A transcriptome atlas of the mouse iris at single-cell resolution defines cell types and the genomic response to pupil dilation.

The iris controls the level of retinal illumination by controlling pupil diameter. It is a site of diverse ophthalmologic diseases and it is … ed and quantified distortions in nuclear morphology associated with iris dilation and clarified the neural crest contribution to the iris by showing that Wnt1-Cre-expressing progenitors contribute to nearly all iris cell types, whereas Sox10-Cre-expressing progenitors contribute only to stromal cells. This work should be useful as a point of reference for inve …

Transcriptional Profiling of Individual Airway Projecting Vagal Sensory Neurons.

Bronchopulmonary sensory neurons are derived from the vagal sensory ganglia and are essential for monitoring the physical and chemical envir … ays and lungs were hierarchically clustered into five types reflecting their developmental lineage (neural crest versus placodal) and putative function (nociceptors versus mechanoreceptors). The purinergic receptor subunit P2rx2 is known to display restricted expression in placodal-derived nodose neurons, and we demonstrate that the gene profiles defining cell …

Crucial and Overlapping Roles of Six1 and Six2 in Craniofacial Development.

SIX1 and SIX2 encode closely related transcription factors of which disruptions have been associated with distinct craniofacial syndromes, w … phogenetic protein-4 signaling pathways to pattern the facial structures. Furthermore, we show that neural crest-specific inactivation of Six1 in Six2 -/- embryos resulted in midline facial cleft and frontal bone agenesis. We show that Six1 -/- Six2 -/- embryos exhibit significantly reduced expression of key frontonasal development genes Alx1 and Alx3 as well …

Mandible and Tongue Development.

The tongue and mandible have common origins. They arise simultaneously from the mandibular arch and are coordinated in their development and growth, which is evident from several clinical conditions such as Pierre Robin sequence. Here, we review in detail the molecular networks controlling both mandible and tongue development. We also discuss their mechanical relationship and evolution as well as the potential for stem cell-based therapies for disorders affecting these organs.

Clinical implications of Schwann cell biology.

The neuroglia of the peripheral nervous system (PNS) are derived from the neural crest and are a diverse family of cells. They consist of myelinating Schwann cells, non-myelinating Schwann cells, satellite cells, and perisynaptic Schwann cells. Due to their prominent role in the formation of myelin, myelinating Schwann cells are the best recognised of these cells. However, Schwann cells and the other neuroglia of the PNS have many functions that are independent of myelination and contribute significantly to the functioning of the peripheral nerve in both health and disease. Here we discuss the…

Role of vitamin A in the formation of congenital heart defects.

Retinoic acid, the biologically active form of vitamin A, is a critical player in normal development. The concentration of retinoic acid is … ng cardiac specific genes. This review briefly discusses the role of cardiac mesenchyme and cardiac neural crest in septation of the heart. This is followed by a discussion of vitamin A metabolism and the cardiac defects associated with abnormal levels of retinoic acid. Finally, a mechanism is proposed concerning the ways abnormal levels of retinoic acid lead …

A reassessment of Goodrich's model of cranial nerve phylogeny.

The first detailed comparisons of the organization and development of craniate head structure occurred during the period 1870-1940. Most wor … -one relationship between nerves and mesodermal derivatives. Review of current data on the roles of neural crest, neuromeric, placodal and somitomeric tissue in the development of vertebrate heads refutes Goodrich's contentions and suggests that at least three, possibly four, separate series of cranial nerves exist in vertebrates and that these nerve series ar …

[A new concept of Pierre Robin syndrome and disease: dysneurulation of the rhombencephalon].

Pierre Robin's syndrome, a disorder apparently constituted by peripheral signs, is in fact the result of early major disturbances of ontogenesis of motor and regulatory organization of the fetal rhombencephalon. This is confirmed by the presenting signs in neonates with Pierre Robin's syndrome: --electrophysiological deglutition and sucking disorders as demonstrated on electromyography; --disorders in tone of tongue, pharyngeal and laryngeal muscles; --cardiac and respiratory regulatory disorders as shown by central and obstructive apneas with diminished oxygen pressure and bradycardia of cent

Craniofacial development: genetics, signaling pathways, teratogenic mechanisms, and clinal significance.

