The use of cyst antigen in the serodiagnosis of amoebiasis.

The choice of a specific and pure antigen is one of the many problems in the serodiagnosis of amoebic infection. Microbial associates in amoebic cultures have often given rise to confusing results in these tests. Entamoeba histolytica trophozoites grown axenically provide a useful source of antigen but the axenic medium is expensive and cumbersome to prepare. The authors have successfully investigated the use of E. invadens cysts as a relatively inexpensive source of bacteria-free antigen which can be employed in routine diagnostic work. Indirect haemagglutination tests using the easily prepare…

Experimental amoebic infection in guinea-pigs immunized with low grade amoebic infection.

Guinea-pigs were immunzied with a low grade amoebic infection, either intracaecally or intramesenterically. The immunized animals were challenged with a virulent strain of Entamoeba histolytica. Some of the animals, immunized via the intracaecal route were challenged repeatedly. 10 to 30% of immunized animals developed caecal lesions compared with 92.8% of non-immunized animals. The percentage of protection in animals challenged repeatedly was greater than in those challenged once. Neither immunized nor non-immunized guinea-pigs developed hepatic abscesses. It appears that prior low grade infe…

Protozoa: The Overlooked Frontiers of Natural Antimicrobial Compounds.

Antimicrobial resistance (AMR) is becoming a major problem in the treatment of infectious diseases, increasing the need for new antimicrobial agents. Although bacteria and fungi have been extensively explored in antimicrobial discovery, protozoa remain comparatively underexplored despite their biological diversity and metabolic capabilities. Recent studies have reported that several protozoan species, including Acanthamoeba, Dictyostelium, Naegleria, and Entamoeba, are capable of exhibiting antimicrobial activity, particularly against multidrug-resistant (MDR) bacteria and fungi. These activit…

A review into the recent advances in the world of amoebiasis.

PURPOSE OF REVIEW: Amoebiasis is a parasitic infection caused by Entamoeba histolytica , affecting 10% of the global population. It is a well recognized cause of morbidity and mortality in low-middle-income countries where it is endemic. However, with increased migration and global travel, amoebiasis is now more common in high-income countries, although diagnosis is often delayed or even missed due to lack of awareness of the latest epidemiology and optimal diagnostic testing. This review discusses the evolving prevalence, and the current international guidelines for the investigation and trea…

Amoebiasis vaccine development: A snapshot on E. histolytica with emphasis on perspectives of Gal/GalNAc lectin.

Amoebiasis/amebiasis is a gastrointestinal infection caused by an enteric dwelling protozoan, Entamoeba histolytica. The disease is endemic in the developing world and is transmitted mainly via the faecal-oral route (e.g., in water or food) and may or may not be symptomatic. This disease of socio-economic importance worldwide involves parasite adherence and cytolysis of human cells followed by invasion that is mediated by galactose-binding (Gal/GalNAc) surface lectin. Disruption of the mucus layer leads to invasive intestinal and extraintestinal infection. Gal-lectin based vaccinations have co…

Host-microbe interactions and defense mechanisms in the development of amoebic liver abscesses.

Amoebiasis by Entamoeba histolytica is a major public health problem in developing countries and leads to several thousand deaths per year. The parasite invades the intestine (provoking diarrhea and dysentery) and the liver, where it forms abscesses (amoebic liver abscesses [ALAs]). The liver is the organ responsible for filtering blood coming from the intestinal tract, a task that implies a particular structure and immune features. Amoebae use the portal route and break through the sinusoidal endothelial barrier to reach the hepatic parenchyma. When faced with systemic and cell-mediated defen…

Purification of the antigen specific for amoebiasis.

Two methods used for the purification of specific antigen for amoebiasis were affinity chromatography and agarose gel electrophoresis. The IgG fraction of the serum from a patient with amoebic liver abscess showing a specific band in the IEP test was coupled to CNBr-activated Sepharose 4B. After passage through the immunoadsorbent column of the crude or partially purified extract of Entamoeba histolytica, followed by acid dissociation, specific antigen was obtained. This antigen gave only one band in the IEP test against the serum which gave multiple bands upon testing with the crude antigen. …
