Effect of intrinsic and extrinsic factors on global and regional cortical thickness.

Global and regional cortical thicknesses based on T1-weighted magnetic resonance images acquired at 1.5 T and 3 T were measured on a relatively large cohort of 295 subjects using FreeSurfer software. Multivariate regression analysis was performed using Pillai's trace test to determine significant differences in cortical thicknesses measured at these two field strengths. Our results indicate that global cortical thickness is not affected by the field strength or gender. In contrast, the regional cortical thickness was observed to be field dependent. Specifically, the cortical thickness in regio

Neural Activation During Cognitive Effort-Based Decision Making: Associations With Avolition in Veterans With Schizophrenia and Bipolar Disorder.

BACKGROUND: Effort-based decision making (EBDM) is a key component of motivation. Impairments in EBDM have been consistently linked to amotivation in individuals with schizophrenia (SZ). Similar deficits are seen in SZ and bipolar disorder (BD), despite striking differences in motivational profiles between the two disorders. Similar task behavior, but distinct motivational profiles, may arise from functional differences in brain regions supporting EBDM. METHODS: Twenty-eight veterans with SZ, 21 veterans with BD, and 30 healthy control participants completed a cognitive EBDM task during funct

Investigation of the impact of rs3025343 near the dopamine ß-hydroxylase (DBH) gene on reward-related responses within the mesolimbic dopamine system.

OBJECTIVES: Recent research focused on the impact of Neandertal alleles on dopamine-related brain traits. A joint measure of these alleles was reported to be associated with lower dopamine synthesis, presumably leading to reduced activity of the reward system. To isolate single variants, we selected rs3025343 near the dopamine beta hydroxylase gene, which contains a likely introgressed allele. rs3025343 is an established variant for reward system-related psychiatric traits. METHODS: We hypothesised that the rs3025343 minor allele reduces responses to conditioned reward stimuli in the mesolimb

Does a monoclonal antibody targeting immune cells affect glutamatergic level in schizophrenia? A multimodal PET/MRS brain imaging study.

Microglial dysfunction and glutamatergic dysregulation are implicated in the neurobiology of schizophrenia. Microglia regulate brain glutamate levels through mechanisms including the cysteine-glutamate antiporter, raising the possibility that microglial dysfunction could underlie glutamatergic dysregulation in schizophrenia. We tested this using a combined cross-sectional/longitudinal study of individuals with first episode psychosis and healthy controls. We investigated two hypotheses: (1) increase in a positron emission tomography (PET) imaging marker of microglia is associated with increase

Unsupervised Clustering of ACC and Hippocampal Metabolites Identifies Subgroups in Antipsychotic-Naïve First-Episode Psychosis.

BACKGROUND AND HYPOTHESIS: Glutamatergic abnormalities are among the most consistent neurometabolic findings in psychosis spectrum disorders, but individual variability suggests biologically distinct subgroups. Proton magnetic resonance spectroscopy (1H-MRS) enables in vivo quantification of glutamate (Glu) and related metabolites but has rarely been applied to subgroup discovery in early psychosis. We hypothesized that metabolite-based clustering of hippocampal and anterior cingulate cortex (ACC) measures would identify biologically distinct subtypes differing in symptom severity and cognitio

Cerebral schistosomiasis masquerading as a CNS malignancy in a sudanese traveler: A diagnostic challenge and review of literature.

INTRODUCTION: Cerebral schistosomiasis is a rare condition, typically associated with Schistosoma japonicum. Cerebral involvement by S. mansoni is exceptionally uncommon and diagnostically challenging, often mimicking CNS tumors. CASE PRESENTATION: A 31-year-old Sudanese male presented with a simple partial seizure. MRI revealed a left cingulate gyrus mass suggestive of malignancy. Stereotactic biopsy showed granulomas with S. mansoni eggs, confirmed by serology despite negative stool studies. The patient received praziquantel 60 mg/kg/day and prednisolone 50 mg/day for two weeks, resulting i

Brain and muscle chemistry in myalgic encephalitis/chronic fatigue syndrome (ME/CFS) and long COVID: a 7T magnetic resonance spectroscopy study.

Myalgic encephalitis/chronic fatigue syndrome (ME/CFS) is a common debilitating medical condition, whose main symptoms - fatigue, post-exertional malaise and cognitive dysfunction - are also present in many cases of long COVID. Magnetic resonance spectroscopy (MRS) allows the insight into their pathophysiology through exploration of a range of biochemicals putatively relevant to aetiological processes, in particular mitochondrial dysfunction and energy metabolism. 24 patients with ME/CFS, 25 patients with long COVID and 24 healthy controls (HC) underwent brain (pregenual and dorsal anterior ci

Deep brain stimulation of the anterior cingulate cortex reduces opioid addiction in preclinical studies.

