Plasmodium falciparum and Plasmodium berghei: effect of magnesium on the development of parasitemia.

The in vitro growth of Plasmodium falciparum was reduced by 35 and 43% through high concentrations (5 mmole/liter) of magnesium in RPMI medium and magnesium-free medium, respectively, after 48 hr, whereas no significant inhibition could be observed under these conditions after 24 hr cultivation in the respective medium. Levels of magnesium between 0.5 and 3 mmole/liter showed no inhibitory effect on the in vitro growth of P. falciparum even after long-term exposure for 7 days. The 50 and 90% chloroquine inhibitory concentrations of the chloroquine-resistant strain K1 after 24 hr were reduced t

Quantitative characterization of hemozoin in Plasmodium berghei and vivax.

The incidence and global distribution of chloroquine resistant (CR) Plasmodium vivax infection has increased since emerging in 1989. The mechanism of resistance in CR P. vivax has not been defined. The resistance likely relates to the formation and disposition of hemozoin as chloroquine's primary mechanism of action involves disruption of hemozoin formation. CR P. berghei strains, like CR P. vivax strains, are confined to reticulocyte host cells and reportedly they do not accumulate appreciable intraerythrocytic hemozoin. Reports comparing hemozoin production between P. vivax strains and CR to

Plasmodium berghei: cloning of the circumsporozoite protein gene.

A DNA fragment encoding the carboxy terminal 80% of the Plasmodium berghei circumsporozoite protein was selected from a genomic DNA expression library. Sequencing revealed that the P. berghei circumsporozoite protein was similar in overall structure to circumsporozoite proteins from other malaria species, although the central repeat region was unique in comprising two different blocks of tandem peptide repeats: 11 eight amino acid repeats with predominant sequence DPAPPNAN were followed by 16 two amino repeats, predominantly PQ. The P. berghei circumsporozoite protein exhibited limited, but ab

Host sphingolipids support Plasmodium berghei liver stage development.

During the asymptomatic liver stage, Plasmodium resides within a parasitophorous vacuole (PV) that protects the parasite from immune clearance while also restricting nutrient exchange with its host cell. Although it is known that Plasmodium must scavenge resources from its environment, the specific nutrients sequestered and the mechanisms for transporting them to the PV are poorly understood, particularly during the liver stage. In this study, we investigated the role of host lipids and discovered that sphingolipids are critical for both Plasmodium berghei liver stage development and invasion.

Plasmodium berghei: eosinophilic depression of infection in mice.

The effects of eosinophilia on the course of Plasmodium berghei infection in mice were studied. Eosinophilia was induced by intravenous injection of Ascaris suum body fluid into the mice. Results indicated that eosinophils may play a role in the suppression of murine malaria. A significant reduction in parasitemias and increased survival time in eosinophilic mice occurred compared to mice not treated with A. suum body fluid. Reduction of parasitemia was effectively achieved when the mice were challenged with P. berghei, only after the level of eosinophils reached at least 10% of total white ce

Plasmodium berghei: physiopathological changes during infections in mice.

Using infections of Plasmodium berghei in laboratory mice, the following physiopathological changes were observed during the seven days of the infection: reduction in haematocrit, increases in parasitized erythrocytes, pulmonary oedema, hypothermia, formation of prostaglandin-like substances in the central nervous system, increases and decreases in plasma bradykininogen levels and leucocytosis, as evidenced by neutrophilia, lymphocytosis and monocytosis. We found no changes in total plasma protein levels and albumin/globulin ratio.

Thymic atrophy induced by Plasmodium berghei ANKA and Plasmodium yoelii 17XL infection.

The thymus is the anatomical site where T cells undergo a complex process of differentiation, proliferation, selection, and elimination of autorreactive cells which involves molecular signals in different intrathymic environment. However, the immunological functions of the thymus can be compromised upon exposure to different infections, affecting thymocyte populations. In this work, we investigated the impact of malaria parasites on the thymus by using C57BL/6 mice infected with Plasmodium berghei ANKA and Plasmodium yoelii 17XL; these lethal infection models represent the most severe complica

Aspects of immunosuppression during Plasmodium berghei infection in rats.

