[Occupational allergies to cellulases].

In the 1960s an increasing number of occupational allergies in the detergent industry against the proteolytic enzyme Subtilisin were observed. In this context the high sensitization potential of enzyme dusts attracted attention. The present evaluation of literature data confirms that this is also true for cellulases. These enzymes--mostly of fungal origin--are frequently used in biotechnology. Several cases of specific airway sensitization caused by cellulases are verified by a number of studies. As symptoms, results of skin prick tests, detection of specific IgE-antibodies and results of spec

The medical dissolution of phytobezoars using cellulase.

Phytobezoars are plant fibre concretions occurring in the stomach as a complication following some types of gastric surgery. Symptoms include distension, pain and vomiting. Ulceration, gastric bleeding and even perforation can occur. If untreated, they result in a significant mortality. Dissolution of the phytobezoar can be achieved without operative intervention. Enzymes and especially cellulase help to break up the mass and it can then be aspirated or allowed to pass on. Five patients with phytobezoars were thus treated and the cellulase method was confirmed to be simple, safe and effective.

Rapid tube test for detecting fungal cellulase production.

A simple procedure is described for the detection of cellulolytic activity in fungi, utilizing dyed cellulose powder as substrate. The powder, incorporated into a basal medium containing agar, mineral salts, and growth factors, is layered onto basal medium minus cellulose. Cellulase production results in release of the dye which diffuses into the lower layer.

Enhanced cellulase production in mutants of Thermomonospora curvata.

Thermomonospora curvata, a thermophilic actinomycete, secretes multiple forms of endo-beta 1-4-glucanase (EG)when grown on cellulose-mineral salts liquid medium. The EG activity(measured as carboxymethyl cellulose hydrolysis) was separated by ion exchange chromatography into three distinct components which differ in their kinetic properties. Exposure of Thm. curvata to ultraviolet light, N-nitrosoguanidine, or ethane methyl sulfonate produced mutants with enhanced EG production. Selection of colonies which cleared cellulose agar plant containing 2-deoxtglucose of glycerol yielded mutants havin

Characterization of an immunogenic cellulase secreted by Cryptococcus pathogens.

Members of the C. neoformans/C. gattiii species complex are an important cause of serious humans infections, including meningoencephalitis. We describe here a 45 kDa extracellular cellulase purified from culture supernatants of C. neoformans var. neoformans. The N-terminal sequence obtained from the purified protein was used to isolate a clone containing the full-length coding sequence from a C. neoformans var. neoformans (strain B-3501A) cDNA library. Bioinformatics analysis indicated that this gene is present, with variable homology, in all sequenced genomes of the C. neoformans/C. gattii sp

Effect of methylxanthines on production of cellulases by Penicillium echinulatum.

AIM: In this work, the effect of supplementing liquid cellulase production media (CPM) with methylxanthines (aminophylline, caffeine and theophylline), with and without the addition of glucose, on the secretion of cellulases by Penicillium echinulatum strain 2HH (wild-type) and the derived mutant strain 9A02S1 was studied. METHODS AND RESULTS: When compared with unsupplemented CPM, both strains produced higher beta-glucosidase and filter paper activities (FPAs) in CPM supplemented with 1 micromol l(-1) of caffeine but lower activities with 5 micromol l(-1) of caffeine. With theophylline only,

Factors influencing the production of cellulases by Sporotrichum thermophile.

Cellulase production and growth of a strain of Sporotrichum thermophile were studied by using a mineral salts medium supplemented with yeast extract and insoluble cellulose. The effects of cultural conditions, such as pH, nitrogen source, substrate concentration, and temperature, were examined. Maximum production of C1 and CX cellulases occurred at 45 C in 2 to 4 days, in the presence of 1% Solka/Floc as substrate, when NaNO3 or urea used as sources of nitrogen. Under these conditions, cellulolytic activity of culture filtrates appeared to be similar to that reported for Trichoderma viride gro

Multifunctional cellulases are potent, versatile tools for a renewable bioeconomy.

Enzyme performance is critical to the future bioeconomy based on renewable plant materials. Plant biomass can be efficiently hydrolyzed by multifunctional cellulases (MFCs) into sugars suitable for conversion into fuels and chemicals, and MFCs fall into three functional categories. Recent work revealed MFCs with broad substrate specificity, dual exo-activity/endo-activity on cellulose, and intramolecular synergy, among other novel characteristics. Binding modules and accessory catalytic domains amplify MFC and xylanase activity in a wide variety of ways, and processive endoglucanases achieve a

Bacterial cellulase from the intestinal tract of the sugarcane borer.

