Functional role of Nav1.8 channels in action potentials of mouse CGRP-lineage dorsal root ganglion neurons.

Primary sensory neurons that sense noxious stimuli (nociceptors) are unusual in possessing two major components of voltage-dependent sodium channel current with different voltage dependence and kinetics. We used pharmacology to characterize the roles of defined sodium channel types in generating action potentials and controlling repetitive firing in putative peptidergic nociceptors, using mice in which CGRP-lineage neurons are fluorescently labelled. We found that the major components of sodium current were carried by Nav1.8 channels, identified by sensitivity to VX-548 (suzetrigine), and by N

Inhibition by non-steroidal anti-inflammatory drugs of compound action potentials in frog sciatic nerve fibers.

AIMS: Although antinociception produced by non-steroidal anti-inflammatory drugs (NSAIDs) is partly attributed to nerve conduction inhibition, this has not been thoroughly examined yet. The aim of the present study was to reveal quantitatively how various types of NSAIDs affect compound action potentials (CAPs), a measure of nerve conduction. MAIN METHODS: CAPs were recorded from the frog sciatic nerve by using the air-gap method. KEY FINDINGS: Soaking the sciatic nerve with acetic acid-based NSAIDs (diclofenac and aceclofenac) reduced the peak amplitude of CAP in a concentration-dependent m

Contribution of action potentials to the extracellular field potential in the nucleus laminaris of barn owl.

Extracellular field potentials (EFP) are widely used to evaluate in vivo neural activity, but identification of multiple sources and their relative contributions is often ambiguous, making the interpretation of the EFP difficult. We have therefore analyzed a model EFP from a simple brainstem circuit with separable pre- and postsynaptic components to determine whether we could isolate its sources. Our previous papers had shown that the barn owl neurophonic largely originates with spikes from input axons and synapses that terminate on the neurons in the nucleus laminaris (NL) (Kuokkanen PT, Wagn

Neurophysiological characteristics of human leg muscle action potentials evoked by transcutaneous magnetic stimulation of the spine.

The objectives of this study were to establish the neurophysiological properties of the compound muscle action potentials (CMAPs) evoked by transcutaneous magnetic stimulation of the spine (tsMSS) and the effects of tsMSS on the soleus H-reflex. In semi-prone seated subjects with trunk semi-flexed, the epicenter of a figure-of-eight magnetic coil was placed at Thoracic 10 with the handle on the midline of the vertebral column. The magnetic stimulator was triggered by monophasic single pulses of 1 ms, and the intensity ranged from 90% to 100% of the stimulator output across subjects. CMAPs were

Discrimination of spinal and bulbar muscular atrophy from amyotrophic lateral sclerosis using sensory nerve action potentials.

INTRODUCTION: Spinal and bulbar muscular atrophy (SBMA) and amyotrophic lateral sclerosis (ALS) are motor neuron diseases. Sensory impairment is sometimes observed, and electrophysiological involvement has been described in the sensory nerves of SBMA patients. We hypothesized that a sensory nerve conduction study (NCS) could be used to discriminate SBMA from ALS. METHODS: We compared the results from NCSs in a total of 120 SBMA cases confirmed by genetic analysis, 188 ALS cases, and 50 normal subjects. RESULTS: Sensory nerve action potential (SNAP) amplitudes of the SBMA group were significa

Olivocochlear reflex assays: effects of contralateral sound on compound action potentials versus ear-canal distortion products.

The strength of the olivocochlear reflex has been assayed by comparing ipsilateral cochlear responses with and without contralateral sound. In humans, ipsilateral cochlear responses have usually been inferred by measuring otoacoustic emissions (OAEs), whereas, in animal work, they have been assessed by measuring compound action potentials (CAPs). Thus reports that the reflex strength is smaller in humans than in animals cannot be interpreted until the differences between the two tests are better understood. The present study directly compares reflex assays using distortion-product (DP) OAE and

Reversible depression of action potentials and force production in frog single muscle fibres by calmodulin-inhibitors.

The effects of the calmodulin-inhibitors trifluoperazine, thioridazine and zaldaride maleate on the responses to electrical stimulation in isolated frog skeletal muscle fibres were investigated. All three drugs initially reduced the amplitude of the action potentials but potentiated twitch force. This was followed by a total loss of action potentials and force production. However, the resting membrane potential was not changed. The effects were completely reversible upon removal of the drugs. These results suggest that an intact calmodulin system is required for normal function of the sarcolem

Comparative action of methylmercury and divalent inorganic mercury on nerve terminal and intraterminal mitochondrial membrane potentials.

Both methylmercury (MeHg) and inorganic divalent mercury (Hg++) alter the flux of ions and small molecules across nerve terminal membranes by mechanisms that may involve membrane depolarization. We compared the effects of MeHg and Hg++ on plasma (psi p) and mitochondrial membrane potentials (psi m) in synaptosomes using the potentiometric carbocyanine dye 3,3'-diethylthiadicarbocyanine iodide [diS-C2(5)]. Both mercurials (1-20 microM) produced concentration-dependent increases in dye fluorescence after 5 min of exposure which were not altered by removal of Ca++ from the medium. To determine di

Conduction of sensory action potentials across the posterior fossa in infratentorial space-occupying lesions in man.

