Thiamin biofortification of crops.

Thiamin is essential for human health. While plants are the ultimate source of thiamin in most human diets, staple foods like white rice have low thiamin content. Therefore, populations whose diets are mainly based on low-thiamin staple crops suffer from thiamin deficiency. Biofortification of rice grain by engineering the thiamin biosynthesis pathway has recently been attempted, with up to 5-fold increase in thiamin content in unpolished seeds. However, polished seeds that retain only the starchy endosperm had similar thiamin content than that of non-engineered plants. Various factors such as

Biofortification of chromium in fenugreek seeds.

BACKGROUND: Fenugreek and chromium are known to have anti-diabetic properties and this has been well demonstrated by multiple studies. Researches have been undertaken to study thebiofortification of chromium (Cr3+) in fenugreek. Some of the researchers have studied the biofortification from the soil irrigated with tannery waste water or from soil amended by tannery-sludge, with a view of enhancing the anti-diabetic effect of fenugreek plants. The present research work was also undertaken to increase the chromium content of fenugreek seeds, but through direct treatment of chromium picolinate so

Folate biofortification in food crops.

Folates are essential vitamins in the human diet. Folate deficiency is still very common, provoking disorders such as birth defects and anemia. Biofortification via metabolic engineering is a proven powerful means to alleviate folate malnutrition. A variety of metabolic engineering approaches have been successfully implemented in different crops and tissues. Furthermore, ensuring folate stability is crucial for long-term storage of crop products. However, the current strategies, shown to be successful in rice and tomato, will need to be fine-tuned to enable adequate biofortification of other s

Carotenoid Biofortification of Crops in the CRISPR Era.

Carotenoids are micronutrients important for human health. The continuous improvements in clustered regularly interspaced short palindromic repeats (CRISPR)-based genome-editing techniques make rapid, DNA/transgene-free and targeted multiplex genetic modification a reality, thus promising to accelerate the breeding and generation of 'golden' staple crops. We discuss here the progress and future prospects of CRISPR/Cas9 applications for carotenoid biofortification.

Combined biofortification of carrot with iodine and selenium.

The aim of a three-year study was to assess the effect of combined biofortification with I and Se in carrot. Four cultivars ('Askona' F1, 'Samba' F1, 'Kazan' F1 and 'White Satin') were grown in soil fertilized with KI (4 kg I ha-1) and Na2SeO4 (0.25 kg Se ha-1). The I + Se fertilization did not affect yield but the plants of all cultivars accumulated both elements in leaves and roots. On average, the I and Se contents in roots increased 7.7-times for I and 4.9-times for Se as well as the average I:Se molar ratio was 0.28:1. The contents of both elements in roots remained well below the hazard

Potential impacts of iron biofortification in India.

Iron deficiency is a widespread nutrition and health problem in developing countries, causing impairments in physical activity and cognitive development, as well as maternal mortality. Although food fortification and supplementation programmes have been effective in some countries, their overall success remains limited. Biofortification, that is, breeding food crops for higher micronutrient content, is a relatively new approach, which has been gaining international attention recently. We propose a methodology for ex ante impact assessment of iron biofortification, building on a disability-adju

Prospects for Carotenoid Biofortification Targeting Retention and Catabolism.

Due to the ongoing prevalence of vitamin A deficiency (VAD) in developing countries there has been a large effort towards increasing the carotenoid content of staple foods via biofortification. Common strategies used for carotenoid biofortification include altering flux through the biosynthesis pathway to direct synthesis to a specific product, generally β-carotene, or via increasing the expression of genes early in the carotenoid biosynthesis pathway. Recently, carotenoid biofortification strategies are turning towards increasing the retention of carotenoids in plant tissues either via alteri

Carotenoid biofortification in crop plants: citius, altius, fortius.

Carotenoids are indispensable for human health, required as precursors of vitamin A and efficient antioxidants. However, these plant pigments that play a vital role in photosynthesis are represented at insufficient levels in edible parts of several crops, which creates a need for increasing their content or optimizing their composition through biofortification. In particular, vitamin A deficiency, a severe health problem affecting the lives of millions in developing countries, has triggered the development of a series of high-provitamin A crops, including Golden Rice as the best-known example.

Folate biofortification strategy by elicitation in kale baby leaves.

