Immunohistochemical demonstration of acetaldehyde-modified epitopes in human liver after alcohol consumption.

Acetaldehyde, the toxic product of ethanol metabolism in the liver, covalently binds to a variety of proteins. Recent studies indicate that such binding can stimulate the production of antibodies against the acetaldehyde adducts. We raised rabbit antibodies which recognized various protein-acetaldehyde conjugates but not the corresponding control proteins. Such antibodies were used in immunohistochemical studies to find out whether acetaldehyde-generated epitopes can be detected from liver specimens of 13 human subjects with different degrees of alcohol consumption. While the specimens obtaine

Acetaldehyde dehydrogenase 2 activation attenuates sepsis-induced brain injury through NLRP3 inflammasome regulation.

OBJECTIVE: Acetaldehyde dehydrogenase 2 (ALDH2) plays an important part in neuroprotection; however, its effect on sepsis-induced brain injury is nuclear. Our aim is to investigate the potential effect and mechanism of ALDH2 in this condition. METHODS: We established an animal model using cecal ligation and perforation (CLP). Twenty-four rats were divided into sham group (n = 6), CLP group (n = 6), CLP + Alda-1 group (n = 6) and CLP + Cyanamide (CYA) group (n = 6). Vital signs were monitored, and arterial blood gas analysis, hippocampal histological staining and ALDH2 activity analysis were c

Formaldehyde and Acetaldehyde Exposure in "Non-Traditional" Occupational Sectors: Bakeries and Pastry Producers.

INTRODUCTION: Formaldehyde, a colorless and highly irritating substance, causes cancer of the nasopharynx and leukemia. Furthermore, it is one of the environmental mutagens to which humans are most abundantly exposed. Acetaldehyde was recently classified as carcinogen class 1B and mutagen class 2 in Annex VI EC regulation. Occupational exposure to the two aldehydes occurs in a wide variety of occupations and industries. The aim of this study is to deepen exposure to the two aldehydes in the non-traditional productive sectors of bakeries and pastry producers. METHODS: The evaluation of exposur

Sensory Specificities Involving Acetaldehyde and Diacetyl in Wines Produced without Added Sulfur Dioxide.

Nowadays, the development of naturality concept is illustrated in the oenological field by the development of wine produced without the addition of SO2. Among its chemical properties, SO2 is able to react with carbonyl compounds to form carbonyl bisulfites. Acetaldehyde and diacetyl are the main carbonyl compounds of red wines, which could influence product perception. The goal of this paper was to evaluate their chemical and sensory impact in red wines produced without any addition of SO2. A first quantification approach revealed a lower concentration of these compounds in wines without added

Zipper phosphorene as sensing element towards formaldehyde and acetaldehyde - A first-principles insight.

We ascertained the structural stability of zipper phosphorene nanosheet (zP-NS) and studied the adsorption behaviour of toxic aldehyde compounds including formaldehyde (FD) and acetaldehyde (AD) on zP-NS based on first-principles calculation. Considerably, zP-NS reveal a semiconducting character with band gap of 1.35 eV. Especially, four distinct favourable adsorption positions including bridge-, hollow-, top- and valley-site of FD and AD vapours on zP-NS were investigated. Furthermore, the calculated binding-energy of prominent adsorption sites are observed to be in the scope of -0.143 eV to

Triacetin Enhances Levels of Acrolein, Formaldehyde Hemiacetals, and Acetaldehyde in Electronic Cigarette Aerosols.

The health effects of inhaled electronic cigarette (e-cigarette) flavoring compounds are largely unknown. Earlier reports of their chemical reactivity have been conflicting, with some claiming, for example, that the degradation of flavoring chemicals in e-cigarettes to aldehydes is statistically insignificant. It is thus important to understand how these molecules react to afford enhanced aerosol products. The purpose of the current study was to investigate the origin of formaldehyde, acrolein, and acetaldehyde in e-cigarettes that contain the popular additive, triacetin (TA). By using 13C lab

Protective role of ALDH2 against acetaldehyde-derived DNA damage in oesophageal squamous epithelium.

