The many shapes of mitochondrial membranes.

The roles of mitochondria in cell death and in aging have generated much excitement in recent years. At the same time, however, a quiet revolution in our thinking about mitochondrial ultrastructure has begun. This revolution started with the use of vital dyes and of green fluorescent protein fusion proteins, showing that mitochondria are very dynamic structures that constantly move, divide and fuse throughout the life of a cell. More recently, some of the first proteins contributing to these various processes have been discovered. Our view of the internal structures of mitochondria has also ch

CLIC4 localizes to mitochondrial-associated membranes and mediates cardioprotection.

Mitochondrial-associated membranes (MAMs) are known to modulate organellar and cellular functions and can subsequently affect pathophysiology including myocardial ischemia-reperfusion (IR) injury. Thus, identifying molecular targets in MAMs that regulate the outcome of IR injury will hold a key to efficient therapeutics. Here, we found chloride intracellular channel protein (CLIC4) presence in MAMs of cardiomyocytes and demonstrate its role in modulating ER and mitochondrial calcium homeostasis under physiological and pathological conditions. In a murine model, loss of CLIC4 increased myocardi

Development of mitochondrial membranes in anaerobically grown yeast cells.

Biochemical analyses of mitochondrial marker substances, especially cardiolipin and oligomycin-sensitive ATPase [EC 3.6.1.3], as well as electron microscopic observations were carried out to eludicate the process of mitochondrial development in annaerobic yeast cells. Cardiolipin was found to be localized in the mitochondria in anaerobic cells. Its cellular content was a little higher in the stationary phase than in the exponential phase in glucose-grown cells and increased further in galactose-grown cells. The lipid content of the mitochondrial preparation obtained from glucose-grown stationa

Mitochondrial antibodies in primary biliary cirrhosis. I. Localization of the antigen to mitochondrial membranes.

The antigen reacting with complement-fixing antibodies in the sera of patients with primary biliary cirrhosis was localized predominantly in the mitochondrial fraction of tissue homogenates obtained by differential centrifugation. Purified mitochondrial preparations had a high content of the antigen whereas purified lysosomes failed to fix complement with PBC sera. Analysis of a number of fractionation experiments showed a high correlation between antigen content and the mitochondrial enzyme succinic dehydrogenase in all fractions. There was much poorer correlation with lysosomal and micrososo

FUNDC1-dependent mitochondrial-ER membranes mediate mitochondrial dysfunction and melanocyte damage under oxidative stress.

BACKGROUND: Mitochondria play a pivotal role in oxidative stress-induced melanocyte destruction in vitiligo. FUN14 domain containing 1 (FUNDC1), a mitochondrial outer-membrane protein, has an important role in mitochondrial function by regulating mitophagy and mitochondria-associated endoplasmic reticulum membranes (MAM). However, the role of FUNDC1 in melanocyte damage under oxidative remains unclear. OBJECTIVES: To determine the contribution of FUNDC1 to oxidative stress-triggered melanocyte damage in vitiligo. METHODS: We treated human melanocyte cell line PIG1 with H2O2 to establish an o

Lighting up mitochondrial membranes: Reading and writing the physical state of the inner mitochondrial membrane.

The mitochondrial inner membrane (IMM) is depicted as a scaffold housing the enzymes of the electron transport chain, allowing electron transfer and generation of the electrochemical gradient that drives respiration and adenosine triphosphate (ATP) synthesis. However, evidence from imaging, structural biology, and novel chemical probes reveals that the IMM is not a mere passive backdrop: its physical properties change with the metabolic state, and together these changes influence mitochondrial function. Here, we summarize how IMM microviscosity, lipid packing, and mesoscale organization might

[Specificity of interaction of hexokinase isozyme II with mitochondrial membranes].

Study on the mechanism of hexokinase isozyme II adsorption on mitochondrial membranes in the presence of 10 mM MgCl2 demonstrated that 0.16% of the total proteins of the soluble fraction and the total hexokinase pool are capable of reversible binding to the membrane. The plot for the dependence of the degree of enzyme adsorption on Mg2+ concentration is hyperbolic. Under these conditions, hexokinase competes favourably for the binding sites with lactate dehydrogenase and creatine kinase. Analysis of the adsorption capacity of natural and artificial phospholipid membranes showed that hexokinase…

Coenzyme Q10 Enhances Resilience of Mitochondrial-like Membranes Against Amyloidogenic Peptides.

