Insulin and islet cell transplant--effect on intestinal uptake of lipids.

A previously validated in vitro technique was used to determine the effect of once-daily injections of NPH insulin (NPH) and islet cell transplant (ICT) on the enhanced uptake (Jd) of lipids into the intestine of diabetic rats. Three months after ICT, the urine volume, urine glucose, and fasting blood sugars were near normal. The Jd of lauryl alcohol was used to measure the effective resistance of the unstirred water layer (UWL); NPH was associated with an increase in UWL in the jejunum when the bulk phase was stirred or unstirred and a rise in UWL in the ileum when the bulk phase was unstirre

Effect of composite resin containing antibacterial filler on sugar-induced pH drop caused by whole saliva bacteria.

STATEMENT OF PROBLEM: Secondary caries around restorations is a major problem and can be attributed to bacteria invading microgaps formed at the tooth-restoration interface. An antibacterial composite resin containing quaternary ammonium silica (QASi) filler has been reported to inhibit enamel demineralization in situ. However, whether the prevention of enamel demineralization by QASi-containing composite resin is because of the reduced metabolic activity of acid-producing saliva bacteria is unclear. PURPOSE: The purpose of this study was to compare the effects of QASi-containing composite re

Combined application of plant growth-promoting bacteria and iron oxide nanoparticles ameliorates the toxic effects of arsenic in Ajwain (Trachyspermum ammi L.).

Soil contamination with toxic heavy metals [such as arsenic (As)] is becoming a serious global problem because of the rapid development of the social economy. Although plant growth-promoting bacteria (PGPB) and nanoparticles (NPs) are the major protectants to alleviate metal toxicity, the study of these chemicals in combination to ameliorate the toxic effects of As is limited. Therefore, the present study was conducted to investigate the combined effects of different levels of Providencia vermicola (5 ppm and 10 ppm) and iron oxide nanoparticles (FeO-NPs) (50 mg/l-1 and 100 mg/l-1) on plant gr

2-buten-4-olide, an endogenous feeding suppressant, improves spatial performance through brain acidic fibroblast growth factor in mice.

Endogenous sugar acid 2-buten-4-olide, a satiety substance, has been shown to increase the blood glucose, norepinephrine, and glucocorticoid concentrations that are known to modulate learning and memory processes. The glucose-induced release of acidic fibroblast growth factor facilitated the hippocampus-dependent memory function. In the present study, we investigated the effect of 2-buten-4-olide on the spatial performance of male DDY mice undergoing the water maze task. The intraperitoneal injection of 2-buten-4-olide (5 mg/kg) facilitated the spatial performance, which was indicated by a red

Techniques for the recovery and identification of Cryptosporidium oocysts from stool specimens.

Due to increasing numbers of patients with documented infections with Cryptosporidium and other coccidia, it is important for the physician and clinical laboratory to be aware of the appropriate diagnostic techniques necessary for organism recovery and identification. Although Cryptosporidium is found in the gastrointestinal tract, tissue biopsies may be insufficient for organism recovery; the examination of stool specimens is a noninvasive procedure and will provide better overall opportunities for organism recovery. Human clinical specimens were examined from 45 patients with confirmed crypt

Lanthanoid tagging via an unnatural amino acid for protein structure characterization.

Lanthanoid pseudo-contact shift (PCS) provides long-range structural information between a paramagnetic tag and protein nuclei. However, for proteins with native cysteines, site-specific attachment may only utilize functional groups orthogonal to sulfhydryl chemistry. Here we report two lanthanoid probes, DTTA-C3-yne and DTTA-C4-yne, which can be conjugated to an unnatural amino acid pAzF in the target protein via azide-alkyne cycloaddition. Demonstrated with ubiquitin and cysteine-containing enzyme EIIB, we show that large PCSs of distinct profiles can be generated for each tag/lanthanoid com

An inositol receptor Itr4 coordinates carbon catabolite regulation and virulence in Cryptococcus neoformans.

