In the Vavoua human trypanosomiasis focus (forest zone of Ivory Coast) four tsetse fly catching series, of nine days each, were made during the rainy season using eighty five biconical traps. The dispersion of flies in the study area was studied by the mark-release-recapture method. Two series of two sets of capture-mark-release were carried out. In the first set flies were captured, marked and released in their point of origin and in the second set captured and marked in the village periphery but released in coffee plantations and vice versa. A total of 10198 tsetse flies were marked and rele…

Acoziborole is an oxaborole 6-carboxamide active against Trypanosoma brucei gambiense and rhodesiense, the parasites responsible for human African trypanosomiasis. This open-label, phase I study in six healthy male participants evaluated the mass balance, pharmacokinetics, metabolism pathways, and excretion of a single oral 960 mg dose of [14C]-acoziborole. Blood, plasma, and feces were collected for 120 days and urine for 16 days. The excretion balance and systemic exposure of total circulating radioactivity were determined using accelerator mass spectrometry. Metabolism profiling was perform…

Climate variability, particularly global warming, is expected to have a significant impact on the interactions between pathogens, vectors, and animal and human hosts. Trypanosomiasis, a neglected zoonotic disease, is particularly sensitive to changes in climate, as increased temperatures can accelerate vector development and expand geographical range. This disease remains prevalent in Zimbabwe, with the Binga District being disproportionately affected. This study aimed to explore the effects of climate variability on the epidemiology of trypanosomiasis in the Binga District of Zimbabwe from 20…

BACKGROUND: Passive screening of gambiense human African trypanosomiasis (HAT) is based on rapid diagnostic tests (RDT), but sensitivity of the currently commercialised RDTs has hardly been assessed prospectively. In view of the increasing importance of remote testing for HAT, the diagnostic performance of reference laboratory tests also needs further documentation.
METHODOLOGY/PRINCIPAL FINDINGS: The study is registered in ClinicalTrials.Gov under identifier NCT03356665. Clinical suspects in 29 health facilities in DR Congo were screened consecutively between October 2017 and December 2020 w…

We conducted a retrospective study on mortality trends and risk factors in 781 naïve cases of advanced stage-2 sleeping sickness admitted between 1989 and 2012 at the National Reference Center for Human African Trypanosomiasis (HAT), Department of Neurology, Kinshasa University, Democratic Republic of Congo (DRC). Death was the outcome variable whereas age, gender, duration of disease, location of trypanosomes in body fluids, cytorachy, protidorachy, clinical status (assessed on a syndromic and functional basis) on admission, and treatment regimen were predictors in logistic regression models …

The extraction of rough antigens (Ag) from trypanosomes came across a major problem getting of a high number of parasites. Moreover several … cted Cricetomys. In order to set up a specific and sensitive serological diagnosis of gambian human trypanosomiasis, the different rough Ag were used comparatively by ELISA and Latex Agglutination tests. Confirming previous works, the results indicate a high sensitivity and a doubtful specificity for both techniques.

The Cellognost indirect haemagglutination (IHA) test was compared with conventional techniques, namely, wet blood film examination, stained thin and thick films, animal inoculation and the microhaematocrit centrifugation, for detecting sleeping sickness cases. Sixty-four out of 245 serum samples collected from suspected and some diagnosed cases of the disease were positive by the Cellognost screening test, while only three cases were detected by stained thin film or microhaematocrit centrifugation. Of the 64 cases 45 strongly positive samples were subjected to quantitative assay. Twenty-five o

Cattle were vaccinated with Brucella abortus (S19) vaccine during acute (25 days) and chronic (25 weeks) Trypanosoma congolense and chronic Trypanosoma vivax (25 weeks) infections in order to determine the effect of such infections on the antibody response to the vaccine. It was found that the specific antibody responses of IgG1 and IgG2 sub-classes were profoundly depressed (80%) in both the acute and chronic infections with T. congolense. Whereas IgM antibody response was also profoundly depressed (90%) in cattle with the acute infection, it was only 50% depressed in those with chronic infec

