[Gastrointestinal hormones].

Recently, knowledge about the physiological and physiopathological role of some already known gastrointestinal hormones has been increased. At the same time, new biologically active substances (peptides) have been isolated from the gastrointestinal tract and pancreas. Some hormones that were thought to be restricted to the digestive system have been identified in the brain and some peptides localized in the nervous terminals in the myenteric plexuses, actively participate in the biological process of the gut. These findings have made the comprehension of this field more complicated. As a conse

Gastrointestinal hormones.

The availability of pure intestinal hormones and the development of radioimmunoassays for their measurement has expedited research into many aspects of gastrointestinal endocrinology. A complex balance evidently exists between the different intestinal hormones and also the rest of the endocrine system. Polyendocrinopathies have been described, and, so far, two diseases due to intestinal hormone excess (Zollinger-Ellison syndrome and the syndrome of watery diarrhea, hypokalemia and achlorhydria) elucidated. It seems likely that many more gastrointestinal endocrine diseases await discovery.

Measurement of Gastrointestinal Hormones.

Towards the end of the 20th century, the number of subjects with diabetes and obesity rose exponentially. The discoveries of insulin- and appetite-modulating chemical signals, including glucagon-like peptide-1 (GLP-1), secreted from the gastrointestinal system, led to development of a new group of drugs which now are being used for glucose-lowering therapy and weight loss. Understanding of the physiology of gut derived signals and their pathophysiologi-cal importance requires accurate measurements of their circulat-ing levels. However, the assessment of these gut-derived hor-mones has been ham

[Gastrointestinal hormones--clinical significance].

The author gives an account of clinical syndromes which develop as a result of overproduction of gastrointestinal hormones. From the various diagnostic approaches, which are not always available or are expensive, the author summarizes the importance of thin-needle biopsy under sonographic control, the argentaffine technique (Grimelius) and histoenzymatic examination for neuron specific enolase. In addition to surgical treatment treatment with streptozotocine, 5-FU, dimethyl triazenoimidazole carboxamide and somatostatin is possible. The author draws attention to the possibility of using somato

[Gastrointestinal hormones. I. Hormones of the gastrin group].

Gastrin is a peptide hormone originating from G-cells of the antrum, the duodenum and the proximal jejunum. From extracts of gastrinomas and from sera of hypergastrinaemic subjects several gastrin molecules could be isolated which were nominated as "mini gastrin" (G13), "little gastrin" (G17), "big gastrin" (G34) and "big big gastrin". Antisera used for radioimmunological gastrin determinations should be characterized with respect to their specificity, as differeing affinity towards the various gastrins and towards CCK-PZ influences the results of the assay and thus the comparability with valu

Effects of gastrointestinal hormones on pancreatic growth.

This article discusses experiments demonstrating that the gastrointestinal hormones, gastrin, secretin, and CCK (cholecystokinin), stimulate the growth of the exocrine pancreas. Exogenous gastrin, secretin, and CCK increase pancreatic weight DNA, RNA and protein content of the rat pancreas. Antrectomy, which removes most endogenous gastrin, decreases pancreatic growth. The effects of antrectomy are prevented by exogenous gastrin. Infusion of HCl into the duodenum to release secretin and infusion of amino acids into the duodenum to release CCK also stimulate pancreatic growth. These results pro

Nutritional control of thyroid morphogenesis through gastrointestinal hormones.

Developing animals absorb nutrients either through the placenta or from ingested food; however, the mechanisms by which embryos use external nutrients for individual organ morphogenesis remain to be elucidated. In this study, we assessed nutrient-dependent thyroid follicle morphogenesis in Xenopus laevis and investigated the role of secreted gastrointestinal (GI) hormones post-feeding. We found that feeding triggers thyroid follicle formation, and the thyroid cells showed transient inactivation of cell proliferation after feeding. In addition, the thyroid cells with multi-lumina were frequentl

Sex hormones and functional gastrointestinal disorders in menopausal women.

Functional gastrointestinal diseases (FGIDs)/disorders of gut-brain interaction (DGBI) that manifest during menopause are characterized by chronic recurrent gastrointestinal (GI) symptoms associated with fluctuations in sex hormones. Current research indicates that the regulatory influence of sex hormones on GI function is multifaceted, involving a complex endocrine regulatory network across multiple organs and systems. Various types of sex hormones exert both direct and indirect effects on GI function through their specific receptors located within the GI tract, the central nervous system, pa

Structural similarities among gastrointestinal hormones and related active peptides.

The amino acid sequences of gastrointestinal hormones were compared in fragments of variable spans. Similarities within each of three peptide groups are extensive, but non-unique alignments were also noticed in the glucagon group. Use of different spans demonstrated that structural similarities are unevenly distributed in the gastrin family. Correct phasing was detected even for proteins with few identities and multi-shifted alignments (alcohol dehydrogenases). Tests for alignments among different groups of peptides revealed similarities between bombesin and glucagon or secretin, as well as be

The coordination of gastrointestinal hormones and the autonomic nerves.

We have conducted investigations on the relationship between gastrointestinal hormones and the autonomic nervous system. In this paper are described main themes of our investigations where a close association between gastrointestinal hormones and the autonomic nervous system was shown from the functional and morphological points of view. The "nervism" proposed by Pavlov, where the regulation of various organs in the body was thought to be accomplished wholly by the nervous system, was based on the findings on the physiology of the digestive glands. In the meantime, the discovery of secretin fr

The effect of gastrointestinal hormones on the gastric microcirculation.

The purpose of the present study was to evaluate the effects of several gastrointestinal hormones on the gastric submucosal arterioles using an in vivo microscopy technique. Alteration in diameter of submucosal arterioles, 40 to 90 mu in diameter, in response to infusion of different agents into the celiac axis, was measured in the anesthetized cat. Pentagastrin, the octapeptide of cholecystokinin, natural secretin, and histamine produced arteriolar dilation. Only with histamine and the octapeptide of cholecystokinin did this occur with doses that might be considered physiological. Synthetic s

Gastrointestinal hormones and the dialogue between gut and brain.

The landmark discovery by Bayliss and Starling in 1902 of the first hormone, secretin, emerged from earlier observations that a response (pancreatic secretion) following a stimulus (intestinal acidification) occurred after section of the relevant afferent nerve pathway. Nearly 80 years elapsed before it became clear that visceral afferent neurons could themselves also be targets for gut and other hormones. The action of gut hormones on vagal afferent neurons is now recognised to be an early step in controlling nutrient delivery to the intestine by regulating food intake and gastric emptying. I