Craniofacial development is a highly coordinated and stage-specific process involving neural crest cell migration, proliferation, differentiation, and epithelial-mesenchymal transition. Genetic mutations, signaling pathway abnormalities, and exposure to environmental teratogens can disrupt normal development. These anomalies can lead to cleft palate, craniosynostosis, and other common congenital craniofacial malformations. Although significant progress has been made in understanding craniofacial development in recent years, the systematic integration of these factors remains limited. Here, we …

Novel PHOX2B mutations in congenital central hypoventilation syndrome.

BACKGROUND: Congenital central hypoventilation syndrome (CCHS) is caused by mutation of paird-like homeobox 2B (PHOX2B). Approximately 90% of patients were found to carry polyalanine repeat expansion mutation (PARM), and the remaining 10% had non-PARM (NPARM). In PARM, the length of the polyalanine expansion correlates with clinical disease severity. Most patients with NPARM have hypoventilation symptoms in the neonatal period and complications of Hirschsprung disease, dysregulation of autonomic nervous system, and tumors of neural crest origin. Data on the genotype-phenotype association may c…

A novel cytokine pathway suppresses glial cell melanogenesis after injury to adult nerve.

The neural crest gives rise to numerous cell types, including Schwann cells, neurons, and melanocytes. The extent to which adult neural crest-derived cells retain plasticity has not been tested previously. We report that cutting adult mouse sciatic nerve induces pigmentation around nerve fascicles, among muscle bundles, and in the hypodermis. Pigmented cells are derived from adult nerve, because pigmentation occurs even when nerve fragments are grafted into tyrosinase null albino mice. Pigmentation defects are pervasive in patients with neurofibromatosis type 1 (NF1). Mice hemizygous for Nf1 m…

Primitive neuroectodermal tumor of the esophagus with metastasis in the pineal gland.

Primitive neuroectodermal tumors (PNET) are very rare tumors that belong to a family of malignant neoplasms of tiny round cells which are derived from the neural crest. This report discusses a rare case of an adult woman with esophageal PNET, confirmed by immunohistochemistry, that presented with metastasis to the pineal gland. To our knowledge, this is the first case report of a PNET with these features. Despite surgery and chemotherapeutic treatment, our case has shown disease progression.

Giant interaortocaval pheochromocytoma.

Tumors arising from chromaffin cells, which are derived embryologically from neural crest cells, are known as pheochromocytomas or paragangliomas. These tumors in the retroperitoneum are usually located in close proximity to the aorta and inferior vena cava. Radical surgical resection is the treatment of choice for these tumors. We present a rare case of a postmenopausal woman who presented to us with a symptomatic giant retroperitoneal mass situated in the interaortocaval region that we managed with surgical resection.

Vasoactive intestinal peptide producing neuroblastoma.

The Verner-Morrison syndrome has been described in 19 previous patients with ganglioneuroma and ganglioneuroblastoma but never neuroblastoma. Its occurrence following treatment of a neuroblastoma with chemotherapy with maturation of the tumor has only been reported on one previous occasion. Our case suggests that vasoactive intestinal polypeptide may be used not only as a diagnostic indicator for the presence of a neural crest tumor but also as a marker to monitor maturation of the tumor and indicate an improving prognosis.

Grainyhead-like 2 downstream targets act to suppress epithelial-to-mesenchymal transition during neural tube closure.

The transcription factor grainyhead-like 2 (GRHL2) is expressed in non-neural ectoderm (NNE) and Grhl2 loss results in fully penetrant crani … NE, including aberrant vimentin expression and increased cellular dynamics that affects the NNE and neural crest cells. The resulting loss of NNE integrity contributes to an inability of the cranial neural folds to move toward the midline and results in NTD. Further, we identified Esrp1, Sostdc1, Fermt1, Tmprss2 and Lamc2 as novel NNE-expressed genes that are …

Phase separation and condensates in craniofacial development.

Biomolecular condensates formed by liquid-liquid phase separation (LLPS) have emerged as a major mode of intracellular organization, and their roles in craniofacial development are only beginning to be defined. In this review, we synthesize evidence that phase separation and condensate biology influences craniofacial morphogenesis at multiple levels, from transcriptional control in neural crest-derived lineages to signaling integration, mechanosensing, and the formation of mineralized tissues. Nuclear condensates formed by transcription factors and chromatin regulators are implicated in clefti…
