Epigenetic regulation of virulence and the transcription of ribosomal protein genes involves a YEATS family protein in Cryptococcus deneoformans.

Epigenetic marks or post-translational modifications on histones have important regulatory roles in gene expression in eukaryotic organisms. The epigenetic regulation of gene expression in the pathogenic yeast Cryptococcus deneoformans remains largely undetermined. The YEATS domain proteins are readers of crotonylated lysine residues in histones. Here, we reported the identification of a single-copy gene putatively coding for a YEATS domain protein (Yst1) in C. deneoformans. To define its function, we created a mutant strain, yst1Δ, using CRISPR-Cas9 editing. yst1Δ exhibited defects in phenoty

An O-GlcNAcylomic Approach Reveals ACLY as a Potential Target in Sepsis in the Young Rat.

Sepsis in the young population, which is particularly at risk, is rarely studied. O-GlcNAcylation is a post-translational modification involved in cell survival, stress response and metabolic regulation. O-GlcNAc stimulation is beneficial in adult septic rats. This modification is physiologically higher in the young rat, potentially limiting the therapeutic potential of O-GlcNAc stimulation in young septic rats. The aim is to evaluate whether O-GlcNAc stimulation can improve sepsis outcome in young rats. Endotoxemic challenge was induced in 28-day-old rats by lipopolysaccharide injection (E. C

Precision Protein Modification via Ligation and Bioconjugation at Selenocysteine.

Selenocysteine (Sec), often referred to as the 21st amino acid, is a rare yet essential amino acid found in a small number of naturally occurring selenoproteins that play critical roles in redox regulation, antioxidant defence, and hormone metabolism. Owing to its distinct chemical properties, namely enhanced nucleophilicity, lower reduction potential, and versatile redox behaviour of the side chain selenol/diselenide moiety relative to the thiol/disulfide of Cys, Sec has recently been leveraged for a range of applications in protein science. In this account, we summarize our endeavors towards

Chronic treatment of C6 glioma cells with antidepressant drugs results in a redistribution of Gsalpha.

Previous studies have demonstrated that chronic treatment of C6 glioma cells with the antidepressants desipramine and fluoxetine increases the Triton X-100 solubility of the G protein Gsalpha (Toki et al., 1999). The antidepressants also caused a 50% decrease in the amount of Gsalpha localized to caveolae-enriched membrane domains. In this study, laser scanning confocal microscopy reveals that Gsalpha is localized to the plasma membrane as well as the cytosol in both treated and control cells. However, striking differences are seen in the distribution of Gsalpha in the long cellular processes

TFAM Lys76 2-Hydroxyisobutyrylation Modulates Oxidative Stress and Energy Metabolism via the TFAM-ROS-AMPK Axis in Postoperative Atrial Fibrillation.

Postoperative atrial fibrillation (POAF) is strongly linked to mitochondrial oxidative stress, yet the upstream post-translational mechanisms that couple redox imbalance to atrial vulnerability remain unclear. We investigated atrial lysine 2-hydroxyisobutyrylation (Khib) remodeling and tested whether site-specific Khib regulates mitochondrial energy-redox signaling in POAF. Quantitative Khib proteomics was performed in right atrial appendage tissue from coronary artery bypass grafting patients. Differentially modified proteins were analyzed by functional enrichment and protein-interaction netw

Hydrogen Gas Enhances Salinity Tolerance in Tomato Seedlings by Regulating the S-Nitrosylation of MEK1.

Hydrogen gas (H2) effectively alleviates abiotic stress in horticultural plants. Protein S-nitrosylation, a key post-translational modification, serves as a critical mechanism for nitric oxide (NO) to exert its biological function under adverse conditions. However, the relationship among H2, NO and S-nitrosylation in response to salt stress remains poorly understood. In this study, we demonstrate that NO participated in H2-enhanced salt tolerance in tomato seedlings (Solanum lycopersicum cv. Micro-Tom). H2 triggered an increase in NO levels and S-nitrosothiol (SNO) content under salt stress, e

Mass-Ratio-Controlled Organ-Selective Phosphatidyl Polymer Carrier for In Vivo Targeted mRNA Delivery.

