Quantification of gait in dystonic Gunn rats.

Spontaneously jaundiced Gunn rats exposed to sulfadimethoxine develop bilirubin encephalopathy (kernicterus) with hearing loss and dystonia, closely resembling the human syndrome. We recently characterized the electromyographic activity in this animal model supporting our clinical impression of dystonia. The objective of this study was to develop a simple, non-invasive method to quantify the motor deficits in dystonic rodents. On postnatal day 16, Gunn rats were treated with 100mg/kg of sulfadimethoxine or saline. On postnatal day 31, the ventral view of the animals was videotaped while the an

Diminished hepatic triiodothyronine production in Gunn rats.

The liver is a major site of conversion of thyroxine (T4) to the more active thyroid hormone 3,5,3'-triiodothyronine (T3). Hepatic T4 to T3 conversion is altered by a variety of pathological processes and pharmacological agents. We studied T4 to T3 conversion in glucuronyl transferase deficient homozygous Gunn rats because they have a hepatic enzyme abnormality which leads to hyperbilirubinaemia, and also because they have been reported to have alterations in thyroid hormone metabolism. An in vitro incubation system employing the 10,000 X g supernatant of liver homogenate was used, and T3 prod

Hepatotoxicity of phenacetin and paracetamol in the Gunn rat.

Both phenacetin and paracetamol produce acute centrilobular liver necrosis in the homozygous Gunn rat. Paracetamol is more hepatotoxic than phenacetin, and both are more hepatotoxic to the homozygous Gunn rat than to the heterozygous Gunn rat or to the albino rat. These findings have relevance to the role of the compounds in the clinical syndromes of paracetamol induced liver necrosis and analgesic nephropathy.

Thyroid hormone metabolism and nuclear binding in Gunn rats.

We examined the serum concentrations of total, free thyroid hormones and TSH, activity of hepatic T4 5'-deiodinase, and T3 binding to hepatic nuclei in homozygous (j/j) and heterozygous (j/+) Gunn rats. Both total T3 and free T3 (FT3) concentrations in sera from j/j rats were significantly lower than those of j/+ rats on 5-10, 15-20, and 25-30 days after birth. Both total T4 and free T4 (FT4) concentrations in j/j and j/+ rat sera were not significantly different on 5-10 days. However, in j/j rats they were significantly higher than those of j/+ rats on days 15-20 and 25-30. Serum reverse T3 (

Pharmacokinetic studies of the enterohepatic circulation in Gunn rats.

Pharmacokinetic investigations of bilirubin resorption by the intestine of Gunn rats have shown that the excretion of bilirubin is markedly dependent upon the frequency of defaecation. The serum bilirubin concentration is strongly affected by obstipation or diarrhea, so therefore the influence of diet upon this parameter in Gunn rats was investigated. Not only natural breast milk but also adapted milk strongly inhibited the resorption of bilirubin. This cannot be attributed to an increased defaecation frequency. Instead it is postulated that the resorption of bilirubin is hindered due to a mas

Experimental pyelonephritis and papillary necrosis in the Gunn rat.

Homozygous and heterozygous female Gunn rats show increased susceptibility to experimental urinary infection. The strain develops pyelonephritis after intravesical inoculation of Proteus mirabilis in numbers which fail to induce the lesion in albino rats, and severe pyelonephritis is frequently complicated by papillary necrosis. The basis for this enhanced susceptibility has not been defined, but the occurrence of the phenomenon in both homozygous and heterozygous rats indicates that it is not caused primarily by high plasma levels of unconjugated bilirubin or by the deposition of bilirubin in

The Gunn rat: an experimental model for central deafness.

The Gunn rat which develops neonatal hyperbilirubinemia has been used as an experimental model to evaluate the effect of bilirubin on the auditory system. Electrocochleographic and morphological studies (including light microscopy, surface preparations and transmission electron microscopy) did not reveal any cochlear abnormality in homozygous Gunn rats. Brainstem auditory evoked potentials showed morphological and amplitude changes suggesting a functional damage in the brainstem auditory pathways. These results suggest that hearing loss, when observed in kernicterus, is primarily due to neuron

Reproductive abnormalities in the prepubertal jaundiced Gunn rat.

