Paradoxical Effects of Erucic Acid-A Fatty Acid With Two-Faced Implications.

Erucic acid (EA) is a monounsaturated fatty acid primarily consumed as rapeseed oil and mustard oil (MO). The consumption of EA-rich food has been reported to have adverse effects on health, particularly myocardial lipidosis and hepatic steatosis. Consequently, several countries, including the United States, European countries, New Zealand, and Australia, set limits on their daily intake. However, EA-rich MO (30%-50%) is still consumed in Asia. In contrast, limited studies on humans have reported a protective role of MO in acute myocardial infarction, ischemic heart disease, and neurologic dis

Mixed Oleic Acid-Erucic Acid Liposomes as a Carrier for Anticancer Drugs.

BACKGROUND: Liposomes are mostly known to be prepared from phospholipids and lipids and have a remarkable capacity to encapsulate both lipophobic and lipophilic molecules. However, there is little research on developing fatty acid liposomes for chemotherapy. OBJECTIVE: We have successfully prepared mixed fatty acid liposomes from two monounsaturated fatty acids, namely oleic acid and erucic acid, which stabilised by DOPEPEG2000. The Critical Vesicular Concentration (CVC) of liposomes was found to be within 0.09 to 0.21 mmol dm-3, with an average particle size of 400 nm. METHODS: Encapsulatio

Erucic acid impairs male fertility by suppressing retinoic acid synthesis in sertoli cells.

Although dietary factors are increasingly implicated as crucial determinants of male fertility, specific dietary risk factors and their metabolic mechanisms remain poorly understood. In this study, patients with non-obstructive azoospermia (NOA) or severe oligospermia (EO) demonstrated significantly elevated erucic acid (EA) levels, with a nearly 3-fold increase in serum (P < 0.0001) and a 27 % increase in semen (P = 0.0147) compared to controls, suggesting a potential dose-dependent negative impact of EA exposure on spermatogenesis. Mice fed a high-EA diet exhibited a significant 52.5 % reduc

Review of the potential pharmacological role of erucic acid: a monounsaturated omega-9 fatty acid.

This comprehensive review aims to provide an overview of the pharmacological properties of erucic acid (EA) and highlight areas that require further research. EA is an omega-9 fatty acid found in certain vegetable oil, such as rapeseed oil has demonstrated favourable effects in rodents, including ameliorating myocardial lipidosis (fat accumulation in the heart muscle), congestive heart disease, hepatic steatosis (fat accumulation in the liver), and memory impairments. These findings have prompted regulatory bodies to establish limits on EA content in food oils. The studies were performed on ro

Purification of erucic acid by preparative high-performance liquid chromatography and crystallization.

Erucic acid (C22:l fatty acid) has been found to be useful in the treatment of adrenoleukodystrophy (ALD). It appears to work by reducing the blood levels of very-long-chain fatty acids (VLCFAs) which destroy the myelin sheaths of the nerves. Erucic acid was purified by reversed-phase high-performance liquid chromatography (HPLC) on columns packed with YMC C18 (10-20 microns, 120 A). Using ethanol-water as the mobile phase, the recovery of erucic acid was 69% and the purity was more than 97% as measured by gas chromatography. The amount of saturated VLCFAs was found to be within the limits spe

Erucic acid, a nutritional PPARδ-ligand may influence Huntington's disease pathogenesis.

Increasing recent evidence suggests a key role of oligodendroglial injury and demyelination in the pathophysiology of Huntington's Disease (HD) and the transcription factor PPARδ is critical for oligodendroglial regeneration and myelination. PPARδ directly involves in the pathogenesis of HD and treatment with a brain-permeable PPARδ-agonist (KD3010) alleviates its severity in mice. Erucic acid (EA) is also a PPARδ-ligand ω9 fatty acid which is highly consumed in Asian countries through ingesting cruciferous vegetables such as rapeseed (Brassica napus) and indian mustard (Brassica juncea). Erucic acid, eicosapentaenoic acid, and docosahexaenoic acid consumption affect hepatic steatosis in mice fed a western diet.

