White matter correlates of cognitive flexibility in youth with bipolar disorder and typically developing children and adolescents.

Prior studies using behavioral tasks and neuroimaging have shown that children and adolescents with bipolar disorder (BD) have deficits in cognitive flexibility (CF)-defined as adaptation to changing rewards and punishments. However, no study, to our knowledge, has examined the white matter microstructural correlates of CF in youth with BD. To address this gap, we examined the relationship between CF assessed with the Cambridge Neuropsychological Testing Automated Battery (CANTAB)'s Intra-Extra Dimensional Set Shift task (ID/ED) and diffusion tensor imaging analyzed with FSL's preprocessing to

Solving the Credit Assignment Problem With the Prefrontal Cortex.

In naturalistic multi-cue and multi-step learning tasks, where outcomes of behavior are delayed in time, discovering which choices are responsible for rewards can present a challenge, known as the credit assignment problem. In this review, I summarize recent work that highlighted a critical role for the prefrontal cortex (PFC) in assigning credit where it is due in tasks where only a few of the multitude of cues or choices are relevant to the final outcome of behavior. Collectively, these investigations have provided compelling support for specialized roles of the orbitofrontal (OFC), anterior

Implicit Valuation of the Near-Miss is Dependent on Outcome Context.

Gambling studies have described a "near-miss effect" wherein the experience of almost winning increases gambling persistence. The near-miss has been proposed to inflate the value of preceding actions through its perceptual similarity to wins. We demonstrate here, however, that it acts as a conditioned stimulus to positively or negatively influence valuation, dependent on reward expectation and cognitive engagement. When subjects are asked to choose between two simulated slot machines, near-misses increase valuation of machines with a low payout rate, whereas they decrease valuation of high pay

Impaired anticipatory vision and visuomotor coordination affects action planning and execution in children with hemiplegic cerebral palsy.

BACKGROUND: Action-planning and execution deficits in children with hemiplegic cerebral palsy (HCP) are potentially due to deficits in the integration of sensory information, such as vision, with motor output. AIMS: To determine differences in anticipatory visual patterns in children with HCP compared to typically developing (TD) children, and to assess visuomotor coordination in children with HCP. METHODS AND PROCEDURES: We included 13 children with HCP (Age = 6.8 + 2.9 yrs) and 15 TD children (Age = 5.8 + 1.1 yrs). The experimental task used in this study is a valid action-planning task, w

Inactivation of the orbitofrontal cortex reduces irrational choice on a rodent Betting Task.

Cognitive biases may play a significant role in disorders of decision making such as pathological gambling and addiction. Understanding the neurobiology of these biases could lead to more effective pharmacological and therapeutic treatments for disorders in which aberrant decision making is prominent. The rodent Betting Task (rBT) was designed to measure one commonly observed decision-making heuristic in rodents, namely "escalation of commitment" in which subjects become more risk averse as the stakes increase, even if the odds of success remain constant. In the rodent task, the animal is pres

Non-linear effects of transcranial direct current stimulation as a function of individual baseline performance: Evidence from biparietal tDCS influence on lateralized attention bias.

Transcranial direct current stimulation (tDCS) is a well-established technique for non-invasive brain stimulation (NIBS). However, the technique suffers from a high variability in outcome, some of which is likely explained by the state of the brain at tDCS-delivery but for which explanatory, mechanistic models are lacking. Here, we tested the effects of bi-parietal tDCS on perceptual line bisection as a function of tDCS current strength (1 mA vs 2 mA) and individual baseline discrimination sensitivity (a measure associated with intrinsic uncertainty/signal-to-noise balance). Our main findings

High-resolution mass spectrometry-based background subtraction for identifying protein modifications in a complex biological system: detection of acetaminophen-bound microsomal proteins including argininosuccinate synthetase.

The detection and characterization of low-level protein modifications in a complex system without a methodology for modification enrichment is a very challenging task. This study describes a high-resolution LC/MS-based background subtraction methodology for the unbiased detection and identification of acetaminophen-bound proteins formed in incubations with mouse liver microsomes. The microsomal incubations were conducted using both acetaminophen and [(13)C2,(15)N]acetaminophen at a drug concentration of 200 μM. After tryptic digestion and high-resolution LC/MS analysis, data from the two drug

The neural representation of sensorimotor transformations in a human perceptual decision making network.

Humans can quickly engage a neural network to transform complex visual stimuli into a motor response. Activity from a key region within this network, the intraparietal sulcus (IPS), has been associated with evidence accumulation and motor planning, thus implicating it in sensorimotor transformations. If such transformations occur within a brain region, a key and untested prediction is that neural activity reflecting both the parametric amount of evidence available and the timing of motor planning can be independently manipulated. To investigate these ideas, we constructed a dot motion discrimi

A developmental shift from positive to negative connectivity in human amygdala-prefrontal circuitry.

Recent human imaging and animal studies highlight the importance of frontoamygdala circuitry in the regulation of emotional behavior and its disruption in anxiety-related disorders. Although tracing studies have suggested changes in amygdala-cortical connectivity through the adolescent period in rodents, less is known about the reciprocal connections within this circuitry across human development, when these circuits are being fine-tuned and substantial changes in emotional control are observed. The present study examined developmental changes in amygdala-prefrontal circuitry across the ages o

Intermanual interactions during initiation and production of rhythmic and discrete movements in individuals lacking a corpus callosum.

