Endogenous psychoactive tryptamines reconsidered: an anxiolytic role for dimethyltryptamine.

The presence of the potent hallucinogenic psychoactive chemical N,N-dimethyltryptamine (DMT) in the human body has puzzled scientists for decades. Endogenous DMT was investigated in the 1960s and 1970s and it was proposed that DMT was involved in psychosis and schizophrenia. This hypothesis developed from comparisons of the blood and urine of schizophrenic and control subjects. However, much of this research proved inconclusive and conventional thinking has since held that trace levels of DMT, and other endogenous psychoactive tryptamines, are insignificant metabolic byproducts. The recent dis

Modification of natural tryptamines for the treatment of neuropsychiatric diseases.

The last decade represented a period of unprecedented interest in, and development of, traditional psychedelic substances for the treatment of a variety of neuropsychiatric disorders. While early clinical trials of classical psychedelics, several of the major ones being tryptamines, have demonstrated robust efficacy in patients with minimal adverse effects, many of these molecules have properties that may limit their broad utility in clinical practice or require more complex methods of delivery. The functional role and importance of a "psychedelic experience" for therapeutic efficacy remain en

Pharmacological and behavioural effects of tryptamines present in psilocybin-containing mushrooms.

BACKGROUND AND PURPOSE: Demand for new antidepressants has resulted in a re-evaluation of the therapeutic potential of psychedelic drugs. Several tryptamines found in psilocybin-containing "magic" mushrooms share chemical similarities with psilocybin. Early work suggests they may share biological targets. However, few studies have explored their pharmacological and behavioural effects. EXPERIMENTAL APPROACH: We compared baeocystin, norbaeocystin and aeruginascin with psilocybin to determine if they are metabolized by the same enzymes, similarly penetrate the blood-brain barrier, serve as liga

Efficacy and Safety of Psychoactive Tryptamines in Addiction: A Systematic Review.

BACKGROUND: Psychedelics such as lysergic acid diethylamide (LSD) and psilocybin have shown a beneficial effect on substance use disorder (SUD) symptoms. In this systematic review, we aimed to assess the efficacy and safety of psychoactive tryptamines in patients with an SUD or non-substance-related disorder (i.e., gambling disorder) in order to provide a comprehensive overview of the available evidence and identify potential research gaps. METHODS: A systematic literature search was performed in eight different databases up to February 2024. Clinical trials were included that assessed the ef

Tryptamines and Mental Health: Activating the 5-HT Receptor for Therapeutic Potential.

Tryptamines, a class of 3-aminoethyl-indoles that activate the serotonin receptor, show potential for novel mental health treatments. The FDA has granted "breakthrough therapy designation" to psilocybin and MDMA for treatment-resistant depression, major depressive disorder, and post-traumatic stress disorder, sparking global research efforts. Various clinical trials are currently investigating the therapeutic value of psilocybin for several mental health disorders. Results thus far indicate significant improvements in patient-reported outcomes via reductions in experiential avoidance. These ad

Ready Access to Benzodiazocines from Tryptamines: A Druggable Scaffold toward Antiviral Agents.

Herein we report the first synthesis of the proposed structure of a benzodiazocine natural product, peganutonin A. Considering the druggable nature of benzodiazocine and its limited exploration in the field of medicinal chemistry, we generalized the approach and created a library of compounds useful for various biological activities. Key steps in present approach include tryptamine cyclization facilitated by tert-butyl hypochlorite and cleavage of the fused indole ring using ozonolysis. Preliminary screening of the synthesized compounds resulted in potent antiviral compounds against the SARS-C

Assessment of Bioactivity-Modulating Pseudo-Ring Formation in Psilocin and Related Tryptamines.

Psilocybin (1) is the major alkaloid found in psychedelic mushrooms and acts as a prodrug to psilocin (2, 4-hydroxy-N,N-dimethyltryptamine), a potent psychedelic that exerts remarkable alteration of human consciousness. In contrast, the positional isomer bufotenin (7, 5-hydroxy-N,N-dimethyltryptamine) differs significantly in its reported pharmacology. A series of experiments was designed to explore chemical differences between 2 and 7 and specifically to test the hypothesis that the C-4 hydroxy group of 2 significantly influences the observed physical and chemical properties through pseudo-ri

New World Tryptamine Hallucinogens and the Neuroscience of Ayahuasca.