Nutrient sensing and utilization are critical for microbes to adapt to their environment and evade predators. The encapsuled yeast Cryptococcus neoformans has a unique ability to sense and utilize the sugar inositol as both a signaling molecule and as a carbon source. This trait is advantageous not only in the natural environment but also to promote its pathogenesis, including invasion of the central nervous system. In this study, we have identified a novel inositol transporter-like receptor (transceptor) Itr4 that coordinates inositol and glucose metabolism by regulating inositol transporters

Study on the cutting technology of chayote (Sechium edule) and the physiological mechanism of rooting regulation by exogenous hormones.

BACKGROUND: 'Tuershao' is a new chayote (Sechium edule) variety whose tuberous roots and tender shoots are edible. These parts are rich in beneficial substances, including amino acids, proteins, phenolic compounds, flavonoids, cucurbitacin, and selenium, demonstrating their unique value. However, its ovary cannot develop into a mature fruit, and cutting techniques help to achieve the breeding of 'Tuershao'. RESULTS: This study found that using peat soil as the substrate and treating semi-hard branches with 1.0 g·L⁻¹ indole-3-butyric acid (IBA) achieved the highest rooting rate of 94.33%, with

Hanseniaspora species can create a privileged niche for Oenococcus oeni during triple-strain simultaneous alcoholic-malolactic fermentation based on metabolomics analysis.

Previous studies have demonstrated that non-Saccharomyces yeasts can enhance malolactic fermentation (MLF) efficiency during co-fermentation. However, the underlying mechanisms remain unclear. Here, we demonstrate how specific strains create favorable metabolic niches through tripartite fermentation assays involving 16 strains from 8 species co-cultured with Saccharomyces cerevisiae F5 and Oenococcus oeni SD-2a, Hanseniaspora spp. emerged as optimal partners, reducing MLF duration by 50 % (≤48 h vs. 96 h in controls) while increasing O. oeni biomass by > 10-fold. Surface Charge Heterogeneity and Mechanisms of Organic Binding Modes on an Iron Oxyhydroxide.

Mineral-associated organic matter involving iron oxyhydroxide minerals is important to the preservation and transformation of organic matter in soils and sediments. Largely lacking is a quantitative evaluation of different binding mechanisms in relation to the mineral surface charges. Here, with ferrihydrite, we investigated complexes with organic compounds of various charges and structures, including a ribonucleotide, a sugar, a phenolic acid, and amino acids with different side chains. After constructing model ferrihydrite nanoparticles using reported iron-oxygen coordination, we mapped theo

Finding the Sweet Spot for the Treatment of B Cell Malignancies.

The glycan profile of cells comprises a high variety of sugar moieties that are attached to proteins (glycoproteins) and lipids (glycolipids) via a process called 'glycosylation'. Cancer cells commonly carry aberrant glycans, which may be of interest for cancer diagnosis, prognosis, as well as the development of novel therapeutic strategies. This review focuses on the differential glycosylation patterns on malignant B cells, including both B cell lymphoma and leukemia. Well-known aberrant glycan profiles on malignant B cells include acquired high mannose N-glycans in the B cell receptor (BCR)

Four new resin glycosides from Ipomoea muricata seeds: muricatins XIV-XVII.

Ipomoea muricata (L.) Jacq. seeds (Convolvulaceae) are used as a traditional laxative and carminative medicine. Muricatins XIV (1), XV (2), XVI (3), and XVII (4), were isolated from I. muricata seeds as four new resin glycosides, along with seven known compounds, three of which were isolated for the first time as natural products; their structures were determined using MS and NMR spectroscopy. Compounds 1-4 are macrolactones (jalapins); the sugar moieties of 1, 2, and 4 are partially acylated with 2S-methylbutyric acid, while that of 3 is esterified with 2S-methylbutyric and 2S-methyl-3S-hydro
