Caledochytrium aldermanii gen. Et sp. nov.; a new Labyrinthulomycete displaying a novel reproductive strategy.

The morphology, reproduction, molecular phylogeny and chemical physiology of a new genus and species of thraustochytrid, Caledochytrium aldermanii gen. et sp. nov is described here. Caledochytrium aldermanii has, amongst its means of reproduction, a novel method not previously reported, in which a mature cell vacuolates and produces secondary cell(s) within the vacuole. Daughter cells are released by rupture of the mother cell. The daughter cell may already contain a tertiary cell within it. Caledochytrium aldermanii displays a range of dispersal mechanisms from sporangia, including unflagella

Chemotaxis of zoospores of Aurantiochytrium limacinum SR21.

Chemotactic responses of Aurantiochytrium limacinum SR21 to 90 distinct compounds including sugars, organic acids, alcohols, amino acids, amines, aromatic compounds, sulfoxides, vitamins, nucleosides, and inorganic compounds were analyzed. While the zoospores were significantly attracted to several amino acids, they did not respond to their typical nutrients such as sugars or organic acids, suggesting that the chemotaxis may not be directly involved in the search for their nutrition. Zoospores were also attracted to vanillin, which is a component of lignin and a major component of land plant c

ICTV Virus Taxonomy Profile: Bacilladnaviridae 2026.

Bacilladnaviridae is a family of viruses with single-stranded, circular DNA genomes that range from 3.4 to 6 kb and contain a short double-stranded region of 0.5-1 kb. The genome is replicated by the rolling-circle mechanism. Bacilladnavirids encode a jelly-roll capsid protein which forms non-enveloped icosahedral capsids with a T=3 symmetry. All cultured bacilladnavirids infect diatoms, a group of unicellular algae (Stramenopiles), and cause lysis of the host cells. This is a summary of the International Committee on Taxonomy of Viruses (ICTV) Report on the family Bacilladnaviridae, which is

Lethal effects of ichthyotoxic raphidophytes, Chattonella marina, C. antiqua, and Heterosigma akashiwo, on post-embryonic stages of the Japanese pearl oyster, Pinctada fucata martensii.

The inimical effects of the ichthyotoxic harmful algal bloom (HAB)-forming raphidophytes Heterosigma akashiwo, Chattonella marina, and Chattonella antiqua on the early-life stages of the Japanese pearl oyster Pinctada fucata martensii were studied. Fertilized eggs and developing embryos were not affected following exposure to the harmful raphidophytes; however, all three algal species severely affected trochophores and D-larvae, early-stage D-larvae, and late-stage pre-settling larvae. Exposure to C. marina (5×102cellsml-1), C. antiqua (103cellsml-1), and H. akashiwo (5×103cellsml-1) resulted

An improved protocol for the isolation of total genomic DNA from Labyrinthulomycetes.

Many protocols have been used for extraction of DNA from Thraustochytrids. These generally involve the use of CTAB, phenol/chloroform and ethanol. They also feature mechanical grinding, sonication, N2 freezing or bead beating. However, the resulting chemical and physical damage to extracted DNA reduces its quality. The methods are also unsuitable for large numbers of samples. Commercially-available DNA extraction kits give better quality and yields but are expensive. Therefore, an optimized DNA extraction protocol was developed which is suitable for Thraustochytrids to both minimise expensive

Hydrogen-assisted nannochloropsis sp. microalgae biodiesel to operate cleaner diesel engines.

The need to find sustainable energy sources has taken the center stage in the light of the growing energy global requirements. The renewable energy sources have also taken much interest in biofuels, as they promise to add renewable and environmentally benign sources of energy. The feedstock used in biodiesel is usually a wide range of inexpensive feedstock. In the current research, a new synthesis method using a feedstock with a green microalga, namely, Nannochloropsis sp. is defined. According to the authors, bio-diesel blends based on this microalgal source would help in producing less toxic

Sustainable production of eicosapentaenoic acid-rich emulsion from Nannochloropsis gaditana by nitrogen regulation and physical cell disruption.

Nannochloropsis gaditana is a promising novel food ingredient; however, its utilization remains limited due to its rigid bilayer cell wall. In this study, co-enrichment of eicosapentaenoic acid (EPA) and protein was achieved by supplementing with 1.76 mmol/L NaNO3, yielding biomass enriched with functional nutrients. N. gaditana emulsions were prepared using three cell disruption methods: high-pressure microfluidization (HPM), ultrasonication (US), and bead milling (BM). HPM-treated emulsions showed the highest release of soluble protein (2.52 g/100 mL), lipids (0.53 g/100 mL), and EPA (0.27 g

Interactive effects of temperature and salinity on the growth and cytotoxicity of the fish-killing microalgal species Heterosigma akashiwo and Pseudochattonella verruculosa.

Fish-killing blooms of Heterosigma akashiwo and Pseudochattonella verruculosa have been devastating for the farmed salmon industry, but in Southern Chile the conditions that promote the growth and toxicity of these microalgae are poorly understood. This study examined the effects of different combinations of temperature (12, 15, 18 °C) and salinity (10, 20, 30 psu) on the growth of Chilean strains of these two species. The results showed that the optimal growth conditions for H. akashiwo and P. verruculosa differed, with a maximum rate of 0.99 day-1 obtained at 15 °C and a salinity of 20 psu f

Growth and alkaline phosphatase activity of Chattonella marina and Heterosigma akashiwo in response to phosphorus limitation.

