Going deep: phylogenomic analyses incorporating museomic data reveal divergence and character evolution in Decapodiformes (Mollusca: Cephalopoda).

Decapodiformes, including cuttlefishes and squid, are ecologically diverse cephalopods widely distributed throughout the world's oceans, yet the evolutionary history of their morphological and ecological traits remains poorly understood. Here, we developed a phylogenetic workflow for decapodiforms by integrating transcriptomic and genome skimming data from 48 freshly collected and historical specimens. The resulting dataset comprised over 6000 loci and covered all major extant lineages. Phylogenetic analyses consistently recovered Idiosepida as the sister lineage to all other decapodiforms. Am

Application of chitosan as a novel antimicrobial dental material -Effects of form, molecular weight, and deacetylation degree on its antimicrobial activity against Candida albicans.

Oral candidiasis is a significant health concern, especially for elderly individuals. The present study examined chitosan as a potential antimicrobial dental material for Candida albicans treatment. Four types of chitosan products purified from Chionoecetes opilio and Decapodiformes, with varying weight average molecular weight and degree of deacetylation (DAC) were used. Antimicrobial activity was assessed with fungal viability assays, culture turbidity measurements, and live/dead cell staining, while mechanism of action was determined by examining extracellular adenosine triphosphate (ATP) l

An anatomical map of squid lateral lines.

Survival of aquatic animals requires sensing water movement. In both aquatic vertebrates and cephalopods, sensory cells arranged in 'lateral lines' on the surface of the animal detect water motion relative to the animal's body with mechanosensitive hair bundles1. Cephalopod lateral lines differ from those of fishes in hair bundle cytoskeletal composition, morphology, and receptor cell location. Fish have lateral lines arrayed on the head and body, whereas squid lateral lines, also termed epithelial lines or lateral line analogs, have only been described in detail on the head and arms2,3; it is

Cephalopod learning and memory.

Cephalopod molluscs are renowned for their unique central nervous system - a donut-shaped brain organised around the oesophagus. This brain supports sophisticated learning and memory abilities. Between the 1950s and 1980s, these cognitive abilities were extensively studied in octopus (Figure 1A) - a now leading model for the study of memory and its neural substrates (approximately 200 papers during this period). The focus on octopus learning and memory was mainly due to their curious nature and the fact that they adapt to laboratory-controlled conditions, making them easy to test and maintain

The inhibitory mechanism of 2-amino-3,8-dimethylimidazo [4,5-f] quinoxaline (MeIQx) formation by ultraviolet-gallic acid (UV-GA) during the oil-frying process of squid.

The inhibitory effect of ultraviolet-gallic acid (UV-GA) on carbonyl valence and intermediates and precursors of 2-amino-3,8-dimethylimidazo [4,5-f] quinoxaline (MeIQx) was investigated to futher clarify the inhibitory mechanism for safety control the quality of oil-fried squid. Ultraviolet C-treated gallic acid (UVC-GA) and ultraviolet B-treated gallic acid (UVB-GA) were produced by ultraviolet 225 nm of band C and 300 nm of band B, respectively. The MeIQx contents in oil-fried squid were significantly higher, and UVC-GA and UVB-GA could significantly inhibit the MeIQx formation and the forma

Comparative bioactivity profile of phospholipids from three marine byproducts based on the zebrafish model.

Phospholipids (PLs) are important components of physiological metabolism in animals and plants, and they have been widely used in clinical treatment, cosmetics, and industry. With the development of marine resources, marine PLs rich in polyunsaturated fatty acids have attracted increasing attention. As important marine resources, shrimp heads (SH), codfish roe (CR), and squid gonads (SG) contain a high PL content. The antithrombotic, antistroke, anti-inflammatory, pro-angiogenic, and cardioprotective activities of PLs from SH, CR, and SG were evaluated and compared using the in vivo zebrafish

Microplastics presence in cultured and wild-caught cuttlefish, Sepia officinalis.

Amongst cephalopods microplastics have been reported only in jumbo squid gut. We investigated microplastics in the digestive system of wild cuttlefish (Sepia officinalis) as they are predators and prey and compared the stomach, caecum/intestine and digestive gland (DG) of wild and cultured animals, exposed to seawater from a comparable source. Fibers were the most common type (≈90% of total count) but were ≈2× higher in relation to body weight in wild vs. cultured animals. Fibers were transported to the DG where the count was ≈2× higher /g in wild (median 1.85 fibers/g) vs. cultured. In wild-c

Infection of two cestode larvae, Nybelinia enterika sp. nov. and Phoreiobothrium sp. in oval squid Sepioteuthis lessoniana species complex.

Recently, oval squid of Sepioteuthis lessoniana species complex has gained importance in aquaculture due to its successful breeding in semi-intensive and intensive aquaculture systems. However, there is a lack of information regarding pathogens and diseases that can affect this species, prompting the need for further research. Two cestode species, Nybelinia enterika sp. nov. and Phoreiobothrium sp., were identified as members of the Trypanorhyncha and Onchoproteocephalidea orders, respectively, using both morphological and molecular diagnostic markers in wild-caught oval squids (Sepioteuthis l

Networks of linked radial muscles could influence dynamic skin patterning of squid chromatophores.

