Aberrant fumarate metabolism links interferon release in diffuse systemic sclerosis.

BACKGROUND: Systemic Sclerosis (SSc) is an idiopathic rheumatic inflammatory disease that is characterised by inflammation and skin fibrosis. Type I interferon is significantly elevated in the disease. OBJECTIVE: The objective of this study is to determine the role of the TCA cycle metabolite fumarate in SSc. METHODS: CD14 + cells were isolated from 12 SSc patients and healthy controls. Fumarate hydratase and Interferon dependant genes were quantified by qPCR. In vitro inhibition of STING using a small molecule STING inhibitor and enforced mitophagy was induced in vitro and IFN-β release was

Targeting the cGAS-STING pathway for therapeutic gain in autoimmune inflammatory cutaneous diseases with a focus on systemic lupus erythematosus and systemic sclerosis.

The cGAS-STING [cyclic GMP-AMP (cGAMP) synthase (cGAS)-stimulator of interferon genes (STING)] pathway is an evolutionarily conserved pathway for sensing and responding to cytosolic double-stranded (ds)DNA, whether pathogen-associated or endogenous. Detection and binding of dsDNA by cGAS enzymatically generates the second messenger cGAMP, which in turn binds the endoplasmic reticulum-resident signalling protein STING, activating IRF3 (interferon regulatory factor 3), leading to expression of type I interferons. This pathway is necessary for both viral infection and antitumoral responses. Vario

MiR-34a deficiency enhances nucleic acid sensing and type I IFN signaling in a mouse model of Alzheimer's disease.

BACKGROUND: A number of microRNAs are implicated in aging, cell senescence, inflammation, and neurodegenerative diseases. Particularly, miR-34a levels in the brain are increased in Alzheimer's disease (AD) but its mechanistic role in AD pathogenesis is unknown. METHODS: In order to investigate the role of miR-34a in AD, we produced an AD mouse model, Tg-SwDI mice, with whole body/constitutive miR-34a knockout (KO). Their cognitive function was evaluated by Morris water maze. Immunohistochemistry and immunofluorescence were used for neuropathological evaluation. Bulk RNA-seq followed by bioinf

ISG15 mRNA transcript level in circulating leucocytes prognostic of overall survival in hepatocellular carcinoma patients and correlated with quality of life disturbances involved in anorexia-cachexia.

BACKGROUND: An increasing number of immuno-therapeutic agents have proven efficacy in hepatocellular carcinoma (HCC). Inflammatory markers (c-reactive protein, interleukin-8 and inflammatory score) have been found to be prognostic factors in HCC patients. These inflammatory markers have demonstrated correlations with quality of life (QOL) disturbances in fatigue, appetite loss and nutritional concern. Type I interferon response triggered by HCC could be responsible for an inflammatory state and anorexia-cachexia syndrome leading to these specific QOL impairment. Peripheral blood Interferon Sti

The SKIV2L RNA exosome limits activation of the RIG-I-like receptors.

Sensors of the innate immune system that detect intracellular nucleic acids must be regulated to prevent inappropriate activation by endogenous DNA and RNA. The exonuclease Trex1 regulates the DNA-sensing pathway by metabolizing potential DNA ligands that trigger it. However, an analogous mechanism for regulating the RIG-I-like receptors (RLRs) that detect RNA remains unknown. We found here that the SKIV2L RNA exosome potently limited the activation of RLRs. The unfolded protein response (UPR), which generated endogenous RLR ligands through the cleavage of cellular RNA by the endonuclease IRE-

A New Phenotype in Candida-Epithelial Cell Interaction Distinguishes Colonization- versus Vulvovaginal Candidiasis-Associated Strains.

Vulvovaginal candidiasis (VVC) affects nearly 3/4 of women during their lifetime, and its symptoms seriously reduce quality of life. Although Candida albicans is a common commensal, it is unknown if VVC results from a switch from a commensal to pathogenic state, if only some strains can cause VVC, and/or if there is displacement of commensal strains with more pathogenic strains. We studied a set of VVC and colonizing C. albicans strains to identify consistent in vitro phenotypes associated with one group or the other. We find that the strains do not differ in overall genetic profile or behavio

Mycobacterium tuberculosis Rv0158 negatively regulates the cGAS-STING pathway mediated type I IFN production and enhances intracellular survival.

