Carbohydrate-binding modules: powerful tools in the research and development of carbohydrates - a review.

Carbohydrate-binding modules (CBMs) are independent domains generally appended in carbohydrate-active enzymes (CAZymes) with carbohydrate-binding functions. The primary roles of CBMs in CAZymes include targeting, proximity, disruption, and adhesion effects. Due to their capacity to recognize and bind to specific carbohydrate structures, CBMs have emerged as valuable tools with wide applications, greatly facilitating the research and development of carbohydrates. This review summarizes the recent research progress and applications of CBMs, with particular emphasis on their applications as: (i)

The unique evolution of the carbohydrate-binding module CBM20 in laforin.

Laforin catalyses glycogen dephosphorylation. Mutations in its gene result in Lafora disease, a fatal progressive myoclonus epilepsy, the hallmark being water-insoluble, hyperphosphorylated carbohydrate inclusions called Lafora bodies. Human laforin consists of an N-terminal carbohydrate-binding module (CBM) from family CBM20 and a C-terminal dual-specificity phosphatase domain. Laforin is conserved in all vertebrates, some basal metazoans and a small group of protozoans. The present in silico study defines the evolutionary relationships among the CBM20s of laforin with an emphasis on newly id

Binding of sperm to somatic cells via HLA-DR. Modulation by sulfated carbohydrates.

We have shown previously that human sperm bind and enter leukocytes expressing surface HLA class II molecules. In the present study, mutant B lymphoblastoid cells and HLA-DR-transfected murine 3T3 fibroblasts are used to confirm that HLA class II molecules are somatic cell receptors for sperm. Further, for isolated HLA-DR expressed on murine cells, we show that sperm receptor activity requires the presence of sulfated carbohydrates. As carriers of multiple HLA-DR binding ligands, sperm may 1) mimic the target cell-activating effects of anti-DR antibody and 2) bind HIV through CD4-like or alter

Structural characterization of the carbohydrate-binding module of NanA sialidase, a pneumococcal virulence factor.

BACKGROUND: Streptococcus pneumoniae Neuraminidase A (NanA) is a multi-domain protein anchored to the bacterial surface. Upstream of the catalytic domain of NanA is a domain that conforms to the sialic acid-recognising CBM40 family of the CAZY (carbohydrate-active enzymes) database. This domain has been identified to play a critical role in allowing the bacterium to promote adhesion and invasion of human brain microvascular endothelial cells, and hence may play a key role in promoting bacterial meningitis. In addition, the CBM40 domain has also been reported to activate host chemokines and neu

Synergic action of an inserted carbohydrate-binding module in a glycoside hydrolase family 5 endoglucanase.

Most known cellulase-associated carbohydrate-binding modules (CBMs) are attached to the N- or C-terminus of the enzyme or are expressed separately and assembled into multi-enzyme complexes (for example to form cellulosomes), rather than being an insertion into the catalytic domain. Here, by solving the crystal structure, it is shown that MtGlu5 from Meiothermus taiwanensis WR-220, a GH5-family endo-β-1,4-glucanase (EC 3.2.1.4), has a bipartite architecture consisting of a Cel5A-like catalytic domain with a (β/α)8 TIM-barrel fold and an inserted CBM29-like noncatalytic domain with a β-jelly-rol

Carbohydrate-binding domain of the POMGnT1 stem region modulates O-mannosylation sites of α-dystroglycan.

The dystrophin glycoprotein complex, which connects the cell membrane to the basement membrane, is essential for a variety of biological events, including maintenance of muscle integrity. An O-mannose-type GalNAc-β1,3-GlcNAc-β1,4-(phosphate-6)-Man structure of α-dystroglycan (α-DG), a subunit of the complex that is anchored to the cell membrane, interacts directly with laminin in the basement membrane. Reduced glycosylation of α-DG is linked to some types of inherited muscular dystrophy; consistent with this relationship, many disease-related mutations have been detected in genes involved in O

Activity of a Carbohydrate-Binding Module Therapy, Neumifil, against SARS-CoV-2 Disease in a Hamster Model of Infection.

