The bacterial antiphage protein Tiamat is an ATPase and DNase whose homologs are found across the tree of life.

Bacteria and archaea encode hundreds of antiviral defense systems, some with homologs found in eukaryotic innate immunity. GHL ATPases like plant MORCs and prokaryotic Tiamat defend against viruses and phage, though their architecture and biochemical activities have remained unclear. Searching thousands of proteomes, we find Tiamat is rare in prokaryotes yet enriched across eukaryotes, including green plants, aquatic chordates, sponges, placozoans, and water molds. We resolve the central DUF3684 and flanking region into three predicted folds (TPR-like repeats, an uncharacterized conserved fold

Functional analysis of NtWRKY70 regulating tobacco defense against Rhizoctonia solani AG3-TB.

Rhizoctonia solani AG3-TB is a destructive necrotrophic pathogen causing tobacco target spot disease. WRKY70 acts as a key regulator integrating salicylic acid (SA) and jasmonic acid (JA) signals in plant immunity. Our previous study found that NtWRKY70 in Nicotiana tabacum L. is significantly induced by R. solani AG3-TB infection; however, its functional mechanism remains elusive. In this study, bioinformatics analysis showed that NtWRKY70 is a nuclear localized protein with conserved domains and transcriptional autoactivation activity. Further investigations using virus-induced gene silencin…

Nanotechnology-Enabled Approaches in Poultry Coccidiosis Management: Advances, Challenges, and Future Perspectives.

Avian coccidiosis, a disease resulting from infection by various Eimeria species, is a major constraint to global poultry production, causing considerable economic losses and affecting animal performance. Traditional approaches to coccidiosis control, such as anticoccidials and live vaccines, face limitations in their use owing to resistance, safety concerns, and species-specific immunity. In this regard, subunit, DNA, and multi-epitope vaccines have been tested against coccidiosis, but their efficacy is hindered by the instability of the vaccine antigens and a lack of cross-protection. Howeve

Ecological and methodological insights from genetic and coprological profiling of gastrointestinal communities in wild howler monkeys.

The gastrointestinal tract hosts a complex community of microorganisms and helminth parasites that collectively contribute to host health and fitness. Analysis of these communities provides insight into diverse aspects of host dietary ecology, immunity, nutrition, and host-parasite interactions. However, research methodologies, such as sample preservation and sequencing approach, can influence how we understand and characterize these features. Here, we profiled the gastrointestinal microbial and helminth communities in different groups of wild Costa Rican mantled howler monkeys (Alouatta palli

A PR1-Like Protein Stabilizes a Barwin-Like Protein to Enhance Wheat Resistance to Leaf Rust.

Pathogenesis-related 1 (PR1) proteins contribute to plant immunity through antimicrobial activity and the regulation of target protein stability and condensation. In wheat, TaPR1-2 has been widely used as a molecular marker of immune responses to rust fungi. However, it remains unclear whether other members of the PR1 family are involved in wheat resistance to leaf rust. Here, we show that the transcript levels of TaPR1-11 are induced following inoculation with Puccinia triticina (Pt). Overexpression of TaPR1-11 significantly enhanced wheat resistance to Pt, accompanied by reduced fungal growt…

The Impact of Consuming Probiotics and Following a Vegetarian Diet on the Outcomes of Acne.

Acne vulgaris is a common skin condition that affects millions of people worldwide. While the exact cause of acne is not fully understood, it is believed to be influenced by various factors such as the skin microbiome, host immunity, hormones, genetics, and possibly diet. There are several treatment options for acne, including antibiotics and vitamin A derivatives (retinoids). However, these treatments can have side effects, such as dryness, redness, and peeling of the skin. The relationship between diet and acne remains somewhat controversial. Studies have found that Western societies have a

Potential Effects of Garlic (Allium sativum L.) on the Performance, Immunity, Gut Health, Anti-Oxidant Status, Blood Parameters, and Intestinal Microbiota of Poultry: An Updated Comprehensive Review.

