therapeutic potential of antipsoriatic agents like calcipotriol (Cal) hinges on the precise and efficient delivery to DCs. Herein, a nanovehicle fabricated using keratinocyte-derived apoptotic vesicles (apoEVs) is proposed to enhance drug accumulation in DCs and synergistically modulate DC function. Specifically, the phosphatidylserine exposed on the apoEV surface serves as an "eat-me" signal, facilitating specific recognition and engulfment by DCs. This process mimics the natural effer

BACKGROUND: SLE features aberrant type I interferon production, with the stimulator of interferon genes (STING) pathway acting as a key upstream driver. However, its expression patterns in patients with SLE and contributions to clinical phenotypes and tissue injury remain insufficiently defined.
METHODS: The proportion of STING-positive cells in peripheral blood mononuclear cells (PBMCs) from 28 patients with SLE and 15 healthy controls was analysed alongside clinical and serological features. Glomerular STING expression and podocyte injury were assessed in kidney biopsies from 39 patients wi

Blockade of inhibitory PD-1 signaling on T cells is a cornerstone of cancer immunotherapy, with current strategies targeting PD-1 or its ligand PD-L1. However, PD-1 engages an alternative ligand, PD-L2, whose role in tumor immunity remains poorly defined. Here, we show that PD-L2 is upregulated on intratumoral CCR7⁺ conventional dendritic cells (cDCs) in both mouse and human melanoma. Using genetic mouse models enabling selective ablation of PD-L1 or PD-L2 in cDCs, we identify a division of labor between these ligands: PD-L1 controls the size of the progenitor CD8⁺ T cell pool in tumor-drainin

Neuronal dendrites can be injured by a number of insults, but the cellular mechanism by which dendrites respond to tissue injury and undergo repair is poorly understood. Much of the field's progress has evaluated dendrite regeneration following laser injury. While precise, laser injury does not accurately model the real-world damage to surrounding tissue that would accompany neuronal injury. Here, we modify a pinch injury technique to injure both the dendrites and their surrounding tissues in both female and male Drosophila melanogaster larvae to better model what is observed following real-wo…

Cyclic GMP-AMP synthase (cGAS) is critical for dendritic cell (DC) maturation. This study investigates how cGAS suppression in DCs influences graft immune tolerance. Bioinformatics analysis assessed cGAS involvement in transplant immunity. Immature DCs were transduced with an adenoviral vector to knockdown cGAS and divided into three groups: cGAS-shRNA-DCs, EGFP-DCs and PBS (control). These were administered intravenously before transplantation to establish a mouse model. Graft survival and histopathology were evaluated. Splenic T cell subsets were analyzed by flow cytometry. After lipopolysac

The dendritic cell-targeting peptide (DCP) trimer sequence has previously shown to enhance specific IgG induction. This study aims to investigate the expression of rabies virus glycoprotein (RVG) in eukaryotic cells through cloning in the pCDNA3.1 + vector, alongside the fusion of a DCP. The DCP trimer sequence was fused to the C-terminal of the RVG to improve the immune response. The construct was designed to include a His-tag for protein purification and an enterokinase (EK) cleavage site for separation of the tag from the recombinant protein. The RVG gene was amplified from the rabies virus

Dendritic cells (DCs), as the most protent professional antigen-presenting cells (APCs), possess an exceptional capacity to initiate and modulate adaptive immune responses, rendering DC-based vaccines a highly promising approach in tumor immunotherapy. This review explores the biological properties of DCs, establishing a foundation for the rational development of DC vaccine. We further provide a comprehensive overview of current mainstream DC vaccine platforms, with analysis of three representative strategies: (i) DC vaccines loaded with tumor lysates; (ii) personalized neoantigen-pulsed DC va

Follicular dendritic cell sarcoma (FDCS) is a rare malignant neoplasm of follicular dendritic cell origin. It is occasionally associated with unicentric Castleman disease (UCD), particularly the hyaline-vascular (HV) variant (HV-UCD). We report a 56-year-old woman with FDCS arising in the parapharyngeal space in the background of HV-UCD. The patient presented with a painless right neck mass and extensive cervical lymphadenopathy without systemic symptoms. Surgical resection of the parapharyngeal mass revealed FDCS in the lymph nodes with features of HV-UCD. Immunohistochemistry confirmed the f

Triple-negative breast cancer (TNBC) is an aggressive malignancy with high mortality and limited treatment options. While surgical resection removes the primary tumor, it often fails to prevent recurrence or metastasis, and despite the promise of immunotherapy, response to immune checkpoint blockade remains poor. Here, a cryogel-based dendritic cell (DC) immunotherapy is developed incorporating gold nanodot-lipopolysaccharide (AuLPS)-loaded DCs, doxorubicin (Dox), and PD-1 immune checkpoint blockade (aPD-1+Dox+AuLPS@DC) to enhance post-surgical antitumor immunity. The AuLPS nanoparticles (NPs)

