A randomized double-blind placebo-controlled study to assess the effect of oral contraceptive pills on the outcome of medical abortion with mifepristone and misoprostol.

This was a randomized double-blind placebo-controlled trial to determine the effect of oral contraceptive (OC) pills taken immediately after medical abortion on the duration of bleeding and complete abortion rate. Two hundred women in the first 49 days of pregnancy were given 200 mg mifepristone orally followed by 400 microg misoprostol vaginally 48 h later. One day later, they were randomized to receive either OC pills (30 microg of ethinyl oestradiol and 0.15 mg of levonorgestrel per tablet) or placebo for 21 days. The complete abortion rates were 98% in the OC group and 99% in the placebo g

Effect of cloricromene on intermittent claudication. A randomized, double-blind, placebo-controlled trial in patients treated with aspirin: effect on claudication distance and quality of life. CRAMPS Investigator Group. Cloricromene Randomized Arteriopathy Multicenter Prospective Study.

The main aim of medical treatment for intermittent claudication (IC) is the reduction of mortality and morbidity from ischemic cardiovascular disease. However, symptomatic treatment with the aim of improving exercise performance and the overall quality of life may also be an important target of the clinical management of patients with intermittent claudication. Cloricromene, a drug with antithrombotic and anti-ischemic activities, has previously shown some promising results in patients with claudication. We have carried out a clinical trial to assess the effect of cloricromene on the claudicat

McKenzie Method of Mechanical Diagnosis and Therapy was slightly more effective than placebo for pain, but not for disability, in patients with chronic non-specific low back pain: a randomised placebo controlled trial with short and longer term follow-up.

BACKGROUND: The McKenzie Method of Mechanical Diagnosis and Therapy (MDT) is one of the exercise approaches recommended by low back pain (LBP) guidelines. We investigated the efficacy of MDT compared with placebo in patients with chronic LBP. METHODS: This was a prospectively registered, two-arm randomised placebo controlled trial, with a blinded assessor. A total of 148 patients seeking care for chronic LBP were randomly allocated to either MDT (n=74) or placebo (n=74). Patients from both groups received 10 treatment sessions over 5 weeks. Patients from both groups also received an education

Vascular effects of dietary nitrate supplementation in older adults with coronary artery disease: A randomized, placebo-controlled, crossover pilot trial.

Coronary artery disease (CAD) is characterized by endothelial dysfunction, arterial stiffening, and abnormal central hemodynamics, which are independent predictors of adverse cardiovascular events. Dietary nitrate supplementation using beetroot-derived products is a promising strategy, but its vascular effects in adults with CAD remain unclear. This randomized, double-blind, placebo-controlled, crossover pilot trial evaluated the safety, feasibility, and vascular effects of dietary nitrate supplementation to inform the development of a full-scale trial. Eight older adults with confirmed CAD (6

Recent cannabis exposure and acute nicotine effects: insights from randomized, double-blind, placebo-controlled intravenous nicotine studies.

OBJECTIVE: Cannabis-nicotine co-use is common and may affect cessation outcomes, yet whether recent cannabis exposure alters nicotine's acute effects in humans remains unclear. We examined whether recent, non-daily cannabis exposure modulates the acute subjective effects of intravenous (IV) nicotine. METHODS: We pooled data from 2 randomized, double-blind, placebo-controlled studies using IV nicotine to isolate nicotine's pharmacodynamics in adults who smoke tobacco cigarettes (N = 60). Participants completed 3 sessions (placebo, 0.1 mg, 0.2 mg nicotine/70 kg). Recent cannabis exposure was de

Effects of vitamin D3 supplementation on cardiac, muscle, and immune responses after a marathon race: a single-blind, placebo-controlled trial.

BACKGROUND: Strenuous endurance exercise imposes substantial physiological stress on the cardiovascular system and has been associated with transient elevations in cardiac biomarkers. Vitamin D₃ has been suggested to influence oxidative stress and immune responses. In this study, we investigated the effects of vitamin D₃ supplementation on biomarkers of cardiac, muscle, and immune responses following a marathon race. METHODS: Twenty-one amateur runners were allocated to either a vitamin D₃ supplementation group (receiving vitamin D₃ for 8 weeks) or a placebo group. All participants completed

Perioperative Antimicrobial Prophylaxis for Invasive Dental Procedures: A Systematic Review and Random-Effects Meta-Analysis of Randomized and Placebo-Controlled Studies.

Prophylactic antimicrobials are frequently administered before invasive dental procedures (including implant placement and extractions) to reduce early infectious complications and procedure-related bacteremia; however, clinical benefit remains debated and must be balanced against ecological disruption and antimicrobial resistance. Evidence from experimental work indicates that even a single prophylactic dose of amoxicillin may transiently perturb the oral microbiome and select resistant strains. A Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA)-guided systematic re

BASTA trial: protocol for a randomised, double-blind, placebo-controlled, crossover trial investigating the effects of atorvastatin on bile acid diarrhoea.

