The effect of vasopressin on renal blood flow and its distribution in the rat.

Anaesthetized Brattleboro rats with hereditary diabetes insipidus were infused with vasopressin at three different doses (1.3, 13 or 130 mu./hr) in order to study the effect of the hormone on renal blood flow and its distribution. Radioactive microspheres were used to determine intrarenal blood flow. The plasma vasopressin level during infusion of the lowest dose was calculated to be within the physiological range. At this dose vasopressin was antidiuretic but was without effect on arterial blood pressure or solute excretion, whereas the two higher doses were both pressor and natriuretic. All

Effects of saralasin on blood pressure, renin and renal blood flow in unilateral renovascular hypertension.

The effect of saralasin, a competitive inhibitor of angiotensin II, was assessed in hypertensive patients with unilateral renal artery stenosis after furosemide application. A significant fall of systemic arterial blood pressure, an increase of renal venous renin activity, significantly on the stenosed side in patients without arteriosclerosis of the contralateral kidney, and an almost equal decrement of renal blood flow in both kidneys were observed. Conceivably saralasin exerts different sodium-dependent effects on peripheral angiotensin II and specific intrarenal vascular receptors.

Effects of Norepinephrine on Renal Cortical and Medullary Blood Flow in Atherosclerotic Rabbits.

OBJECTIVE: The aim of this study was to explore the effect of norepinephrine (NE) on renal cortical and medullary blood flow in atherosclerotic rabbits without renal artery stenosis. METHODS: Atherosclerosis was induced in 21 New Zealand white rabbits by feeding them a cholesterol-rich diet for 16 weeks. Thirteen healthy New Zealand white rabbits were randomly selected as controls. After atherosclerosis induction, standard ultrasonography was performed to confirm that there was no plaque or accelerated flow at the origin of the renal artery. Contrast-enhanced ultrasound (CEUS) was performed a

Effects of prophylactic indomethacin on renal and intestinal blood flows in premature infants.

BACKGROUND: Prophylactic indomethacin reduces severe intraventricular hemorrhage and symptomatic patent ductus arteriosus in premature infants. The purpose of this study was to investigate the effects of prophylactic low-dose indomethacin on renal and intestinal blood flow. METHODS: Subjects were 19 extremely low-birthweight infants admitted to our hospital and enrolled in a multicenter randomized control trial to study the efficacy and complications of prophylactic low-dose indomethacin in the reduction of severe intraventricular hemorrhage and patent ductus arteriosus (indomethacin and plac

The effect of the inhibition of prostaglandin synthesis on renal blood flow in fetal sheep.

Treatment with prostaglandin synthesis inhibitors has been associated with oligohydramnios in the fetus. The presumed mechanism is a reduction in fetal renal blood flow. We examined the effect of meclofenamate administration on renal blood flow in chronically catheterized fetal sheep during normoxia and during moderate and severe hypoxia. Ten fetal sheep were made hypoxic twice at least 4 days after surgery: once in the presence and once in the absence of meclofenamate infusion. Renal blood flow and combined ventricular output were measured with radioactive microspheres. Prostaglandin synthesi

Effect of ACE inhibition on the fetal kidney: decreased renal blood flow.

The kidneys of nine fetuses whose mothers were chronically hypertensive were examined microscopically. Three of these mothers used antihypertensive agents throughout pregnancy including one who used an angiotensin-converting enzyme (ACE) inhibitor. The tubular defects found in these kidneys were compared to the kidneys of 20 normal controls, 13 fetuses with various multiple malformation syndromes and six cases of the twin to twin transfusion syndrome. Evidence from these cases as well as the literature suggest that the primary mechanism by which ACE inhibitors affect development of the fetal k

Effect of controlled ventilation on renal and splanchnic blood flows during nicotine.

The present study was undertaken to evaluate the influence of respiratory condition [free breathing (FB) vs. controlled ventilation (CV)] and of anesthetic [pentobarbital (PA) vs. chloralose (CA)] on nicotine-induced vasomotor responses in the renal cortex and splanchnic beds. Nicotine (36 micrograms . kg-1 . min-1 iv) was infused in four groups of dogs: group I, PA and CV; group II, PA and FB; group III, CA and CV; group IV, CA and FB. Regional vascular conductances (VC) were calculated from regional blood flows measured with 15-microns radioactive microspheres. In group I, VC fell in renal c

Effects of indomethacin on fetal renal function, renal and umbilicoplacental blood flow and lung liquid production.