Dendritic computation based on the spatiotemporal integration for excitatory and inhibitory synaptic inputs plays a pivotal role in performing sophisticated information processing. It is vital to develop artificial dendritic devices with a flexible spatiotemporal integration capacity for excitatory and inhibitory inputs driven by the inputs with the same polarity. Here, a dual-gate configuration dendristor (abbreviated as DGD) is demonstrated with p-type and n-type memtransistor characteristics based on Na+-doped MoS2. The excitatory and inhibitory synaptic responses are achieved on the DGD un

The endoplasmic reticulum (ER) is the largest intracellular Ca2+ store, serving as the source and sink of intracellular Ca2+ The ER Ca2+ store is continuous yet organized into distinct subcompartments with spatial and functional heterogeneity. In cerebellar Purkinje cells (PCs), glutamatergic inputs trigger Ca2+ release from specific ER domains via inositol 1,4,5-trisphosphate receptors (IP3Rs) or ryanodine receptors (RyRs). Upon ER store depletion, refilling occurs through store-operated Ca2+ entry mediated by stromal interaction molecule-1 (STIM1). Although the significance of STIM1-mediated

BACKGROUND: Graves' disease (GD) and orbitopathy (GO) are common systemic autoimmune diseases targeting the human thyrotropin receptor. We carried out detailed immune cell investigations in GD/GO.
METHODS AND RESULTS: For the first time, we describe inflammatory and tolerogenic dendritic cells (tolDCs) derived from GD/GO patient monocytes. GD/GO tolDCs show reduced inflammatory marker CD86 expression, but higher CD11c levels. CD86 expression on DCs correlates to anti-TSHR antibody titers in the patients. Release of interleukin-10 from GD/GO tolDCs is increased, and their phagocytotic capacity

Regarding the global increase in dental caries, the development of high-quality resin-based materials is essential. However, it is still a challenge to alleviate the polymerization shrinkage of methacrylate monomers. Herein, we constructed the first- and second-generation betulin-based multimethacrylate dendritic monomers (D1, D2, and D3) via the esterification reaction, which were differed primarily in the number and position of the terminal methacrylate groups. These photo-polymerizable dendritic monomers were introduced as the partial (10 wt%) and total (50 wt%) replacements of bisphenol A

Dendrites grow by stochastic branching, elongation, and retraction. A key question is whether such a mechanism is sufficient to form highly branched dendritic morphologies. Alternatively, does dendrite geometry depend on signals from other cells or from the topological hierarchy of the growing network? To answer these questions, we developed an isotropic and homogenous mean-field model in which branch dynamics depends only on average lengths and densities: that is, without external influence. Branching was modeled as density-dependent nucleation so that no tree structures or network topology w…

Sleep is critically involved in strengthening memories. However, our understanding of the morphological changes underlying this process is still emerging. Recent studies suggest that specific subsets of dendritic spines are strengthened during sleep in specific neurons involved in recent learning. Contextual memories associated with traumatic experiences are involved in post-traumatic stress disorder (PTSD) and represent recent learning that may be strengthened during sleep. We tested the hypothesis that dendritic spines encoding contextual fear memories are selectively strengthened during sle

BACKGROUND: Tolerogenic dendritic cells (toDCs) are critical for maintaining immune homeostasis and preventing autoimmune disease development, such as type 1 diabetes (T1D). We have previously shown that DCs of non-obese diabetic (NOD) mice expressing active Stat5b (Stat5b-CA.DCs) acquire toDCs signature and protect against diabetes. However, the mechanisms involved in reprogramming DCs to adopt tolerogenic or immunogenic signatures are not fully known. This study investigates for the first time the role of USP7 in DC-mediated immune regulation in T1D using a transgenic NOD mouse model express

We investigated the growth of gold nano/micro-structures by reducing gold ions with ascorbic acid in a mixed solvent of ethanol and water containing hexadecyltrimethylammonium bromide (CTAB). The geometries of the gold structures were influenced by the ratio of ethanol to water and the concentrations of CTAB and NaBr. We observed gold dendrites exceeding 10 μm in size, which were composed of numerous needles of varying sizes, some as small as a few tens of nanometers. The gold dendrites were grown within 20 min, a shorter duration compared to previous reports. The overall size of the gold dend…

BACKGROUND: Mitochondrial dynamics and mitophagy are key mechanisms maintaining mitochondrial quality and homeostasis in inflammatory diseases, though their activation pathways in inflammatory regulation remain unclear. Sestrin2 (Sesn2), a stress-responsive protein critical for cellular homeostasis, was investigated in this study for its regulatory role in mitochondrial dynamics during sepsis and its potential mechanism in dendritic cell (DC) necroptosis.
METHODS: This study evaluated Sesn2-regulated mitochondrial dynamics proteins such as dynamin-related protein 1 (DRP1), mitochondrial fissi

Bottom-up synthetic biology seeks to construct artificial cells with biomimetic or novel functionalities to uncover the fundamental principles of cellular evolution and drive advances in medicine and bioengineering. Among them, membranized coacervate microdroplets (MCM) uniquely combine a molecularly crowded aqueous interior with a surrounding membrane, both hallmarks of eukaryotic cells. Replicating cellular functions requires synthetic cells to remain structurally stable in biological environments, where ionic strength presents a significant threat to the integrity of complex coacervates.