This study explored whether erucic acid (EA) intake at levels typical of fatty fish consumption aggravates non-alcoholic fatty liver disease (NAFLD) or affects the benefits of marine n-3 polyunsaturated fatty acids (PUFAs). N-3 PUFAs, particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), enhance beta-oxidation and suppress lipogenesis, reducing hepatic triglyceride accumulation. Sixty-six C57BL/6N mice were fed a chow diet, western diet (WD), or WDs supplemented with EA alone or combined with EPA or DHA for 13 weeks. Dietary EA, EPA, and DHA levels matched average intake fro

Multi-omics of the nasturtium (Tropaeolum majus) and the parallel evolution of the erucic acid biosynthesis in angiosperms.

Erucic acid is a widely used raw material in industry, and it can be synthesized in the seeds of a number of plants, including Tropaeolum majus (nasturtium), an herbaceous vine in Tropaeolaceae. In this study, using a newly assembled chromosome-level nasturtium genome, we screened candidate genes encoding the rate-limiting enzymes involved in the nasturtium erucic acid biosynthesis. After genome-wide identification and expression profiling, nine out of 25 3-ketoacyl-CoA synthase (KCS) genes were found to be expressed in nasturtium seed, while further expression pattern clustering divided the n

The protective effects and mechanisms of Erucic acid in allergic rhinitis via the MyD88/NF-κB pathway.

Allergic rhinitis (AR), a local inflammatory disease mediated by IgE in the nasal mucosa, presents a significant challenge to global health. Erucic acid (EA) is a monounsaturated fatty acid. Current research indicates EA possesses diverse biological activities, including anti-inflammatory, antioxidant, and immunomodulatory properties. However, its effects on AR remain unexplored. This study aims to investigate the therapeutic action and mechanism of EA in AR. The efficacy and mechanisms of EA were evaluated on the AR mouse model. Therapeutic outcomes were assessed through behavioral observatio

Mortality and production characteristics of laying chickens fed high- and low-erucic acid rapeseed oils.

Laying pullets were fed a diet supplemented with three percent of either high-erucic acid rapeseed (HEAR) oil or low-erucic acid rapeseed (LEAR) oil for 39 weeks. Egg production for the period was 78.8 and 80.1% by the birds fed the respective oils. Average egg weights were 56.8 and 58.7 g. respectively. Gains in body weight, liver weight and adrenal gland weight were similar with the two types of oil fed. Feed consumption was similar for the two groups of birds. The efficiency of utilization of the diet containing HEAR oil was accordingly lower than that of the diet containing LEAR oil. In th

Mediator subunit MED25 represses ABI5-dependent activation of erucic acid biosynthetic gene FAE1 in Brassica napus.

ABSCISIC ACID INSENSITIVE 5 (ABI5) is a key regulator that represses seed germination, flowering, and seedling growth in Arabidopsis thaliana and functions in abscisic acid (ABA) signal transduction. However, whether ABI5 is involved in other biological processes and its interacting partners remain largely unknown. Here, we show that BnaABI5, an ortholog of AtABI5, positively regulates erucic acid synthesis and the transcript level of the marker gene FAE1 in Brassica napus. Expression patterns of BnaABI5 and BnaFAE1 overlap during specific stages of seed development, as indicated by the BnIR d

Resistance to erucic acid as a selectable marker for peroxisomal activity: isolation of revertants of an infantile Refsum disease cell line.

A system based on the ability of cells to oxidize very long-chain fatty acids (VLCFA) was developed to select in vitro normal human fibroblasts from fibroblasts of patients suffering from peroxisomal disorders with multienzymatic deficiencies: Zellweger syndrome, neonatal adrenoleukodystrophy, infantile Refsum disease (IRD). Cells treated with various concentrations of erucic acid (C22:1 n-9) revealed an enhanced toxicity of this fatty acid for the fibroblasts of patients compared with normal cells. This differential toxicity is correlated with variable accumulations of C22:1 n-9 and the absen

Early Neuroprotective Effects of Erucic Acid and Gentisic Acid on Hippocampal Glutamate Concentrations and Alzheimer-like Molecular Alterations in an Intracerebroventricular Streptozotocin Rat Model.