INTRODUCTION: Bile acid diarrhoea (BAD) is a chronic disease with typical symptoms of daily watery stools with increased frequency and urgency. BAD arises due to pathologically high levels of bile acids in the colon, which may be due to impaired reabsorption or increased synthesis. Evidence indicates that atorvastatin administration suppresses bile acid synthesis in healthy individuals, likely due to reduced substrate availability. The Bile Acid diarrhoea STAtin Trial aims to investigate whether atorvastatin treatment reduces bile acid synthesis in patients with BAD and consequently alleviates

Effects of a Multi-Component Nutraceutical (NUT2) on the Lipid Profile in Adults with Low-to-Moderate Cardiovascular Risk: A Randomized Placebo-Controlled Trial.

Background: Lifestyle intervention is recommended as first-line management for individuals with low-to-moderate cardiovascular (CV) risk and elevated low-density lipoprotein cholesterol (LDL-C), although dietary intervention alone often produces only modest lipid reductions. NUT2 is a proprietary multi-component nutraceutical formulated to target complementary pathways involved in lipid metabolism, oxidative stress and cardiometabolic risk. This study evaluated its efficacy and safety as an adjunct to dietary intervention. Methods: In this randomized, double-blind, placebo-controlled, parallel

A randomized, double-blind, placebo-controlled, multiple dose, parallel study to investigate the effects of a cathepsin S inhibitor in celiac disease.

Celiac disease is a chronic, immune-mediated enteropathy with symptoms triggered by exposure to dietary gluten in genetically predisposed individuals. The only available management option is lifelong adherence to a gluten-free diet. This randomized, double-blind, placebo-controlled, parallel-group, single-center study tested the effects of the cathepsin S inhibitor RO5459072 on the immune response to a 13-day gluten challenge in 19 participants with celiac disease (ClinicalTrials.gov: NCT02679014). Nine participants in the RO5459072 arm received 100 mg study drug b.i.d. (200 mg daily); 10 rece

Placebo effects improve sickness symptoms and drug efficacy during systemic inflammation: a randomized controlled trial in human experimental endotoxemia.

BACKGROUND: Systemic inflammation triggers a wide range of sickness symptoms, including bodily discomfort and affective symptoms, which are relevant to numerous health conditions. While extensive research in the placebo field demonstrates that positive expectations can improve symptoms, it remains unclear if interventions designed to augment positive treatment expectations can alleviate sickness symptoms in the context of immunomodulatory drug therapies. METHODS: In this randomized, controlled, fully balanced 2 × 2 factorial placebo design, N = 124 healthy volunteers received either active ib

A within-subject, double-blind, placebo-controlled randomized evaluation of the combined effects of cannabidiol and hydromorphone in a human laboratory pain model.

Preclinical and epidemiological evidence supports that cannabinoids may have opioid-sparing properties and could be one strategy to decrease opioid use and associated harms like overdose and extramedical use. The objective of this within subjects, double-blind, double-dummy, randomized human laboratory trial was to examine whether cannabidiol (CBD) increases opioid analgesic effects and whether there are corresponding increases in other opioid mediated effects. Healthy participants (N = 31) attended 5 outpatient sessions where they received the following drug conditions: (1) placebo + placebo,

Assessing the effects of naltrexone-bupropion on hepatic steatosis and fibrosis in patients with T2DM and overweight or obesity: Insights from a placebo-controlled trial.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is prevalent among individuals with overweight or obesity and type 2 diabetes (T2DM), increasing the risk for liver-related and cardiovascular complications. Lifestyle changes to reduce body weight are the primary intervention to decrease the risk of MASLD and liver fibrosis. This study aimed to evaluate the impact of a pharmacological intervention with naltrexone/bupropion (NB) on hepatic steatosis and fibrosis risk in patients with T2DM using non-invasive tests. MATERIALS AND METHODS: In this post hoc analysis of a

Effects of Probucol on plasma amyloid-β transport in patients with hyperlipidemia: a 12-week randomized, double-blind, placebo-controlled trial.

BACKGROUND: Although dyslipidemia has been acknowledged as a risk factor for Alzheimer's disease (AD), the effects of lipid-lowering drugs on AD have not been determined. The primary pathophysiological hallmark of AD is the deposition of amyloid-β (Aβ) plaques in the brain. Plasma Aβ levels are influenced by the transport of Aβ from the central nervous system to the peripheral blood. This study investigates the effects of Probucol, a lipid-lowering and antioxidant drug, on plasma Aβ transport. METHODS: A total of 120 hyperlipidemic patients with normal cognition were randomly assigned (1:1 ra

The effects of empagliflozin in patients with type 1 diabetes: Results of a 12-week, double-blind, randomized, placebo-controlled clinical trial.

BACKGROUND: Sodium-glucose cotransporter-2 (SGLT-2) acts as a key element in the reabsorption of glucose in the kidney. Currently, SGLT-2 inhibitors are FDA-approved for the treatment of type 2 diabetes. It is suggested that the mechanism of action may operate in the treatment of type 1 diabetes mellitus (T1DM), as well. This study aimed to evaluate the application of empagliflozin as an adjunctive to insulin in patients with T1D. METHODS: In this double-blind placebo-controlled randomized clinical study, 60 type 1 diabetic patients were randomly assigned to have either once-daily empaglifloz