BACKGROUND: Type I interferons (IFN) play an important role in the host defense against Mycobacterium tuberculosis (M. tb) infection and disease pathogenesis. Although M. tb has evolved several mechanisms to evade host immune surveillance, the mechanism used to regulate type I IFN expression remains unclear. METHODS: In this study, genome-wide high-throughput loss-of-function screening was performed to screen M. tb determinants that regulate the Type I IFN pathway, and the role for M. tb Rv0158 in inhibiting type I IFN responses was identified in vitro and in vivo. RESULTS: The M. tb coding

An Antiviral Role for TRIM14 in Ebola Virus Infection.

Ebola virus (EBOV) is a highly pathogenic virus that encodes 7 multifunctional structural proteins. Multiple host factors have been reported to interact with the EBOV proteins. Here, we found that tripartite motif-containing 14 (TRIM14), an interferon-stimulated gene that mediates cellular signaling pathways associated with type I interferon and inflammatory cytokine production, interacts with EBOV nucleoprotein to enhance interferon-β (IFN-β) and nuclear factor-κB (NF-κB) promotor activation. Moreover, TRIM14 overexpression reduced viral replication in an infectious but biologically contained

Type I interferonopathies in pediatric rheumatology.

Defective regulation of type I interferon response is associated with severe inflammatory phenotypes and autoimmunity. Type I interferonopathies are a clinically heterogenic group of Mendelian diseases with a constitutive activation of this pathway that might present as atypical, severe, early onset rheumatic diseases. Skin vasculopathy with chilblains and livedo reticularis, interstitial lung disease, and panniculitis are common. Recent studies have implicated abnormal responses to nucleic acid stimuli or defective regulation of downstream effector molecules in disease pathogenesis. As observ

Inhibition of phosphorylated-STAT1 nuclear translocation and antiviral protein expression in human brain vascular adventitial fibroblasts infected with varicella-zoster virus.

In varicella-zoster virus (VZV)-infected primary human brain vascular adventitial fibroblasts (BRAFs), levels of beta interferon (IFN-β,) STAT1, and STAT2 transcripts as well as STAT1 and STAT2 protein were decreased. IFN-α transcript levels were increased but not secreted IFN-α protein levels. Compared to IFN-α-treated control results, in VZV-infected BRAFs, phosphorylated STAT1 did not translocate to the nucleus, resulting in impaired downstream expression of interferon-inducible antiviral Mx1. Overall, VZV interference with the type I interferon pathway may promote virus persistence in cere

[Alpha interferon in condylomata acuminata and juvenile diabetes mellitus].

Persistent condylomata acuminata in a 21-year-old patient with diabetes mellitus were treated with highly purified interferon-alpha (IFN-alpha) obtained by recombinant DNA technology. Daily dose was 1.5 X 10(6) IU, given subcutaneously. Already during treatment the condylomata regressed. Two weeks after the end of therapy, i.e. after a total dose of 10.5 X 10(6) IU IFN-alpha, all condylomata had completely receded. Blood glucose levels remained constant with concomitant insulin therapy. Toxic side-effects or antibodies to IFN-alpha were not observed.

Induction of IFN-alphabeta enables Listeria monocytogenes to suppress macrophage activation by IFN-gamma.

Production of type I interferon (IFN; IFN-alphabeta) increases host susceptibility to Listeria monocytogenes, whereas type II IFN (IFN-gamma) activates macrophages to resist infection. We show that these opposing immunological effects of IFN-alphabeta and IFN-gamma occur because of cross talk between the respective signaling pathways. We found that cultured macrophages infected with L. monocytogenes were refractory to IFN-gamma treatment as a result of down-regulation of the IFN-gamma receptor (IFNGR). The soluble factor responsible for these effects was identified as host IFN-alphabeta. Accor

Single cell transcriptomics shows that malaria promotes unique regulatory responses across multiple immune cell subsets.