Cellulolytic enzymes have wide use in several industrial segments (e.g. biofuels, pulp and paper, food, and cosmetics). However, one of the challenges is their large-scale production with high specific activity to eliminate the dependence of the purchase of enzymatic cocktails produced by commercial parties. The aims of this study were (1) isolation, selection, and partial characterization of bacterial cellulases present in the intestinal tract of the sugarcane borer and (2) to identify cellulase-producing bacteria by analyzing the 16S rDNA gene. Cellulase production and purification assays re

Occupational asthma caused by cellulase and lipase in the detergent industry.

Three employees from two different detergent companies were investigated for occupational asthma, using skin prick tests, serum specific IgE, and specific bronchial challenge. Two were challenged with lipase and one with cellulase. All three cases had immunological evidence of sensitisation to the detergent enzymes with which they worked. Bronchial challenge in each provoked a reproducible dual asthmatic response, which reproduced their work related symptoms. These are the first reported cases of occupational asthma attributable to cellulase and lipase in the detergent industry. Four of the mo

Termite cellulases: potential target for screening of natural termiticides.

Among the methods tested for termite eradication, termiticides derived from plants are particularly noteworthy. These termiticides represent an effective strategy for insect management, causing minimal environmental harm and posing less risk to human health than widely used chemical controls. The present study aims to obtain crude protein extracts from termites and to characterize their cellulolytic activities for their use in the bioprospecting of natural termiticides. Adult individuals were collected in wood fragments, identified, selected, and sectioned to obtain two protein extracts: one s

Cello-oligosaccharide Nanoarchitectonics: Drug Carriers with High Responsiveness to Cellulase.

Stimulus-responsive delivery systems enable precise control over the release of therapeutic and agricultural agents. Among various triggers, cellulase has recently attracted increasing attention because of its unique biological distribution; it is absent in humans but produced by specific micro-organisms and insects. Herein, we report a cellulase-responsive drug delivery carrier based on terminally alkylated cello-oligosaccharide assemblies. Among the assemblies examined, terminally hexylated cello-oligosaccharides with the cellulose I allomorph (CELI-C6) exhibited the highest loading capacity

Effect of cellulase on antioxidant activity and flavor of Rosa roxburghii Tratt.

Cellulase can increase the soluble dietary fiber (SDF) content in Rosa roxburghii Tratt (RRT), but the effects on polyphenol content, bioactivity, and flavor are unknown. This study analyzed the changes in SDF content, total phenolic content, antioxidant activity and flavor before and after cellulase treatment. Cellulase treatment increased the SDF and total phenolic content of RRT by 13 % (P < 0.05) and 25.68 % (P < 0.05), respectively, and increased the antioxidant activity. HS-GC-IMS identified a total of 42 volatile compounds present, and ROAV analysis revealed that the characteristic arom

Optimization of the Cellulase Free Xylanase Production by Immobilized Bacillus Pumilus.

BACKGROUND: The extracellular xylanase secreted by microorganisms is a hydrolytic enzyme, which arbitrarily cleaves the β-1, 4 backbone of the polysaccharide xylan; an enzyme used in the food processing, bio-pulping and bio-bleaching. The commercial production of the xylanase is limited because of a higher cost involvement, which can be overcome by the cost-effective production of the xylanase through immobilization of the microbial cell by the non-toxic substances. OBJECTIVES: In this work, the optimization of the extra-cellular cellulase free xylanase production by the immobilized cell of t

Porous biochar/chitosan composites for high performance cellulase immobilization by glutaraldehyde.

In this work, porous biochar derived from sugarcane bagasse was prepared and then coated with different amounts of chitosan (C@CS) for cellulase immobilization. Cellulase was covalently immobilized on the support by using glutaraldehyde as a linker. The chemical characteristics and morphology of the samples were determined by SEM, BET, FT-IR and XPS. The properties of immobilized enzyme were evaluated by activity recovery, optimum pH value and temperature, and recyclability. The results showed that all the three kinds of immobilized cellulase did not change the optimum pH value of 4 and temper
