Paracrine communication between mechanically stretched myocytes and fibroblasts.

Mechanical stretch is a major factor for myocardial hypertrophy and heart failure. Stretch activates mechanical sensors from cardiac myocytes, leading to a series of signal transduction cascades, which can result in cell malfunction and remodeling. It is well known that mechanical stretch also induces the release of paracrine factors from cardiac fibroblasts, as well as myocytes. Due to complicated circumstance of heart tissue, it is difficult to fully investigate the characteristics of these factors in situ. Here we describe static stretch and conditioned medium experiments as methods to exam

Loss of GTPase activating protein neurofibromin stimulates paracrine cell communication via macropinocytosis.

Neurofibromin, the protein product of the neurofibromatosis type 1 (NF1) tumor suppressor gene, is a negative regulator of Ras signaling. Patients with mutations in NF1 have a strong predisposition for cardiovascular disease, which contributes to their early mortality. Nf1 heterozygous (Nf1+/-) bone marrow to wild type chimeras and mice with heterozygous recombination of Nf1 in myeloid cells recapitulate many of the vascular phenotypes observed in Nf1+/- mutants. Although these results suggest that macrophages play a central role in NF1 vasculopathy, the underlying mechanisms are currently unk

Pig conceptuses utilize extracellular vesicles for interferon-gamma-mediated paracrine communication with the endometrium†.

Interferon-gamma (IFNG) is a pro-inflammatory cytokine secreted by the porcine conceptus (embryo and extra-embryonic membranes) during the peri-implantation period of pregnancy. IFNG modifies the endometrial inflammatory immune response and is required for the implantation and survival of the conceptus. It is not known how IFNG from the conceptus trophectoderm is transported across the endometrial luminal epithelium (LE). In the present study, immunofluorescence analyses detected immunoreactive IFNG protein in both the trophectoderm and endometrial LE on Day 15 of pregnancy, while our previous

Oxidative stress induced by DMSA-IONPs impairs breast cancer cell migration and paracrine cell communication.

BACKGROUND: The migration of tumor cells requires a complex interplay of these cells with their microenvironment. Significantly, these migrating (metastatic) cells undergo metabolic and oxidative changes that render them more sensitive to shifts in their redox homeostasis. Nanoparticle (NP)-based pro-oxidant therapies are strategies that can potentially influence cell viability and migration. Among these, iron oxide NPs (IONPs) stand out due to their biocompatibility and the fact they induce ROS production through the Fenton Reaction. Here we investigate the possibility of using dimercaptosucc

Rab27a-Dependent Paracrine Communication Controls Dendritic Spine Formation and Sensory Responses in the Barrel Cortex.

Rab27a is an evolutionarily conserved small GTPase that regulates vesicle trafficking, and copy number variants of RAB27a are associated with increased risk of autism. However, the function of Rab27a on brain development is unknown. Here, we identified a form of paracrine communication that regulates spine development between distinct populations of developing cortical neurons. In the developing somatosensory cortex of mice, we show that decreasing Rab27a levels in late-born pyramidal neurons destined for layer (L) 2/3 had no cell-autonomous effect on their synaptic integration but increased e

Transcriptional network analysis of PTEN-protein-deficient prostate tumors reveals robust stromal reprogramming and signs of senescent paracrine communication.

Among the extensive genomic alterations in prostate cancer, phosphatase and tensin homolog (PTEN) deletion stands out as one of the most consistently observed events. PTEN loss in prostate tumors is primarily associated with cancer-cell proliferation and survival through the activation of the phosphoinositide 3-kinase (PI3K)-protein kinase B (AKT)-mechanistic target of rapamycin (mTOR) (PI3K-AKT-mTOR) signaling pathway. However, the use of PTEN as a robust biomarker in clinical practice is hampered by its complex epigenetic, transcriptional and post-translational regulation. In situ protein as

Extracellular vesicles are involved in the paracrine communication between epithelial cells in different regions of the domestic cat epididymis†.

Sperm maturation depends on exposure to microenvironments within the different segments of the epididymis, but mechanisms underlying how these microenvironments are produced or maintained are not well understood. We hypothesized that epididymal extracellular vesicles could play a role in the process of maintaining microenvironments in different regions of the epididymis. Specifically, we tested whether the extracellular vesicles from different regions of the epididymis can ensure paracrine communication between cells in different segments. Domestic cat tissues were used to develop a reproducib

Extracellular Vesicle Communication at the Neural Graft-Host Interface: From Bidirectional Paracrine Signaling to Graft Fate and Neural Circuit Integration.

Neural cell transplantation is progressing from nonspecific trophic support toward regionally defined cell replacement and functional circuit reconstruction. Yet transplanted cells do not develop in isolation; they remain embedded within a dynamic host environment composed of neurons, glia, vascular-associated cells, cerebrospinal fluid, and systemic signals. Extracellular vesicles (EVs) may provide a membrane-enclosed route for this reciprocal communication by transferring nucleic acids, proteins, lipids, and organelle-associated cargo across the graft-host interface. Moving beyond EVs as cel

Functional genomics of the dopaminergic system in hypertension.

