StoS is a hybrid histidine kinase that, to date, has been investigated only in Xanthomonas oryzae pathovar oryzae. In this study, we elucidated the function and transcriptional regulation of the stoS gene in X. campestris pathovar campestris (Xcc). An stoS deletion mutant and its complemented strain were constructed to assess phenotypic characteristics. Inactivation of StoS markedly decreased tolerance to heat, sodium dodecyl sulfate, and hydrogen peroxide, impaired membrane integrity, and reduced stationary-phase survival, whereas complementation restored these phenotypes. The Gram-negative bacterium Xanthomonas campestris pv. campestris is the etiological agent of black rot, a widespread and destructive disease affecting cruciferous plants. In this study, a lon mutant was obtained by EZ-Tn5 transposon mutagenesis of the X. campestris pv. campestris. The lon gene encodes an ATP-dependent protease implicated in protein quality control and stress adaptation across various bacterial species. Functional analysis revealed that lon disruption in X. campestris pv. campestris resulted in diminished extracellular protease activity, reduced virulence, and heightened sensi…

Citrus is an economically important fruit crop that is severely afflicted by citrus canker, a disease caused by Xanthomonas citri ssp. citri (X. citri); thus, new sustainable strategies to manage this disease are needed. Although all Citrus spp. are susceptible to this pathogen, they are resistant to other Xanthomonas species, exhibiting non-host resistance (NHR), for example, to the brassica pathogen X. campestris pv. campestris (Xcc) and a gene-for-gene host defence response (HDR) to the canker-causing X. fuscans ssp. aurantifolii (Xfa) strain C. Here, we examine the plant factors associated

Black rot caused by Xanthomonas campestris pv. campestris was found in 28 sampled cabbage fields in five major cabbage-growing districts in Nepal in 2001 and in four cauliflower fields in two districts and a leaf mustard seed bed in 2003. Pathogenic X. campestris pv. campestris strains were obtained from 39 cabbage plants, 4 cauliflower plants, and 1 leaf mustard plant with typical lesions. Repetitive DNA polymerase chain reaction-based fingerprinting (rep-PCR) using repetitive extragenic palindromic, enterobacterial repetitive intergenic consensus, and BOX primers was used to assess the genet…

The γ-proteobacterium Xanthomonas campestris pv. campestris B100 is the causal agent of black rot disease in a wide range of economically important crops. In addition, X. campestris pv. campestris is commercially relevant due to the synthesis of the exopolysaccharide xanthan. In this work, we first introduce a novel transcriptional regulator, termed Crt1, and present the effect that the deletion of the crt1 gene exerts on growth, xanthan production, virulence, and transcriptome and proteome profiles. Differential transcriptome analysis of the deletion mutant X. campestris pv. campestris B100 Δ…

Phenazine-1-carboxylic acid (PCA), a member of phenazines secreted by microorganisms, inhibits the growth of many bacteria and fungi. Xanthomonas campestris pv. campestris is the causal agent of black rot, the most important disease of cruciferous crops worldwide, and is more tolerant to PCA than other Xanthomonas species. Previous studies reported that reactive oxygen species (ROS) scavenging ability is involved in regulating the PCA tolerance of Xanthomonas species. Additionally, the cytochrome c maturation (CCM) system has been found to play a more important role in tolerance to phenazines …

Xanthomonas campestris pv. campestris, the causal agent of black rot, is one of the most important bacterial pathogens affecting Brassicaceae crops worldwide. Its seed-borne nature, multiple routes of dissemination, and limited availability of resistant cultivars make black rot difficult to manage and underscore the importance of early and reliable pathogen detection. This review summarizes current knowledge on the epidemiology of X. campestris pv. campestris, with emphasis on advances in conventional and molecular approaches used for its detection and characterization, as well as on the curre…

Black rot of crucifers, (Xanthomonas campestris pv. campestris) is the principal yield-limiting and destructive pathogen of cruciferous crop worldwide. In order to validate a bio-based control alternative for this disease, whey, lime sulfur, biofertilizer, Bordeaux mixture or raw milk were applied to kale (Brassica oleracea var. acephala) plants. The disease control was achieved by most of the tested products. Milk-based products (raw milk and whey) and biofertilizer reduced the severity by 44 and 56% in the field. Antioxidants, crude fibber, crude protein and lipid contents and kale yield wer…

In this study, a Cu2O/TiO2 (CuTi) visible-light photocatalytic composite was employed for the treatment of Xanthomonas campestris and X. campestris-infected Brassica napus seedlings. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values against X. campestris were determined to be 8 and 32 μg ml-1, respectively. Transmission electron microscopy analysis demonstrated a direct correlation between the extent of bacterial cell damage and the concentration of CuTi. Noteworthily, a bactericidal rate of 100% was achieved at a concentration of 150 μg ml-1 over a…

In Xanthomonas campestris pv. campestris (Xcc), the chromosomally encoded class A β-lactamase (Bla(xcc)) is expressed at a high basal level in the absence of an inducer and its expression is inducible by ampicillin. Like most of the class A β-lactamases, Bla(xcc) cannot digest the β-lactam ring of cefoxitin. However, Xcc exhibits high basal resistance to cefoxitin. A promoter activity assay with P(blaxcc)-lacZ transcriptional fusion from a plasmid and western blotting using anti-Bla(xcc) polyclonal antibodies demonstrated that a sublethal concentration of cefoxitin can induce expression of the…