The visibility of cephalopod chromatophore organs is regulated dynamically by rosettes of obliquely striated radial muscles that dilate or relax the diameter of a central pigmented sacculus in 100-300 ms. Each of the several dozen muscles has a flared proximal end that adheres tightly to its pigmented sacculus and an extremely elongated distal end which branches into single fibrils that anchor into the dermis. This geometry provides ample opportunity for overlap of the many muscles from neighboring chromatophores. The temporal activity of these muscles has been believed to be patterned exclusi

Isolated Axoplasm from Squid and Pathogenic Mechanisms in Amyotrophic Lateral Sclerosis.

The squid giant axon is an experimental model that has been instrumental in providing answers to many fundamental questions in cell biology, including the discovery of the motor protein kinesin and probing molecular mechanisms of neurodegenerative diseases such as amyotrophic lateral sclerosis (ALS). The giant axon offers scale and ease of access that is not possible in mammalian models. The power of this model stems from the fact that one can introduce reagents and proteins that would not cross a plasma membrane at defined concentrations in a buffer environment that is closely modeled on norm

Changes in feeding behavior of longfin squid (Doryteuthis pealeii) during laboratory exposure to pile driving noise.

Anthropogenic noise can cause diverse changes in animals' behaviors, but effects on feeding behaviors are understudied, especially for key invertebrate taxa. With the offshore wind industry expanding, concern exists regarding potential impacts of pile driving noise on squid and other commercially and ecologically vital taxa. We investigated changes in feeding and alarm (defense) behaviors of squid, Doryteuthis pealeii, predating on killifish, Fundulus heteroclitus, during playbacks of pile driving noise recorded from wind farm construction within squids' habitat. Fewer squid captured killifish

Lighting the way: how the squid-Vibrio model can inform thermal impacts on symbiotic dynamics.

As global temperatures are shifting, so too is the landscape of organismal fitness and, by extension, the role of the symbiotic microbes they house. As these host-microbe partnerships grapple with changing environments, current research struggles to keep pace with the complexity of microbial symbioses acclimating, adapting and evolving as environmental conditions change around them. Wild-caught organisms have been used to test adaptation to extreme environments, but extrapolating and interpreting data on how separate partners within a symbiosis respond to detrimental conditions is difficult. Peracetic acid treatment of squid eggs infected with parasitic copepod (Ikanecator primus gen. et sp. nov.).

Having been successfully bred in semi-intensive and intensive aquaculture systems, oval squids of the Sepioteuthis lessoniana species complex are emerging as promising candidates for research and industry. Nevertheless, information about pathogens and diseases that may affect squid aquaculture remains sparse. In this study, we identify new parasitic copepod species that causes squid mortality and decreases squid hatching rates, and we also offer a solution to eliminate the pathogen during incubation of squid eggs. The newly discovered copepod Ikanecator primus gen. et sp. nov. was identified o

Feeding ecology of broadbill swordfish (Xiphias gladius) in the California current.

The feeding ecology of broadbill swordfish (Xiphias gladius) in the California Current was described based on analysis of stomach contents collected by fishery observers aboard commercial drift gillnet boats from 2007 to 2014. Prey were identified to the lowest taxonomic level and diet composition was analyzed using univariate and multivariate methods. Of 299 swordfish sampled (74 to 245 cm eye-to-fork length), 292 non-empty stomachs contained remains from 60 prey taxa. Genetic analyses were used to identify prey that could not be identified visually. Diet consisted mainly of cephalopods but a

A newly identified enzyme from Japanese common squid Todarodes pacificus has the ability to biosynthesize d-aspartate.

Amino acids exist in two chiral forms, namely L and D. Although l-amino acids are predominant in vivo, certain limited circumstances have reported the usage of d-amino acids. d-aspartate (Asp), among them, plays crucial physiological roles in living organisms and is biosynthesized from L-Asp by the enzyme named aspartate racemase (AspRase). D-Asp is known to accumulate in large amounts in the nervous system of cephalopods. To understand the function of D-Asp in nervous system in more detail, it is necessary to elucidate its metabolic pathway; however, AspRase gene has not been identified in ce

Jumbo squid beaks: inspiration for design of robust organic composites.

The hard tissues found in some invertebrate marine organisms represent intriguing paradigms for robust, lightweight materials. The present study focuses on one such tissue: that comprising the beak of the jumbo squid (Dosidicus gigas). Its main constituents are chitin fibers (15-20wt.%) and histidine- and glycine-rich proteins (40-45%). Notably absent are mineral phases, metals and halogens. Despite being fully organic, beak hardness and stiffness are at least twice those of the most competitive synthetic organic materials (notably engineering polymers) and comparable to those of Glycera and N

Anisakidae larvae found in marine fishes and squids from the Gulf of Tongking, the East China Sea and the Yellow Sea.

Marine fishes and squids caught in the Gulf of Tongking, the East China Sea and the Yellow Sea were dissected, and many Anisakidae larvae were found in them. The larvae of Anisakis simplex (Anisakis Type I larvae), the most important causative agent of anisakiasis, was found in about 30% of fishes and squids caught in the Gulf of Tongking and in about 60% of those caught in the East China Sea and the Yellow Sea. Larvae belonging to three other genera than Anisakis--Raphidascaris or Raphidascaroides, Hysterothylacium, and Terranova (Cannon Type I)--were also detected.