The effects of indomethacin (10 mg/kg i.v. to the ewe and 12 mg/kg i.v. to the fetus) were examined in 8 chronically catheterized fetal sheep (117-138 days gestation). These doses suppressed fetal 6-keto-prostaglandin F1 alpha and thromboxane B2 levels. Fetal arterial PO2 increased (P < 0.01); PCO2 (P < 0.001) and pH fell (P < 0.001) and arterial pressure did not change. Placental blood flow increased in 4 of the 5 fetuses in which blood flows were measured. Lung liquid flow rate fell (P < 0.001). Fetal renal blood flow did not change but its distribution did, i.e. flow to the inner part of th

Effects of vibration, noise and restraint on heart rate, blood pressure and renal blood flow in the pig.

Pigs and other farm animals are usually transported at least once during their lives; for example, all meat animals are finally taken to the abattoir for slaughter. The vibration, noise and handling associated with such transportation is usually novel to the animals, and therefore constitutes a potential 'stressor'. Such adverse stimuli may trigger physiological and psychological changes which rapidly produce profound changes in blood flow to peripheral organs, particularly the kidney, whose blood vessels are richly supplied with sympathetic nerve fibres. In the present study, ultrasonic techn

[A method of determining separate effective renal blood flow by dynamic scintigraphy of the kidneys using I-131-hippurate].

The authors present a mathematical model for determining the separated effective renal blood flow (ERB) and a computer program which was used to determine separate parameters of the ERB in 56 patients including 35 with a unilateral lesion of the kidneys (20 patients with renovascular hypertension and 15 with unilateral pyelonephritis) and 21 patients with symmetrical renal damage (12 patients with chronic diffuse glomerulonephritis and 9 with essential hypertension). The patients with a unilateral renal lesion exhibited a significant decrease in the ERB parameters on the affected side whilst t

Differential effect of frusemide on renal medullary and cortical blood flow in the anaesthetised rat.

In addition to its known effect on renal tubular transport, frusemide (furosemide) has been shown to affect renal circulation. This study in the anaesthetised rat examined the influence of frusemide (bolus 0.25 or 0.5 mg kg(-1) I.V., then infusion delivering the same dose over 1 h) on renal cortical and medullary circulation measured as laser-Doppler blood (cell) flux. The responses were compared with simultaneously measured changes in renal excretion and in the tissue admittance, an index of medullary ionic hypertonicity of the interstitium. Renal vascular responses to frusemide were signific

Different effects of various vasodilators on autoregulation of renal blood flow in anesthetized dogs.

In order to examine whether the autoregulation of renal blood flow is equally influenced by all kinds of vasodilators, kidney perfusion experiments were performed in anesthetized dogs. The perfused kidney usually showed excellent autoregulation of blood flow over the perfusion pressure between 120 and 200 mmHg. Renal blood flow was increased by the renal arterial infusion of diltiazem (100 micrograms/min), papaverine (10 mg/min) or nicorandil (300 micrograms/min) (at the basal perfusion pressure of 100 mmHg) and was maintained at an increased level while the infusion was continued. Inhibitory effects of trifluoperazine and chlorpromazine, calmodulin inhibitor, on autoregulation of renal blood flow.

An intra-arterial infusion of a calmodulin inhibitor trifluoperazine (3 mg/min) or chlorpromazine (5 mg/min) caused obvious inhibition of autoregulation of renal blood flow in dogs. Simultaneous infusion of CaCl2 (30 mg/min) or Ca channel activator BAY K 8644 (5 micrograms/min) with these calmodulin inhibitors reduced renal blood flow, but could not block the inhibitory activity of calmodulin inhibitors on the autoregulation. The present experiment shows the contribution of the Ca-calmodulin system to the mechanism of autoregulation of renal blood flow.

Effects of diltiazem on renal blood flow and renal function during angiotensin II infusion in anesthetized dogs.

Effects of diltiazem on renal blood flow and renal function were studied under angiotensin II-loaded condition in anesthetized dogs. Intravenous infusion of angiotensin II at a rate of 0.25 microgram/kg/min produced an increase in blood pressure by 30 to 40 mmHg and a decrease in renal blood flow by 30 to 40%, resulting in a 100% increase in renal vascular resistance. Under this condition, diltiazem exhibited a dose related increase in renal blood flow not only by intrarenal administration (1-30 micrograms/kg) but also by intravenous administration (10-200 micrograms/kg). The effect was greatl