Substance Use Disorder (SUD) is a medical condition where an individual compulsively misuses drugs or alcohol despite knowing the negative consequences. The anterior cingulate cortex (ACC) has been implicated in various types of SUDs, including nicotine, heroin, and alcohol use disorders. Our research aimed to investigate the effects of deep brain stimulation (DBS) in the ACC as a potential therapeutic approach for morphine use disorder. Additionally, we measured c-Fos protein expression as an indicator of neural activity in the nucleus accumbens (NAc) and prefrontal cortex (PFC). Our findings

Role of the rostral anterior cingulate cortex in emotion processing in Treatment Resistant Depression.

The rostral anterior cingulate cortex (rACC) has been identified as a key region in treatment-resistant depression (TRD), potentially influencing the adaptive interplay between the default mode network and other critical neural networks. This study aims to further uncover the role of the rACC in TRD by investigating its differential connectivity during tasks that evoke conscious and non-conscious emotional responses. Thirty-one TRD patients, 35 treatment-sensitive depression (TSD) patients, and 37 healthy controls (HC) underwent 3T fMRI while performing tasks designed to elicit emotional respo

Neural responses to stress and alcohol cues in individuals with pain with and without alcohol use disorder.

Pain and alcohol use disorder (AUD) frequently co-occur, but the underlying neurobiology is not well-understood. Although many studies have reported disruptions in stress and reward cue-elicited neural reactivity and heightened alcohol craving in individuals with AUD, little is known about these constructs among patients who experience pain. Here, individuals with pain (Pain+, n = 31) and without pain (Pain-, n = 37) completed a well-validated functional magnetic resonance imaging (fMRI) paradigm involving stress (S), alcohol (A) and neutral (N) cue exposure with repeated alcohol craving asses

Evaluation of the Glymphatic System in Schizophrenia Spectrum Disorder Using Proton Magnetic Resonance Spectroscopy Measurement of Brain Macromolecule and Diffusion Tensor Image Analysis Along the Perivascular Space Index.

BACKGROUND AND HYPOTHESIS: The glymphatic system (GS), a brain waste clearance pathway, is disrupted in various neurodegenerative and vascular diseases. As schizophrenia shares clinical characteristics with these conditions, we hypothesized GS disruptions in patients with schizophrenia spectrum disorder (SCZ-SD), reflected in increased brain macromolecule (MM) and decreased diffusion-tensor-image-analysis along the perivascular space (DTI-ALPS) index. STUDY DESIGN: Forty-seven healthy controls (HCs) and 103 patients with SCZ-SD were studied. Data included 135 proton magnetic resonance spectro

Intrinsic Functional Connectivity between the Anterior Insular and Retrosplenial Cortex as a Moderator and Consequence of Cocaine Self-Administration in Rats.

While functional brain imaging studies in humans suggest that chronic cocaine use alters functional connectivity (FC) within and between key large-scale brain networks, including the default mode network (DMN), the salience network (SN), and the central executive network (CEN), cross-sectional studies in humans are challenging to obtain brain FC prior to cocaine use. Such information is critical to reveal the relationship between individual's brain FC and the subsequent development of cocaine dependence and brain changes during abstinence. Here, we performed a longitudinal study examining func

Anterior cingulate cortex-related functional hyperconnectivity underlies sensory hypersensitivity in Grin2b-mutant mice.

Sensory abnormalities are observed in ~90% of individuals with autism spectrum disorders (ASD), but the underlying mechanisms are poorly understood. GluN2B, an NMDA receptor subunit that regulates long-term depression and circuit refinement during brain development, has been strongly implicated in ASD, but whether GRIN2B mutations lead to sensory abnormalities remains unclear. Here, we report that Grin2b-mutant mice show behavioral sensory hypersensitivity and brain hyperconnectivity associated with the anterior cingulate cortex (ACC). Grin2b-mutant mice with a patient-derived C456Y mutation (

Impact of NPSR1 gene variation on the neural correlates of phasic and sustained fear in spider phobia-an imaging genetics and independent replication approach.