Depression in immunological responsiveness was manifested in phase with parasitaemia in rats infected with Plasmodium berghei. The spleen was the most affected organ. The response of spleen cells to phytohaemagglutinin (PHA) and the number of plaque forming cells among spleen cells of rats injected with sheep red blood cells (SRBC), were reduced especially at peak parasitaemia. At the onset of the disease the spleen was activated and the responses were amplified. Antibody titres in the serum revealed basically the same picture. Malaria changed also the dose response to antigens so that an over

C-Mannosyltransferase Is Essential for Malaria Transmission in Plasmodium berghei.

C-Mannosylation of the thrombospondin type I repeat (TSR) domains is one of the most important factors involved in their function. It occurs on the first tryptophan of the WXXWXXC conserved motif where the tryptophan is usually surrounded by arginine or lysine forming the ligand-binding stretch of this sticky domain. It is found in its canonical or modified forms in many Plasmodium proteins. TSR containing proteins such as thrombospondin-like anonymous protein (TRAP), circumsporozoite protein (CSP), CSP and TRAP related protein (CTRP), and secreted protein with altered thrombospondin repeat (S

Effects and Mechanism of Action of Artemisinin on Mitochondria of Plasmodium berghei.

OBJECTIVE: To study the antimalarial effects and mechanisms of artemisinin (Qinghaosu in Chinese, QHS) on mitochondria in mice infected with Plasmodium berghei. METHODS: A total of 108 C57 mice infected with Plasmodium berghei were randomly divided into 3 groups by weight: the control group, 200 and 400 mg/kg QHS groups. The two QHS treatment groups were further divided into 4 sub-groups with 12 animals each time according to the treatment time, 0.5, 1, 2, and 4 h. Normal saline was intragastrically (i.g.) administered to the control group. The other two groups received different doses of QHS

Development of a bsd-blasticidin selection system in Plasmodium berghei.

Plasmodium berghei is used as a rodent model for the study of malaria. However, multiple genetic manipulations are restricted by the paucity of selectable markers. The bsd-blasticidin selection system is widely used for eukaryotic cells; however, it could not previously be used for P. berghei due to toxicity to the rodent host. Here, we report the application of this selection system in P. berghei using an in vitro selection method. The desired bsd-integrated mutants are enriched by more than 90% within 2 weeks when using this system. Furthermore, the bsd marker can be used sequentially with e

Plasmodium berghei: inhibition of the sporogonous cycle by alpha-difluoromethylornithine.

alpha-Difluoromethylornithine (DFMO), an enzyme inhibitor of ornithine decarboxylase, inhibits the sporogonous cycle of the malaria parasite Plasmodium berghei in the mosquito vector Anopheles stephensi. DFMO was administered to the mosquitoes dissolved either in the sugar solution at their disposal in the cages or through blood meals taken from treated mice. The mice subsequently bitten by mosquitoes treated with DFMO by both routes of administration did not contract malaria.

The influence of riboflavin deficiency on Plasmodium berghei infection in rats.

Two experiments were done in which rats in various stages of riboflavin deficiency were infected with Plasmodium berghei. Various control groups were included to compare the influence of food restriction on the P. berghei infection with that of riboflavin deficiency, namely, pair-fed (PF), weight-matched (WM) and ad libitum-fed (C-AL) control groups. Riboflavin deficiency depressed maximum parasite counts by comparison with all control groups and the degree of depression was inversely related to the riboflavin status. Survival of animals with P. berghei infection was approximately 10 to 14 day

Plasmodium berghei-induced malaria decreases pain sensitivity in mice.

Various types of pain were reported by people with Plasmodium falciparum and were mostly attributed to a symptom of malarial infection. Neural processes of pain sensation during malarial infection and their contributions to malaria-related death are poorly understood. Thus, these form the focus of this study. Swiss mice used for this study were randomly divided into two groups. Animals in the first group (Pb-infected group) were inoculated with Plasmodium berghei to induce malaria whilst the other group (intact group) was not infected. Formalin test was used to assess pain sensitivity in both