The use of antibiotics as growth promoters or for the prevention of some poultry diseases has faced global concern and serious criticism. Their addition to poultry feed has shown hazardous effects, including the development of antimicrobial resistance and a potentially harmful effect on human health. To eliminate these threats, there is increasing interest in natural alternatives. Plant derivatives such as garlic (Allium sativum L.) and its derivatives are presently extensively used in the poultry production system. The dietary supplementation of broilers and layers with garlic induced improve

Tea tree oil nanoemulsion targets AgrA protein potentiates amoxicillin efficacy against methicillin-resistant Staphylococcus aureus.

The excessive utilization of antibiotics gives rise to the development of bacterial resistance, the deterioration of animal immune functions, the increase in mortality rates, and the undermining of human immunity. Therefore, there is an urgent necessity to explore new antimicrobial agents or alternatives to tackle bacterial resistance. We investigated tea tree oil (TTO), a pure natural plant essential oil extracted from Melaleuca leaves, which exerted efficient antibacterial activities. However, the poor solubility and high volatility of TTO limited the clinical application. Therefore, tea tre

The regulating effect of Tibet Opuntia ficus-indica (Linn.) Mill. polysaccharides on the intestinal flora of cyclophosphamide-induced immunocompromised mice.

The stem of Opuntia species, a traditional medicinal plant, is widely used as food and functional raw material because of its rich polysaccharide content. There have been many studies on the immune function of polysaccharides from Opuntia stem, but only few have examined this function with respect to intestinal microbes. In this study, the effects of different concentrations of Opuntia stem polysaccharides on the immunity and intestinal microflora of cyclophosphamide (CTX)-induced immunocompromised mice were explored. The results showed that Tibet Opuntia ficus-indica (Linn.) Mill. polysacchar

Construction of a fusion anti-caries DNA vaccine in transgenic tomato plants for PAcA gene and cholera toxin B subunit.

Chronic bacterial infections in the oral cavity influence the development of dental caries. Mutans streptococci are the major pathogenic cause of dental caries. The World Health Organization (WHO) ranks dental caries, cancer, and cardiovascular diseases as the three major global diseases that need urgent preventative and curative measures. However, substantial evidence suggests that traditional prevention and treatment strategies are inefficient in reducing the prevalence of dental caries. For protection against caries, it is important to develop effective vaccines that induce anticolonizing i

Peucedanum japonicum-Derived Exosome-like Nanovesicles Alleviate Contact Dermatitis.

Contact dermatitis is a common inflammatory skin disorder triggered by exposure to allergens or irritants and characterized by erythema, swelling, and immune cell infiltration. In this study, we investigated the anti-inflammatory effects of exosome-like nanovesicles derived from Peucedanum japonicum Thunb. (PjELNs) using a 2,4,6-trinitrochlorobenzene (TNCB)-induced mouse model of contact dermatitis. Intraperitoneal administration of PjELNs markedly reduced ear swelling and histopathological damage and decreased infiltration of inflammatory immune cells, particularly neutrophils. Moreover, PjEL

Understanding the intricate impacts and mechanism of actions of adaptogens on reproductive function.

Adaptogens, comprising plants and mushrooms, modulate the immune system, energy balance, and various physiological processes, including reproduction. Despite their potential benefits, the impact of adaptogens on reproductive function remains understudied. This review examines the effects of common adaptogens on male and female reproductive functions, highlighting their regulation of neuro-endocrine-immune interactions crucial for reproduction. While existing literature reveals varying impacts on reproductive function, most adaptogens exhibit beneficial effects, modulating neuroimmunology and p

Antifungal and antibiofilm activities of chromones against nine Candida species.

The persistence of Candida infections is due to its ability to form biofilms that enable it to resist antifungals and host immune systems. Hence, inhibitions of the biofilm formation and virulence characteristics of Candida sp. provide potential means of addressing these infections. Among various chromone derivatives tested, four chromone-3-carbonitriles showed antifungal, antibiofilm, and antivirulence activities against several Candida species. Their mode of action has been partially revealed, and their toxicity is reported here using nematode and plant models.

An Improved Immunochromatographic Strip Based on Plant-Derived E2 for Detection of Antibodies against Classical Swine Fever Virus.