Central conduction time following median nerve stimulation has been recorded in 28 patients with infratentorial space occupying lesions. Fourteen of these (50%) showed a prolonged central conduction time, which invariably involved that segment of the conduction time between the dorsal column nuclei generators and the generation site for P15, which we believe to be the upper brain stem or thalamus. Three other cases showed prolongation of the N14-P15 segments though total central conduction time was within normal limits. One case with marked hydrocephalus showed prolongation both before and aft

The response of Ca-mediated action potentials and contractile activity in mammalian ventricular myocardium towards alkalosis.

Alkalosis (pH 7.8) produced by reduction of CO2 concentration augmented both upstroke velocity of Ca action potentials and isometric contractile force of mammalian heart muscle. If the increase of pH to 7.8 was achieved by a raise of HCO3 concentration (with simultaneous reduction of CO2 concentration), the positive inotropic response was not accompanied by an augmented Ca current. Obviously, the well-known positive inotropic effect of alkalosis does not only depend upon the enhancement of transmembrane Ca influx during excitation, but can be mediated alone by affecting intracellular Ca moveme

Antiviral properties of phages: potential mechanisms of actions.

Bacteriophages (phages) have long been known to treat bacterial infections, although some early studies showed that phages also have potential antiviral mechanisms. This review provides a descriptive summary of ideas on how phages might have a significant role in inhibiting viral infections. Phages are known to directly modulate the host immunity that subsequently influences the immune responses against viral infections. It is also reasonable to explore the phage potential to directly inhibit viral infection in humans, including herpes simplex virus (HSV) and severe acute respiratory syndrome

Computational modeling of the human compound action potential.

The auditory nerve (AN) compound action potential (CAP) is an important tool for assessing auditory disorders and monitoring the health of the auditory periphery during surgical procedures. The CAP has been mathematically conceptualized as the convolution of a unit response (UR) waveform with the firing rate of a population of AN fibers. Here, an approach for predicting experimentally recorded CAPs in humans is proposed, which involves the use of human-based computational models to simulate AN activity. CAPs elicited by clicks, chirps, and amplitude-modulated carriers were simulated and compar

The potential therapeutic actions of melatonin in colorectal cancer.

Colorectal cancer (CRC) is the third most common cancer and lethal disease worldwide. Melatonin, an indoleamine produced in pineal gland, shows anticancer effects on a variety of cancers, especially CRC. After clarifying the pathophysiology of CRC, the association of circadian rhythm with CRC, and the relationship between shift work and the incidence of CRC is reviewed. Next, we review the role of melatonin receptors in CRC and the relationship between inflammation and CRC. Also included is a discussion of the mechanism of gene regulation, control of cell proliferation, apoptosis, autophagy, a

Parsing the Potential Neuroendocrine Actions of FGF21 in Primates.

Fibroblast growth factor (FGF) 21, a unique, largely liver-derived endocrine member of the FGF superfamily, is often thought of as a fasting factor owing to its induction in rodents during starvation. However, FGF21 is not increased by fasting for periods of <7 days in humans; instead, it rises sharply after acute alcohol and sugar intake and also after several days of overfeeding, suggesting another role in states of positive energy balance. Recent studies suggest that in the postingestive state, FGF21 may regulate energy intake and discourage consumption of alcohol and sugars, most likely th

CCN proteins as potential actionable targets in scleroderma.

Systemic sclerosis (SSc) is a complex autoimmune connective tissue disease combining inflammatory, vasculopathic and fibrotic manifestations. Skin features, which give their name to the disease and are considered as diagnostic as well as prognostic markers, have not been thoroughly investigated in terms of therapeutic targets. CCN proteins (CYR61/CCN1, CTGF/CCN2, NOV/CCN3 and WISP1-2-3 as CCN4-5-6) are a family of secreted matricellular proteins implicated in major cellular processes such as cell growth, migration, differentiation. They have already been implicated in key pathophysiological pr

Quantification of motor unit action potential energy.

OBJECTIVE: Motor unit action potentials (MUAPs) recorded by needle electrode reflect the functional state of the motor unit and its force-generating capacity, and are usually described morphologically (e.g. amplitude, duration). However, since the purpose of motor unit activation is force generation, MUAP energy seems a more physically meaningful measurement. METHODS: MUAPs were obtained by multi-MUAP decomposition of real interference patterns taken from human patients with neurological diseases. The energy content of each MUAP was measured from a time-frequency representation (TFR), specifi

Steroid inhibitors of androgen-potentiated actions on skin.

Antiandrogens, such as cyproterone acetate, and oestrogens both inhibit sebaceous secretion in rats and have a potentiality for the treatment of hirsutism and acne in the human female. However, they act at different points. In castrated rats treated with testosterone, 3 micrograms/day oestradiol produced a greater decrease in sebum secretion than a dose of cyproterone acetate over 1000 times larger; moreover the antiandrogen reduced the incidence of sebaceous mitoses whereas the oestradiol did not. In hirsute women, oral administration of 100 mg of cyproterone acetate daily caused a 40% reduct

Interactions of neurotoxins with the action potential NA + ionophore.

Four neurotoxins that activate the action potential Na+ionophore of electrically excitable neuroblastoma cells interact with two distinct classes of sites, one specific for the alkaloids veratridine, batrachotoxin, and aconitine, and the second specific for scorpion toxin. Positive heterotropic cooperativity is observed between toxins bound at these two classes of sites. Tetrodotoxin, a specific inhibitor of the action potential Na+ current, inhibits activation by each of these toxins in a noncompetitive manner (KI=4-8 nM). These results suggest the existence of three functionally separable co
