Auxin and Cytokinins-Mediated Regulation of Adventitious Shoot Regeneration via the RpARR12/RpARF19-RpWOX4-RpLBD4 Pathway in Robinia pseudoacacia.

Woody plants are fast-growing and ecologically important, with widespread applications in urban landscaping and ecological restoration. However, limited understanding of the molecular mechanisms underlying adventitious shoot regeneration has hindered advances in tissue culture and genetic improvement of woody species. Here, we present a comprehensive framework for elucidating adventitious shoot regeneration in Robinia pseudoacacia. Histological analyses delineated key developmental stages, while RNA-sequencing and hormone profiling highlighted the critical roles of auxin and cytokinins. WGCNA

Millets for a sustainable future.

Our current agricultural system faces a perfect storm-climate change, a burgeoning population, and unpredictable outbreaks such as COVID-19 which disrupt food production, particularly for vulnerable populations in developing countries. A paradigm shift in agriculture practices is needed to tackle these issues. One solution is the diversification of crop production. While ~56% of the plant-based protein stems from three major cereal crops (rice, wheat, and maize), underutilized crops such as millets, legumes, and other cereals are highly neglected by farmers and the research community. Millets

ATP-Binding Cassette G Transporters SGE1 and MtABCG13 Control Stigma Exsertion.

Stigma exsertion is an important agricultural trait that facilitates the application of heterosis in crop breeding. Although several quantitative trait loci associated with stigma exsertion have been fine-mapped or cloned, the underlying genetic basis, particularly in legumes, remains unclear. In this study, we identified and characterized the exserted stigma mutant stigma exsertion1 (sge1) in the model legume Medicago truncatula The exserted stigma phenotype of sge1 is mainly caused by physical interaction between floral organs, in which normal petal and stamen elongation are inhibited due to

Mutations in the miR396 binding site of the growth-regulating factor gene VvGRF4 modulate inflorescence architecture in grapevine.

Bunch rot caused by Botrytis cinerea infections is a notorious problem in grapevine cultivation. To produce high quality fruits, grapevine plants are treated with fungicides, which is cost intensive and harmful to the environment. Conversely, loose cluster bunches show a considerably enhanced physical resilience to bunch diseases. With the aim to identify genetic determinants that modulate the development of bunch architecture, we have compared loose and compact 'Pinot noir' clones. Loose cluster architecture was found to be correlated with increased berry size, elongated rachis and elongated

β-Glucosidase activity in almond seeds.

Almond bitterness is the most important trait for breeding programs since bitter-kernelled seedlings are usually discarded. Amygdalin and its precursor prunasin are hydrolyzed by specific enzymes called β-glucosidases. In order to better understand the genetic control of almond bitterness, some studies have shown differences in the location of prunasin hydrolases (PH, the β-glucosidase that degrades prunasin) in sweet and bitter genotypes. The aim of this work was to isolate and characterize different PHs in sweet- and bitter-kernelled almonds to determine whether differences in their genomic

Early Response QTLs and DEGs Underlying Oil Palm Resistance to Ganoderma boninense in Two Breeding Populations Evaluated in Pre-Nursery Trials.

Ganoderma boninense, a soil-borne pathogenic fungus causing basal stem rot (BSR) disease in oil palm, is the major threat to cultivation in Southeast Asia, with the potential to reduce oil production by up to 80% in severely affected plantations. With a view to marker-assisted selection (MAS) of resistant planting material, we combined genetics and transcriptomics approaches to investigate the defense mechanisms of oil palm against infection by G. boninense. We first performed quantitative trait locus (QTL) mapping of BSR resistance using a multi-locus Bayesian variable selection approach appl

Salt stress and the wheat curl mite (Aceria tosichella) infestation reprograms barley nitrogen metabolism via nitric oxide signaling.

