The immune adjuvant response of polysaccharides from Atractylodis macrocephalae Koidz in chickens vaccinated against Newcastle disease (ND).

Build on our previous research, polysaccharides from the rhizome of Atractylodis macrocephalae Koidz (RAMPS), RAMPStp and RAMPS60c were prepared and the structural characterization and immune response of ND vaccine in chicken were investigated. Immune organ index, Lymphocyte proliferation, antibody titers, cell cycle distribution, and percentages of CD4(+) and CD8(+) cells were determined. GPC analysis showed that the Mn of RAMPS with two peaks were 1.29×10(5) and 1.74×10(3), respectively. GC-MS analysis revealed that RAMPS was composed of glucose, mannose, arabinose, galactose, xylose, d-Ribo

The role of SLC2A1 mutations in myoclonic astatic epilepsy and absence epilepsy, and the estimated frequency of GLUT1 deficiency syndrome.

The first mutations identified in SLC2A1, encoding the glucose transporter type 1 (GLUT1) protein of the blood-brain barrier, were associated with severe epileptic encephalopathy. Recently, dominant SLC2A1 mutations were found in rare autosomal dominant families with various forms of epilepsy including early onset absence epilepsy (EOAE), myoclonic astatic epilepsy (MAE), and genetic generalized epilepsy (GGE). Our study aimed to investigate the possible role of SLC2A1 in various forms of epilepsy including MAE and absence epilepsy with early onset. We also aimed to estimate the frequency of G

Reduced Myelination and Increased Glia Reactivity Resulting from Severe Neonatal Hyperbilirubinemia.

Bilirubin-induced neurologic dysfunction (BIND) and kernicterus has been used to describe moderate to severe neurologic dysfunction observed in children exposed to excessive levels of total serum bilirubin (TSB) during the neonatal period. Here we use a new mouse model that targets deletion of the Ugt1 locus and the Ugt1a1 gene in liver to promote hyperbilirubinemia-induced seizures and central nervous system toxicity. The accumulation of TSB in these mice leads to diffuse yellow coloration of brain tissue and a marked cerebellar hypoplasia that we characterize as kernicterus. Histologic studi

The SLC37 family of phosphate-linked sugar phosphate antiporters.

The SLC37 family consists of four sugar-phosphate exchangers, A1, A2, A3, and A4, which are anchored in the endoplasmic reticulum (ER) membrane. The best characterized family member is SLC37A4, better known as the glucose-6-phosphate (G6P) transporter (G6PT). SLC37A1, SLC37A2, and G6PT function as phosphate (Pi)-linked G6P antiporters catalyzing G6P:Pi and Pi:Pi exchanges. The activity of SLC37A3 is unknown. G6PT translocates G6P from the cytoplasm into the lumen of the ER where it couples with either glucose-6-phosphatase-α (G6Pase-α) or G6Pase-β to hydrolyze intraluminal G6P to glucose and P

GLUT1 endothelial reactivity distinguishes hepatic infantile hemangioma from congenital hepatic vascular malformation with associated capillary proliferation.

Hepatic vascular lesions in pediatric patients have overlapping definitions and a plethora of confusing terminology. The so-called hepatic infantile hemangioendothelioma (IHE) frequently coexists and shares some biological features with cutaneous juvenile hemangioma (CJH). To clarify the nature of hepatic vascular lesions in pediatric patients and to investigate the association between IHE and CJH, we reviewed the clinical features, imaging findings and histopathology of 19 cases of hepatic vascular lesions diagnosed at our institution over the last 33 years. Large-scale purification, dissociation and functional reassembly of the maltose ATP-binding cassette transporter (MalFGK(2)) of Salmonella typhimurium.

The maltose ATP-binding cassette (ABC) transporter of Salmonella typhimurium is composed of a membrane-associated complex (MalFGK(2)) and a periplasmic substrate binding protein. To further elucidate protein-protein interactions between the subunits, we have studied the dissociation and reassembly of the MalFGK(2) complex at the level of purified components in proteoliposomes. First, we optimized the yield in purified complex protein by taking advantage of a newly constructed expression plasmid that carries the malK, malF and malG genes in tandem orientation. Incorporated in proteoliposomes, t

Genetic determinants of jaundice and gallstones in haemoglobin E beta thalassaemia.

Chronic hyperbilirubinaemia, gallstone formation, and gall bladder disease are unusually common in people with haemoglobin E beta thalassaemia in Sri Lanka. To determine whether this has a genetic basis we compared the bilirubin levels and frequency of gallstones in patients with different alleles of the UGT*1 gene. There was a significantly higher bilirubin level in those with the 7/7 genotypes compared with 6/6 and 6/7 genotype (p=0.032 and 0.0015 respectively), who also appeared more prone to gallstone formation. These results suggest that the UGT*1 genotpe is of importance in the genesis o

Protection by tetracyclines against ion transport disruption caused by nystatin in human airway epithelial cells.

Polyene antifungal antibiotics like nystatin form monovalent cation pores on the plasma membrane that perturb the intracellular electrolyte milieu, resulting in cell damage. In the present study, we investigated the effects of tetracyclines (minocycline and tetracycline) on ion transporters disrupted by nystatin in cultured human airway Calu-3 cells. Apical application of nystatin (50 microM) on a monolayer of the cells stimulated Na(+)-K(+) pump activity as estimated by ouabain (1 mM)-sensitive short-circuit current (I(sc)). The nystatin-potentiated I(sc) was inhibited by minocycline (IC(50)

Nocardia veterana sp. nov., isolated from human bronchial lavage.