Acetaldehyde is an ethanol-derived definite carcinogen that causes oesophageal squamous cell carcinoma (ESCC). Aldehyde dehydrogenase 2 (ALDH2) is a key enzyme that eliminates acetaldehyde, and impairment of ALDH2 increases the risk of ESCC. ALDH2 is produced in various tissues including the liver, heart, and kidney, but the generation and functional roles of ALDH2 in the oesophagus remain elusive. Here, we report that ethanol drinking increased ALDH2 production in the oesophagus of wild-type mice. Notably, levels of acetaldehyde-derived DNA damage represented by N(2)-ethylidene-2'-deoxyguanos

Voluntary ethanol consumption decreases after the inactivation of central acetaldehyde by d-penicillamine.

Acetaldehyde, the first metabolite of ethanol, may mediate some ethanol-induced effects. Previous research in our laboratory has shown that D-penicillamine, an inactivation agent for acetaldehyde, is effective in decreasing locomotor stimulation and conditioned place preference induced by ethanol in mice. In the present study, the effects of D-penicillamine on the voluntary consumption of ethanol were assessed. Male rats were offered ethanol under restricted access, without food or water deprivation. Daily availability of ethanol was limited to a 15-min period in the home cages. When the respo

In vitro exposure to malondialdehyde-acetaldehyde adducted protein inhibits cell proliferation and viability.

BACKGROUND: Circulating antibodies against malondialdehyde-acetaldehyde (MAA) haptenated proteins are increased significantly in patients with alcohol-induced cirrhosis and hepatitis and are associated with severity of liver damage. Additionally, MAA haptenated proteins are highly immunogenic without the use of adjuvant and have been suggested to induce autoreactive responses. The mechanism of this immunogenicity is currently unknown but may be mediated by cell death in a similar manner as other autoimmune diseases such as systemic lupus erythematosus, mixed connective tissue disease, myositis

Alcoholic cardiomyopathy--acetaldehyde poisoning rat: myocardial and serum enzyme changes in acute exposure.

In order to clarify the pathogenesis of alcoholic cardiomyopathy, acetaldehyde (Ach) was administered to rats using the inhalation method. Serum enzyme changes and structural alterations of the heart were observed at various time intervals after 2 ml of Ach exposure for 2 hours. Possible myocardium-related enzymes, such as glutamic oxaloacetic transaminase, lactic dehydrogenase and creatine phosphokinase, were elevated within 24 hours and then returned almost to the previous levels. The most significant change seen using light microscopy was a prominent contraction band scattered throughout th

Acetaldehyde concentration in rat blood and brain during the calcium carbimide--ethanol interaction.

Ethanol was administered to rats by oral intubation 3 h after intraperitoneal administration of calcium carbimide. Increased blood acetaldehyde concentration and the presence of acetaldehyde in brain were found for the calcium carbimide--ethanol interaction. There was no detectable brain acetaldehyde for ethanol administration following saline treatment. The magnitude of the acetaldehyde concentration in blood and brain during the calcium carbimide--ethanol interaction was directly dependent on the doses of calcium carbimide (0.7, 3.5, 7.0 mg/kg) and ethanol (0.5, 1.0, 1.5 g/kg). There was an

Induction of liver acetaldehyde dehydrogenase: possible role in ethanol tolerance after exposure tobarbiturates.

Mice were injected twice a day for 4 days with saline or phenobarbital or ethanol. Treatment with phenobarbital, but not ethanol, increased the amount of liver acetaldehyde dehydrogenase activity. More rapid removal of acetaldehyde, which is a toxic metabolic intermediate of ethanol, may contribute to the alcohol tolerance exhibited by persons who use barbiturates regularly.

Differences in the serum levels of acetaldehyde and cytotoxic acetaldehyde-albumin complexes after the consumption of red and white wine: in vitro effects of flavonoids, vitamin E, and other dietary antioxidants on cytotoxic complexes.

After the consumption of ethanol, acetaldehyde levels increase in the serum, and the serum develops a nondialyzable cytotoxic activity caused by the formation of unstable acetaldehyde-albumin complexes. The concentration of acetaldehyde in the serum and the cytotoxic activity in serum albumin 8.5 hr after six healthy volunteers began to drink 94 g of ethanol were significantly less when the ethanol was consumed as red wine than as white wine. The serum acetaldehyde was measured by a fluorigenic HPLC assay, and the cytotoxic activity in albumin was determined using two different assays based on

Steam Reforming of Ethanol to Acetaldehyde and Acetone Over Al-Doped ZnO Catalysts.

