BACKGROUND: Lung neuroendocrine neoplasms (NENs) represent about 20% of all lung cancers. Few therapeutic options are available for atypical carcinoids (ACs). Single-agent temozolomide (TEM) is active in lung NENs, but whether the addition of capecitabine (CAPTEM) is associated with improved outcomes, is unknown. We sought to investigate the TEM-based therapies (TEM or CAPTEM) in patients with advanced AC.
MATERIAL AND METHODS: This was a retrospective analysis of prospectively collected data from patients with AC of the lung referred to our institution from January 2003 to January 2023 who h

A 62-year female received radiotherapy over six weeks with daily 75 mg/m2 Temozolomide (TMZ) for Glioblastoma (GB). At the last week of radiotherapy, her liver enzymes and serum bilirubin started deteriorating. TMZ was discontinued. The histopathology demonstrated the features of acute cholestasis and focal parenchymal inflammation. A range of investigations failed to show any other contributory cause of hepatitis. She required in-hospital care for a prolonged period for a grade three hepatic failure. The liver functions very slowly recovered over 40 weeks, but her general condition continues

Temozolomide is an oral alkylating agent recently approved for the treatment of glioblastoma multiforme. It has a favorable side effect profile and is generally well tolerated. Although mild respiratory symptoms have been described, pulmonary toxicity that requires discontinuation of therapy is rare. To our knowledge, this is the first case of temozolomide-associated organizing pneumonitis.

Temozolomide (TMZ) is an oral alkylating agent used to treat glioblastoma multiforme (GBM) and astrocytomas. However, at least 50% of TMZ treated patients do not respond to TMZ. This is due primarily to the over-expression of O6-methylguanine methyltransferase (MGMT) and/or lack of a DNA repair pathway in GBM cells. Multiple GBM cell lines are known to contain TMZ resistant cells and several acquired TMZ resistant GBM cell lines have been developed for use in experiments designed to define the mechanism of TMZ resistance and the testing of potential therapeutics. However, the characteristics o

BACKGROUND: The optimal salvage therapy for recurrent MGMT-methylated glioblastoma (GBM), IDH wildtype, remains undefined. While lomustine is often used in clinical trials and considered standard-of-care, cumulative toxicity precludes its use in patients previously treated with lomustine/temozolomide. The role of temozolomide rechallenge in this setting is unclear.
METHODS: This monocentric retrospective study included 70 patients with MGMT-methylated GBM, IDH wildtype, who received lomustine/temozolomide as first-line therapy. Descriptive data on second-line therapies were collected, and the

A 64-year-old woman was previously treated for Cushing's disease with trans-sphenoidal surgery, external beam radiotherapy and bilateral adrenalectomy. Progression of an aggressive corticotroph adenoma was evident 3 years post-adrenalectomy; involvement of the clivus was treated with surgery and gamma knife radiosurgery. Tumour spread through the skull base, occiput and left ear with persistent facial pain and left ear discharge; progression continued despite second gamma knife treatment. ACTH levels peaked at 2472 and 2265 pmol/l pre- and post-hydrocortisone respectively. Treatment with temoz

The EORTC-NCIC regimen for glioblastoma requires different dosing of temozolomide (TMZ) during radiation and maintenance therapy. This complexity is exacerbated by the availability of multiple TMZ capsule strengths. TMZ is an alkylating agent and the major toxicity of this class is dose-related myelosuppression. Inadvertent overdose can be fatal. The websites of the Institute for Safe Medication Practices (ISMP), and the Food and Drug Administration (FDA) MedWatch database were reviewed. We searched the MedWatch database for adverse events associated with TMZ and obtained all reports including

PURPOSE: Temozolomide (TMZ) is a widely used oral alkylating agent that has been associated with the development of severe hematologic adverse events (HAEs). Limited clinical information about HAEs is available.
METHODS: We searched the FDA MedWatch database for TMZ and obtained all MedWatch reports on TMZ submitted to the FDA from November 1, 1997 to September 3, 2008. We defined major HAEs, namely agranulocytosis, aplasia, aplastic anemia (AA), leukemia (various), myelodysplastic syndrome (MDS), and lymphoma, and several minor HAEs.
RESULTS: A total of 5,127 reports on 3,490 patients were

BACKGROUND: Temozolomide (TMZ) is an effective oral alkylating agent used in treating glioblastoma multiforme (GBM) and high-grade gliomas. It works by introducing methyl groups into DNA, inhibiting cell division. A case of blepharoconjunctivitis linked to the administration of TMZ is detailed in this report.
CASE PRESENTATION: We present a case of a 58-year-old African-American man diagnosed with GBM. Following adjuvant TMZ treatment, he developed blepharoconjunctivitis, characterized by eyelid and conjunctival inflammation. Symptoms included eyelid swelling, crusting, and conjunctival disch

Successful management of a heavily pretreated 58-year-old woman with metastatic esthesioneuoblastoma using temozolomide is reported. There is no standard treatment of this tumors with extra- and intracranial manifestations. The response, long term stability and high quality of life using temozolomide for this tumor entity should be recognized.