Mitochondria possess a double-membrane envelope which is susceptible to insult by pathogenic intracellular aggregates of amyloid-forming peptides, such as the amyloid-beta (1-42) (Aβ42) peptide and the human islet amyloid polypeptide (hIAPP). The molecular composition of membranes plays a pivotal role in regulating peptide aggregation and cytotoxicity. Therefore, we hypothesized that modifying the physicochemical properties of mitochondrial model membranes with a small molecule might act as a countermeasure against the formation of, and damage by, membrane-active amyloid peptides. To investiga

Aspirin, acetaminophen and proton transport through phospholipid bilayers and mitochondrial membranes.

Mechanisms of proton transport were investigated in planar phospholipid bilayer membranes exposed to aspirin (acetylsalicylic acid), acetaminophen (4-acetamidophenol), benzoic acid and three aspirin metabolites (salicylic acid, gentisic acid and salicyluric acid). The objectives were to characterize the conductances and permeabilities of these weak acids in lipid bilayer membranes and then predict their effects on mitochondrial membranes. Of the compounds tested only aspirin, benzoate and salicylate caused significant increases in membrane conductance. The conductance was due mainly to proton …

Apoptotic Bax at Oxidatively Stressed Mitochondrial Membranes: Lipid Dynamics and Permeabilization.

Mitochondria are crucial compartments of eukaryotic cells because they function as the cellular power plant and play a central role in the early stages of programmed cell death (apoptosis). To avoid undesired cell death, this apoptotic pathway is tightly regulated by members of the Bcl-2 protein family, which interact on the external surface of the mitochondria, i.e., the mitochondrial outer membrane (MOM), and modulate its permeability to apoptotic factors, controlling their release into the cytosol. A growing body of evidence suggests that the MOM lipids play active roles in this permeabiliz

Molecular mechanism of mitochondrial membrane fusion.

Mitochondrial fusion requires coordinated fusion of the outer and inner membranes. This process leads to exchange of contents, controls the shape of mitochondria, and is important for mitochondrial function. Two types of mitochondrial GTPases are essential for mitochondrial fusion. On the outer membrane, the fuzzy onions/mitofusin proteins form complexes in trans that mediate homotypic physical interactions between adjacent mitochondria and are likely directly involved in outer membrane fusion. Associated with the inner membrane, the OPA1 dynamin-family GTPase maintains membrane structure and

Effect of tricyclic drugs on mitochondrial membrane.

The effects of tricyclic drugs (clomipramine, imipramine, chlorpromazine and promethazine) on isolated liver mitochondria of rats were examined. All the drugs tested accelerated state 4 respiration. Their stimulative potency at concentrations below 100 microM was in the order of chlorpromazine greater than clomipramine greater than imipramine, promethazine. On state 3 respiration, the chlorine containing drugs had an inhibitive effect at high concentrations, while the other drugs seemed to have a slightly stimulative effect. These drugs stimulated latent ATPase activity of mitochondria. Clomip

TMEM126A is a mitochondrial located mRNA (MLR) protein of the mitochondrial inner membrane.

BACKGROUND: Hereditary optic neuropathies (HONs) are a heterogeneous group of disorders that affect retinal ganglion cells (RGCs) and axons that form the optic nerve. Leber's Hereditary Optic Neuropathy and the autosomal dominant optic atrophy related to OPA1 mutations are the most common forms. Nonsyndromic autosomal recessive optic neuropathies are rare and their existence has been long debated. We recently identified the first gene responsible for these conditions, TMEM126A. This gene is highly expressed in retinal cellular compartments enriched in mitochondria and supposed to encode a mito

Aggregation and membrane activity of mutant A30P alpha-synuclein on mitochondrial membranes.

The A30P point mutation in the α-synuclein (α-syn) protein is linked to a familial form of Parkinson's disease (PD), the most common motor neurodegenerative disorder. The association of α-syn with lipids in cellular membranes plays a key role in modulating the aggregation behavior of the protein, as well as its toxicity. Here, we studied the impact of the A30P mutation on the interaction of α-syn with mitochondrial membranes, since the interplay between α-syn aggregation and mitochondrial dysfunction is central for the pathogenic process in PD, but still not completely understood. Experiments …