Salinity (NaCl) and wheat curl mite infestation profoundly alter nitric oxide (NO) metabolism in barley, with NaCl dose-dependent responses indicating that nitrogen metabolism is fine-tuned through NO-signaling pathways. Nitric oxide (NO) serves as a multifaceted regulator in plants' responses to environmental stress, acting as both a signaling molecule and a protective agent, while also exhibiting potential harmful effects. Our study investigated NO metabolism in barley (Hordeum vulgare L.) plants under salinity (50 mM and 100 mM NaCl) and wheat curl mite (WCM) infestation, exposed to either

The nature of gain curves.

Gain curves have been a staple of sex allocation theory for decades. They represent patterns in which fitness is obtained from resource investments in reproductive functions. The monotonic forms that have been used for gain curves can represent fitness accrual by individuals, but only on the assumption that sufficient mates are always available to allow the stipulated monotonic pattern of reproductive success to occur. However, sexual populations do not have external banks of mating opportunities that lie outside the dynamics of the population (such opportunities would, by definition, be part

The SeABI5 transcription factor couples ABA signaling with flavonoid antioxidant production to confer cadmium tolerance in Salicornia europaea.

Understanding the extraordinary stress tolerance mechanisms of extremophile plants represented by Salicornia europaea is crucial for isolating genetic tools to improve crop resilience. However, the detailed mechanisms of S. europaea to cadmium (Cd) remain poorly understood. Here, combining transcriptomic and metabolomic profiling, we deciphered the adaptive response of S. europaea to Cd stress. Cd stress triggered extensive metabolic and transcriptional reprogramming, with 237 metabolites and 1677 genes upregulated, among which flavonoid biosynthetic genes (SeCHS and SeF3H) and their correspon

Characterization of BrSTM16 from Brassica rapa and functional analysis of its role in regulating cold tolerance.

The SHOOTMERISTEMLESS (STM) gene family controls shoot apical meristem homeostasis and participates in plant development and abiotic stress adaptation in higher plants. While the conserved roles of STM in maintaining meristem stability are well documented, the functions and regulatory mechanisms of BrSTM genes in Brassica rapa cold tolerance remain elusive, hindering our understanding of stress mediated meristem plasticity. Here, we systematically characterized the Brassica rapa STM gene family and identified 31 BrSTM members, all harboring the conserved KNOXⅠ domain and belonging to the class

Cloning of CgWRKY53 from Cymbidium goeringii and Functional Analysis of Its Negative Regulatory Role in Response to Cold Stress.

Background:Cymbidium goeringii, one of China's traditional and valuable orchids, possesses significant ornamental and economic value. However, it is relatively sensitive to low temperature and other abiotic stresses, which severely restrict its application in landscaping and industrial development. WRKY transcription factors play important roles in plant responses to abiotic stresses, yet related research in C. goeringii remains limited. Methods: In this study, based on transcriptome data of C. goeringii under four different stresses, we identified and cloned the WRKY transcription factor gene

Sustained assimilate supply determines grain development in wheat under post-anthesis heat.

Short episodes of post-anthesis heat restrict grain filling by accelerating leaf senescence, reducing photosynthetic capacity, and limiting assimilate supply to developing grains. We explored the contribution of assimilates, derived from both current photosynthesis and remobilisation, for sustaining wheat yield under post-anthesis heat. Two recombinant inbred lines, SB062 and SB003, were subjected to 7-day heat treatment during different post-anthesis phases. High temperature during the early to mid-grain filling phases (7-14 or 14-21 days after anthesis (DAA)) accelerated leaf senescence, wit

Utilizing genetic variation in perennial sorghum to improve host plant resistance to aphids.

With growing concerns over the sustainability of conventional farming systems, perennial crops offer an environmentally friendly and resilient alternative for long-term agricultural production. Perennial grain crops provide numerous benefits, such as low input investment, reduced tillage, soil conservation, better carbon sequestration, sustainable yields, and enhanced biodiversity support. Sorghum (Sorghum bicolor) is the fifth most-grown cereal crop grown for food, fuel, and food grain in the world. The development of perennial sorghum offers a substitute for traditional annual sorghum crops

Unraveling Cross-Cellular Communication in Cannabis sativa Sex Determination: A Network Ontology Transcript Annotation (Nota) Analysis.

