We have investigated IgG antibodies to microfilament-associated proteins in sera from 29 normal controls and from 172 patients with autoimmu … and myosin light chain. Results shows that normal as well as autoimmune sera are polyreactive with microfilament proteins. The specificity of reactivity against a particular microfilament-associated protein was demonstrated by inhibition experiments with the specific protein. No correlation was observed between immunoblot and immunofluorescence results. Of the 201 sera …

BACKGROUND: Assembly of recombinant capsid proteins into virus-like particles (VLPs) still represents an interesting challenge in virus-based nanotechnologies. The structure of VLPs has gained importance for the development and design of new adjuvants and antigen carriers. The potential of Tobacco etch virus capsid protein (TEV CP) as adjuvant has not been evaluated to date.
FINDINGS: Two constructs for TEV CP expression in Escherichia coli were generated: a wild-type version (TEV-CP) and a C-terminal hexahistidine (His)-tagged version (His-TEV-CP). Although both versions were expressed in th

Corneal stromal fibroblasts play an important role in wound healing. Proteins from all three cytoskeletal classes (microfilament, microtubule, and intermediate filament) are involved in the control of various cellular events, such as motility, cell adhesion, shape changes, intracellular transport, and mitosis. By epifluorescent light microscopy, we studied the intracellular distributions of actin (microfilament), tubulin (microtubule), and vimentin (intermediate filament), as well as vinculin (a junctional protein connecting microfilaments to the cell membrane), in cultured corneal fibroblasts

Acanthamoeba are free-living amoebae found in most environments that can cause brain and corneal infections. To infect humans, these pathogens must interact with host cells and the extracellular matrix (ECM). In order to define the mode by which amoebae recognize ECM components and process this recognition, we analyzed Acanthamoeba culbertsoni attachment and invasion, respectively, on collagen I and laminin-1 and on tridimensional collagen I and matrigel matrices. We determined that amoebae surface proteins are involved in adhesion, that exogenous sugars can decrease adhesion and invasion, and

Programmed cell death (PCD), one of the main reasons of cell viability changes after cryopreservation. The microfilament cytoskeleton serves as a core modulator of cold stress responses and PCD signaling cascades, but the molecular mechanism linking microfilament cytoskeletal dynamics to PCD during seed cryopreservation has not yet been elucidated. Therefore, in this study, two types of Paeonia lactiflora seeds with different viability changes after cryopreservation were used as experimental materials. Taking the microfilament cytoskeleton as insertion point, we explored the mechanism of its a

Hepatic perisinusoidal cells (PSCs) proliferate and are thought to be the principal source of extracellular matrix proteins during the development of liver fibrosis. We have studied the classical model of carbon tetrachloride induced liver fibrosis in order to evaluate the possible modulation of PSCs into a synthetically active and contractile cell: the myofibroblast (MF). At the ultrastructural level, this modulation was characterized by reduction of lipid vacuoles and appearance of a developed rough endoplasmic reticulum as well as of microfilament bundles. On investigating the cytoskeletal

Crumbs homolog 3 (or Crumbs3, CRB3) is a polarity protein expressed by Sertoli and germ cells at the basal compartment in the seminiferous epithelium. CRB3 also expressed at the blood-testis barrier (BTB), co-localized with F-actin, TJ proteins occludin/ZO-1 and basal ES (ectoplasmic specialization) proteins N-cadherin/β-catenin at stages IV-VII only. The binding partners of CRB3 in the testis were the branched actin polymerization protein Arp3, and the barbed end-capping and bundling protein Eps8, illustrating its possible role in actin organization. The tubule-like cells (TLC) of the parietal layer of the Bowman's capsule of the renal corpuscle in normal male mice are provided with bundles of thin microfilaments (50-70 A). Moreover, the ultrastructural observations demonstrated that similar thin filaments (50-70 A) are located in the flattened cells of the parietal layer of the Bowman's capsule and in the tubular cells. All the above cells bind antimyosin-like antibodies. Considering the correspondence between the immunochemical and ultrastructural findings, it is suggested that the microfilaments located in TLC contain a 'myosin-like' pr

Ectoplasmic specialization (ES) is an actin-rich adherens junction in the seminiferous epithelium of adult mammalian testes. ES is restricted to the Sertoli-spermatid (apical ES) interface, as well as the Sertoli cell-cell (basal ES) interface at the blood-testis barrier (BTB). ES is typified by the presence of an array of bundles of actin microfilaments near the Sertoli cell plasma membrane. These actin microfilament bundles require rapid debundling to convert them from a bundled to branched/unbundled configuration and vice versa to confer plasticity to support the transport of 1) spermatids