Abnormalities in dopamine production and receptor function have been described in human essential hypertension and rodent models of genetic hypertension. Under normal conditions, D(1)-like receptors (D(1) and D(5)) inhibit sodium transport in the kidney and intestine. However, in the Dahl salt-sensitive and spontaneously hypertensive rats (SHRs) and in humans with essential hypertension, the D(1)-like receptor-mediated inhibition of epithelial sodium transport is impaired because of an uncoupling of the D(1)-like receptor from its G protein/effector complex. The uncoupling is receptor specific

Paracrine signaling mediators of vascular endothelial barrier dysfunction in sepsis: implications for therapeutic targeting.

Vascular endothelial barrier disruption is a critical determinant of morbidity and mortality in sepsis. Whole blood represents a key source of paracrine signaling molecules inducing vascular endothelial barrier disruption in sepsis. This study analyzes whole-genome transcriptome data from sepsis patients' whole blood available in the NCBI GEO database to identify paracrine mediators of vascular endothelial barrier dysfunction, uncovering novel insights that may guide drug repositioning strategies. This study identifies the regulated expression of paracrine signaling molecules TFPI, MMP9, PROS1

Do androgens influence hair growth by altering the paracrine factors secreted by dermal papilla cells?

Androgens regulate many aspects of human hair growth in both sexes. After puberty they transform tiny vellus follicles in many areas, e.g. the face, to terminal ones producing long, thick, pigmented hairs. In genetically predisposed individuals, androgens also cause the reverse transformation of terminal scalp follicles into vellus ones, causing balding. In the current hypothesis for androgen action, androgens control most follicular cells indirectly acting via the mesenchyme-derived dermal papilla which regulates many aspects of follicular activity. In this model androgens binding to androgen

Autocrine and paracrine regulation of pituitary endocrine cells.

The anterior pituitary gland is an important endocrine organ composed of a variety of key endocrine cells that regulate the functions of peripheral targeted organs, such as growth, development, behavior, lactation, reproduction and the stability of the internal environment and metabolism. This gland also regulates the functions of targeted peripheral endocrine organs, such as the thyroid, adrenal gland and gonads. In recent years, H&E and immunohistochemical staining, electron microscopic observation, pituitary endocrine cell line establishment and transplantation, flow cytometry, immunoblotti

Brain-heart interaction after acute ischemic stroke.

Early detection of cardiovascular dysfunctions directly caused by acute ischemic stroke (AIS) has become paramount. Researchers now generally agree on the existence of a bidirectional interaction between the brain and the heart. In support of this theory, AIS patients are extremely vulnerable to severe cardiac complications. Sympathetic hyperactivity, hypothalamic-pituitary-adrenal axis, the immune and inflammatory responses, and gut dysbiosis have been identified as the main pathological mechanisms involved in brain-heart axis dysregulation after AIS. Moreover, evidence has confirmed that the

The autocrine/paracrine regulation of thyrotropin secretion.

Peptides originally described in other tissues have been located in the anterior pituitary gland. Detection of their encoding mRNAs and specific receptors, together with demonstration of peptide local action led to the postulation of the existence of a paracrine/autocrine regulation of pituitary function. Direct evidence for the role of endogenous peptides has come from studies aiming to block their action through immunoneutralization or pharmacologic blockade. Here we review evidence of pituitary produced peptides as potential candidates as local regulators of thyrotropin secretion. Few studi

Large extracellular vesicles regulate endothelial angiogenic potential via paracrine and autocrine signaling.

Angiogenesis, a process associated with tumor growth and development, is often linked to advanced disease and poor clinical outcomes. Tumor cells establish a proangiogenic microenvironment through the release of paracrine signaling mediators, including extracellular vesicles (EVs). EVs have been shown to facilitate intercellular communication and encompass a diverse range of secreted vesicles, including small EVs, which range in size from ∼60 to 100 nm, and large EVs (L-EVs), which are even more diverse and range from 200 nm to >1 μm in size. Despite advancements in anti-angiogenic cancer ther

Design strategies for hydrogels with paracrine effects and their ability to promote wound healing: status and development.

Wound healing is a complex process involving various stages such as hemostasis, inflammation, cell proliferation, and tissue remodeling. If a wound fails to heal promptly, it can lead to ulcers, necrosis, and even limb amputation, resulting in significant losses. Over the years, many clinical studies have explored the role of dressings in wound healing. Although numerous dressings have been introduced to the market, traditional ones typically influence only external factors such as temperature, humidity, and pH, often causing issues like excessive moisture or dryness, allergic reactions, secon

Paracrine Regulation of Aldosterone Secretion in Physiological and Pathophysiological Conditions.

Aldosterone secretion by the zona glomerulosa of the adrenal cortex is controlled by circulating factors including the renin angiotensin system (RAS) and potassium. Mineralocorticoid production is also regulated through an autocrine/paracrine mechanism by a wide variety of bioactive signals released in the vicinity of adrenocortical cells by chromaffin cells, nerve endings, cells of the immune system, endothelial cells and adipocytes. These regulatory factors include conventional neurotransmitters and neuropeptides. Their physiological role in the control of aldosterone secretion is not fully

The Different Faces of the Pancreatic Islet.

Type 1 diabetes (T1D) patients who receive pancreatic islet transplant experience significant improvement in their quality-of-life. This comes primarily through improved control of blood sugar levels, restored awareness of hypoglycemia, and prevention of serious and potentially life-threatening diabetes-associated complications, such as kidney failure, heart and vascular disease, stroke, nerve damage, and blindness. Therefore, beta cell replacement through transplantation of isolated islets is an important option in the treatment of T1D. However, lasting success of this promising therapy depen