The functional neuropeptide S receptor 1 (NPSR1) gene A/T variant (rs324981) is associated with fear processing. We investigated the impact of NPSR1 genotype on fear processing and on symptom reduction following treatment in individuals with spider phobia. A replication approach was applied [discovery sample: Münster (MS) nMS = 104; replication sample Würzburg (WZ) nWZ = 81]. Participants were genotyped for NPSR1 rs324981 [T-allele carriers (risk) versus AA homozygotes (no-risk)]. A sustained and phasic fear paradigm was applied during functional magnetic resonance imaging. A one-session virtu

Amyloid-PET Levels in the Precuneus and Posterior Cingulate Cortices Are Associated with Executive Function Scores in Preclinical Alzheimer's Disease Prior to Overt Global Amyloid Positivity.

BACKGROUND: Global amyloid-β (Aβ) deposition in the brain can be quantified by Aβ-PET scans to support or refute a diagnosis of preclinical Alzheimer's disease (pAD). Yet, Aβ-PET scans enable quantitative evaluation of regional Aβ elevations in pAD, potentially allowing even earlier detection of pAD, long before global positivity is achieved. It remains unclear as to whether such regional changes are clinically meaningful. OBJECTIVE: Test the hypothesis that early focal regional amyloid deposition in the brain is associated with cognitive performance in specific cognitive domain scores in pAD

Disrupted myelination network in the cingulate cortex of Parkinson's disease.

The cingulate cortex is part of the conserved limbic system, which is considered as a hub of emotional and cognitive control. Accumulating evidence suggested that involvement of the cingulate cortex is significant for cognitive impairment of Parkinson's disease (PD). However, mechanistic studies of the cingulate cortex in PD pathogenesis are limited. Here, transcriptomic and regulatory network analyses were conducted for the cingulate cortex in PD. Enrichment and clustering analyses showed that genes involved in regulation of membrane potential and glutamate receptor signalling pathway were up

Changes of functional connectivity of the subgenual anterior cingulate cortex and precuneus after cognitive behavioral therapy combined with fluoxetine in young depressed patients with suicide attempt.

This single-center, randomized, single-blind, parallel-controlled study aimed to analyze the changes in resting-state functional connectivity (RSFC) in young patients with a suicide attempt caused by depression before and after cognitive-behavioral therapy (CBT) combined with fluoxetine or fluoxetine alone by functional magnetic resonance imaging (fMRI). Before treatment, functional connectivity of the right subgenual anterior cingulate cortex (R-sgACC), left subgenual anterior cingulate cortex (L-sgACC) and right precuneus (R-PCu) was lower in depressed patients with a suicide attempt than th

Insula reactivity mediates subjective isolation stress in alexithymia.

The risk for developing stress-related disorders is elevated in individuals with high alexithymia, a personality trait characterized by impaired emotional awareness and interpersonal relating. However, it is still unclear how alexithymia alters perceived psychosocial stress and which neurobiological substrates are mechanistically involved. To address this question, we examined freshmen during transition to university, given that this period entails psychosocial stress and frequently initiates psychopathology. Specifically, we used a functional magnetic resonance imaging emotional face matching

Changes in Brain Glutamate on Switching to Clozapine in Treatment-Resistant Schizophrenia.

It has been suggested that the antipsychotic clozapine may modulate brain glutamate, and that this effect could contribute to its efficacy in treatment-resistant schizophrenia (TRS). The aim of this study was to examine the effects of clozapine on brain glutamate in TRS longitudinally. This study examined individuals with TRS before and 12 weeks after switching from a non-clozapine antipsychotic to treatment with clozapine as part of their normal clinical care. Proton magnetic resonance spectroscopy (1H-MRS) measured concentrations, corrected for voxel tissue content, of glutamate (Glucorr), a

Acute Hyperglycemia Increases Brain Pregenual Anterior Cingulate Cortex Glutamate Concentrations in Type 1 Diabetes.

The brain mechanisms underlying the association of hyperglycemia with depressive symptoms are unknown. We hypothesized that disrupted glutamate metabolism in pregenual anterior cingulate cortex (ACC) in type 1 diabetes (T1D) without depression affects emotional processing. Using proton MRS, we measured glutamate concentrations in ACC and occipital lobe cortex (OCC) in 13 subjects with T1D without major depression (HbA1c 7.1 ± 0.7% [54 ± 7 mmol/mol]) and 11 healthy control subjects without diabetes (HbA1c 5.5 ± 0.2% [37 ± 3 mmol/mol]) during fasting euglycemia followed by a 60-min +5.5 mmol/L h