The growth and alkaline phosphatase activity (APA) of two raphidophyceae species Chattonella marina and Heterosigma akashiwo were investigated in response to P-limitation and subsequent addition of dissolved inorganic phosphorus (DIP, NaH2PO4) and two dissolved organic phosphorus (DOP) compounds: guanosine 5-monophosphate (GMP) and triethyl phosphate (TEP). APA levels increased greatly after P-starvation as the decrease of the cellular phosphorus quotes (Qp). C. marina responded to P-limitation quickly and strongly, with 10-fold increase in APA within 24 hr after P-starvation. The larger diffe

The Myb-like transcription factor phosphorus starvation response (PtPSR) controls conditional P acquisition and remodelling in marine microalgae.

Phosphorus (P) is one of the limiting macronutrients for algal growth in marine environments. Microalgae have developed adaptation mechanisms to P limitation that involve remodelling of internal phosphate resources and accumulation of lipids. Here, we used in silico analyses to identify the P-stress regulator PtPSR (Phaeodactylum tricornutum phosphorus starvation response) in the diatom P. tricornutum. ptpsr mutant lines were generated using gene editing and characterised by various molecular, genetics and biochemical tools. PtPSR belongs to a clade of Myb transcription factors that are conser

The influence of carbon limitation on growth of Heterosigma akashiwo: A case study in fatty acids composition.

Heterosigma akashiwo is an algal blooms species, and thus selected as the target microalgae in this work. This study attempted to investigate the influence of carbon limitation on the growth of H. akashiwo. Experiments were carried out in CO2-unlimited and CO2-limited systems (both include three nutritional groups). The stable isotope signatures and compound-specific stable three nutritional groups carbon isotopic composition of fatty acids were measured. Here we hypothesized that the carbon limitation could lead to the enriched of stable isotope ratios in the H. akashiwo. The results showed t

Blastocystis sp. Subtype Diversity in Wild Carnivore Species from Spain.

The occurrence and molecular diversity of the stramenopile eukaryote Blastocystis sp. was investigated by PCR and sequencing (Sanger and NGS) methods in 380 faecal specimens of free-living carnivores in Spain. Blastocystis sp. was confirmed in 1.6% (6/380) of the specimens analysed. Two samples from a common genet and a fox were successfully subtyped as ST7 by Sanger. Using NGS, ST14 was found in a fox and a European polecat, ST7 in a fox, and two additional foxes presented mixed infections of ST1/ST2/ST4 and ST1/ST2/ST7, respectively. Wild carnivore species could act as carriers of zoonotic B

Response of Hemolytic and Photosynthetic Activity of Chattonella marina Complex Under Variable N:P Stoichiometry.

Chattonella marina is an ichthyotoxic, bloom-forming raphidophyte known for its hemolytic activity. However, the mechanisms by which nitrogen (N) and phosphorus (P) limitation influence this hemolytic toxicity remain poorly understood. In this study, both N and P limitation reduced growth, photosynthetic efficiency (Fv/Fm, YII, rETRmax), and the expression of nutrient-uptake, tetrapyrrole/chlorophyll biosynthesis genes. Nevertheless, the two nutrients produced opposite effects on toxicity: N limitation lowered hemolytic activity and ROS levels to near zero, whereas P limitation kept both relat

Combining high pressure and electric fields towards Nannochloropsis oculata eicosapentaenoic acid-rich extracts.

Nannochloropsis oculata is naturally rich in eicosapentaenoic acid (EPA). To turn this microalga into an economically viable source for commercial applications, extraction efficiency must be achieved. Pursuing this goal, emerging technologies such as high hydrostatic pressure (HHP) and moderate electric fields (MEF) were tested, aiming to increase EPA accessibility and subsequent extraction yields. The innovative approach used in this study combined these technologies and associated tailored, less hazardous different solvent mixtures (SM) with distinct polarity indexes. Although the classical

Efficient solvothermal wet in situ transesterification of Nannochloropsis gaditana for biodiesel production.

In situ transesterification of wet microalgae is a promising, simplified alternative biodiesel production process that replaces multiple operations of cell drying, extraction, and transesterification reaction. This study addresses enhanced biodiesel production from Nannochloropsis gaditana at elevated temperatures. Compared with the previously reported in situ transesterification process of conducting the reaction at a temperature ranging from 95 to 125 °C, the present work employs higher temperatures of at least 150 °C. This relatively harsh condition allows much less acid catalyst with or wi

Sulfadiazine selection enables high-efficiency transformation in Nannochloropsis and Microchloropsis.

Nannochloropsis and Microchloropsis are important as eustigmatophyte model organisms and the production of high-value lipids. Efficient selectable markers are indispensable for genetic engineering to study and improve production yield of these species. Here, we establish sulfadiazine resistance mediated by the bacterial sul1 gene as an efficient and robust selection system. Growth assays identified 300 μg·mL-1 sulfadiazine as an effective selection concentration. Mitochondrial targeting of Sul results in transformation efficiencies and false-positive rates superior to commonly used markers aph

Techno-economic analysis of bioactive-rich oil production from Nannochloropsis oculata.

Carotenoids and polyunsaturated fatty acids (PUFAs) from microalgae have applications in food, pharmaceuticals, cosmetics, and aquaculture. However, the large-scale use of these compounds is limited by high production costs associated with microalgae harvest and extraction. Microalgae such as Nannochloropsis oculata are also recognized for their capacity to capture CO₂ and remove nutrients from wastewater, positioning them as promising tools for pollution mitigation within circular bioeconomy models. In this article, a techno-economic analysis was performed for three plant sizes (720 kg h-1, 3