Vaccination is an effective method to control the spread of classical swine fever virus (CSFV), which is a major cause of economic losses to the swine industry. Although serological detection assays are commonly used to assess immune status, current methods for monitoring of antibodies (Abs) are time-consuming, expensive, and require cell culture and virus manipulation. To address these problems, the E2 protein of CSFV was expressed in transgenic rice seeds as a labeled antigen for the development of an immunochromatographic test strip (ICTS) for rapid, precise, and cost-effective detection of

Novel molecular components involved in callose-mediated Arabidopsis defense against Salmonella enterica and Escherichia coli O157:H7.

BACKGROUND: Food contamination with Salmonella enterica and enterohemorrhagic Escherichia coli is among the leading causes of foodborne illnesses worldwide and crop plants are associated with > 50% of the disease outbreaks. However, the mechanisms underlying the interaction of these human pathogens with plants remain elusive. In this study, we have explored plant resistance mechanisms against these enterobacteria and the plant pathogen Pseudomonas syringae pv. tomato (Pst) DC3118, as an opportunity to improve food safety. RESULTS: We found that S. enterica serovar Typhimurium (STm) transcript

Antifungal peptides: From modes of action to synergistic and immunologic potential.

Fungal infections pose a significant global health threat with rising morbidity and mortality rates. However, the repertoire of effective antifungal drugs remains narrow, a challenge that is further exacerbated by the increasing emergence of (multi)drug-resistant strains. This underscores the urgent need for novel therapeutic strategies. Among them, antifungal peptides (AFPs) have emerged as a promising alternative. AFPs are small, naturally occurring peptides produced by a wide range of organisms, including plants, animals, fungi, and bacteria, as part of their innate immune defense. In addit

Systemic proteome remodeling in soybean during early nodulation: distinct responses in roots and leaves.

Here, we aimed to deepen our understanding of the integration between root and shoot signaling during early nodulation by jointly analyzing proteome-level dynamics in roots and leaves of the wild type (WT) and the hypernodulating autoregulation-of-nodulation (AON) mutant, nark. To this end, we performed a time-course shotgun proteomics experiment in soybean roots and leaves of the same plants inoculated with Bradyrhizobium diazoefficiens. In WT roots, we observed proteome changes linked to calcium-dependent signaling, cellular reorganization, and nutrient allocation. In the leaves, isoflavonoi

Crocodile defensin (CpoBD13) antifungal activity via pH-dependent phospholipid targeting and membrane disruption.

Crocodilians are an order of ancient reptiles that thrive in pathogen-rich environments. The ability to inhabit these harsh environments is indicative of a resilient innate immune system. Defensins, a family of cysteine-rich cationic host defence peptides, are a major component of the innate immune systems of all plant and animal species, however crocodilian defensins are poorly characterised. We now show that the saltwater crocodile defensin CpoBD13 harbors potent antifungal activity that is mediated by a pH-dependent membrane-targeting action. CpoBD13 binds the phospholipid phosphatidic acid

Defensin-barbed innate immunity: clinical associations in the pediatric population.

Defensins and related antimicrobial peptides serve a central role in innate immunity in all species of plants and animals. In humans, defensins are widely expressed, including in neutrophils, skin, and mucosal epithelia. Most defensins are potent antibiotics, and some have chemotactic and toxin-neutralizing activities. Results of recent studies on the homeostatic and disease-fighting activities of human defensins point to a key relevance in several pediatric disorders. Inherited variation in defensin gene expression may contribute to susceptibility to several diseases, including psoriasis and

[The characteristics of the clinical manifestations and the pathogenesis of foot mycoses complicated by candidiasis in metallurgists].

Mycologic examinations of patients with mycoses of the soles, working at 5 metallurgical plants in the town of Zaporozhye, have detected T. rubrum in 71.3 percent and T. interdigitale in 28.7 percent of cases. In 110 (18.3 percent) patients C. albicans were isolated from foci of involvement in parallel with Trichophyton. As a rule these patients were engaged in 'hot' shops and suffered from dysfunctions of various vital organs and from metabolic disorders. Mycoses of the soles complicated with Candida infection were characterized by marked exudation and dissemination with symptoms of eczema, i