Cannabis sativa L. (marijuana, hemp) is a dioecious angiosperm species currently evolving sex chromosomes. Genetic mechanisms, primarily controlled by an XY chromosome system, appear to dictate sex expression in C. sativa. However, sexual expression is also governed by the interplay of hormone regulatory gene networks, influenced by both genetic and environmental factors. Within the species, some populations exhibit dioecy, monoecy, or a gradient of both. Dioecious individuals produce exclusively male or female flowers, while monoecious plants bear both male and female flowers. However, throug

BcAMT1;2 interacts with BcLBD41 and BcMAMYB transcription factors during nitrogen metabolism in flowering Chinese cabbage.

Chinese cabbage is an important vegetable in southern China. Excessive nitrogen fertilizer application can lead to the accumulation of nitrate in edible organs, which affects food value. Hence, the cultivation of varieties with high nitrogen utilization efficiency (NUE) and low nitrate accumulation is essential for molecular breeding. In flowering Chinese cabbage, Ammonium transporter 1;2 (AMT1;2,XM_009113156.2) significantly promotes plant growth, while reducing the nitrate content and ultimately improving the nutritional value of the crop; however, the exact underlying regulatory mechanisms

Eco-geographically divergent diploids, Caucasian clover (Trifolium ambiguum) and western clover (T. occidentale), retain most requirements for hybridization.

BACKGROUND AND AIMS: DNA sequence similarities and hybridization patterns in Trifolium (clovers) section Trifoliastrum suggest that rapid radiation from a common ancestral source led to this complex of diverse species distributed across Europe, western Asia and North Africa. Two of the most geographically and ecologically divergent of these species are the rhizomatous T. ambiguum from high altitudes in eastern Europe and western Asia and the stoloniferous T. occidentale from sea level in western Europe. Attempts were made to hybridize these species to ascertain whether, despite this separation

Genome-wide identification of the aldo-keto reductase gene family and functional characterization of CsAKRIVB2 conferring cold tolerance in tea plant (Camellia sinensis).

Aldo-keto reductases (AKRs) are a superfamily of NADPH-dependent oxidoreductases that play a crucial role in detoxifying reactive carbonyl species and maintaining redox homeostasis under abiotic stresses. However, systematic characterization of AKRs and their functional roles in cold stress responses in tea plants (Camellia sinensis) remain largely unexplored. We identified 48 CsAKR genes genome-wide and classified them into four groups. Structural and promoter analyses pointed to conserved protein backbones alongside diversified regulatory architectures. Expression profiling and RT-qPCR scree

The durable resistance gene Tm-22 remains partially resistant to tomato brown rugose fruit virus.

The tomato Tm-22 gene is a highly effective, and durable resistance gene in agriculture that has protected tomato production against viruses of the Tobamovirus genus, such as tomato mosaic virus (ToMV) and tobacco mosaic virus (TMV) for over 60 years. This dominant R gene, originally sourced from wild tomato species (Solanum peruvianum), acts by recognizing the viral movement protein (MP) and triggering an immune response, often resulting in extreme resistance (ER). However, this durable protection is challenged by a recently emerged new tobamovirus named tomato brown rugose fruit virus (ToBRF

CRISPR/Cas9-Mediated Editing of Bsr-d1 and Pi21 Enhances Blast Resistance in a High-Quality Rice Maintainer Line.

Rice (Oryza sativa L.) is a staple food crop worldwide, and improving disease resistance is a core target in rice breeding. In this study, we employed CRISPR/Cas9 genome editing to modify the coding sequence (CDS) of two susceptibility genes, Bsr-d1 and Pi21, in the elite maintainer line Gengxiang B to enhance its blast resistance. We generated Bsr-d1/Pi21 double homozygous mutants via Agrobacterium-mediated genetic transformation. Quantitative RT-PCR revealed significantly suppressed transcript accumulation of both target genes in the edited lines compared with the wild type Gengxiang B. Upon