Xanthomonas campestris pv. incanae (Xci) is known to cause systemic infections in the stem of Matthiola incana. Prior research observed that the pathogen can invade the vascular system and extend into the seed peduncles of infected plants, suggesting a likely mean for internal contamination of the seeds. However, this and other potential pathways of seed infection and seed-to-seed transmission, have not been sufficiently investigated. Using both culture-based and metagenomic approaches, we evaluated the potential for and efficiency of seed infection by Xci after vascular and floral inoculation…

The bacterium Xanthomonas campestris pv. campestris (Xcc) causes black rot disease in cruciferous crops, resulting in severe yield loss worldwide. The excessive use of chemical pesticides in agriculture to control diseases has raised significant concern about the impact on the environment and human health. Nanoparticles have recently gained significant attention in agriculture owing to their promising application in plant disease control, increasing soil fertility and nutrient availability. In the current study, we synthesized thymol-loaded chitosan nanoparticles (TCNPs) and assessed their ant…

Bacteriophages have emerged as promising alternatives to pesticides for controlling bacterial pathogens in crops. Black rot, caused by Xanthomonas campestris pv. campestris (Xcc), is one of the most destructive bacterial diseases affecting cruciferous crops worldwide. In this study, a novel lytic bacteriophage, X1, infecting Xcc 8004, was isolated from sediment collected in an oligotrophic karst cave. Phage X1 formed clear plaques and exhibited myovirus-like morphology with an icosahedral head and a contractile tail. Biological characterization revealed productive infection of the tested Xcc s…

Polymerase chain reaction (PCR) primers (BXW-1 and BXW-3) for conventional PCR were developed from conserved sequences in the hrpB operon of the hrp gene cluster from Xanthomonas campestris pv. musacearum, the causative agent of banana Xanthomonas wilt (BXW). All 50 strains of X. campestris pv. musacearum, isolated from Uganda, Rwanda, and Tanzania, produced a 214-bp amplicon when whole cells, bacterial ooze from infected tissue, and genomic DNA purified from bacterial ooze or infected tissue were used as template. The BXW primers also detected strains of X. axonopodis pv. vasculorum isolated …

The avirulence gene avrBs3 from Xanthomonas campestris pv. vesicatoria was cloned and found to be localized on a self-transmissable plasmid. Genetic analysis of an avrBs3 insertion mutation revealed that avrBs3 constitutes a single locus, specifying the resistant phenotype on pepper plants. Southern blot experiments showed that no DNA sequences homologous to avrBs3 were present in other races of X. c. pv. vesicatoria, which are unable to induce a hypersensitive reaction on ECW-30R. However, the DNA of several different pathovars of X. campestris hybridized to the avrBs3 probe. A deletion analy…

Response regulators (RRs) of two-component signalling systems (TCSs) containing tandem receiver (REC) domains are widespread in bacteria, yet their functions and regulatory mechanisms remain poorly understood. In our previous study, DrdR, one such RR in the cruciferous black rot disease pathogen Xanthomonas campestris pv. campestris (Xcc) was demonstrated to positively regulate pilus-dependent motility and negatively regulate flagellum-dependent motility. We showed that DrdR modulates the ATPase activities of pili motor proteins PilT and PilB, thereby enhancing bacterial pilus-dependent swarmi…

Bacterial adaptation is facilitated by the presence of mobile genetic elements and horizontal gene transfer of genes, such as those coding for virulence factors or resistance to antimicrobial compounds. A hybrid assembly of Nanopore MinIon long-read and Illumina short-read data was produced from a copper-resistant Xanthomonas campestris pv. campestris strain isolated from symptomatic broccoli leaves in Mauritius. We obtained a 5.2-Mb high-quality chromosome and no plasmid. We found four genomic islands, three of which were characterized as integrative conjugative elements or integrative mobili…

Alternative host plants are important in the survival and perpetuation of several crop pathogens and have been suspected to play a role in the survival of Xanthomonas campestris pv. musacearum (Xcm) and perpetuation of Xanthomonas wilt (XW) disease of banana and enset. This study determined the potential risk posed by two weeds (Canna spp. and wild sorghum) and common banana intercrops (maize, millet, sorghum, taro, and sugarcane) as alternative hosts to Xcm. The study employed screenhouse experiments, laboratory procedures and diagnosis of banana fields in XW-affected landscapes. Typical XW s…

Like many other bacterial organisms, Xanthomonas campestris pv. campestris (Xcc) employs two motility appendages, a single polar flagellum and type IV pili, for sensing environmental cues and traversing liquids and solid surfaces. However, compared to the flagellum, the molecular mechanisms underlying the function and regulation of the pilus in Xcc and other xanthomonads are not fully understood. In this study, we demonstrated that a special conserved orphan response regulator DrdR, which contains double receiver (REC) domains but lacks an output domain, modulates the activities of motor prote…

Bacterial resistance to bacteriophages represents a major limitation for the durable use of phage-based biocontrol strategies in agriculture. Here, we used a phage-resistance derivative of Xanthomonas campestris pv. campestris (Xcc) as a selective host to isolate an additional phage with activity against resistant bacteria. A spontaneous mutant, designated Xcc 8004R, was isolated following exposure of Xcc 8004 to phage X1. Xcc 8004R exhibited a growth profile comparable to that of the wild-type strain, but differed in colony appearance, X1 adsorption efficiency, and motility phenotypes. Whole-…