Ethanol steam reforming with a water to ethanol molar ratio of 7:1 was investigated over pure ZnO and Al-doped ZnO catalysts with up to 10 mol% Al3+ synthesized via coprecipitation. This synthesis route yielded wurtzite ZnO, with Al being incorporated into the ZnO lattice at low doping levels. Al doping was found to alter the reaction pathway of ethanol steam reforming by suppressing the consecutive acetaldehyde conversion to acetic acid and further to acetone. Continuous kinetic experiments using a plug flow reactor resulted in almost full conversion and an acetone selectivity of 53% at 450°C

Proline Amide Catalyzes Formation of Toxic Crotonaldehyde from Acetaldehyde Under Physiologically Relevant Conditions.

Crotonaldehyde is a human toxin that reacts with nucleophilic groups on DNA and proteins. Putative crotonaldehyde-derived adducts on DNA are reported in cells and patients after ethanol exposure, which implies that crotonaldehyde is formed in cells. Here, we show that proline amide, which is a model of N-terminal proline-containing proteins, catalyzes the aldol condensation of the ethanol metabolite acetaldehyde to crotonaldehyde under physiologically relevant conditions. This reaction is more efficient at neutral pH than under acidic or basic conditions, but is inhibited by competing imidazol

Electronic cigarette solvents, pulmonary irritation, and endothelial dysfunction: role of acetaldehyde and formaldehyde.

After more than a decade of electronic cigarette (E-cig) use in the United States, uncertainty persists regarding E-cig use and long-term cardiopulmonary disease risk. As all E-cigs use propylene glycol and vegetable glycerin (PG-VG) and generate abundant saturated aldehydes, mice were exposed by inhalation to PG-VG-derived aerosol, formaldehyde (FA), acetaldehyde (AA), or filtered air. Biomarkers of exposure and cardiopulmonary injury were monitored by mass spectrometry (urine metabolites), radiotelemetry (respiratory reflexes), isometric myography (aorta), and flow cytometry (blood markers).

Asymmetric synthesis of D-ribo-phytosphingosine from 1-tetradecyne and (4-methoxyphenoxy)acetaldehyde.

An asymmetric synthesis of d-ribo-phytosphingosine (1) was achieved by utilizing the ProPhenol (12)-catalyzed alkynylation of unsaturated aldehyde 8 to afford allylic propargylic alcohol (S)-6 followed by asymmetric epoxidation and opening of propargylic epoxy alcohol anti-5 with NaN(3)/NH(4)Cl. Deprotection and reduction of the resulting acyclic azide 3 then gave 1. Alkyne-azide 3 was subjected to an intramolecular click reaction, generating a bicyclic triazole, which was found to have unexpected vicinal coupling constants. Application of the advanced Mosher method verified the configurations

Salivary acetaldehyde concentration according to alcoholic beverage consumed and aldehyde dehydrogenase-2 genotype.

BACKGROUND: Acetaldehyde is suspected of playing a critical role in cancer development in the upper aerodigestive tract (UADT). The high salivary acetaldehyde levels after alcohol drinking are partly due to acetaldehyde production by oral bacteria. Some alcoholic beverages, especially Calvados and shochu, contain very high levels of acetaldehyde. Inactive heterozygous aldehyde dehydrogenase-2 (ALDH2) increases the risk of UADT cancer in drinkers. METHODS: In a randomized cross-over design study, 19 healthy Japanese volunteers ingested 0.6 g ethanol/kg body weight in the form of 13% ethanol Ca

T cell proliferative responses to malondialdehyde-acetaldehyde haptenated protein are scavenger receptor mediated.

Malondialdehyde-acetaldehyde (MAA) haptenated proteins have been described in disease processes related to prolonged oxidative stress (via malondialdehyde production), such as alcohol liver disease (ALD), non-alcoholic non-steatohepatitis (NASH) and atherosclerosis. Experimentally, high titer IgG1 antibody responses are seen after immunization without adjuvant; however, T cell proliferative responses and the role of scavenger receptors in this immunogenicity has not previously been described. In this study, T cell proliferative responses to the carrier protein, but not the MAA hapten itself, w
