Pigkeepers' knowledge and perceptions regarding African swine fever and the control measures in Estonia.

African swine fever (ASF) has been present in Estonia since 2014, and pigkeepers play a major role in controlling the spread of viral infection in domestic pigs. Participatory epidemiology may provide insight into livestock keepers' existing attitudes and compliance with implementing control strategies. By conducting focus group discussions and using participatory epidemiology tools, this study aimed to reveal pigkeepers' knowledge and awareness of ASF clinical signs, transmission routes, and preventive measures. Furthermore, opinions including trust in stakeholders in ASF control, acceptabili…

[Alternative approaches to the diagnosis of African swine fever in the Russian Federation in 2017-2021].

INTRODUCTION: Prevention and control of African swine fever (ASF) transmission on the territory of the Russian Federation requires monitoring based on testing of samples from pigs and wild boars. Specific anti-ASFV antibodies are rarely detected in samples during routine serological diagnostics. Although, ASF isolates with weakened virulence were confirmed in Russia and neighboring countries. The aim of this work was to determine the possibility of using alternative samples for ASF diagnosis and evaluate the effectiveness of the diagnostic methods used on the territory of Russia. MATERIALS AND…

Relationships between the defensive systems of Iberian-breed swine and the European vector of African swine fever, Ornithodoros erraticus.

To discover whether the immune system of Iberian-breed pigs exerts any adverse action on Ornithodoros erraticus, 3 pairs of pigs were subjected to a weekly infestation over 12 wk with 1,000 larvae, 500 nymphs-1, or 200 adults. Each pair was bitten by only 1 developmental stage. Batches of parasites identical to the foregoing ones were fed weekly on control swine. In none of the 10 parameters studied for each of the batches fed weekly was any significant difference found that could be attributed to the state of sensitization of the animals in which, in a previous study, the presence of high tit

Potential economy-wide impacts of an African swine fever outbreak in the United States.

INTRODUCTION: African swine fever (ASF) poses a major threat to the U.S. pork sector, with potentially large spillovers through domestic markets and international trade. To inform preparedness and policy design, we quantify the economy-wide consequences of a hypothetical ASF outbreak under alternative scenarios of production losses and export restrictions. METHODS: We use a computable general equilibrium (CGE) model to simulate four hypothetical ASF scenarios that vary by outbreak size and export restrictions. The model endogenously captures adjustments in production, prices, bilateral trade …

Live Attenuated Vaccines for African Swine Fever: Perspectives on Their Use and Strategic Directions.

African swine fever (ASF) continues to cause substantial economic and production losses in affected countries. Although biosecurity and conventional control measures remain essential, the challenges of ASF control underscore the need for safe and effective vaccines as additional tools, particularly in endemic settings. Live attenuated vaccines (LAVs) are the most advanced ASF vaccine platform, providing the most consistent protection to date and remaining the only ASF vaccines to reach commercial deployment. Field experience with licensed products has shown encouraging safety, immunogenicity, …

Establishment of an immunoperoxidase monolayer assay for the detection of African swine fever virus antibodies.

African swine fever (ASF) is a lethal infectious disease affecting domestic and wild pigs, caused by the African swine fever virus (ASFV), with a mortality rate of up to 100 %. The evolving prevalence and variation of ASFV has led to the emergence of low-virulence strains, which induced chronic infections and posed challenges in nucleic acid-based diagnostics due to potential false negatives. This underscores the urgent need for reliable antibody monitoring to facilitate early diagnosis. In this study, based on a highly attenuated BK2258 cell-adapted strain HLJ18/BK33, we established an immuno…

Emergence of Microvariants of African Swine Fever Virus Genotype II in the Asia-Pacific.

African swine fever virus (ASFV) is a highly stable DNA virus showing little genetic variation among genomes. This genetic stability has often posed challenges in tracking variants and identifying potential transmission chains as ASFV spreads into new regions. While mutations within individual sequenced genes are infrequent, the application of whole-genome sequencing enables the identification of neutral and functional mutations across the entire genome, contributing to a deeper understanding of ASFV evolution and epidemiology. In this study, we sequenced whole genomes from 25 ASFV positive sa…

Fluorescence immunochromatographic detection of antibodies to the p72 protein of African swine fever virus.

The African swine fever virus (ASFV), a highly contagious pathogen responsible for African swine fever (ASF), causes significant economic losses in the global pork industry. Due to its large and complex structure, ASFV remains refractory to commercial vaccine development, necessitating the creation of rapid, sensitive, and specific diagnostic tools for disease control. In this study, quantum dots were conjugated to ASFV p72 protein to establish a fluorescent immunochromatographic assay for detecting ASFV-specific antibodies. The assay test strips contained four adjacent pads arranged sequentia…

Dataset for the analysis of stock price responses to African Swine Fever Outbreaks in China.

The African Swine Fever (ASF) outbreaks in China since 2018 caused a more than 100 million decline in its hog inventory. Leveraging publicly available announcements of ASF outbreaks and daily stock prices data from 25 major publicly listed hog companies from China and eight major hog exporting countries, we use the event study method to estimate firm-level abnormal stock price responses to China's ASF outbreak announcements for both Chinese and foreign hog companies. This article describes the data used in the research article "A Fortune from misfortune: Evidence from hog firms' stock price re…

African Swine Fever Virus as a Difficult Opponent in the Fight for a Vaccine-Current Data.