The female, jaundiced Gunn rat has been previously reported to be infertile. This study describes some abnormalities in the reproductive system of the prepubertal jaundiced female rat. Vaginal opening occurred at the time of weaning, which was approximately 7 days earlier when compared to the nonjaundiced female littermates. Following vaginal opening, the jaundiced rat showed persistent vaginal cornification. The estimated age at first ovulation in the jaundiced rat was 52 days. The uteri of jaundiced rats were significantly larger than those of nonjaundiced rats by 17 days after birth. The ac

Impaired fertility in the jaundiced female (Gunn) rat.

The hyperbilirubinemic female Gunn rat has been reported to have impaired fertility. A total of 267 jaundiced (j/j) and 91 nonjaundiced (+/j) female Gunn rats were used in a series of experiments to characterize the nature of this reduced fertility. Sixteen percent of the jaundiced females mated and delivered litters which were characteristically small in number (4.5 pups). A comparison of the gross observations at necropsy of jaundiced and nonjaundiced pregnant rats indicated that the number of implantation sites and live fetuses were significantly lower in the jaundiced females. The number o

Effect of hyperbilirubinemia on the inner ear in Gunn rats.

The effect of hyperbilirubinemia on the inner ear was studied in fifteen jaundiced homozygous Gunn rats. Seven non-jaundiced heterozygous littermates served as controls. Histopathological examination of the temporal bone in these animals showed normal sensory and neuronal structures of the inner ear. This cannot be taken as conclusive evidence that a central, rather than a peripheral, lesion is the cause of hearing loss in hyperbilirubinemia.

Effect of intermittent phototherapy on bilirubin dynamics in Gunn rats.

To determine whether continuous phototherapy is necessary to control neonatal jaundice, groups of jaundiced (Gunn) rats were exposed to four blue light regimens: continuous light, 30 min light/30 min dark, 6 min light/6 min dark, and 6 min light/18 min dark. An exponential decrease in serum bilirubin concentration (SBC) was found with all regimens. A logarithmic dose response curve was obtained with a 50% light dose exhibiting 75% efficacy, and a 25% light dose exhibiting 59% efficacy compared to continuous phototherapy. The time constants of the decrease in serum bilirubin concentration also

Salicylate nephropathy in the Gunn rat: potential role of prostaglandins.

We examined the potential role of prostaglandins in the development of analgesic nephropathy in the Gunn strain of rat. The homozygous Gunn rats have unconjugated hyperbilirubinemia due to the absence of glucuronyl transferase, leading to marked bilirubin deposition in renal medulla and papilla. These rats are also highly susceptible to develop papillary necrosis with analgesic administration. We used homozygous (jj) and phenotypically normal heterozygous (jJ) animals. Four groups of rats (n = 7) were studied: jj and jJ rats treated either with aspirin 300 mg/kg every other day or sham-treated

Hepatic excretion of circulating bilirubin photoproducts in the Gunn rat.

To investigate the origin and metabolism of the intermediates that occur in blood during phototherapy of neonatal jaundice, serum from irradiated homozygous Gunn rats was injected intravenously into other homozygous Gunn rats fitted with bile fistulas, and the excretion of pigment in the bile of the recipient rats was studied. In some experiments the donor rats were labeled with [14C]bilirubin; in others the recipient rats were labeled. Injection of donor serum from irradiated rats caused a transient burst of pigment excretion in the bile of the recipient rats. However, simultaneous bursts of

The effect of Na benzoate on serum bilirubin of the Gunn rat.

The administration of Na benzoate to grown and suckling Gunn rats in single doses of 7 and 35 mg/kg failed to significantly alter serum bilirubin concentrations. These doses are comparable to quantities of Na benzoate contained in injectable diazepam used therapeutically for newborn infants. Repeated doses of 7 mg/kg in the grown rat showed no effect, as well. A single dose of 100 or 200 mg/kg of Na benzoate and repeated doses of 35 mg/kg resulted in depressed serum bilirubin concentrations. The higher concentrations of Na benzoate, however, greatly exceed amounts contained in doses of diazepa