Organ-selective mRNA transfection enables precise regulation of gene expression in specific tissues and represents a pivotal strategy for advancing mRNA therapeutics toward multiorgan and multi-indication applications. However, prevailing delivery systems rely on tissue-specific ligands or polyplex chemical modifications, limiting the modularity, scalability, and clinical translatability of delivery systems. Herein, we report a mass-ratio-controlled organ-selective (MACO) mRNA delivery platform based on phosphatidyl polyethylenimine derivatives (PEI-PPs). Impressively, the MACO platform enable

The Measurement of Reversible Redox Dependent Post-translational Modifications and Their Regulation of Mitochondrial and Skeletal Muscle Function.

Mitochondrial oxidative stress is a common feature of skeletal myopathies across multiple conditions; however, the mechanism by which it contributes to skeletal muscle dysfunction remains controversial. Oxidative damage to proteins, lipids, and DNA has received the most attention, yet an important role for reversible redox post-translational modifications (PTMs) in pathophysiology is emerging. The possibility that these PTMs can exert dynamic control of muscle function implicates them as a mechanism contributing to skeletal muscle dysfunction in chronic disease. Herein, we discuss the signific

Modification of SV40 T antigen by poly ADP-ribosylation.

The SV40 large T antigen is a multifunctional protein presumed to represent a single translation product of the early viral genes. A wide range of biological controls including the regulation of viral DNA replication, transcription and cell transformation has been attributed to T antigen. Previous evidence has indicated that large T antigen is modified in at least two ways, N-terminal acetylation and amino acid phosphorylation. In this study, we demonstrate a novel modification of a population of SV40 T antigen molecules by poly ADP-ribosylation. The covalent linkage of this oligonucleotide si

β-Hydroxybutyrate upregulates hepatic histone β-hydroxybutyrylation modification, promotes the expression of PPARα, and alleviates the hepatic steatosis in MASLD.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) stands as the most widespread chronic liver disorder globally. Histone β-hydroxybutyrylation (Kbhb)-a novel post-translational modification of histones driven by β-hydroxybutyrate (BHB)-has recently been recognized as a key epigenetic modulator. Our study aimed to explore how BHB influences the expression of hepatic lipid metabolism-associated genes in MASLD, and to determine whether histone Kbhb acts as the mechanistic mediator underlying these effects. METHODS: For in vivo experiments, db/db mice fed a high-fat die

Identification of ubiquitination-related hub genes in chronic myeloid leukemia cell by bioinformatics analysis.

Purpose: Chronic myeloid leukemia stem cells (CML-LSCs) are posited as the primary instigators of resistance to tyrosine kinase inhibitors (TKIs) and recurrence of CML. Ubiquitination, a post-translational modification, has been implicated in the worsening process of CML. A more detailed understanding of their crosstalk needs further investigation. Our research aims to explore the potential ubiquitination-related genes in CML-LSC using bioinformatics analysis that might be the target for the eradication of LSCs. Methods: The ubiquitination modification-related differentially expressed genes (U

Exercise-induced endothelial Mecp2 lactylation suppresses atherosclerosis via the Ereg/MAPK signalling pathway.

BACKGROUND AND AIMS: Lactylation, a recently identified post-translational modification (PTM), plays a central role in the regulation of multiple physiological and pathological processes. Exercise is known to provide protection against cardiovascular disease. However, whether exercise-generated lactate changes lactylation and is involved in the exercise-induced attenuation of atherosclerotic cardiovascular disease (ASCVD) remains unclear. The purpose of this study was to investigate the effects and mechanisms of exercise-induced lactylation on ASCVD. METHODS AND RESULTS: Using the high-fat di

Protein lactylation in atherosclerosis - Mechanisms and therapeutic potential: A narrative review.

