Amelogenesis imperfecta refers to a group of hereditary diseases that affect dental enamel, often leading to a wide range of clinical manifestations and aesthetic concerns. This case report describes a female patient diagnosed with amelogenesis imperfecta associated with a skeletal open bite. The treatment approach was multidisciplinary. Periodontal therapy was initiated, followed by orthodontic treatment using brackets on temporary crowns to expand and coordinate the maxillary and mandibular arches. Subsequently, orthognathic surgery, including a LeFort I osteotomy and genioplasty, was perfor

BACKGROUND: Amelogenesis Imperfecta (AI) is a disorder of tooth development characterized by abnormal enamel formation. In order to detect other dental and jawbone abnormalities that could be associated with AI, a retrospective and analytic study was conducted comparing panoramic radiographs of AI and non-AI patients.
MATERIAL AND METHODS: Digital panoramic radiographs of 60 AI and 60 non-AI patients were examined. Abnormalities in dental number, size, shape, eruption, and in the shape of the dental arches were checked and blindly recorded by two experimented observers. Descriptive statistics

Amelogenesis imperfecta (AI) is a heterogeneous group of rare inherited diseases presenting with enamel defects. More than 30 genes have been reported to be involved in syndromic or non-syndromic AI and new genes are continuously discovered (Smith et al., 2017). Whole-exome sequencing was performed in a consanguineous family. The affected daughter presented with intra-uterine and postnatal growth retardation, skeletal dysplasia, macrocephaly, blue sclerae, and hypoplastic AI. We identified a homozygous missense mutation in exon 11 of SLC10A7 (NM_001300842.2: c.908C>T; p.Pro303Leu) segregating

Patients with Amelogenesis imperfecta (AI) can present with rapid tooth loss or fractures of enamel as well as alterations in enamel thickness, color, and shape; factors that may compromise aesthetic appearance and masticatory function. The aim was to explore the experiences and perceptions of adolescents and young adults living with AI and receiving early prosthetic therapy. Seven patients with severe AI aged 16 to 23 years who underwent porcelain crown therapy participated in one-to-one individual interviews. The interviews followed a topic guide consisting of open-ended questions related to

OBJECTIVE: To analyze the ultrastructure and chemical compositions of two teeth affected by amelogenesis imperfecta (AI) with specific FAM83H mutations.
STUDY DESIGN: Mutation analysis was performed by sequencing the candidate FAM83H gene. Two extracted teeth were collected: one from a new family and the other from a previously reported family with FAM83H mutation (Q452X). The specimens were prepared for ultrastructure detection using scanning electron microscopy (SEM) and elemental analysis by energy dispersive X-ray spectrometry (EDX).
RESULTS: An FAM83H mutation R325X was identified in th

This clinical report describes a male with autosomal recessive generalized hypoplastic amelogenesis imperfecta. This case is unusual in coronal resorptions prior to tooth eruption. This finding has been reported in some cases of autosomal recessive, autosomal dominant and X linked amelogenesis imperfecta (AI). In reported cases, the defects were usually small and occurred in a maximum of 2 teeth per person. In our case, pre-eruptive coronal resorptions affected three second molar teeth from both jaws. On the other hand; congenitally missing teeth and malocclusion were present in this case. Rec

In this survey we summarize data on mineralization of enamel mostly obtained in organ culture experiments in our laboratory. Historically, the enzyme alkaline phosphatase has been proposed to stimulate mineralization by supplying phosphate or by splitting away inorganic pyrophosphate PPi, a potent inhibitor of mineralization. Localization of alkaline phosphatase in developing teeth by enzyme histochemistry shows that cells of the stratum intermedium contain extremely high levels of alkaline phosphatase but secretory ameloblasts that are engaged in deposition of the matrix and in transport of m

Inherited dental malformations constitute a clinically and genetically heterogeneous group of disorders. Here, we report on four families, three of them consanguineous, with an identical phenotype, characterized by significant short stature with brachyolmia and hypoplastic amelogenesis imperfecta (AI) with almost absent enamel. This phenotype was first described in 1996 by Verloes et al. as an autosomal recessive form of brachyolmia associated with AI. Whole-exome sequencing resulted in the identification of recessive hypomorphic mutations including deletion, nonsense and splice mutations, in

