Tropomyosin is a highly conserved α-helical coiled-coil protein that is involved in maintaining the structural integrity of the sarcomere and regulating contraction of striated muscle. Mutations in the TPM1 gene encoding cardiac α-tropomyosin (Tpm1.1) are associated with left ventricular noncompact cardiomyopathy and congenital heart defects (CHD). We investigated the structural and functional properties of Tpm1.1 with the I130V and S229F mutations associated with CHD. We found that both mutations decreased the Tpm affinity for F-actin and differently affected the sliding velocity of F-actin-T

Background: Advanced glycation end-products (AGEs) are formed and deposited in tissues, contributing to various disorders, including diabetes, other metabolic diseases, and aging. A new epitope, AGE10, was identified in human and animal tissues using a monoclonal antibody raised against synthetic melibiose-derived glycation end-products (MAGE), which were synthesized under anhydrous conditions with bovine serum albumin or myoglobin. The biology of the AGE10 epitope, particularly its role in diseases and in cancer tissues, is not well understood. Methods: The study was aimed at investigating th

As a tropomyosin-binding component, troponin T (TnT) is essential for the Ca2+ regulation of striated muscles' contraction and locomotion activity, but its impacts on the growth and development of insects have rarely been reported. In this study, TnT was identified and functionally characterized in Tribolium castaneum by RNA interference (RNAi) and transcriptome analysis. The TnT of T. castaneum contained a 1152 bp open reading frame encoding 383 amino acids. It displayed the highest expression in late pupae and was highly expressed in the integument and CNS. Both the larval and early pupal in

Nitric oxide (NO) binding to heme-proteins is a physiologically relevant molecular process, contributing to regulate blood pressure and flow to provide an adequate O2 supply to all body compartments. This appears to be relevant for energy-demanding tissues, characterized by high O2-consuming rates, such as the eye's retina and striated muscles under physical effort. The reaction of NO with the ferrous heme-Fe atom (heme-Fe(II)) is characterized by very fast association rate constants and very slow dissociation kinetics, which drastically differ from those observed for other gaseous ligands, su

Four genera of the Boidae family are found in Brazil: Eunectes, Corallus, Epicrates, and Boa. Male copulatory organs in snakes are located inside the tail and are called hemipenes. They are double structures in an inverted position that are exposed during copulation. This study describes the macroscopic and histological aspects of hemipenes in Boidae snakes. One Boa constrictor, three Epicrates cenchria, one Corallus hortulanus hemipenes, and one fragment of the muscular retractor penis magnus (m. retractor penis magnus) from Eunectes murinus were included in this study. The structures were ev

BACKGROUND: Stress incontinence in men is a rare, usually iatrogenic condition. Its prevalence can be expected to rise in the future because of the increasingly common performance of radical prostatectomy. Most men who have undergone prostatectomy experience a transient disturbance of urinary continence. Such disturbances are only rarely due to structural damage to the sphincter apparatus and therefore have a good prognosis for spontaneous recovery.
METHOD: Selective literature review.
RESULTS: Pelvic floor training and/or pharmacotherapy can be used for more rapid restoration of subjectivel

The migratory stage of Trichinella spiralis, the newborn larva, travels along the pulmonary microvascular system on its way to the striated muscle cells. In the present study, an important inflammatory reaction was observed on days 5 and 14 post-infection (p.i.) in the lungs of infected rats. This inflammation was characterized by a Th2 cell phenotype of hyperplastic bronchus-associated lymphoid tissue and by goblet cell hyperplasia. Among the inflammatory cells were eosinophils and mast cells scattered over the pulmonary parenchyma. On day 5 p.i. the number of IgE(+), CD4(+) and CD5(+) cells

The laminin alpha2-chain is a component of merosin, a member of the laminin family molecules, which is mainly expressed in the basement membranes of striated muscle. It is known that laminin alpha2 gene (lama2) null mutant mice (dy3k/dy3k) exhibit congenital muscular dystrophy (CMD). Because the laminin alpha2-chain is also expressed in the thymus, the role of merosin in the thymus was examined. In association with the onset of muscular dystrophy, CD4+ CD8+ double-positive (DP) thymocytes disappear by apoptotic cell death, while CD4+ CD8- or CD4- CD8+ thymocytes remain. In order to study the m

Following the discovery of a second gene containing a CGRP-like sequence, we demonstrated that "beta-CGRP" was indeed translated as a 37-amino acid peptide in vivo and was the predominant form of CGRP produced by the enteric nervous system. The presence of CGRP in the islet has been reported by several groups. We now show that beta-CGRP is again the major form. Another 37-amino acid peptide was recently isolated from islet amyloid deposits and found to have approximately 50% amino acid sequence homology with CGRP. Islet amyloid polypeptide, or amylin, is co-localized with insulin to the beta-c