A nocardioform bacterium was isolated from the bronchoscopic lavage of a 78-year-old man with a past history of tuberculous pleurisy, who presented with bilateral upper lobe lesions at Austin and Repatriation Medical Centre, Heidelberg, Australia. The strain was aerobic, Gram-positive, produced beige substrate mycelium and scant white aerial mycelium. It showed chemotaxonomic markers which were consistent with the classification of Nocardia: i.e. meso-diaminopimelic acid, N-glycolylmuramic acid, arabinose and galactose as diagnostic sugars; phospholipids phosphatidylinositol mannosides, phosph

Type-1c glycogen storage disease is not caused by mutations in the glucose-6-phosphate transporter gene.

Glycogen storage disease type 1 (GSD-1) is a group of autosomal recessive disorders caused by deficiencies in glucose-6-phosphatase (G6Pase) and the associated substrate/product transporters. Molecular genetic studies have demonstrated that GSD-1a and GSD-1b are caused by mutations in the G6Pase enzyme and a glucose-6-phosphate transporter (G6PT), respectively. While kinetic studies of G6Pase catalysis predict that the index GSD-1c patient is deficient in a pyrophosphate/phosphate transporter, the existence of a separate locus for GSD-1c remains unclear. We have previously shown that the G6Pas

Neurological changes induced by stress in streptozotocin diabetic rats.

Previous studies from our laboratory demonstrated that chronic stress produces molecular, morphological, and ultrastructural changes in the rat hippocampus that are accompanied by cognitive deficits. Glucocorticoid impairment of glucose utilization is proposed as a causative factor involved in stress-induced changes. Current studies have examined the neurological changes induced by stress in rats with a preexisting strain upon their homeostatic load--namely, in streptozotocin (stz)-diabetic rats. Administration of stz (70 mg/kg, i.v.) produced diabetic symptoms such as weight loss, polyuria, p

Reduced glucose transporter at the blood-brain barrier and in cerebral cortex in Alzheimer disease.

We studied the hexose transporter protein of the frontal and temporal neocortex, hippocampus, putamen, cerebellum, and cerebral microvessels (which constitute the blood-brain barrier) in Alzheimer disease and control subjects by reversible and covalent binding with [3H]cytochalasin B and by immunological reactivity. In Alzheimer disease subjects, we found a marked decrease in the hexose transporter in brain microvessels and in the cerebral neocortex and hippocampus, regions that are most affected in Alzheimer disease, but there were no abnormalities in the putamen or cerebellum. Hexose transpo

Euterpe oleracea Mart. (açaí) seeds show in vitro prebiotic potential: Integrative evidence from human colonic fermentation.

Açaí seeds (Euterpe oleracea Mart.) are an abundant agro-industrial by-product rich in bioactive compounds, remain unexplored for their gut microbiota modulating potential. This study investigated the modulation of gut microbiota composition and metabolism by discarded açaí seeds using an in vitro colonic fermentation model. The seeds were characterized by a high insoluble fiber (70.40 g/100 g), and mannose as the major monosaccharide. Colonic fermentation with discarded açaí seeds (AS48) preserved microbial diversity relative to the fermentation control (CT48) and was associated with higher a

BACKGROUND: The rising incidence of immune-related diseases has increased healthcare expenditures. Currently, conventional drugs are limited by high costs, increased infection risk, off-target effects, and other adverse effects. Natural polysaccharides offer an alternative due to their diverse sources, safety, and remarkable immunomodulatory activities. A systematic review of their sources, extraction methods, mechanisms, and structure-activity relationships is lacking. SCOPE AND METHODS: This paper reviews the progress in research on natural polysaccharides with immunomodulatory properties,

Extracellular polymeric substances in aerobic granular sludge under increasing salinity conditions.

The long-term effects of environmental conditions, such as seawater salinity, on the extracellular investigated EPS changes during a stepwise increase in salinity (0-4%), renewing over 90% of biomass at each condition. Stable granulation, complete anaerobic acetate uptake, and phosphate removal were maintained throughout. FT-IR of granules showed significant changes in glycans (1025 cm⁻¹) and sialic acid (1730 cm⁻¹), which were reflected in the EPS. Lectin microarray revealed that increasing salinity reduced glycan diversity in EPS glycoproteins, while increasing negatively charged groups, inc

Injectable Arnebia Euchroma Polysaccharide-Based Hydrogel as CpG Oligonucleotide Delivery System with Dual Immunomodulatory Activities.

This study developed novel immunostimulatory hydrogels composed of aldehyde-modified Arnebia euchroma polysaccharide (oxidized ARP, O-ARP) and gelatin (GE) for oligonucleotide delivery. Structural characterization of three O-ARP derivatives with varying degrees of oxidation confirmed reduced molecular weight, a preserved yet altered molar ratio of monosaccharide composition, and triple-helical conformation. Hydrogels (GE-O1, GE-O2, GE-O3) were formed via a Schiff base reaction between aldehyde and amino groups. Molecular dynamics simulations showed that O-ARP and gelatin can form a stable thre

Structure and Wound-Healing Activity of a Branched Levan-Type Fructan from Cyathula officinalis Roots.

Cyathula officinalis Kuan, a medicinal plant used in traditional medicine, remains underexplored as a source of structurally defined wound-repair polysaccharides. In this study, a water-soluble polysaccharide fraction, CoPS, was isolated from C. officinalis roots and structurally characterized using methylation analysis and 1D/2D NMR spectroscopy. Purified CoPS had a total carbohydrate content of 94.8%, a weight-average molecular weight (Mw) of 7.491 kDa, and a narrow dispersity (Mw/Mn = 1.04). Monosaccharide composition analysis showed that CoPS was mainly composed of fructose and glucose at
