Plant-Derived Polynucleotides/Polydeoxyribonucleotides in Skin Biomaterials: Delivery Platforms and Bioactivity Attribution.

In the fields of dermatology and skin biomaterials, polynucleotides (PN) and polydeoxyribonucleotides (PDRN) typically refer to deoxyribonucleic acid (DNA)-based polymers or heterogeneous DNA-fragment mixtures rather than ribonucleic acid (RNA) polynucleotides. Recently, plant-derived PN/PDRN preparations, sourced from callus, adventitious roots, and plant cell culture systems, have emerged as a promising animal-free material class. However, current evidence remains predominantly preclinical and should not be construed as demonstrating clinical equivalence to conventional animal-derived PDRN.

Polydeoxyribonucleotides (PDRNs) From Skin to Musculoskeletal Tissue Regeneration via Adenosine A2A Receptor Involvement.

Polydeoxyribonucleotides (PDRNs) are low molecular weight DNA molecules of natural origin that stimulate cell migration and growth, extracellular matrix (ECM) protein production, and reduce inflammation. Most preclinical and clinical studies on tissue regeneration with PDRNs focused on skin, and only few are about musculoskeletal tissues. Starting from an overview on skin regeneration studies, through the analysis of in vitro, in vivo, and clinical studies (1990-2016), the present review aimed at defining the effects of PDRN and their mechanisms of action in the regeneration of musculoskeletal

The Role of Polydeoxyribonucleotide as a Biotherapy for Musculoskeletal Disorders.

Polydeoxyribonucleotide (PDRN) is a DNA-derived biological therapy gaining prominence in the treatment of musculoskeletal disorders. It functions as an adenosine A2A receptor (A2AR) agonist and serves as a substrate in the nucleotide salvage pathway. These mechanisms drive anti-inflammatory effects, stimulate angiogenesis via vascular endothelial growth factor, and promote collagen synthesis. In osteoarthritis, PDRN reduces cartilage degradation and promotes the chondrogenic differentiation of stem cells. PDRN has shown therapeutic potential in tendinopathies, including Achilles and rotator cu

Viscoelastic interactions between polydeoxyribonucleotide and ophthalmic excipients.

This study investigated the interaction between polydeoxyribonucleotide (PDRN) and several ionic and nonionic isotonic agents, thickeners and a preservative that were employed as excipients in ophthalmic preparations. Interaction of each individual excipient and PDRN aqueous solution was evaluated by analyzing their rheological properties. Rheological properties of PDRN solutions were evaluated by dynamic oscillatory shear tests and values of elastic modulus (G'), viscous modulus (G″) and loss tangent (tan δ) were used to assess the relative changes in viscoelastic properties. In vitro polydeoxyribonucleotide effects on human pre-adipocytes.

OBJECTIVES: Adipose tissue is the most abundant and accessible source of adult stem cells. Human processed lipoaspirate contains pre-adipocytes that possess one of the a characteristic pathways of multipotent adult stem cells and are able to differentiate in vitro into mesenchymal and also neurogenic lineages. Because stem cells have great potential for use in tissue repair and regeneration, it would be significant to be able to obtain large amounts of these cells in vitro. As demonstrated previously, purine nucleosides and nucleotides mixtures can act as mitogens for several cell types. The a

Polydeoxyribonucleotide Treatment in Genital Lichen Sclerosus in Males.

It has been reported that treatment with polydeoxyribonucleotide (PDRN) in males affected by lichen sclerosus significantly improves the overall conditions of treated patients. It confirms that PDRN has a potent cytoprotective action as in other pathological conditions. We believe that multicentric randomized clinical trials are necessary for testing the benefits of this promising agent.

Effect of Polydeoxyribonucleotide Injection in a Patient With Carpal Tunnel Syndrome.

Ultrasound-guided corticosteroid injection into the carpal tunnel is widely performed for the management of carpal tunnel syndrome. However, this injection may worsen the condition in patients with type 2 diabetes mellitus. Polydeoxyribonucleotide was recently recognized as a substitute for corticosteroid because of its anti-inflammatory effects. A 41-yr-old woman with type 2 diabetes mellitus was admitted to our rehabilitation clinic with a 1-mo history of numbness and tingling in her right hand and first three fingers tips. She declined corticosteroid treatment because of a previous episode

Air-Botulinum Neurotoxin and Polydeoxyribonucleotide Injections for Acne Scar Treatment.

INTRODUCTION: Botulinum toxin injections, commonly used for wrinkle reduction, have been found to improve skin quality, including elasticity and texture, when used intradermally. Recent studies suggest that combining botulinum toxin with other treatments, like polydeoxyribonucleotide (PDRN), may enhance the effectiveness of scar treatments. This case study explores the efficacy of combining air-botulinum toxin injections with PDRN for treating acne scars. METHODS: A prospective, open-label, randomized study involved 17 participants with Grade 2-5 acne scars. They were divided into 3 groups: G

Engineering zein/polydeoxyribonucleotide nanocomposite particles as stable and multifunctional carriers.

Reactive oxygen species (ROS)-induced oxidative stress impedes efficient wound healing, while the therapeutic application of hydrophobic bioactive compounds is severely limited by poor water solubility, stability, and bioavailability. Herein, naturally derived zein/polydeoxyribonucleotide (Zein/PDRN) nanocomposite particles were fabricated through an antisolvent method to stabilize curcumin and enhance its biological activity. PDRN regulated zein self-assembly in a concentration-dependent manner, producing homogeneous nanocomposite particles with enhanced colloidal stability under diverse envi

Beneficial Effect of Polydeoxyribonucleotide in the Treatment of Actinic Cheilitis: A Case Report.

Actinic cheilitis (AC) is considered a potentially malignant disorder characterized by epithelial atrophy, chronic inflammation, and the progressive deterioration of the tissue architecture of the vermilion border of the lip. Currently, there is no effective minimally invasive treatment to reverse the progression of this pathology. The use of polydeoxyribonucleotide (PDRN) has demonstrated benefits across various dermatologic and wound-healing contexts due to its regenerative, angiogenic, and anti-inflammatory properties. This study aims to evaluate the healing effect of PDRN on AC lesions. Th

Genital Lichen Sclerosus in Male Patients: A New Treatment with Polydeoxyribonucleotide.

Lichen sclerosus (LS) is an inflammatory and chronic disease that causes itching, pain, dysuria, urinary retention, dyspareunia and sexual dysfunction, in both men and women. The first line pharmacological treatment is based on the use of topical steroids, which have proved their efficacy in 60-70% of cases but with a high rate of relapses in time (50-80% of the patients of both sexes). The purpose of our non-randomised prospective pilot study was to evaluate the efficacy and tolerability of a new loco regional therapy with polydeoxyribonucleotides (PDRN) in the treatment of male genital LS.