The rapid global spread of severe acute respiratory coronavirus 2 (SARS-CoV-2) has resulted in an urgent effort to find efficacious therapeutics. Broad-spectrum therapies which could be used for other respiratory pathogens confer advantages, as do those based on targeting host cells that are not prone to the development of resistance by the pathogen. We tested an intranasally delivered carbohydrate-binding module (CBM) therapy, termed Neumifil, which is based on a CBM that has previously been shown to offer protection against the influenza virus through the binding of sialic acid receptors. Exploring carbohydrate binding module fusions and Fab fragments in a cellulose-based lateral flow immunoassay for detection of cystatin C.

This paper presents a lateral flow assay (LFA) for the quantitative, fluorescence-based detection of the kidney biomarker cystatin C that features conjugates of capture antibodies and fusions of carbohydrate binding modules (CBM) with ZZ domains anchored on cellulose deposited over nitrocellulose (NC). The ZZ-CBM3 fusion provides a biomolecular interface between the cellulose layer and the Fc portion of the capture antibodies. By resorting to detection Fab fragments that lack the Fc portion we overcome the observed interference of full-length detection antibodies with the ZZ-CBM3 fusion at the

Identification, molecular characterization, and expression analysis of a DOMON-like type 9 carbohydrate-binding module domain-containing protein of Coccidioides posadasii.

Previously, we investigated the effect of N-acetylglucosamine (GlcNAc) on Coccidioides posadasii chitinolytic enzymes during in vitro spherule-endospore (S/E) phase culture. During those studies, sodium dodecyl sulfatepolyacrylamide gel electrophoresis analysis of supernatants from S/E phase cultures grown in Converse medium with or without added GlcNAc revealed a ∼ 28-kDa band (CFP28), whose abundance was increased by GlcNAc in parallel with the chitinolytic enzymes. 1H, 15N and 13C backbone and side-chain resonance assignments of a family 32 carbohydrate-binding module from the Clostridium perfringens NagH.

The Gram-positive anaerobe Clostridium perfringens is an opportunistic bacterial pathogen that secretes a battery of enzymes involved in glycan degradation. These glycoside hydrolases are thought to be involved in turnover of mucosal layer glycans, and in the spread of major toxins commonly associated with the development of gastrointestinal diseases and gas gangrene in humans. These enzymes employ multi-modularity and carbohydrate-binding function to degrade extracellular eukaryotic host sugars. Here, we report the full (1)H, (15)N and (13)C chemical shift resonance assignments of the first f

Interactions between the engineering family 3 carbohydrate-binding module (CBM3) and different types of nanocelluloses revealed by single-molecule force spectroscopy (SMFS).

Carbohydrate-binding modules (CBMs) exhibit specific binding affinity for carbohydrate substrates, making them invaluable in the recognition, utilization, and modification of various polysaccharides. Herein, we investigate the nanocellulose-CBM interaction, highlighting a specific engineered family 3 CBM (CBM3). There is a distinct polymorph- and surface morphology-dependency on CBM3's binding preferences, in accordance with the single-molecule force spectroscopy (SMFS) investigation for nanocellulose with different allomorphs (I and II) and in various forms (nanocrystals, CNC, and nanofibrils

Sensing ligand binding to a clinically relevant lectin by tryptophan fluorescence anisotropy.

Increasing insights into the involvement of endogenous lectins in disease processes fuel the interest to develop potent inhibitors. As a consequence, robust assay procedures are required. Due to their activity as adhesion/growth-regulatory effectors this study focussed on galectins. The human proto-type galectin-1 was selected as representative of this family with conserved presence of a tryptophan moiety in the binding site. This structural feature was taken advantage of to establish its use as reporter for ligand contact measuring polarized fluorescence emission. The experimentally determine

Exploring the effect of tandem non-native non-F-type lectin carbohydrate-binding domains on Streptosporangium roseum α-L-Fucosidase activity.

Microbial carbohydrate-active enzymes often contain non-catalytic carbohydrate-binding modules, which enhance the activity of the catalytic domain by proximity, targeting, or disruptive effects. We have previously biochemically characterized a GH29 α-L-fucosidase from Streptosporangium roseum, demonstrated that its C-terminal F-type lectin domain (FLD) improves the enzyme activity of its tandem N-terminal α-L-fucosidase domain towards fucooligosaccharides, and proposed a "bind and jump" model to explain this FLD-driven improvement of α-L-fucosidase activity. Here, we have explored the use of n

Comparative genomic analysis of hemicellulose-degrading potential in bacterial isolates from the anterior intestine of Eisenia andrei (Bouché, 1972).