Prevention and control of African swine fever virus (ASFV) in Europe, Asia, and Africa seem to be extremely difficult in view of the ease with which it spreads, its high resistance to environmental conditions, and the many obstacles related to the introduction of effective specific immunoprophylaxis. Biological properties of ASFV indicate that the African swine fever (ASF) pandemic will continue to develop and that only the implementation of an effective and safe vaccine will ensure a reduction in the spread of ASFV. At present, vaccines against ASF are not available. The latest approaches to …

The Role of Male Reproductive Organs in the Transmission of African Swine Fever-Implications for Transmission.

African swine fever (ASF) has evolved from an exotic animal disease to a threat to global pig production. An important avenue for the wide-spread transmission of animal diseases is their dissemination through boar semen used for artificial insemination. In this context, we investigated the role of male reproductive organs in the transmission of ASF. Mature domestic boars and adolescent wild boars, inoculated with different ASF virus strains, were investigated by means of virological and pathological methods. Additionally, electron microscopy was employed to investigate in vitro inoculated sper…

An immunohistochemical study of the tonsils in pigs with acute African swine fever virus infection.

An immunohistochemical study of the tonsils was carried out to gain further insight in the pathogenesis of acute African swine fever (ASF). Twenty-one pigs were inoculated by intramuscular route with a highly virulent isolate of ASF virus and painlessly killed at 1-7dpi. Viral antigen was highly distributed in the tonsil from 3 to 4dpi and an increase in the number of monocyte-macrophages was very evident at the same days post inoculation. This phenomenon was observed together with an increase of the expression of proinflammatory cytokines (Tumour necrosis factor alpha and Interleukin-1 alpha)…

Subcellular changes in the tonsils of pigs infected with acute African swine fever virus.

A study of the pathogenesis of acute African swine fever (ASF) was carried out in pigs inoculated with a highly virulent strain of ASF virus to determine the sequential development of the subcellular changes in a particular lymphoepithelial organ, the tonsil. The apoptosis of the lymphocytes and the inhibition of lymphocyte proliferation were the main changes that occurred in the tonsillar lymphoid structures. This may explain the early lymphopenia observed in acute ASF. Moreover, vascular changes, consisting of increased vascular permeability, activation of endothelial cells and loss of these…

African swine fever interference with foot-and-mouth disease infection and seroconversion in pigs.

Initial oral infection of pigs with either highly virulent (L-60) or moderately virulent (DR-2) African swine fever virus (ASFV), followed in 3 days with exposure to foot-and-mouth disease virus (FMDV) (tongue inoculation and contact), failed to cause FMDV infection or seroconversion in 18 of 22 L-60-infected pigs and 13 of 34 DR-2-infected pigs. Of the 13 DR-2-infected pigs remaining free of foot-and-mouth disease (FMD), 2 pigs survived to 24 days without antibody to FMDV, despite constant contact with clinically infected pigs with FMD. Three other DR-2-infected pigs never developed FMD lesio…

High ex vivo recombination rate of African swine fever virus in porcine alveolar macrophages.

Using fluorescently labeled African Swine Fever Virus (ASFV) strains, a high (~0.2) quasi-recombinant frequency (qRF3-4) was detected ex vivo during 3-4 replication cycles in naïve porcine alveolar macrophages (PAM). While co-infection and recombination frequencies remained low in PAM, they increased sharply between the second and fourth viral cycles. Infection multiplicity and synchronicity significantly influenced ASFV recombination frequency, which correlated positively with genetic distance. Crossing the ASFV/Lv17/WB/Rie1 mutants yielded a saturation curve resembling the Haldane mapping fu…

Characterization of African swine fever outbreaks in Hong Kong SAR, winter 2023 to 2024.

Hong Kong veterinary authorities responded to a series of suspected outbreaks of African swine fever in domestic pigs in the New Territories of the Hong Kong Special Administrative Region from November 2023 to January 2024. Cases were identified on 10 farms, with one farm having a single pig with an inconclusive result. Subsequent genetic analysis suggested a more complicated picture than initially considered. Full genome sequencing identified a genotype II virus with a novel deletion in the left of the genome, as well as a genotype I/II hybrid virus similar to those previously reported in the…

African Swine Fever Virus pD345L Suppresses JAK-STAT Signaling by Selectively Triggering STAT1 Degradation.

African swine fever (ASF) is a highly lethal viral disease of pigs caused by the African swine fever virus (ASFV). The mortality rate is nearly 100%. Currently, it is known that the ASFV has a complex structure, and its genome encodes various immune escape proteins. However, the pathogenic mechanism of ASFV remains to be studied. This study found that ASFV pD345L significantly inhibits the activation of the ISRE promoter triggered by interferon (IFN) β and the production of downstream IFN-stimulated genes (ISG). We further reveal that pD345L may degrade STAT1 via the autophagy pathway and impe…

Correlates of protection against African swine fever virus identified by a systems immunology approach.

African swine fever virus (ASFV) causes a fatal hemorrhagic disease in domestic pigs and wild boars, which poses severe threats to the global pork industry. Despite the promise of live attenuated vaccines (LAVs), their narrow margin between efficacy and residual virulence presents major safety challenges. This study bridges a critical knowledge gap in ASF vaccinology by identifying innate and adaptive correlates of protection. This was achieved by using an established model with two groups of pigs differing in baseline immunological status (farm and specific pathogen-free [SPF]). The animals w…