INTRODUCTION: Humans cannot synthesize folates, so they must be acquired through dietary sources, with plants serving as a primary source. To enhance folate levels in vegetables, this study investigated the pre-harvest application of elicitors on kale ('Cavolo Nero') baby leaves under greenhouse-autumn conditions in Southern Italy. METHODS: Phenylalanine (0.37 µM) and salicylic acid (250 µM) were applied three times and compared to a water control. Folate content, alongside biometric traits (yield, leaf area and number, dry matter), bio-physiological parameters (chlorophyll content; measureme

Pathway Editing Targets for Thiamine Biofortification in Rice Grains.

Thiamine deficiency is common in populations consuming polished rice as a major source of carbohydrates. Thiamine is required to synthesize thiamine pyrophosphate (TPP), an essential cofactor of enzymes of central metabolism. Its biosynthesis pathway has been partially elucidated and the effect of overexpression of a few genes such as thi1 and thiC, on thiamine accumulation in rice has been reported. Based on current knowledge, this review focuses on the potential of gene editing in metabolic engineering of thiamine biosynthesis pathway to improve thiamine in rice grains. Candidate genes, suit

Biofortification of Sprouts and Microgreens with Trace Elements and Calcium.

A large proportion of the world's population struggles to meet the body's requirements for certain minerals. It raises interest in methods of enhancing their levels in food raw materials. Sprouts and microgreens are highly promising raw materials for biofortification. Mineral accumulation in these raw materials may be influenced by genetic factors, as well as by concentration, form and method of mineral application, light conditions, and the plant growth period. In some cases, studies have reported several dozen-fold increases in the content of the applied mineral compared with control samples

The Role of Sulfur in Agronomic Biofortification with Essential Micronutrients.

Sulfur (S) is an essential macronutrient for plants, being necessary for their growth and metabolism and exhibiting diverse roles throughout their life cycles. Inside the plant body, S is present either in one of its inorganic forms or incorporated in an organic compound. Moreover, organic S compounds may contain S in its reduced or oxidized form. Among others, S plays roles in maintaining the homeostasis of essential micronutrients, e.g., iron (Fe), copper (Cu), zinc (Zn), and manganese (Mn). One of the most well-known connections is homeostasis between S and Fe, mainly in terms of the role o

Present and future of folate biofortification of crop plants.

Improving nutritional health is one of the major socio-economic challenges of the 21st century, especially with the continuously growing and ageing world population. Folate deficiency is an important and underestimated problem of micronutrient malnutrition affecting billions of people worldwide. More and more countries are adapting policies to fight folate deficiency, mostly by fortifying foods with folic acid. However, there is growing concern about this practice, calling for alternative or complementary strategies. In addition, fortification programmes are often inaccessible to remote and po

Maize Biofortification Approaches for Fighting Malnutrition in Sub-Saharan Africa.

Maize is a staple food in sub-Saharan Africa (SSA); however, commonly used maize varieties are deficient in essential nutrients, including lysine, tryptophan, vitamin A, and minerals such as iron (Fe) and zinc (Zn). As a result, millions of people in this region suffer from one or more nutrient deficiencies that commonly lead to malnutrition-related diseases. Especially children and nursing mothers are seriously affected. Over 32% of children under 5 years of age are stunted, and 37% of women aged 15-49 years are affected by anemia in SSA. These indicate that the severity of malnutrition remai

Accelerating iron biofortification in millets: progress, challenges, and future prospects.

Integrating conventional breeding, omics, and CRISPR-based genome editing can overcome genetic and antinutrient constraints, enabling efficient iron biofortification of millets for sustainable and nutrition-secure food systems. Iron (Fe) deficiency remains one of the most widespread forms of micronutrient malnutrition. Biofortification of staple crops has emerged as a particularly sustainable and scalable strategy to combat this issue. Millets are nutrient-dense staple cereals with exceptional nutritional quality and climate resilience. However, genetic variations and the presence of antinutri

Iodine Fortification of Edible Legume Sprouts: A Pilot Biofortification Study.

Iodine has an essential role in the human body; however, its insufficiency is still a challenge. Therefore, the search for new strategies to increase iodine intake in the daily diet is fully justified, with sprouts as a preferred and interesting candidates for biofortification. The aim of this pilot study was to investigate the effect of different methods of iodine enrichment of legume sprouts (yellow lupine, lentil, red and white clover, and common vetch) as well as to identify the most promising species for iodine bioaccumulation. The iodine content in mineralized sprout extracts was determi