Bladder Paraganglioma (BPG) is a rare form of pheochromocytoma and paraganglioma (PPGL) characterized by elevated blood pressure when urinating, headache, dizziness, palpitations, and hematuria. Surgical resection is the most commonly used treatment for BPG. Here, we reporte a man who initially presented with hypogastralgia and hematuria and was diagnosed as BPG at local hospital. The size of mass was too huge to receive partial cystectomy at diagnosis. Then this patient was referred to our medical center. After evaluation, we chose temozolomide as neoadjuvant therapy to reduce the tumor size.

Temozolomide is a relatively new chemotherapeutic agent frequently associated with selective CD4+ T-lymphocytopenia. Patients with cell-mediated immune defects are at higher risk for acquiring infections with Salmonella species. We describe the first case of disseminated salmonellosis in a patient treated with temozolomide.

PURPOSE: Temozolomide (TMZ) with concomitant radiation is the standard therapy for treating glioblastomas (GBM) and is dosed based on body surface area (BSA) using a goal dose of 75 mg/m2. Neurooncologists have different methods of dosing patients, with some dosing within 5 mg of the calculated dose and others rounding to reduce patient burden. We aimed to determine the effect of rounded doses of TMZ on survival.
METHODS: We conducted a single-center retrospective review of 380 patients with GBM from 2013 to 2024. Relevant data was extracted from electronic medical records. Kaplan-Meier curve

Temozolomide (TMZ) is a cytotoxic agent of the imidazotetrazine class, chemically related to dacarbazine. Its use poses higher risks of lymphopenia and opportunistic infections. Prophylaxis for Pneumocystis jiroveci must be considered up to 12 months after treatment discontinuation. The due literature (MEDLINE) makes no mention of a possible connection between the use of TMZ and tuberculosis (TB). A female patient, aged 59, featuring glioblastoma multiforme and having undergone solely a brain biopsy, was submitted to TMZ along with radiotherapy. After the first TMZ maintenance cycle, the refer

Gliomas are the most common primary intracerebral tumours and over 60% of these are malignant. Standard treatment in the UK for patients with a good performance status consists of surgery and postoperative radiotherapy, however, recurrence is almost inevitable. Treatment of recurrent malignant gliomas (MG) is limited to further surgery, chemotherapy and novel biological therapies. The response rate to standard chemotherapy protocols for recurrent MG is less than 30%. Temozolomide (Temodar-US, Temodal-Rest of World) is an oral alkylating agent with a similar chemical structure to dacarbazine, a

BACKGROUND: Glioblastoma (GBM) is the most common adult primary brain malignancy. Recent studies demonstrate that temozolomide (TMZ) facilitates the persistence of quiescent glioma stem cells (GSCs), which are responsible for GBM recurrence. An ideal therapy should eradicate both proliferating cells and GSCs. Abexinostat (Abx), a histone deacetylase inhibitor, was identified through connectivity mapping to target the specific GBM signature. Here, we demonstrate the anti-proliferative effect of Abx on both differentiated cells and GSCs.
METHODS: Using patient-derived tumor cultures (PDCs) to t

Glioblastoma (GBM) remains a devastating disease for which standard temozolomide chemotherapy is limited by O6-methylguanine-DNA methyltransferase (MGMT)-mediated DNA damage repair. By targeting epidermal growth factor receptor (EGFR), a major oncogenic driver of GBM, Guo, Habib, and colleagues demonstrate that EGFR inhibition prior to temozolomide treatment induces an adaptive response that downregulates MGMT, enhancing therapeutic efficacy.

BACKGROUND: Gliomas are the most common malignant primary brain tumors, and Temozolomide (TMZ) remains a key component of their management. Despite its efficacy, TMZ is associated with adverse drug reactions (ADRs) that may affect quality of life and treatment compliance. This study evaluated the pattern and frequency of ADRs in South Indian glioma patients receiving TMZ, examined demographic associations, and reviewed management strategies.
METHODS: A retrospective observational study was conducted at KMC Hospital, Mangalore, India, over the past three years. Thirty-eight patients with histo

BACKGROUND: Temozolomide (TMZ) is an alkylating agent used in chemoradiotherapy and adjuvant chemotherapy regimens for treatment of newly diagnosed or recurrent glioblastoma. In Germany alone, 900,000 daily doses of the drug are prescribed each year. Therefore, all severe side effects of TMZ, even those rarely observed, are relevant to radiotherapists.
MATERIALS AND METHODS: We report a case of severe drug-induced toxic hepatitis that developed during chemoradiotherapy with TMZ in a patient with glioblastoma multiforme.
RESULTS: Transaminase elevation was observed after 5 weeks of TMZ treatm

BACKGROUND: Temozolomide (TMZ) remains the first-line chemotherapeutic agent for glioblastoma (GBM), but its clinical efficacy is often limited by acquired resistance. This study aimed to investigate whether nectin-4 could serve as a target to sensitize GBM cells to TMZ and to determine whether resveratrol enhances TMZ efficacy through downregulation of nectin-4.
METHODS: Nectin-4 expression was evaluated in TMZ-treated GBM cell lines (U87, U251) and in an orthotopic GBM mouse model using qRT-PCR, Western blotting, and immunohistochemistry. Gain- and loss-of-function approaches were performed