Infections of highly susceptible BALB/c mice with virulent strains of Trypanosoma congolense or Trypanosoma brucei result in rapid death (8 days). We have previously shown that this mortality is IFN-gamma dependent. In this study we show that IFN-gamma is produced predominantly by CD3+Thy1.2+TCRbeta+CD4+ T cells shortly before the death of infected mice. Mortality may therefore be dependent on IFN-gamma-producing CD4+ T cells. Surprisingly, infected CD4+/+ and CD4-/- BALB/c mice have similar parasitemia and survival time. In infected CD4-/- mice, the production of both IFN-gamma and IL-10 is v

BACKGROUND: Animal African trypanosomiasis (AAT) poses a significant impediment to livestock production and economic progress in sub-Saharan Africa. To reduce the burden of AAT in Uganda and contribute to the AAT progressive control pathway, there is a need to develop effective AAT and tsetse control measures. In this study, we assessed a combination of chemotherapy and Restricted Insecticide Application Protocol (RAP) as a last mile control option for the progressive control of AAT and acute sleeping sickness in south- eastern Uganda.
METHODS: Cattle from fourteen AAT endemic villages in sou…

Trypanosoma carassii, a typical freshwater fish trypanosome, has recently been identified as the etiological agent of a trypanosomiasis outbreak in cage-cultured large yellow croaker (Larimichthys crocea) in China and has been designated as T. c. larimichthys. To date, publicly available genomic data for trypanosomes have been limited to terrestrial species, particularly those of medical importance. Here, we present a chromosome-level genome assembly of T. carassii, the first genome of an aquatic trypanosome, generated using PacBio HiFi long-read sequencing and Hi-C scaffolding technologies. A…

BACKGROUND: In previous clinical trials, oral fexinidazole treatment showed a favourable safety profile, while being easily administered and effective for most adult and paediatric patients with gambiense human African trypanosomiasis. The aim of this open-label cohort study was to investigate the effectiveness and safety of fexinidazole in a wider population of patients and to explore the feasibility of treating patients with gambiense human African trypanosomiasis outside the hospital setting.
METHODS: In this phase 3b, prospective, open-label, non-randomised, cohort study, fexinidazole was…

OBJECTIVE: To enable the human African trypanosomiasis (HAT) control program of the Democratic Republic of the Congo to generate data on treatment outcomes, an electronic database was developed. The database was piloted in two provinces, Bandundu and Kasai Oriental. In this study, we analysed routine data from the two provinces for the period 2006-2008.
METHODS: Data were extracted from case declaration cards and monthly reports available at national and provincial HAT coordination units and entered into the database.
RESULTS: Data were retrieved for 15 086 of 15 741 cases reported in the tw…

The authors report results obtained after combination of a serological diagnostic test and the early treatment of suspected cases (a person with a positive serological test without parasitological confirmation) in an area in Zaire where sleeping sickness caused by Trypanosoma brucei gambiense is endemic. The serological test used was Testryp CATT, having a very high sensitivity and quite a high specificity, which is easy to handle in the field and which permits results to be obtained on the spot. The treatment employed was a medication active in the first stage of the disease (hematolymphatic

A slide flocculation test for Chagas' disease is described, which uses a lyophilized, stable antigen obtained by formalin and ultrasonic treatment of culture forms of Trypanosoma cruzi. The test was compared with other tests for the serodiagnosis of American trypanosomiasis and showed a high sensitivity, positive results being obtained in every case of acute or chronic Chagas' disease. In sera from blood donors and from normal individuals with negative T. cruzi antigen complement fixation tests a specificity of 96% was found. False positive flocculation tests were seen, especially in cases of …

BACKGROUND: Human African trypanosomiasis (HAT) is a fatal vector-borne disease caused by Trypanosoma brucei. Although HAT incidence has declined, meeting WHO's elimination targets remain difficult, particularly due to limited diagnostic sensitivity for low-parasite load-infections. Arboviruses such as dengue (DENV 1-4), chikungunya (CHIKV), and yellow fever (YFV) virus, present with nonspecific febrile symptoms similar to HAT and are underdiagnosed in Sub-Saharan Africa. Due to this overlap in symptoms and a suspected geographical overlap of vectors and pathogens in the Democratic Republic of…