Glycogen storage disease type IV (GSD IV) is a rare autosomal recessive disorder caused by deficiency of the glycogen-branching enzyme (GBE). The diagnostic hallmark of the disease is the accumulation of a poorly branched form of glycogen known as polyglucosan (PG). The disease is clinically heterogeneous, with variable tissue involvement and age at onset. Complete loss of enzyme activity is lethal in utero or in infancy and affects primarily the muscle and the liver. However, residual enzyme activity as low as 5-20% leads to juvenile or adult onset of a disorder that primarily affects the cen

One of the most common gynecologic complaints is urinary incontinence, which afflicts approximately one third of the female population. In urodynamic units, approximately 80 per cent of women with urinary incontinence have detrusor hyperreflexia as the sole reason for, or as a component of, their incontinence problem. Approximately 80 to 85 per cent of patients with detrusor hyperreflexia have a functional neuromuscular disturbance of their detrusor muscle. One to 2 per cent of the patients seen in our urodynamic unit have detrusor hyperreflexia caused by a neurogenic bladder. As our clinic's

UNLABELLED: Strabismus is a significant cause of a decrease in the functional capabilities of the organ of vision, additionally, it leads to the deteriorating quality of life. Orbital factors and nervous system changes may play an important role in strabismus pathogenesis. There are few reports on binocular vision disorders in radiation-exposed persons.Functions of the external eyeball muscles might be disturbed based on the changed coordination process of subcortical nerve structures and due to altered metabolism. A carefully conducted research is necessary to clarify the possible pathogenesi

Mechanosensing plays an essential role in maintaining tissue functions. Across the human body, several tissues (i.e., striated muscles, bones, tendons, ligaments, as well as cartilage) require mechanical loading to exert their physiological functions. Contrary, mechanical unloading triggers pathological remodeling of these tissues and, consequently, human body dysfunctions. At the cellular level, both mechanical loading and unloading regulate a wide spectrum of cellular pathways. Among those, pathways regulated by oxidants such as reactive oxygen species (ROS) represent an essential node criti

BACKGROUND In the pediatric age group, middle ear tumors are rare. Rhabdomyosarcoma is considered the most common soft-tissue sarcoma in children. It comprises 5% of all pediatric malignant tumors. It is hypothesized to originate from embryonic mesenchymal cells of striated skeletal muscles. These malignant lesions display an aggressive behavior with local and distant metastasis and can be staged as per the Intergroup Rhabdomyosarcoma Study Group, depending on the organs involved, such as the orbit, head, neck, or genitourinary tract. CASE REPORT In this study, a 2-year-old boy with no medical

The forensic evidence of hanging is based on the autopsy demonstration of the traces left by the noose or the ligature on the neck, as well as on the histological assessment of the hanging mark vitality. However, the specific topography of haemorrhagic infiltration in the context of the cervical damaged tissues involved in hanging is not known. We carried out an extensive microscopic examination to identify if haemorrhagic infiltration in hanging appeared in specific topographic locations that could have been considered as elective. From 102 victims of suicidal hanging, a skin fragment was sam

The tayra (Eira barbara) is a mammal belonging to the Mustelidae family that occurs in all Brazilian biomes. The present work aimed to describe the morphology of the tongue of these specimens highlighting their structures and particularities that will serve as a subsidy to elucidate the anatomy of the same and for comparative studies among other species of domestic and wild animals. Five adult male specimens of E. barbara were studied, which were fixed using 10% aqueous formaldehyde solution. The tongue was removed by opening the oral cavity and separating the maxillary/mandible bone complex.

Striated myocytes compose about half of the cells of the heart, while contributing the majority of the heart's mass and volume. In response to increased demands for pumping power, including in diseases of pressure and volume overload, the contractile myocytes undergo non-mitotic growth, resulting in increased heart mass, i.e. cardiac hypertrophy. Myocyte hypertrophy is induced by a change in the gene expression program driven by the altered activity of transcription factors and co-repressor and co-activator chromatin-associated proteins. These gene regulatory proteins are subject to diverse po

Nuclear lamins, namely lamins A, B and C, surround the nucleoplasmic contents in a meshlike network called the nuclear lamina. These intermediate filaments provide a structural framework to the nuclear envelope (NE), play a role in arrangement of the chromatin within the nucleus, in DNA replication and also participate in DNA damage repair. In order for lamins to be involved in these important nuclear processes and to be functionally active, they undergo a series of post-translational modifications (farnesylation, endoproteolytic cleavage, carboxylmethylation etc.), of which farnesylation is t