Directional migration of tumor cells depends on mechanical forces generated by intracellular asymmetry. This study establishes an electric field-induced directional migration model using fluorescence tension probes to visualize microfilament forces and intracellular osmotic pressure dynamics. Electric fields induce protein kinase A (PKA)-dependent PDLIM7 recruitment, polarizing microfilament tension at cell edges via site-specific phosphorylation at serine 190. Concurrently, the electric field induces asymmetric changes in membrane potential, driven by protein nanoparticles and calcium ions, r

Huntington's disease (HD) is one of the inheritable neurodegenerative diseases, and these diseases share several similar pathological characteristics, such as abnormal neuronal morphology. miR-196a is a potential target to provide neuroprotective functions, and has been reported to enhance polymerization of neuronal microtubules in HD. While microtubules and microfilaments are two important components of the neuronal cytoskeleton, whether miR-196a improves neuronal microfilaments is still unknown. Here, we identify insulin-like growth factor 2 mRNA binding protein 3 (IMP3), and show that miR-1

We investigated the distribution of microtubules and microfilaments in rat thyroid follicular epithelial cells by applying an immunofluorescence technique with monoclonal antibodies against tubulin and by staining sections with rhodamine-phalloidin. In normal thyroid cells, microtubules run longitudinally from the apical region to the basal region intersecting with each other. In addition, intense labelling with tubulin antibodies was observed in the apical part of the cell. The ultrastructural examinations showed that microtubules often run along the apical plasma membrane. Dot-like labelling

Membrane microfilaments are found throughout the animal world in situations suggesting that they fulfill a critical role in providing normal cell shape and plasticity. We have hypothesized that hereditary spherocytosis, a congenital hemolytic anemia associated with intrinsically rigid and mishapen erythrocytes, might result from genetically defective microfilaments in erythrocyte membranes. By using three different drugs (vinblastine, colchicine, and strychnine) that share one common attribute-that of potently precipitating purified microfilamentous protein-we have provided support for this hy

Suppression of tropomyosins (TMs), a family of actin-binding, microfilament-associated proteins, is a prominent feature of many transformed cells. Yet it is unclear whether downregulation of TMs occur in human tumors. We have investigated the expression of tropomyosin-1 (TM1) in human breast carcinoma tissues by in situ hybridization and immunofluorescence. TM1 mRNA and protein are readily detectable in normal mammary tissue. In contrast, TM1 expression is abolished in the primary human breast tumors. Expression of other TM isoforms, however, is variable among the tumors. The consistent and pr

Microvilli isolated from 13762 mammary ascites tumor cells contain a major calcium-sensitive protein (AMV-p35) that can be isolated with microvillar microfilament cores prepared by Triton X-100 extraction in the presence but not absence of calcium. AMV-p35 can be readily purified from ethylene glycol bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid extracts of the microfilament cores by chromatography on an anion exchange column, to which it does not bind. Immunoblot analysis indicates that AMV-p35 is related to calpactin I, the pp60src tyrosine kinase substrate. In the presence of calciu

Polycystin-2 (PC2) is the product of the second cloned gene (PKD2) responsible for autosomal dominant polycystic kidney disease and has recently been shown to be a calcium-permeable cation channel. PC2 has been shown to connect indirectly with the actin microfilament. Here, we report a direct association between PC2 and the actin microfilament. Using a yeast two-hybrid screen, we identified a specific interaction between the PC2 cytoplasmic C-terminal domain and tropomyosin-1 (TM-1), a component of the actin microfilament complex. Tropomyosins constitute a protein family of more than 20 isofor

An integral sialoglycoprotein with Mr approximately 130,000 (Sgp 130) and highest expression in adult chicken gizzard smooth muscle has been recently identified as an excellent candidate for classification as a plasma membrane protein natively associated (directly or indirectly) with actin microfilaments (Rogalski, A.A., and S.J. Singer, 1985, J. Cell Biol., 101:785-801). In this study, the relative in situ distributions of the Sgp 130 integral species (a designation that also includes non-smooth muscle molecular forms) and the peripheral protein, vinculin, have been simultaneously revealed fo

WASp family proteins serve as conserved regulators of branched microfilament array formation via the Arp2/3 actin polymerization machinery. We have identified a specific role during spermatogenesis for the Drosophila WASp homolog (Wsp) and associated elements. Spermatogenesis within the fly testis is carried out in cysts, where a pair of somatic cyst cells encloses differentiating sperm. The final phase of the process involves the attachment of matured cysts to a specialized epithelium at the base of the testis, followed by release of individual motile spermatids into the adjoining seminal ves