The combination of recessively inherited cone-rod dystrophy (CRD) and amelogenesis imperfecta (AI) was first reported by Jalili and Smith in 1988 in a family subsequently linked to a locus on chromosome 2q11, and it has since been reported in a second small family. We have identified five further ethnically diverse families cosegregating CRD and AI. Phenotypic characterization of teeth and visual function in the published and new families reveals a consistent syndrome in all seven families, and all link or are consistent with linkage to 2q11, confirming the existence of a genetically homogenou

Cystic fibrosis (CF) is a life-shortening inherited disorder caused by mutations in the gene encoding the Cystic Fibrosis Transmembrane conductance Regulator (CFTR), a protein that controls the transport of chloride ions across cell membranes. Defects in CFTR cause thick mucus, leading to chronic infections and inflammation in multiple organs. Severe mutations, such as G542X, that completely abolish the function of CFTR present major therapeutic challenges. One potential approach is to stimulate an alternative chloride channel, ANO1, which is repressed in patients by the small regulatory molec

Amelogenesis Imperfecta (AI) represents a group of hereditary conditions that manifest tooth enamel defects. Several causative mutations in the WDR72 gene have been identified and patients with WDR72 mutations have brown (or orange-brown) discolored enamel, rough enamel surface, early loss of enamel after tooth eruption, and severe attrition. Although the molecular function of WDR72 is not yet fully understood, a recent study suggested that WDR72 could be a facilitator of endocytic vesicle trafficking, which appears inconsistent with the previously reported cytoplasmic localization of WDR72. UNLABELLED: The treatment of amelogenesis imperfecta (AI) with an anterior open bite (AOB) is a challenge for the clinician and often requires a multidisciplinary team of specialists. Most often, patients suffering from these conditions are young and a good functional and esthetic long-term result must be aspired. This clinical report illustrates the orthodontic, maxillofacial, restorative, and prosthodontic rehabilitation of a 20-year-old woman with a hypoplastic form of AI and an AOB malocclusion, having received treatment for the last 6 years. It included adhesive resin composite restoratio

An adult woman with amelogenesis imperfecta, a skeletal Class III relationship, long face syndrome, and a severe anterior open bite received interdisciplinary treatment (orthodontics, orthognathic surgery, and prosthodontics). Presurgical orthodontic treatment was followed by a maxillary posterior impaction with anterior advancement and a mandibular setback. After surgery, the patient received ceramic crowns. Function and esthetics were successfully re-established.

PURPOSE: Successful bonding of resins to teeth affected by amelogenesis imperfecta (AI) may be highly dependent on how the enamel responds to acid etching. The aim of this study was to determine, using scanning electron microscopy (SEM), the types of etching pattern achieved with 37% phosphoric acid on dental enamel of 5 clinical variants of AI, namely, pitted hypoplastic, smooth hypoplastic, X-linked (male), X-linked (female), and hypomineralized.
METHODS: A normal premolar and primary molar from two healthy patients were used as controls. The enamel was scanned before and after acid etching

Second maxillary molars of hamsters were cultured in the presence or absence of 250 micrograms/ml vitamin C for periods up to 12 days. At various days, cultured explants were studied with histological and biochemical methods to investigate effects of vitamin C-deficiency on matrix production and mineralization in vitro. As biochemical parameters for protein synthesis and mineralization, uptake and incorporation of [3H]-proline, 45Ca and 32PO4 were used. To discriminate between synthesis of collagenous and non-collagenous proteins digestion of the [3H]-proline-labelled material by purified coll

PURPOSE: Amelogenesis imperfecta (AI) is a genetic enamel defect affecting both primary and permanent dentitions, often leading to functional, aesthetic, and psychosocial challenges. Patient-reported outcome measures (PROMs) are increasingly used in paediatric dentistry. This study aimed to explore the impact of AI on oral health-related quality of life in children and adolescents using an AI-specific PROM, and to assess clinician use of the PROM in the UK.
METHODS: This single-centre retrospective service evaluation involved children with AI who completed a nine-question PROM pre- and mid-tr

A female patient is described with clinical symptoms of both microphthalmia with linear skin defects (MLS or MIDAS) and dental enamel defects, having an appearance compatible with X-linked amelogenesis imperfecta (XAI). Genomic DNA was purified from the patient's blood and semiquantitative multiplex PCR revealed a deletion encompassing the amelogenin gene (AMELX). Because MLS is also localized to Xp22, genomic DNA was subjected to array comparative genomic hybridization, and a large heterozygous deletion was identified. Histopathology of one primary and one permanent molar tooth showed abnorma