Plasmodium falciparum malaria drives immunoregulatory responses across multiple cell subsets, which protects from immunopathogenesis, but also hampers the development of effective anti-parasitic immunity. Understanding malaria induced tolerogenic responses in specific cell subsets may inform development of strategies to boost protective immunity during drug treatment and vaccination. Here, we analyse the immune landscape with single cell RNA sequencing during P. falciparum malaria. We identify cell type specific responses in sub-clustered major immune cell types. Malaria is associated with an

Interferon alpha promotes caspase-8 dependent ultraviolet light-mediated keratinocyte apoptosis via interferon regulatory factor 1.

INTRODUCTION: Ultraviolet (UV) light is a known trigger of both cutaneous and systemic disease manifestations in lupus patients. Lupus skin has elevated expression of type I interferons (IFNs) that promote increased keratinocyte (KC) death after UV exposure. The mechanisms by which KC cell death is increased by type I IFNs are unknown. METHODS: Here, we examine the specific cell death pathways that are activated in KCs by type I IFN priming and UVB exposure using a variety of pharmacological and genetic approaches. Mice that overexpress Ifnk in the epidermis were exposed to UVB light and cell

Intracellular ISG-virus interactions determine viral infection severity and persistence.

In innate immune response, type I interferons (IFNs) activate interferon-stimulated genes (ISGs), which suppress viral replication and secretion at the intracellular level. Yet, how these ISG-virus interactions shape infection progression and severity remains poorly understood. Here, we introduce a new viral infection model that explicitly incorporates intracellular ISG-virus dynamics. It structures, for the first time, infected cells based on viral load and ISG expression which offers a computationally efficient and adaptable approach to integrating ISG-virus intracellular dynamics into viral

Interferon-sensitized hematopoietic progenitors dynamically alter organismal immunity.

Inflammation has enduring impacts on organismal immunity. However, the precise mechanisms by which tissue-restricted inflammation conditions systemic responses are poorly understood. Here, we leveraged a highly compartmentalized model of skin inflammation and identified a surprising type I interferon (IFN)-mediated activation of hematopoietic stem/progenitor cells (HSPCs) that results in profound changes to systemic host responses. Post-inflamed mice were protected from atherosclerosis and had worse outcomes following influenza virus infection. This IFN-mediated HSPC modulation was dependent o

Interferons in Pain and Infections: Emerging Roles in Neuro-Immune and Neuro-Glial Interactions.

Interferons (IFNs) are cytokines that possess antiviral, antiproliferative, and immunomodulatory actions. IFN-α and IFN-β are two major family members of type-I IFNs and are used to treat diseases, including hepatitis and multiple sclerosis. Emerging evidence suggests that type-I IFN receptors (IFNARs) are also expressed by microglia, astrocytes, and neurons in the central and peripheral nervous systems. Apart from canonical transcriptional regulations, IFN-α and IFN-β can rapidly suppress neuronal activity and synaptic transmission via non-genomic regulation, leading to potent analgesia. IFN-

Lambda interferon inhibits human immunodeficiency virus type 1 infection of macrophages.

The newly identified type III interferon (IFN-lambda) has antiviral activity against a broad spectrum of viruses. We thus examined whether IFN-lambda has the ability to inhibit human immunodeficiency virus type 1 (HIV-1) infection of blood monocyte-derived macrophages that expressed IFN-lambda receptors. Both IFN-lambda1 and IFN-lambda2, when added to macrophage cultures, inhibited HIV-1 infection and replication. This IFN-lambda-mediated anti-HIV-1 activity is broad, as IFN-lambda could inhibit infection by both laboratory-adapted and clinical strains of HIV-1. Investigations of the mechanism

Modeling hamsters for evaluating West Nile virus therapies.

A hamster model infected with a New York crow brain isolate of West Nile virus (WNV) was characterized for evaluating potential antiviral therapies. Older hamsters (7-11 weeks old) had a lower mortality of approximately 50% and more apparent disease signs as compared to >90% mortality in younger hamsters and mice. Disease signs such as limb strength, lacrimation, front limb tremors, somnolence, and deficiencies in neurological responses were noted at different times after viral injection. Weight loss was a marker for WNV disease signs, whereas, the ability to climb up an inclined ramp was asso