Earthworms and their associated microbiota can degrade various types of lignocellulosic residues, but the enzymes, carbohydrate-binding modules, and sugar transporters involved in this process remain imperfectly understood. The present study aimed to identify genes and characterize hemicellulose degradation strategies of bacterial strains with high xylanolytic activity from the earthworm gut. The strains, originally isolated from the anterior intestine of earthworms fed on a diet of palm fiber (Streptomyces thermophilus PF5-2S and Niallia circulans PF7-2S) and coffee husk (Cellulosimicrobium c

MgCBP1 Synergistically Enhances MgENG2-Mediated Cellulose Degradation to Promote Parasitism in Meloidogyne graminicola.

Plant-parasitic nematodes secrete effector proteins, including cell wall-degrading enzymes (CWDEs) and cell wall-modifying proteins (CWMPs), to facilitate infection. However, whether cellulose-binding proteins (CBPs), which are a class of CWMPs, can synergize with cellulases (a subgroup of CWDEs) to promote parasitism remains unclear. Here, we identified and characterized a CBP (MgCBP1) and a β-1,4-endoglucanase (MgENG2) from the rice root-knot nematode Meloidogyne graminicola. MgCBP1 contains a conserved cellulose-binding module (CBM) and is specifically expressed in the subventral oesophagea

Proteome-wide prediction of bacterial carbohydrate-binding proteins as a tool for understanding commensal and pathogen colonisation of the vaginal microbiome.

Bacteria use carbohydrate-binding proteins (CBPs), such as lectins and carbohydrate-binding modules (CBMs), to anchor to specific sugars on host surfaces. CBPs in the gut microbiome are well studied, but their roles in the vagina microbiome and involvement in sexually transmitted infections, cervical cancer and preterm birth are largely unknown. We established a classification system for lectins and designed Hidden Markov Model (HMM) profiles for data mining of bacterial genomes, resulting in identification of >100,000 predicted bacterial lectins available at unilectin.eu/bacteria. Biomimetic engineering of a fortified ice composite with enhanced mechanical properties.

This work presents BioPykrete, a high-performance sustainable bio-composite engineered from ice, cellulose nanocrystals (CNC), and a custom chimeric protein designed to facilitate interfacial adhesion. By genetically fusing the ice-binding protein AFPIII with the carbohydrate-binding module CBM3a, we developed a chimeric protein to act as a molecular "bioadhesive" bridging the crystalline reinforcement phase to the ice matrix. During the controlled solidification of the suspension, the CBM3a-AFPIII chimera is proposed to regulate the self-organisation of the CNC fibers into a reinforcing 3D ne

Discovery and phylogeny of a ricin-B-like domain from rice.

Lectins are carbohydrate-binding proteins which play key roles in various biological processes, including cell signaling, pathogen recognition and development. Previous research conducted on ricin-B lectin domains and carbohydrate-binding modules of family 13 (CBM13) illustrated the striking resemblances between these two groups of protein domains. In this study, we report on the discovery, identification and putative biochemical characteristics of a ricin-B-like domain that is unique for GH27 enzymes from land plants, identified in the OsAPSE enzyme from Japanese rice (Oryza sativa L. subsp.

Streptococcus pneumoniae surface protein PfbA is a versatile multidomain and multiligand-binding adhesin employing different binding mechanisms.

Streptococcus pneumoniae, one of the major human respiratory pathogens, uses its repertoire of surface proteins to adhere to the epithelium of the nasopharynx and lungs leading to colonization. PfbA is a conserved surface protein of S. pneumoniae and helps the bacterium to colonize the host by recognizing the extracellular matrix (ECM) molecule fibronectin, as well as blood proteins like plasminogen and human serum albumin. The crystal structure of rPfbA150-607 revealed it to possess a beta-helical region similar to those of carbohydrate-active enzymes as well as a C-terminal segment that rese
