Nifedipine overdose.

A 59-year-old man ingested 900 mg of nifedipine. Profound hypotension, sinus and atrioventricular node dysfunction, and hyperglycemia resulted. The patient's condition responded favorably to aggressive treatment with intravenous fluids, calcium, and dopamine. Similar effects and response to therapy have been reported in 17 cases of verapamil overdose and are briefly summarized. The effects of nifedipine overdose are discussed, and preliminary recommendations about its management are offered. With increasing use of nifedipine and other calcium channel blockers, more incidents of overdose can be…

[Electroanalysis of nifedipine].

Nifedipine is an electrochemically active compound. Reducibility of 2-nitrophenyl group on dropping mercury electrode renders polarographic determination of nifedipine possible. Reductive half-wave potential of nitro group depends on saturation of neighbouring pyridine ring i.e. 1,4-dihydropyridine is reduced on more negative potential than aromatic pyridine derivatives. The different half-wave potentials can be explained with the different stereo structures. On this basis differential pulse polarographic (DPP) method was developed for determination of nifedipine and its photolytic decompositi…

Nifedipine for primary dysmenorrhoea.

BACKGROUND: Dysmenorrhoea (period pain) is a common condition with a substantial impact on the well-being and productivity of women. Primary … evels of prostaglandins by contracting forcefully, causing low oxygen levels and consequently pain. Nifedipine is a calcium channel blocker in widespread clinical use for preterm labour due to its ability to inhibit uterine contractions in that setting. This review addresses whether this effect of nifedipine also helps with relief of the uterine contractions …

[Nifedipine: a novel vasodilator].

Nifedipine was synthesized by Bayer Germany in 1966 and considered for clinical use as a coronary vasodilator in patients with angina pectoris. Japanese investigators played a great part in the pre-clinical and clinical development of nifedipine. Professor Hashimoto demonstrated that nifedipine could increase coronary blood flow even in extremely low dosages, and Professor Kimura found that nifedipine is highly efficacious in preventing anginal attacks in patients with angina pectoris, particularly those with variant form of angina. In early trials it was noted that nifedipine decreased blood …

Hydrotropic solubilization of nifedipine.

Nifedipine (1), a drug practically insoluble in water, has been solubilized employing sodium benzoate (2) and sodium salicylate (3) as hydrotropes. In 30% w/v 2 solution the solubility of 1 increased 85 and 76 fold at 25 +/- 1 degree C and at 37 +/- 1 degree C respectively. The corresponding increase in solubility of 1 in 30% w/v 3 solution was 135 and 107 fold respectively. To study the mechanism of hydrotropic solubilization of 1, the solution properties of 2 and 3 over a concentration range of 1 to 30% w/v were undertaken. The probable mechanism involves a complexation type of interaction a…
