The rise and global dissemination of multidrug-resistant (MDR) Acinetobacter baumannii has become a major public health threat. However, the constant emergence of drug-resistant mutants has resulted in increasingly limited therapeutic options. Tigecycline (TGC) continues to be one of the few antibiotics still effective against many A. baumannii strains. Herein, we discovered that quercetin (Qu) synergizes with TGC, enhancing its therapeutic efficacy against MDR A. baumannii by reducing the minimum inhibitory concentration (MIC) by 4- to 32-fold. Mechanistic analysis revealed that the synergist

INTRODUCTION: Acinetobacter baumannii, a World Health Organization (WHO) critical-priority pathogen, causes difficult-to-treat nosocomial infections. The link between resistance genes, biofilm, and virulence carriage remains poorly understood in clinical isolates of high-burden South Asian settings. This study intended to thoroughly describe and explain linkages among clinical isolates collected in Haryana, India.
METHODS: A total of 200 non-duplicate A. baumannii isolates underwent antimicrobial susceptibility testing by broth microdilution against 17 antibiotics, interpreted according to CL

Carbapenem-resistant Acinetobacter baumannii (CRAB) is characterized as a critical priority pathogen with restricted therapeutic options. To date, the most effective antimicrobial treatment against this difficult-to-treat bacterial strain has not been established. Sulbactam is a β-lactamase inhibitor with intrinsic activity against this pathogen, however, as a β-lactam, it can be hydrolysed by β-lactamases produced by A. baumannii. High-dose, extended-infusion treatment with sulbactam can overcome this hydrolysis by β-lactamases and is considered an effective therapeutic strategy against CRAB.

BACKGROUND: Multidrug-resistant gram-negative bacterial infections pose a serious threat to human health. Given that iron levels affect bacterial pathogenesis, iron homeostasis is a promising target for antimicrobial therapies. We investigated the effects and mechanisms of action of hinokitiol against Acinetobacter baumannii, a notorious opportunistic pathogen that causes nosocomial infections worldwide.
MAIN METHODS: The minimum inhibitory concentration was determined using the broth microdilution method. The inhibition of A. baumannii isolates was determined by growth kinetics analysis and

UNLABELLED: Biological sex is known to affect the incidence and outcomes of infection, varying significantly by pathogen. The bacterial pathogen Acinetobacter baumannii is recognized as a serious global healthcare threat worldwide, yet understanding of how biological sex impacts its infection course is limited. We previously documented in a murine model of acute pneumonia that female mice were more susceptible to infection, but there are no published studies looking at outcomes in humans according to biological sex. We conducted a retrospective cohort study of 220 adult patients with a positiv

With a limited number of traditional virulence factors, the success of the nosocomial pathogen Acinetobacter baumannii is largely attributed to its ability to persist and resist. The niches encountered during infection vary significantly from the more commonly studied laboratory setting, and consequently, the genes responsible for in vivo pathogenesis have yet to be fully elucidated. This study utilized the A. baumannii AB5075-UW transposon mutant library with unbiased genome-wide transposon sequencing to identify the genetic basis for survival and fitness during pneumonia and septicaemia infe

OBJECTIVE: Carbapenem-resistant Acinetobacter baumannii pose a major public health threat. These bacteria often display broad-spectrum antibiotic resistance, rendering them resistant to nearly all classes of antibiotics. Hydrogen sulphide (H2S), a compound naturally produced by many bacterial species, has been proposed to exert a protective effect against certain antibiotics. However, A. baumannii does not produced H2S.
METHODS: To investigate the effect of exogenous H2S on antibiotic resistance, several clinical isolates of carbapenemase-producing A. baumannii were cultured under various con

Multidrug-resistant (MDR) Acinetobacter baumannii poses a significant therapeutic challenge due to its resistance to multiple antibiotics and its ability to form biofilm. This study aimed to characterize MDR A. baumannii isolates for their biofilm-forming capabilities and the presence of common biofilm-related genes at a tertiary care university hospital in Nepal. In addition, it assessed the efficacy of various compounds, particularly essential oils, in inhibiting biofilm formation. Identification and antibiotic sensitivity testing of A. baumannii isolates from clinical specimens were conduct

Acinetobacter baumannii is an opportunistic Gram-negative bacterium representing one of the leading causes of ventilator-associated pneumonia. The development of pneumonia results from a complex interplay between pathogens and pulmonary innate mucosal immunity. Therefore, the knowledge of the host immune responses is pivotal for the development of effective therapeutics to treat A. baumannii infections. Previous studies were conducted using cell lines and animal models, but a comprehensive understanding of the interaction between A. baumannii and primary human immune cells is still lacking. To

Acinetobacter, particularly the Acinetobacter baumannii group, is a major cause of nosocomial infections, and carbapenem-resistant Acinetobacter spp. are important human pathogens. We collected 492 Acinetobacter spp. strains from two hospitals in Taiwan and classified them using MALDI-TOF MS and blaOXA-51-like PCR; 94.5% were A. baumannii, and 5.5% were non-A. baumannii (NAB). We confirmed their identity by rpoB gene sequencing of 239 randomly selected A. baumannii strains and all 27 NAB strains. Our analysis revealed that the rpoB alleles of OXA51-like-negative strains matched those of two NA

Many Gram-negative bacteria deploy a type VI secretion system (T6SS) to inject toxins into target cells to promote their survival and replication in complex environments. Here, we report that Acinetobacter baumannii uses its T6SS to kill fungi and that the effector TafE (ACX60_15365) is responsible for such killing. Although ectopically expressed TafE is toxic to both Escherichia coli and Saccharomyces cerevisiae, deletion of tafE only affects the antifungal activity of A. baumannii. We demonstrate that TafE is a DNase capable of targeting the nuclei of yeast cells and that an Ntox15 domain is