Three individuals lacking a corpus callosum, two due to callosotomy and one agenesis, and three age-matched healthy controls were tested on a bimanual task in which a discrete or rhythmic arm movement was initiated following a visual signal while the other arm produced continuous, rhythmic movements. The control participants initiated the secondary, rhythmic movement in phase with the ongoing rhythmic base movement and the two limbs were coupled in an inphase mode across the duration of the trial. In contrast, the acallosal individuals failed to show phase entrainment at the initiation of the

1 hz rTMS over the right prefrontal cortex reduces vigilant attention to unmasked but not to masked fearful faces.

BACKGROUND: Recent repetitive transcranial magnetic stimulation (rTMS) research in healthy subjects suggests that the emotions anger and anxiety are lateralized in the prefrontal cortex. Low-frequency rTMS over the right prefrontal cortex (PFC) shifts the anterior asymmetry in brain activation to the left hemisphere and reduces anxiety. The same rTMS technique results in enhanced anger-related emotional processing, observed as elevations in attention for angry faces. The current study used low-frequency rTMS over the right PFC and indexed selective attention to fearful faces, hypothesizing a r

Stressful conditions of work: the participative ergonomics approach.

The nature of working stress requires a multi-disciplinary approach for its study. Ergonomics, by its nature multidisciplinary, takes a perspective in which the individual is central and the work and working environment represent the influences to be adjusted. This perspective considers human capacities to have a permissible range of values, within which work demands must be matched. Work demands are more than the physical or mental loads directly related to task performance, since the organisational structure imposes loads. These arise, inter alia, from the relative match between the organisa

CLI-PPO: A Memory-Augmented PPO Framework with Intrinsic Curiosity and Curriculum Learning for Mapless Multi-Obstacle Navigation.

Mapless autonomous navigation in unknown multi-obstacle environments remains challenging for deep reinforcement learning due to limited perception, sparse rewards, and distribution shifts between training and deployment scenarios. Existing methods mainly rely on instantaneous observations or individual enhancement strategies, which often struggle to balance exploration robustness and navigation efficiency. This paper proposes a Curriculum Learning and Intrinsic Curiosity-enhanced Temporal Proximal Policy Optimization framework, referred to as CLI-PPO, for end-to-end autonomous navigation. The

Neuromuscular and behavioural evidence of motor control adaptations following 8 weeks of virtual reality exergaming.

PURPOSE: Virtual reality (VR) exergaming was shown to enhance cognitive performance, yet its effects on neuromuscular function remain unclear. This study investigated whether chronic VR exergaming induces adaptations in neuromuscular coordination and speed-accuracy regulation. METHOD: Twenty-eight healthy young adults (22.8 ± 1.2 years; 9 women) completed an 8-week training programme (two 30-min sessions/week) consisting of either immersive VR exergaming (VR game with rapid and targeted, goal-directed upper-limb movements) or non-immersive video-guided coordination training with Arnis Stick (

Non-invasive somatotopic proprioceptive feedback for closed-loop robotic hand aperture control.

Objective.Current robotic prostheses developed for individuals with transradial amputation often lack physiological feedback, particularly proprioceptive information, which limits control precision and increases reliance on vision.Approach.This study investigates the effect of introducing non-invasive somatotopic feedback using transcutaneous electrical nerve stimulation (TENS) to convey hand aperture information, as an artificial form of proprioceptive feedback. Twenty healthy participants with intact limbs were divided into two groups: one receiving TENS-based feedback and one without feedba

A direction aware predictive offloading framework for energy conscious mobile augmented reality systems.

Mobile Augmented Reality (MAR) applications require efficient and quick real-time object recognition with minimal latency in order to maintain user immersion. However, these applications are severely affected by energy constraints and dynamic user movement. Conventional offloading models often neglecting direction-based user behavior incur unnecessary processing and communication overhead which further leads to inefficient resource utilization and degraded responsiveness. Methods based on reactive task scheduling fail to predict user motion which leads to delayed execution and increased energy

Counterfactual Diffusion Models Provide Interpretable Explanations of Artificial Intelligence Models in Pathology.

UNLABELLED: Deep learning can extract predictive and prognostic biomarkers from histopathology whole-slide images. However, explainable artificial intelligence approaches widely used in digital pathology, such as attention heatmaps and class activation mapping, provide limited insight into the image features associated with classifier outputs. In this study, we developed Morphing histoPathology Diffusion (MoPaDi), a framework for generating counterfactual explanations for histopathology images that help identify morphologic or stain-related features linked to model predictions. MoPaDi combined

Effects of hormonal contraceptives on spatial navigation in gonadally intact female rats.

Rats can either use place memory, response memory or rely on a single cue to navigate their environment. Which navigation strategy a female rat is biased towards is influenced by circulating levels of ovarian hormones. Hormonal contraceptive use is typically associated with changes in circulating levels of endogenous ovarian hormones. The aim of the current study was to examine the effects of synthetic hormones used in hormonal contraceptives on navigation strategies. A total of 61 gonadally intact female rats were tested in two Morris water maze tasks. Rats were administered ethinyl estradiol

Reasoning decline during aging under familiar and unfamiliar physics.

Physical reasoning is the capacity to anticipate how an environment will change as its elements move and interact. This cognitive skill, which is based on humans' intuitive knowledge of physics, underlies everyday tasks that are potentially critical to older adults, such as avoiding collisions. Nevertheless, the effects of aging on physical reasoning remain understudied. Here, we tested physical reasoning among younger (18-35 years) and older (over 65 years) adults as they completed different difficulty levels of a physical reasoning paradigm. Participants viewed object displacements in a virt
