Innocuous temperature sensation arises from the activity of primary afferent thermoreceptors, but how these neurons encode cool and warm at the population level remains unclear. Using two-photon in vivo imaging of the L4 dorsal root ganglion in awake and anesthetized mice, we show that the thermoreceptor population is strongly biased toward activation by cooling and suppression of activity by warming. Although thermoreceptors activated by cool or warm only are present in the population, most show bidirectional responses and encode absolute temperatures in a graded manner. Pharmacological manip…

Cold-sensitive nerve terminals (CSNTs) encode steady temperatures with regular, rhythmic temperature-dependent firing patterns that range from irregular tonic firing to regular bursting (static response). During abrupt temperature changes, CSNTs show a dynamic response, transiently increasing their firing frequency as temperature decreases and silencing when the temperature increases (dynamic response). To date, mathematical models that simulate the static response are based on two depolarizing/repolarizing pairs of membrane ionic conductance (slow and fast kinetics). However, these models fai

The mechanism(s) for the changes in cardiac function during heat stress remain unknown. This study tested two unique hypotheses. First, sympathetic innervation to the heart is required for increases in cardiac systolic function during heat stress. This was accomplished by comparing responses during heat stress between paraplegics versus tetraplegics, with tetraplegics having reduced/absent cardiac sympathetic innervation. Second, stimulation of skin thermoreceptors contributes to cardiovascular adjustments that occur during heat stress in humans. This was accomplished by comparing responses du…

In anaesthetized cats, sensory vagal units were recorded in the nodose ganglion by means of extracellular glass microelectrodes. In the antrum and the duodenum we have found receptors tonically activated by warm (38-51 degrees C with an optimum at 46-49 degrees C) or cold (36-10 degrees C with an optimum at 12-10 degrees C) solutions. These receptors did not respond to mechanical stimuli (compression and distension of the viscera) and to chemical ones (perfusion with glucose and acid solutions). Thus they did not belong to polymodal type, but they must be considered as true thermoreceptors, sp…

Pyroelectric infrared sensors (PIR) are widely used as infrared (IR) detectors due to their basic implementation, low cost, low power, and performance. Combined with a Fresnel lens, they can be used as a binary detector in applications of presence and motion control. Furthermore, due to their features, they can be used in autonomous intelligent devices or included in robotics applications or sensor networks. In this work, two neural processing architectures are presented: (1) an analog processing approach to achieve the behavior of a presynaptic neuron from a PIR sensor. An analog circuit simi

TRPM8 is the main molecular entity responsible for cold sensing. This polymodal ion channel is activated by cold, cooling compounds such as menthol, voltage, and rises in osmolality. In corneal cold thermoreceptor neurons (CTNs), TRPM8 expression determines not only their sensitivity to cold, but also their role as neural detectors of ocular surface wetness. Several reports suggest that Protein Kinase C (PKC) activation impacts on TRPM8 function; however, the molecular bases of this functional modulation are still poorly understood. We explored PKC-dependent regulation of TRPM8 using Phorbol 1

BACKGROUND: The body has a thermoregulation setting where when it gets hot, the body responds by dilating blood vessels or vasodilation, otherwise it gets a cold response, which causes narrowing of blood vessels or vasoconstriction. The thermal conditioning system is a system that can influence the baby in terms of its thermal quality. So that the baby can feel a comfortable environment or not. Skin temperature is a fundamental factor in heat exchange between the body and its environment.
OBJECTIVE: The purpose of this study was to analyze the effect of thermal exposure to carbon fiber filame

PURPOSE: There is a substantial body of evidence indicating that corneal sensory innervation is affected by pathology in a range of diseases. However, there are no published studies that have directly assessed whether the nerve fiber density of the different subpopulations of corneal sensory neurons are differentially affected. The present study explored the possibility that the intraepithelial nerve fiber density of corneal polymodal nociceptors and cold thermoreceptors are differentially affected in mice fed with a high-fat high cholesterol (HFHC; 21% fat, 2% cholesterol) diet and in those t…

The role of TRPA1 in the thermosensitivity of the corneal cold thermoreceptor nerve endings was studied in young and aged mice. The contribution of the TRPA1-dependent activity to basal tearing and thermally-evoked blink was also explored. The corneal cold thermoreceptors' activity was recorded extracellularly in young (5-month-old) and aged (18-month-old) C57BL/6WT (WT) and TRPA1-/- knockout (TRPA1-KO) mice at basal temperature (34 °C) and during cooling (15 °C) and heating (45 °C) ramps. The blink response to cold and heat stimulation of the ocular surface and the basal tearing rate were als…

Cyclosporine A (CsA) is used for the treatment of dry eye (DE) with good clinical results, improving tear secretion and decreasing subjective symptoms. These effects are attributed to the improved tear film dynamics, but there are no data on the effect of CsA on the abnormal sensory nerve activity characteristic in DE. Our purpose was to evaluate the CsA effect on the enhanced activity of corneal cold thermoreceptors in a tear-deficient DE animal model using in vitro extracellular recording of cold thermoreceptors nerve terminal impulses (NTIs) before and in the presence of CsA. NTI shape was …

The effect of altering input from cutaneous thermoreceptors of the face and trunk on the relationship between hypothalamic temperature (Thy) and heat production (HP) was studied in three rats. The signal from cutaneous receptors was altered in two ways: by altering skin temperature (Tsk) and by sectioning nerves supplying cutaneous receptors. It was found that when Tsk was lowered in normal rats Thy threshold for thermoregulatory HP was elevated, but the slope of the relationship between Thy and HP was not significantly altered. After the spinal nerves serving the trunk skin were sectioned, th…

Corneal nerve ablation upregulates TRPM8 mainly in canonical cold thermoreceptors, enhancing their cold and menthol sensitivity, inducing a rise in the ongoing firing activity of TRPM8(+) nerve endings and an increase in basal tearing. Our results suggest that unpleasant dryness sensations, together with augmented tearing rate af …

Pulpitis pain might be triggered by a cold stimulus, yet the cellular mechanisms responsible for this phenomenon are largely unknown. One possible mechanism involves the direct activation of cold-responsive thermoreceptors. The purpose of this study was to evaluate the possible role of the TRPM8 thermoreceptor in cold-mediated noxious pulpal pain mechanisms by comparing expression patterns in pulpal nerves from healthy control molars to cold-sensitive painful molars with irreversible pulpitis. Samples were identically processed with the indirect immunofluorescence method, and images were obtai…

In-sensor computing and logics are the core for advancing machine vision technology, but face challenges in extending such capability into thermal form. Here, a bioinspired thermoreceptor array based on twisted graphene and titanium diselenide (TiSe2) heterostructures is presented. The electron-phonon interaction between graphene and few-layer TiSe2 exhibits strong twist-angle (θ) dependence, as evidenced by varied phonon-induced gap of graphene (114 ± 13 meV for θ = 1° and 67 ± 6 meV for θ = 7°), enabling sensitive thermal reception at temperature range from 20 to 350 K. Integrating such hete