Background: Alterations in glutamatergic homeostasis have been implicated in Alzheimer's disease, but the relationship between total hippocampal tissue glutamate and Alzheimer-like molecular alterations remains unclear. This study evaluated the early neuroprotective potential of gentisic acid (GA) and erucic acid (EA) in an intracerebroventricular streptozotocin (icv-STZ)-induced rat model of sporadic Alzheimer-like pathology. Methods: Sixty-four female Sprague-Dawley rats were randomly assigned to eight groups: control, sham, STZ, STZ + GA (100 or 200 mg/kg/day), STZ + EA (25 or 50 mg/kg/day)

Induction of a reversible cardiac lipidosis by a dietary long-chain fatty acid (erucic acid). Relationship to lipid accumulation in border zones of myocardial infarcts.

Previous studies have demonstrated that cardiac myocytes in the border zone of acute myocardial infarction become markedly overloaded with neutral lipid during the transition from reversible to irreversible injury. To examine directly the role of these changes in neutral lipid metabolism in the development of irreversible cellular injury and associated increases in tissue Ca2+ content, the authors fed rats large amounts of a fatty acid (erucic acid) that is poorly oxidized by the heart and that subsequently accumulates as neutral lipid. Rats fed a high erucic acid (C22:1) diet in the form of 2

Erucic Acid, Derived by Lactobacillus Crispatus, Induces Ferroptosis in Cervical Cancer Organoids Through the PPAR-δ Signaling Pathway.

The microbiome present throughout the human body serves a variety of functions. In this study, 16S rRNA sequencing is employed to uncover differences in the abundance of Lactobacillus within the vaginal microbiota between individuals with cervical cancer and those with healthy cervixes. The research further identifies that the metabolite of Lactobacillus crispatus can induce ferroptosis in cervical cancer cells. This conclusion is reached through targeted bacterial culture, patient-derived organoids (PDO) and single-cell RNA sequencing. Erucic acid, identified as a primary metabolite via untar

Oral Fusobacterium nucleatum resists the acidic pH of the stomach due to membrane erucic acid synthesized via enoyl-CoA hydratase-related protein FnFabM.

BACKGROUND AND OBJECTIVE: Oral bacteria can translocate to the intestine, and their colonization efficiency is influenced by the gastrointestinal tract pH. Understanding how oral bacteria resist acidic environments is crucial for elucidating their role in gut health and disease. METHODS: To investigate the mechanisms of acid resistance in oral bacteria, an in vitro gastrointestinal tract Dynamic pH Model was established. This model was used to simulate the acidic conditions encountered by bacteria during their translocation from the mouth to the intestine. RESULTS: Fusobacterium nucleatum ex

Erucic acid aggravates cardiac fibrosis after myocardial infarction by CD36-mediated metabolic reprogramming with impaired mitochondrial aerobic oxidation.

BACKGROUND: Post-myocardial infarction (MI) cardiac fibrosis is a key driver of heart failure, with dysregulated cardiac metabolism playing a central role. The specific impact of circulating fatty acid metabolites on mitochondrial function and fibrotic remodeling remains unclear. Erucic acid, a very-long-chain fatty acid found in certain edible oils, has a historical association with cardiac lipidosis, yet its causal role and mechanism in post-MI fibrosis are unknown. METHODS: We employed an integrative, two-stage strategy. First, a hypothesis-free two-sample Mendelian Randomization (MR) anal

Combined in vitro effects of temozolomide and erucic acid on U87 MG cell viability, oxidative stress, and expression of genes involved in apoptotic signalling.

Limited efficacy of glioblastoma treatment with temozolomide (TMZ) due to resistance has prompted researchers to look for adjuvants capable of enhancing TMZ-associated cytotoxicity. The aim of our in vitro study was to evaluate such effects of erucic acid when combined with TMZ in human U87 MG cells. Cells were treated for 24 or 72 h with vehicle (control), TMZ (200 ng/mL), or TMZ plus EA (20, 40, 80, or 160 µg/mL). Cell viability was assessed using the MTT assay, membrane integrity with lactate dehydrogenase (LDH) release, and intracellular redox balance by measuring total antioxidant capacit