Protein lactylation, which links cellular metabolism to gene regulation and protein function, may influence atherosclerosis. This narrative review explores its cellular mechanisms and therapeutic potential. We searched PubMed from inception to September 30, 2026, for English-language studies and reviews. The synthesis differentiated lactylation from lactate signaling and distinguished direct evidence in atherosclerosis from findings in other disease models. Predominantly preclinical evidence suggests that both histone and non-histone lactylation are involved in endothelial inflammation, endoth

S100A8 lactylation at K23/K36 reduces RNF5-dependent degradation and promotes colorectal cancer progression.

Colorectal cancer (CRC) progression is deeply influenced by the acidic, lactate-rich tumor microenvironment. While the calcium-binding protein S100A8 is a known oncogenic driver and prognostic biomarker in CRC, the mechanisms governing its protein stability remain incompletely understood. This study investigated the competitive interplay between lactate-induced lactylation and RNF5-mediated ubiquitination of S100A8. Reanalysis of clinical lactylomic datasets and survival analyses revealed that S100A8 is heavily lactylated in colon cancer tissues and its elevated expression correlates with poor

Glutaredoxin 1 promotes intestinal epithelial cell copper toxicity in inflammatory bowel disease.

Protein S-glutathionylation (PSSG), a redox-sensitive post-translational modification, regulates protein conformation and activity, potentially resulting in cell death. PSSG is dynamically reversed by glutaredoxin 1 (GRX1), a critical oxidoreductase. Here, we observed elevated PSSG levels in colonic tissues of ulcerative colitis (UC) patients and experimental mouse models. Quantitative redox proteomics revealed glutathione disulfide oxidoreductase activity as the most significantly altered pathway in UC patients compared to controls. Interestingly, GRX1 undergoes site-specific S-glutathionylat

PRMT2 Aggravates Pressure Overload-induced Cardiac Remodeling by Promoting Endothelial Phenotypic Transition via Snail1 Methylation.

PURPOSE: Endothelial-to-mesenchymal transition (EndMT) is implicated in cardiac remodeling under pathological stress, although its full in vivo occurrence may be context-dependent. Emerging evidence suggests that protein methylation is an important post-translational modification involved in regulating endothelial phenotypic transition (EndMT-like process). However, the role of protein arginine methyltransferase 2 (PRMT2), a key protein arginine methyltransferase, in modulating endothelial phenotypic transition-particularly in the context of cardiac remodeling-remains poorly understood. Host DHFRL1 interacts with MtbEis and increases the intracellular survival of Mycobacterium tuberculosis by inhibiting autophagy.

The incredible ability of Mycobacterium tuberculosis to orchestrate the host cell machinery is achieved by its secretory proteins. M. tuberculosisEnhanced Intracellular Survival (MtbEis) protein is one such virulence-associated secretory acetyltransferase that acetylates aminoglycoside antibiotics, arylalkylamines, mycobacterial proteins and host proteins. To reveal the unknown functions of MtbEis, we used Yeast Two-Hybrid assay to explore its interactions with host proteins. We discovered a novel interaction of MtbEis with mouse Dihydrofolate Reductase (mDHFR) protein and demonstrated that Mt

Bile acids and polyphenols inhibit succinic anhydride-induced protein succinylation and amyloid aggregation: Mechanistic insights.

Lysine succinylation is a major post-translational modification affecting diverse proteins, and its excessive occurrence can lead to protein misfolding and aggregation-hallmarks of various proteinopathies, such as amyloid-β and Tau tangle formation in Alzheimer's disease and islet amyloid polypeptide aggregation in type 2 diabetes. Here, we investigated the inhibitory effects of bile acid metabolites (deoxycholic acid, glycocholic acid, and taurocholic acid sodium) and natural polyphenols (anthocyanin and salidroside) on succinylation and succinylation-induced amyloid aggregation.
