Sensitization to p-phenylenediamine.

p-Phenylenediamine (PPD) from 0.2 mg to 1.6 mg in water or in petrolatum was applied as a patch test to 501 patients with different dermatoses, asthma, hay fever, and venereal diseases. Of these patients 10.6% reacted within the first five days indicating previous hypersensitivity and 14.4% between the sixth and the 16th day indicating the initiation of hypersensitivity to PPD by the patch application (flare-up). The reactions within the first five days were not correlated with the amount of PPD in the tests, but a statistically significant correlation was found between flare-ups and amount, s

Patch testing with hydroxyethyl-p-phenylenediamine sulfate - cross-reactivity with p-phenylenediamine.

BACKGROUND: Adverse reactions to permanent hair dyes are frequent, and primarily result from sensitization to p-phenylenediamine (PPD). OBJECTIVES: To investigate the degree of cross-reactivity to a chemically similar dye, hydroxyethyl-p-phenylenediamine sulfate (HPPS), and whether this might be a dyeing alternative for patients who are sensitive to PPD. METHOD: HPPS was patch tested in two concentrations in a total of 216 patients suspected of having contact dermatitis caused by hair dyes and/or hair cosmetics. A regular use test with a hair dye containing HPPS was suggested to every patien

Penetration and haptenation of p-phenylenediamine.

Although p-phenylenediamine (PPD) has been recognized as an extreme sensitizer for many years, the exact mechanism of sensitization has not been elucidated yet. Penetration and the ability to bind to proteins are the first two hurdles that an allergen has to overcome to be able to sensitize. This review is an overview of studies regarding PPD penetration through skin (analogues) and studies on the amino acids that are targeted by PPD. To complete this review, the auto-oxidation and N-acetylation steps involved in PPD metabolism are described. In summary, under normal hair dyeing exposure condi

p-Phenylenediamine sensitization and occupation.

BACKGROUND: p-Phenylenediamine (PPD) is an extreme delayed-type skin sensitizer, and is relevant in both occupational and non-occupational exposures. OBJECTIVES: To estimate the prevalence of PPD sensitization in a population of consecutive patients with suspected allergic contact dermatitis who attended units of dermatology or occupational medicine in north-eastern Italy and to investigate the association between their PPD sensitization and occupation. PATIENTS/MATERIALS/METHODS: A total of 14,464 patients (67.6% women and 32.4% men) with suspected allergic dermatitis underwent patch testin

Two sensitizing oxidation products of p-phenylenediamine patch tested in patients allergic to p-phenylenediamine.

BACKGROUND: The results from a previous study indicated the presence of several possible sensitizers formed during oxidation of the potent sensitizer p-phenylenediamine (PPD) to which PPD-sensitized patients might react, in various patterns. OBJECTIVES: To extract and analyse a yellow spot from a thin-layer chromatogram with oxidized PPD, to which 6 of 14 (43%) PPD-positive patients had reacted in a previous study, in order to identify potential sensitizer(s) and to patch test this/these substance(s) in the 14 PPD-positive patients. METHODS: The yellow spot was extracted from a thin-layer ch

Ecological risk of substituted p-phenylenediamines and quinones in Chinese freshwaters using QSAR-ICE modeling.

The high ecotoxicity of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPD-Q) has directed global attention to substituted p-phenylenediamines (PPDs) and their quinone derivatives (PPD-Qs). To date, however, comprehensive evaluations of their toxicity and associated ecological risks in aquatic ecosystems remain scarce. This study utilized an integrated approach combining quantitative structure-activity relationship and interspecies correlation estimation (QSAR-ICE) model predictions, species sensitivity distribution analysis, and ecological risk assessment to evaluate the potenti…

Base-catalyzed multicomponent access to quinoxalin-2-thiones from o-phenylenediamines, aryl ketones and sulfur.

3-Arylquinoxaline-2-thiones were conveniently synthesized via three-component oxidative condensation of acetophenones with o-phenylenediamines and sulfur in DMSO in the presence of piperidine as a catalyst. The products could be readily isolated from the reaction mixture by simple precipitation and washing with methanol. This set of reaction conditions applied to higher homologs of acetophenones as well as benzyl phenyl ketones led to 2,3-di-C-substituted quinoxalines. Further functionalization of 3-phenylquinoxaline-2-thione via reaction on the thione group could be readily performed to provi…

Allergic contact dermatitis to para-phenylenediamine.

Exposure to hair dye is the most frequent route of sensitisation to para-phenylenediamine (PPD), a common contact allergen. International studies have examined the profile of PPD, but Australian-sourced information is lacking. Patients are often dissatisfied with advice to stop dyeing their hair. This study examines patients' characteristics, patch test results and outcomes of PPD allergy from a single Australian centre, through a retrospective analysis of patch test data from 2006 to 2013 at the Liverpool Hospital Dermatology Department. It reviews the science of hair dye allergy, examines al

Sensitivity to para-phenylenediamine and intolerance to hydrochlorothiazide.

para-Phenylenediamine (PPD) is a contact allergen that cross-reacts with a variety of medications, including thiazide diuretics and sulfonamides. We present the case of a 52-year-old African American atopic woman who came for evaluation of a severe eyelid dermatitis and recurrent outbreaks of acneiform (follicular-based) pruritic papules on her face, chest, and back. These symptoms presented after the patient was started on hydrochlorothiazide for hypertension. The patient also reported a history of eyelid dermatitis associated with "black hair dye." Patch testing revealed a positive reaction