New World indigenous peoples are noted for their sophisticated use of psychedelic plants in shamanic and ethnomedical practices. The use of psychedelic plant preparations among New World tribes is far more prevalent than in the Old World. Yet, although these preparations are botanically diverse, almost all are chemically similar in that their active principles are tryptamine derivatives, either DMT or related constituents. Part 1 of this paper provides an ethnopharmacological overview of the major tryptamine-containing New World hallucinogens. Part 2 focuses on ayahuasca and its effects on the

Copper-catalyzed cascade reaction of tryptamines with diazo compounds to access hexahydropyrroloindoline derivatives.

A domino reaction sequence of cyclopropanation/ring-opening/iminium cyclization of tryptamine derivatives with donor-acceptor diazo compounds is developed to furnish pyrroloindolines, creating three consecutive stereogenic centers in a single step. The copper-catalyzed reaction provides pyrroloindolines at room-temperature with good substrate scope.

SOME PHARMACOLOGICAL EFFECTS OF A SERIES OF TRYPTAMINE DERIVATIVES.

The methods of synthesis and some pharmacological properties of a number of N-substituted tryptamines are described. Peripheral vasopressor activity decreased with increase in complexity of the substituent and was not related to toxicity or to behavioural activity. Activity in increasing the rectal temperature of rabbits appeared to parallel modifications in the open-field behaviour of rats. Some compounds were investigated for their ability to block a conditioned avoidance-response in rats and to affect operant conditioning in cats. Evidence is presented which suggests that the tryptamine der

Effect of oral tryptamines on the gut microbiome of rats-a preliminary study.

BACKGROUND: Psilocybin and related tryptamines have come into the spotlight in recent years as potential therapeutics for depression. Research on the mechanisms of these effects has historically focused on the direct effects of these drugs on neural processes. However, in addition to such neural effects, alterations in peripheral physiology may also contribute to their therapeutic effects. In particular, substantial support exists for a gut microbiome-mediated pathway for the antidepressant efficacy of other drug classes, but no prior studies have determined the effects of tryptamines on micro

Biosynthesis of pathogen-induced hydroxylated tryptamine derivatives in barley.

Barley (Hordeum vulgare), a major cereal crop, suffers significant yield losses due to pathogen attack. Understanding the chemical defense responses that follow pathogen infection is essential to uncover new resistance targets in barley. In this study, we employed an untargeted metabolomics and mass spectrometry imaging approach to profile the chemical landscape of barley leaves inoculated with the hemibiotrophic fungal pathogen Pyrenophora teres f. teres, the causal agent of net blotch. Pathogen infection triggered a significant chemical defense response, including the induction of many pheny

Rhabdomyolysis after ingestion of "foxy," a hallucinogenic tryptamine derivative.

"Foxy methoxy" (chemical name, 5-methoxy-N,N-diisopropyltryptamine) is a hallucinogenic tryptamine that has been abused with increasing frequency since its appearance in the late 1990s. Like other drugs in this class, foxy frequently produces feelings of euphoria, disinhibition, and auditory as well as visual hallucinations. The drug has been linked to adverse effects, including restlessness, agitation, gastrointestinal distress, and muscle tension. In light of the relatively recent advent of foxy as a drug of abuse and given the inability of commercial toxicologic screening tests to detect th

Tryptamine: a metabolite of tryptophan implicated in various neuropsychiatric disorders.

Although early interest in the biomedical relevance of tryptamine has waned in recent years, it is clear from the above discussion that the study of tryptamine is worthy of serious consideration as a factor in neuropsychiatric disorders. The study of [3H]-tryptamine binding sites indicates an adaptive responsiveness characteristic of functional receptors. The question raised by Jones (1982d) on whether tryptamine is acting centrally as a neurotransmitter or a neuromodulator still remains mostly unanswered, although the evidence cited within this review strongly suggests a modulatory role for t
